Abstract:The mechanism and origins of selectivities in [Ni(NHC)]-catalyzed intramolecular (5 + 2) cycloadditions and homo-ene reactions of vinylcyclopropanes (VCPs) and alkynes have been studied using density functional theory. The preferred mechanism involves oxidative alkyne-alkene cyclization to form a metallacyclopentene intermediate, in contrast to a cyclopropane cleavage pathway in the reaction with Rh(I) catalysts. The selectivity between the (5 + 2) and homo-ene products is determined in the subsequent competing reductive elimination and β-hydride elimination steps. Two similar-sized N-heterocyclic carbene (NHC) ligands, SIPr and ItBu, yielded reversed product selectivity, favoring the (5 + 2) and homo-ene products respectively. This is attributed to the anisotropic steric environment of these NHC ligands, which positions the bulky substituents on the ligand toward different directions and leads to distinct steric control in the reductive elimination and β-hydride elimination transition states.

Abstract:The mechanism, solvent effects, and origins of selectivities in Ru(II)-catalyzed intramolecular (5+2) cycloaddition and ene reaction of vinylcyclopropanes (VCPs) and alkynes have been studied using density functional theory. B3LYP/6-31G(d)/LANL2DZ optimized structures were further evaluated with the M06 functional, 6-311+G(2d,p) and LANL2DZ basis sets, and the SMD solvent model. The favored mechanism involves an initial ene-yne oxidative cyclization to form a ruthenacyclopentene intermediate. This mechanism is different from that found earlier with rhodium catalysts. The subsequent β-hydride elimination and cyclopropane cleavage are competitive, determining the experimental selectivity. In trans-VCP, the cyclopropane cleavage is intrinsically favored and leads to the (5+2) cycloaddition product. Although the same intrinsic preferences occur with the cis-VCP, an unfavorable rotation is required in order to generate the cis-double bond in seven-membered ring product, which reverses the selectivity. Acetone solvent is found to facilitate the acetonitrile dissociation from the precatalyst, destabilizing the resting state of the catalyst and leading to a lower overall reaction barrier. In addition, the origins of diastereoselectivities when the allylic hydroxyl group is trans to the bridgehead hydrogen are found to be the electrostatic interactions. In the pathway that generates the favored diastereomer, the oxygen lone pairs from the substituent are closer to the cationic catalyst center and provide stabilizing electrostatic interactions. Similar pathways also determine the regioselectivities, that is, whether the more or less substituted C-C bond of cyclopropane is cleaved. In the trans-1,2-disubstitued cyclopropane substrate, the substituent from the cyclopropane is away from the reaction center in both pathways, and low regioselectivity is found. In contrast, the cleavage of the more substituted C-C bond of the cis-1,2-disubstituted cyclopropane has steric repulsions from the substituent, and thus higher regioselectivity is found.

Abstract:The mechanisms of N‐heterocyclic carbene (NHC)‐catalyzed dearomatization reaction of alkyl pyridinium have been intensively studied using the density functional theory (DFT) method. The chemoselectivity and stereoselectivity were both analyzed based on the established mechanisms. The computational results showed that the chemoselective C4‐addition occurred before the C2‐addition, which is mainly because the C4 atom was more reactive according to the local reactivity analysis. The stereoselectivity was also investigated, and the C‐C bond formation step was identified as the stereoselectivity‐determining step. The C‐H···F and O‐H···N interactions are identified as the main factors that govern the stereoselectivity by NCI analysis, and the preferred product was the RS‐configuration. This theoretical investigation would provide a case for understanding and predicting the selectivity of the organocatalytic dearomatization reactions of alkyl pyridinium.

Abstract:The mechanisms and chemo- and regioselectivities of Ru(II)-catalyzed decarboxylative C-H alkenylation of aryl carboxylic acids with alkynes were investigated with density functional theory (DFT) calculations. The catalytic cycle involves sequential carboxylate-directed C-H activation, alkyne insertion, decarboxylation and protonation. The facile tether-assisted decarboxylation step directs the intermediate toward the desired decarboxylative alkenylation, instead of typical annulation and double alkenylation pathways. The decarboxylation barrier is very sensitive to the tether length, and only the seven-membered ring intermediate can selectively undergo the designed decarboxylation, suggesting a tether-dependent chemoselectivity. This tether-dependent chemoselectivity also applies to the alkyl tethers. In addition, the polarity of solvent is found to control the chemoselectivity between the decarboxylative alkenylation and [4 + 2] annulation. Solvent with low polarity (toluene) favors the decarboxylation pathway, leading to the decarboxylative alkenylation. Solvent with high polarity (methanol) favors the ionic stepwise C-O reductive elimination pathway, leading to the [4 + 2] annulation. To understand the origins of regioselectivity with asymmetric alkynes, the distortion/interaction analysis was applied to the alkyne insertion transition states, and led to a predictive frontier molecular orbital model. The asymmetric alkynes selectively use the terminal with the larger HOMO orbital coefficient to form the C-C bond in the insertion step.

