Abstract:Biocatalysis is constantly providing novel options for the synthesis of active pharmaceutical ingredients (APIs). In addition to drug development and manufacturing, biocatalysis also plays a role in drug discovery and can support many active ingredient syntheses at an early stage to build up entire scaffolds in a targeted and preparative manner. Recent progress in recruiting new enzymes by genome mining and screening or adapting their substrate, as well as product scope, by protein engineering has made biocatalysts a competitive tool applied in academic and industrial spheres. This is especially true for the advances in the field of nonribosomal peptide synthesis and enzyme cascades that are expanding the capabilities for the discovery and synthesis of new bioactive compounds via biotransformation. Here we highlight some of the most recent developments to add to the portfolio of biocatalysis with special relevance for the synthesis and late-stage functionalization of APIs, in order to bypass pure chemical processes.

Abstract:Biocatalysis holds immense potential for pharmaceutical development as it enables synthetic routes to various chiral building blocks with unparalleled selectivity. Therein, solvent and water use account for a large contribution to the environmental impact of the reactions. In the spirit of Green Chemistry, a transition from traditional highly diluted aqueous systems to intensified non-aqueous media to overcome limitations (e.g., water shortages, recalcitrant wastewater treatments, and low substrate loadings) has been observed. Benefiting from the spectacular advances in various enzyme stabilization techniques, a plethora of biotransformations in non-conventional media have been established. Deep eutectic solvents (DESs) emerge as a sort of (potentially) greener non-aqueous medium with increasing use in biocatalysis. This review discusses the state-of-the-art of biotransformations in DESs with a focus on biocatalytic pathways for the synthesis of active pharmaceutical ingredients (APIs). Representative examples of different enzyme classes are discussed, together with a critical vision of the limitations and discussing prospects of using DESs for biocatalysis.

Current Trends in API Co-Processing: Spherical Crystallization and Co-Precipitation Techniques

Abstract:Active Pharmaceutical Ingredients (APIs) do not always exhibit processable physical properties, which makes their processing in an industrial setup very demanding. These issues often lead to poor robustness and higher cost of the drug product. The issue can be mitigated by co-processing the APIs using suitable solvent media-based techniques to streamline pharmaceutical manufacturing operations. Some of the co-processing methods are the amalgamation of API purification and granulation steps. These techniques also exhibit adequate robustness for successful adoption by the pharmaceutical industry to manufacture high quality drug products. Spherical crystallization and co-precipitation are solvent media-based co-processing approaches that enhances the micromeritic and dissolution characteristics of problematic APIs. These methods not only improve API characteristics but also enable direct compression into tablets. These methods are economical and time-saving as they have the potential for effectively circumventing the granulation step, which can be a major source of variability in the product. This review highlights the recent advancements pertaining to these techniques to aid researchers in adopting the right co-processing method. Similarly, the possibility of scaling up the production of co-processed APIs by these techniques is discussed. The continuous manufacturability by co-processing is outlined with a short note on Process Analytical Technology (PAT) applicability in monitoring and improving the process.

Campos et al. review some of the advantages that have come from recent innovations in synthetic methods. Example of Finishing Technologies as Key Elements for Successful Active Pharmaceutical Ingredient Process Development

Abstract:Little has been published so far to emphasize the role of finishing technologies (crystallization, filtration, drying) as crucial elements for successful process development of active pharmaceutical ingredients (APIs). In the presented example high marketing forecasts stressed the objective for the process to be robust, simple, and tailor-made for high-volume production. The major hurdle was the purification of the reaction mixture of an intermediate containing inorganic salts, polymers, isomers, and colored byproducts that needed to be separated. Systematic application of finishing technologies paved the way to a commercial process. Removal of colored byproducts and isomers was found to be the most challenging task after separation of inorganic salts by quench and of polymers by charcoal treatment. The problem was resolved in three steps. First, solvent screening for the crystallization yielded toluene to be most efficient to separate colored byproducts. Second, solubility measurements of product and isomers provided an operating range of the process parameters for an economic crystallization at an unusually high concentration (60% w/w). Isomers could be separated since they remain soluble at endpoint temperature. To avoid the problem of wall crusts during scale-up, the metastable range was estimated experimentally. And third, near-infrared spectroscopy was implemented as an on-line measurement method for the product concentration in toluene to determine the endpoint of evaporation and thus to facilitate robustness and yield optimization. As further result of proper elaboration of crystallization conditions the crystals exhibited compact cubic morphology and large size. Consequently, filtration and washing of the wet product cake were efficient, and the drying process could be eliminated before continuing with the next synthesis step.

