PGL13, a novel allele of OsCRTISO, containing a polyamine oxidase domain, affects chloroplast development and heat stress response in rice

Abstract:Carotenoids are essential photosynthetic pigments in higher plants, play crucial roles in regulating chloroplast development and adaptation to various environmental stresses. The PGL13 gene encodes a carotenoid isomerase, which functions in regulating panicle germination, hormone responses, and tillering. However, the role of PGL13 in rice's response to abiotic stresses has rarely been studied. In this study, we characterized a rice mutant, pgl13 ( pale green leaf 13 ), which exhibited a pale-yellow color, accompanied by chloroplast structural disintegration, reduced chlorophyll content, and decreased photosynthetic parameters throughout its entire growth period. PGL13 was identified through map-based cloning, and the mutation in the third intron splicing recognition site of the PGL13 gene in the mutant changed from AG to TG, disrupting the normal splicing of the precursor of the PGL13 transcript and resulting in premature termination of translation. Further gene knockout and overexpression experiments confirmed that PGL13 is the gene responsible for controlling the leaf color phenotype. Functional analysis revealed that PGL13 includes a polyamine oxidase (PAO) domain, which was associated with plant stress-resistant. High temperature treatment showed that pgl13 showed sensitivity to high temperatures, with excessive accumulation of reactive oxygen species (ROS). These results indicate that the loss of PGL13 function leads to decreased photosynthesis and thermal sensitivity, where defects in carotenoid synthesis and the absence of the PAO domain are significant factors. This study enriches our understanding of carotenoids in abiotic stress responses and deepens our understanding of their roles in rice growth, development, and responses to abiotic stresses, provides a theoretical foundation for developing rice varieties with high light efficiency and stress resistance.