Abstract:Regio‐ and stereo‐divergent construction of carbon‐fluorine bond remains a challenge in fluorine chemistry, especially reactions involving fluorination at the α‐position of a carbonyl group. Herein, we present a detailed study for predicting the origins of regio‐ and stereo‐selectivity for organocatalyst catalyzed fluorination reaction. Possible mechanisms and origins of selectivities of NHC (N‐heterocyclic carbene)‐catalyzed fluorination reaction of epoxy‐enals were investigated by using DFT (density functional theory) method. The computational results show that the C‐F bond formation step determines stereoselectivity and regioselectivity. LP•••π and C‐H•••F interactions were identified as the main contributors for the stereoselectivity control, in which R‐configured product was preferred. It was found that α‐fluorination is more energetically favorable than γ‐fluorination, which is possibly due to higher reactivity of the α‐carbon; deduced by the local reactivity index analysis. This work would provide valuable insights for predicting origin of the selectivity of fluorination reactions and developing of more effective organocatalyst with high selectivity.

Abstract:The mechanism, chemoselectivity and stereoselectivity in NHC-catalyzed reaction between enal and pyrrole for the synthesis of 5,6-dihydroindolizine were studied using DFT calculations. The cycle for catalytically generation of 5,6-dihydroindolizine proceeds via seven steps: (1) addition of the NHC to enal, (2) formation of Breslow intermediate through [1,2]-proton transfer, (3) oxidation, (4) Michael addition, (5) [2+2] cycloaddition, (6) liberation of NHC and (7) decarboxylation. Our results show that the presence of DMAP·H+ lowers the barrier for [1,2]-proton transfer. In addition, the NHC·H+ plays a key role in decarboxylation. Michael addition which involves forming a new C–C bond was identified to be the chemo- and stereoselectivity-determining step, leading to the experimentally observed 5S,6R-dihydroindolizine. Analysis of the noncovalent interactions revealed that the observed stereoselectivity is attributed to the differential weak interactions (CH···π, LP···π and CH···CH) involved in the transition states during the Michael addition step. The computational results not only rationalize experimental observations but also provide some useful information for future design of new catalytic processes.

Abstract:Transition metal-catalyzed hydroalkenylation is widely applied in organic synthesis to construct carbon-carbon bonds and synthesize substituted alkenes. In this work, the mechanism and regioselectivities of [(NHC)NiH]+-catalyzed intramolecular cycloisomerization of dienes are studied by density functional theory (DFT) calculations. Through an initial hydride insertion, [(NHC)NiH]+ reacts with the diene substrate to generate the alkyl nickel species. This alkyl nickel species is the resting state of the catalytic cycle, and the hydroalkenylation reaction involves sequential olefin insertion and β-hydride transfer. The selectivity between the possible cycloisomerization products is determined by both the olefin insertion and β-hydride transfer steps. The olefin insertion favors the exo-cyclization due to the high ring strain of the insertion transition states for the endo-cyclization. In the β-hydride transfer step, the hydride transfers to the internal olefin position selectively to avoid the steric repulsions between the bulky NHC ligand and the alkyl substituent of the olefin substrate. This selectivity of β-hydride transfer leads to the n-exo cyclization instead of the (n - 1)-exo cyclization. These mechanistic insights will shed light on the future development of transition metal-catalyzed hydroalkenylation reactions.

Abstract:In the present study, the mechanism, origin of chemoselectivity as well as substituent effects have been investigated using the DFT method. Multiple pathways, including the formation of hydrofluorenone, Cloke-Wilson, and cyclopenta-fused products, are studied and compared. The computational results show that the pathway for formation of hydrofluorenone is the most favorable, which contains four processes: nucleophilic substitution to open the three-membered ring, intramolecular Michael addition for the formation of an enolate intermediate, intramolecular [1,5]-proton transfer to give the ylide, and intramolecular Wittig reaction to deliver the final product. For clarifying the chemoselectivity, structural analysis and the local reactivity index analysis are employed. Moreover, the substituent effects are also considered using the QTAIM analysis. The current study would provide useful insights for understanding the phosphine-catalyzed chemoselective reactions.