Abstract:Active pharmaceutical ingredients (APIs) extracted and isolated from traditional Chinese medicines (TCMs) are of interest for drug development due to their wide range of biological activities. However, the overwhelming majority of APIs in TCMs (T-APIs), including flavonoids, terpenoids, alkaloids and phenolic acids, are limited by their poor physicochemical and biopharmaceutical properties, such as solubility, dissolution performance, stability and tabletability for drug development. Cocrystallization of these T-APIs with coformers offers unique advantages to modulate physicochemical properties of these drugs without compromising the therapeutic benefits by non-covalent interactions. This review provides a comprehensive overview of current challenges, applications, and future directions of T-API cocrystals, including cocrystal designs, preparation methods, modifications and corresponding mechanisms of physicochemical and biopharmaceutical properties. Moreover, a variety of studies are presented to elucidate the relationship between the crystal structures of cocrystals and their resulting properties, along with the underlying mechanism for such changes. It is believed that a comprehensive understanding of cocrystal engineering could contribute to the development of more bioactive natural compounds into new drugs.

Abstract:Continuous flow chemistry is an emerging technology in the chemical pharmaceutical industry. Compared with the traditional batch kettle reaction, continuous flow reaction has the advantages of rapid mass and heat transfer, improves the controllability and safety of the reaction process, and can also integrate the online analysis and purification steps into the flexible operation sequence. The new progress of continuous flow reaction technology in the continuous synthesis process from starting raw material to final active pharmaceutical ingredient (API) from 2019 to 2022 is introduced. Starting from the flow chart of continuous flow reaction, the preparation process and technical advantages of continuous flow reaction are described. At the same time, it is pointed out that the fully continuous flow reaction technology still has some challenges in the process connection and coupling treatment of each single step synthesis transformation, which needs to be further improved.

Abstract:A miniaturized multi‐reactor system and knowledge‐based workflow were developed to identify optimal process conditions for controlling the crystal morphology of active pharmaceutical ingredients (APIs). This method, which features minimal material usage, inline imaging, and independent control of temperature T and supersaturation S, can be used to obtain more equant‐shaped crystals of diverse APIs. The morphology of crystalline active pharmaceutical ingredients (APIs) can significantly affect their product properties, so that its control during manufacturing is crucial. To address this, a newly augmented commercial milliliter‐scale facility and knowledge‐based workflow were developed with the aim of identifying optimal process conditions for producing more equant‐shaped crystals. The design includes minimal material usage, inline imaging, and independent temperature and supersaturation control through evaporative crystallization. The methodology enables the identification of process conditions for equant‐shaped crystals across diverse APIs. These findings contribute to advancing pharmaceutical research and development by providing a reliable approach to optimize crystal morphology.

Abstract:Synthesis of molecules containing all-carbon quaternary stereocenters has been a longstanding challenge in organic chemistry. In one of our discovery oncology programs, a key chiral building block bearing an all-carbon quaternary chiral center was of particular interest and was later identified as a core structure for a KRAS G12C inhibitor. Herein, the development of a safer and practical route to the key building block 1 is described. By replacing processes involving the use of an energetic reagent and extensive chromatographic purifications, a scalable process utilizing chemical resolution was developed to access the chiral building block in kilogram quantities, enabling timely delivery of API for preclinical and clinical studies.

Abstract:With the growing problem of drug shortages worldwide, a solution could be a centralized compact pharmaceutical manufacturing platform that produces API and finished drug product. In this paper, the development of a combined crystallization, hybrid filtration-drying-dissolution apparatus to be used in such a compact platform is discussed. Experiments were conducted to evaluate each unit operation. Crystallization experiments using a conventional stirred tank and the newly designed scraped surface crystallizer demonstrated the advantages of the new design in terms of crystallization rates, yields and the ease of automation. Postcrystallization operations were operated stepwise using the custom hybrid device that delivered satisfactory results for each operation, e.g. after filtration, Fluoxetine HCl was dried in less than 20 min, with 99% yield after dissolution in the liquid excipient.