Abstract:We reported a density functional theory (M06–2X) study of the possible mechanisms operating in a chemo- and stereoselective [3+3] annulation reaction of 2-bromoenal with β - ketothioamide catalyzed by a chiral NHC catalyst. DFT computations suggest the catalytic reaction involves nucleophilic addition, [ 1 , 2 ]-proton transfer, debromination, [ 1 , 3 ]-proton transfer, Michael addition, [ 1 , 5 ]-proton transfer, ring closure and catalyst dissociation, in which Michael addition is the chemo- and stereoselectivity-determining step and provides the SR -configured sipro-piperidinone predominantly. This result is consistent with the stereoselectivity observed in experiment. Nonconvalent interaction (NCI) analysis reveals that the origin of stereoselectivity is the different weak interactions (C–H···π, LP···π and π···π). The computational results should deepen our understanding of NHC catalysis, and may thus contribute to the design of new catalytic systems.

Abstract:Ruthenium-catalyzed sigma-bond activation-assisted meta-C-H functionalization has emerged as a useful tool to forge distal C-C bonds. However, given the limited number of mechanistic studies, a clear understanding of the origin of the site-selectivity and the complete reaction pattern is not available. Here, we present systematic computational studies on ruthenium-catalyzed C-H functionalization with primary, secondary, tertiary alkyl bromides and aryl bromides. The C-H scission and the C-C formation were carefully examined. Monocyclometalated ruthenium(II) complexes were identified as the active species, which then underwent inner-sphere single electron transfer (ISET) to activate the organic bromides. The site-selectivity results from the competition between the close-shell reductive elimination and the open-shell radical coupling. Based on this mechanistic understanding, a multilinear regression model was built to predict the site-selectivity, which was further validated by experiments.
Predicting Regioselectivity in Radical C−H Functionalization of Heterocycles Through Machine Learning

Abstract:Radical C-H bond functionalization provides a versatile approach for elaborating heterocyclic compounds. The synthetic design of this transformation relies heavily on the knowledge of regioselectivity, while a quantified and efficient regioselectivity prediction approach is still elusive. Herein, we report the feasibility of using a machine learning model to predict the transition state barrier from the computed properties of isolated reactants. This enables rapid and reliable regioselectivity prediction for radical C-H bond functionalization of heterocycles. The Random Forest model with physical organic features achieved 94.2 % site accuracy and 89.9 % selectivity accuracy in the out-of-sample test set. The prediction performance was further validated by comparing the machine learning results with additional substituents, heteroarene scaffolds and experimental observations. This work revealed that the combination of mechanism-based computational statistics and machine learning model can serve as a useful strategy for selectivity prediction of organic transformations.
Computational Exploration of Mechanism and Selectivities of (Nhc)nickel(ii)hydride-Catalyzed Hydroalkenylations of Styrene with Α-Olefins

Abstract:The [LNiH](+)-catalyzed hydroalkenylation between styrene and alpha-olefins gives distinctive chemo- and regioselectivities with N-heterocyclic carbene (L = NHC) ligands: (a) the reaction with NHC ligands produces the branched tail-to-tail products, whereas the reaction with phosphine ligands (L = PR3) favors the tail-to-head regio-isomers; (b) the reaction stops at heterodimerization with no further oligomerization even with excess alpha-olefin substrates; (c) typical side reactions with alpha-olefins, such as isomerization to internal olefins or polymerization, are either significantly diminished or eliminated. To understand the operating mechanism and origins of selectivities, density functional theory (DFT) calculations were performed, and several additional experiments were conducted. The olefin insertion step is found to determine both the regioselectivity and chemoselectivity, leading to the tail-to-tail heterohydroalkenylation product. With a small NHC ligand (1,3-dimethylimidazol-2-ylidene), the intrinsic electronic effects of ligand favor the tail-to-head regioisomer by about 1 kcal/mol in the olefin insertion step. With bulky NHC ligands (1,3-bis(2,6-dimethylphenyl)imidazol-2-ylidene or SIPr), the steric repulsions between the ligand and the substituent of the inserting alkene override the intrinsic electronic preference, making the tail-to-tail regioisomer favored (about 3 kcal/mol with both ligands). In the competition between homo- and heterodimerization, the insertion of the secondary styrene breaks its pi-conjugation, making the insertion of styrene about 2 kcal/mol less favorable than that of alkyl-substituted alkenes. In addition, the interaction between nickel and phenyl group of styrene stabilizes the resting state and inhibits the side reactions with alpha-olefins, suggesting that styrene, or similar aryl olefins, is not only a substrate, but also an inhibitor for side reactions. This unique effect of styrene is verified by control experiments.