Abstract:Active pharmaceutical ingredients (APIs), frequently delivered to the patient in the solid-state as part of an approved dosage form, can exist in such diverse solid forms as polymorphs, pseudopolymorphs, salts, co-crystals and amorphous solids. Various solid forms often display different mechanical, thermal, physical and chemical properties that can remarkably influence the bioavailability, hygroscopicity, stability and other performance characteristics of the drug. Hence, a thorough understanding of the relationship between the particular solid form of an active pharmaceutical ingredient (API) and its functional properties is important in selecting the most suitable form of the API for development into a drug product. In past decades, there have been significant efforts on the discovery, selection and control of the solid forms of APIs and bulk drugs. This contribution discusses the thermodynamics and kinetics of polymorphic systems, the characterization of polymorphs, and the transformation between polymorphs. The major techniques for polymorph discovery and control developed in the past years are discussed as well.

Abstract:Despite the success in developing various pharmaceutical formulations, most of the active pharmaceutical ingredients (APIs)/drugs, according to the Biopharmaceutics Classification System (BCS), often suffer from various intrinsic limitations of solubility and permeability, substantially hindering their bioavailability in vivo. Regardless of the fact that the availability of different particle fabrication approaches (top-down and bottom-up) towards pharmaceutical manufacturing, the supercritical fluid (SCF) technology has emerged as one of the highly effective substitutes due to the environmentally benign nature and processing convenience, as well as the economically promising character of SCFs. The exceptional features of SCFs have endowed the fabrication of various APIs either solely or in combination with the compatible supramolecular species towards achieving improved drug delivery. Operating such APIs in high-pressure conditions often results in arbitrary-sized particulate forms, ranging from micron-sized to sub-micron/nano-sized particles. Comparatively, these SCF-processed particles offer enhanced tailorable physicochemical and morphological properties (size, shape, and surface), as well as improved performance efficacy (bioavailability and therapy) over the unprocessed APIs. Although the "carrier-based" delivery is practical among diverse delivery systems, the direct fabrication of APIs into suitable particulate forms, referred to as "carrier-free" delivery, has increased attention towards improving the bioavailability and conveying a high payload of the APIs. This review gives a comprehensive emphasis on the SCF-assisted fabrication of diverse APIs towards exploring their great potential in drug delivery. Initially, we discuss various challenges of drug delivery and particle fabrication approaches. Further, different supercritical carbon dioxide (SC-CO2)-based fabrication approaches depending on the character of SCFs are explicitly described, highlighting their advantages and suitability in processing diverse APIs. Then, we provide detailed insights on various processing factors affecting the properties and morphology of SCF-processed APIs and their pharmaceutical applications, emphasizing their performance efficacy when administered through multiple routes of administration. Finally, we provide exciting perspectives based on the lessons learned and moving forward in terms of challenges and opportunities in the scale-up and clinical translation of these drugs using this innovative technology.

Abstract:The pharmaceutical industry has traditionally employed batch processing as a means of synthesizing active pharmaceutical ingredients (APIs) on a commercial scale. Batch manufacturing enables API synthesis in large quantities in order to meet regulations. Yet, this strategy remains cumbersome, is labor‐intensive, and creates complex logistical networks. In recent decades, batch API processing has gradually eroded American economic competitiveness and has led to the off‐shoring of API manufacturing from the United States. Today it is estimated that +80% of APIs are manufactured overseas. Meanwhile, disruptions from the Coronavirus Disease (SARS‐CoV‐2) have exacerbated weaknesses in the global supply chain, culminating with widespread shortages of critical life‐saving drugs. These shortages have emphasized the importance of reshoring drug manufacturing to the U.S. and fostered a regulatory landscape that seeks advanced methods of API manufacturing in order to bolster America's supply chain resiliency. Recently, increased attention has been directed towards continuous drug manufacturing to enable nonstop API production within modular stand‐alone flow systems. Continuous manufacturing (CM) facilitates modular automation, offers new avenues towards process intensification, and even enables in‐line analytical monitoring from raw materials to packaged products. This strategy offers better conversion, increased safety, reduced waste, and overall cost savings versus traditional batch‐style processing. The rise of advanced API manufacturing, coupled with the current regulatory landscape, offers a rare window of opportunity to convert traditional batch pharmaceutical processes into continuous, streamlined production systems to on‐shore API manufacturing and eliminate drug shortages. This review will outline the current state‐of‐the‐art in the CM of APIs and will highlight the translation of chemistry and unit operations from batch processes to modular CM systems.