Abstract:The α,β-unsaturated acyl azolium has emerged as a central reactive intermediate for reaction discovery using N-heterocyclic carbene catalysis during the past decades, whereas, the mechanistic studies on transformations of carbonyl compounds involving α,β-unsaturated acyl azolium as key intermediates are rare. In the present study, the mechanism of activation/transformation of enals catalyzed by NHC is theoretically studied using DFT method. Based on the calculations, Michael-type addition process is the stereoselectivity-determining step with SR-configured product being generated preferentially. Furthermore, NCI and AIM analyses have been performed to disclose the origin of stereoselectivity, by which the larger number and stronger noncovalent interactions are the key for stabilizing the low-energy transition state and thus leading to the stereoselectivity inducing.

Abstract:The density functional theory (DFT) method was used to investigate the mechanism and the origin of stereoselectivity of N-heterocyclic carbene (NHC)-catalyzed [3 + 2] cycloaddition between enals and cyclic imine N-sulfonyl trifluoromethyl ketimines at the M06-2X/SMD/6-311+G(d,p)//M06-2X/SMD/6-31G (d,p) level. The results show that the favorable reaction path consists of five steps: nucleophilic attack, proton transfer, the formation of the C-C bond, the tautomerism of the enol intermediate, the formation of the five-membered ring, and the regeneration of the catalyst. For the process of proton transfer, the base-assisted reaction can reduce the activation free energy and make the reaction easier to occur compared with the direct proton transfer process. The formation of the C-C bond is the crucial step of stereoselectivity, in which two chiral centers and four configurations of intermediates (RR/RS/SR/SS) were generated. The free energy barriers obtained and the noncovalent interaction analysis confirm that the dominant configuration is SS, becoming the final trans-type product observed in experiment. Furthermore, through the analyses of the conceptual DFT and natural atomic charges, it is revealed that NHC acts as a double catalyst, which can not only increase the nucleophilicity of reactants by Lewis base but also activate the C-H bond and promote the proton transfer process. The understanding of the mechanism obtained in this study should be helpful to the other organic catalytic reactions with high stereoselectivity.

Abstract:The mechanisms and origins for the Pd- and Ni-catalyzed regioselective hydrosilylation of allene have been investigated by means of density functional theory (DFT) calculations. The free-energy profiles of Pd- and Ni-catalyzed reactions with small and bulky N-heterocyclic carbene (NHC) ligands are calculated to determine the mechanism for regioselectivities. The calculation results show that different metals (Ni vs Pd) lead to regiochemical reversals for the hydrosilylation of allene. The allylsilane is the major product via palladium catalysis with small NHC ligand, while the vinylsilane is the major product via nickel catalysis with bulky NHC ligand. Both electronic and steric factors play a key role in the regioselectivities for the hydrosilylation of allene via Pd and Ni catalysts. The calculation results are in good agreement with observed regioselectivities and could provide insights into the design of new catalysts for the regioselectivity of hydrosilylation reactions.
Exploring the general mechanistic map on NHC-catalyzed activation/transformation reactions of saturated carboxylic anhydrides

Abstract:A general mechanistic map involving multiple intermediates and pathways has been proposed and systmatically studied for NHC-catalyzed transformation reactions of saturated carboxylic anhydrides. Based on the map, the origin of chemo- and stereo-selectivities has been predicted in a case study of NHC-catalyzed reaction of saturated carboxylic anhydride with benzylidene diketone. The computational results show that the fundamental pathway of this kind of reaction mainly includes the following processes: C-O bond cleavage for formation of acyl azolium, α-H elimination for formation of enolate, β-H elimination for the formation of Breslow intermediate, β-C functionalization with benzylidene diketone, acyl migration, aldol addition, dissociation of catalyst, and decarboxylation. The Michael-type addition process is identified to be the stereoselectivity-determining step, with SR-configured product generating preferentially. The different chemoselective pathways would undergo different intermediates, which is the key for understanding the origin of chemoselectivity. The chemoselectivity is successfully predicted by comparing the energy gap of FMO between the two interactive partners, which would provide a general guideline for determining the general principle behind organocatalytic reactions with various chemoselectivities and suggest a general application of the FMO in predicting the potential chemoselectivities.