Abstract:Ionic liquids (ILs) have been widely used in biomedical and pharmaceutical fields as solvents or permeation enhancers. Recently, more and more researchers focused on optimizing the physicochemical properties of active pharmaceutical ingredient (API) by ILs technology. Converting APIs into ILs (API-ILs) has shown great potential for drug delivery by eliminating polymorphism, tailoring solubility, improving thermal stability, increasing dissolution, controlling drug release, modulating the surfactant properties, enhancing permeability of APIs and modulating cytotoxicity on tumor cells. In addition, API-ILs are also used in various formulations as active ingredients, such as solutions, emulsions, even tablets or nanoparticles. This paper aims to review current status of API-ILs, including the rational and design, preparation and characterization, the improvement on the physicochemical characteristics of APIs, the compatibility of API-ILs with various formulations, and the future prospects of API-ILs in biomedical and pharmaceutical fields.

Abstract:Over the last 20 years, it has become clear that organocatalysis is the third pillar of catalysis. The low reactivity in the early days of organocatalysis has been overcome with the invention of more efficient catalysts, and by harnessing enabling technologies like continuous-flow chemistry and photo-redox catalysis. The main focus of this review is on the development over the last 10–15 years of key APIs using asymmetric organocatalysis. Due to significant engineering advances, and also due to the need for continuous manufacturing, flow and photo-redox approaches are becoming more widespread. Over the last 20 years, organocatalysis has been used on various occasions for accessing chiral drugs. The great advantage of using these catalysts is that the final active pharmaceutical ingredient (API) is metal-free. Also due to their inherent stability in air and water, they are very amenable to recovery via attachment to appropriate solid supports and also application in continuous flow systems. In recent years, more efficient organocatalysts have been developed, which includes the photoredox types, with much potential for chiral API synthesis.

Abstract:This perspective discusses the underexploited potential of renewable glycerol and its derivatives for the preparation of active pharmaceutical ingredients, some of which are on the World Health Organization list of essential medicines. The regulatory challenges faced by industries considering the replacement of petro-based building blocks with renewably sourced ones are described before diving into pharmaceutical ingredients that could potentially incorporate these bio-based atoms. Active Pharmaceutical Ingredients (APIs) are sorted following their therapeutic potential, including entities treating cardiovascular diseases, musculoskeletal drugs and compounds endowed with anti-infective properties. Finally, polymeric drugs and more eclectic substrates such as dietary supplements, radiosensitizers or chemotherapeutical agents are considered in the last two sub-sections. The broad spectrum of presented substrates relying on glycerol or potentially glycerol-derived reagents in their synthetic pathway emphasizes the potential contribution of bio-based substrates in already developed industrial processes. The presented examples hint towards a future chemical development in which APIs may be constructed with increasing percentages of bio-sourced atoms.

Abstract:Polymorphism, which plays an important role in the pharmaceutical industry, is quite common for active pharmaceutical ingredients (API). Owing to the structural differences between polymorphs, they often possess remarkably different physical and chemical properties, thus leading to diversity in curative effects and shelf life. How to precisely control the polymorph of API has attracted the attention of researchers around the world. In this paper, the regulation methods of drug polymorphs are comprehensively reviewed, to give a better view for the future research. The traditional method, including the control of the nucleation and growth process, is systematically introduced and analyzed from the aspects of the action mechanism and their application in recent years. In addition, novel techniques, including templates, microfluidics technology, ultrasound, laser, ionic liquids, and supramolecular gels, are expounded and analyzed in detail. An Automated Dual Modeling Approach to Accelerate Reaction Analysis and Optimization

Abstract:In modern pharmaceutical research, the demand for expeditious development of synthetic routes to active pharmaceutical ingredients (APIs) has led to a paradigm shift towards data-rich process development. Conventional methodologies en-compass prolonged timelines for reaction and analytical model developments. Both method developments are separated into different departments and often require an iterative process to optimize the models. Addressing this issue, we intro-duce an innovative dual modeling approach, seamlessly integrating the development of a Process Analytical Technology (PAT) strategy with reaction optimization. This integrated approach is exemplified in diverse amidation reactions and the synthesis of the API benznidazole. The platform, characterized by a high degree of automation and minimal operator in-volvement, achieves PAT calibration through a “standard addition” approach. Dynamic experiments are executed to screen a broad process space and gather data for fitting kinetic parameters. Employing a Julia-coded software program facilitates rapid kinetic parameter fitting and in-situ optimization within minutes. This highly automated workflow not only expedites the understanding and optimization of chemical processes, but also holds significant promise for time and resource savings within the pharmaceutical industry.
