Abstract:Polysaccharides, due to their remarkable features, have gained significant prominence in the sustainable production of nanoparticles (NPs). High market demand and minimal production cost, compared to the chemically synthesised NPs, demonstrate a drive towards polysaccharide-based nanoparticles (PSNPs) benign to environment. Various approaches are used for the synthesis of PSNPs including cross-linking, polyelectrolyte complexation, and self-assembly. PSNPs have the potential to replace a wide diversity of chemical-based agents within the food, health, medical and pharmacy sectors. Nevertheless, the considerable challenges associated with optimising the characteristics of PSNPs to meet specific targeting applications are of utmost importance. This review provides a detailed compilation of recent accomplishments in the synthesis of PSNPs, the fundamental principles and critical factors that govern their rational fabrication, as well as various characterisation techniques. Noteworthy, the multiple use of PSNPs in different disciplines such as biomedical, cosmetics agrochemicals, energy storage, water detoxification, and food-related realms, is accounted in detail. Insights into the toxicological impacts of the PSNPs and their possible risks to human health are addressed, and efforts made in terms of PSNPs development and optimising strategies that allow for enhanced delivery are highlighted. Finally, limitations, potential drawbacks, market diffusion, economic viability and future possibilities for PSNPs to achieve widespread commercial use are also discussed.

Abstract:Polysaccharides are the most abundant polymers in nature. They exhibit robust biocompatibility, reliable non-toxicity, and biodegradable character; thus, they are employed in multiple biomedical applications. The presence of chemically accessible functional groups on the backbone of biopolymers (amine, carboxyl, hydroxyl, etc.) makes them suitable materials for chemical modification or drug immobilisation. Among different drug delivery systems (DDSs), nanoparticles have been of great interest in scientific research in the last decades. In the following review, we want to address the issue of rational design of nanoparticle (NP)-based drug delivery systems in reference to the specificity of the medication administration route and resulting requirements. In the following sections, readers can find a comprehensive analysis of the articles published by authors with Polish affiliations in the last few years (2016–2023). The article emphasises NP administration routes and synthetic approaches, followed by in vitro and in vivo attempts toward pharmacokinetic (PK) studies. The 'Future Prospects' section was constructed to address the critical observations and gaps found in the screened studies, as well as to indicate good practices for polysaccharide-based nanoparticle preclinical evaluation.

Abstract:Polysaccharides are natural biopolymers that have been recognized to be the most promising hosts for the synthesis of metallic nanoparticles (MNPs) because of their outstanding biocompatible and biodegradable properties. Polysaccharides are diverse in size and molecular chains, making them suitable for the reduction and stabilization of MNPs. Considerable research has been directed toward investigating polysaccharide-based metallic nanoparticles (PMNPs) through host–guest strategy. In this review, approaches of preparation, including top-down and bottom-up approaches, are presented and compared. Different characterization techniques such as scanning electron microscopy, transmission electron microscopy, dynamic light scattering, UV-visible spectroscopy, Fourier-transform infrared spectroscopy, X-ray diffraction and small-angle X-ray scattering are discussed in detail. Besides, the applications of PMNPs in the field of wound healing, targeted delivery, biosensing, catalysis and agents with antimicrobial, antiviral and anticancer capabilities are specifically highlighted. The controversial toxicological effects of PMNPs are also discussed. This review can provide significant insights into the utilization of polysaccharides as the hosts to synthesize MPNs and facilitate their further development in synthesis approaches, characterization techniques as well as potential applications.

Abstract:Biocompatible and biodegradable polysaccharides are abundant and renewable natural materials. Polysaccharides and their derivatives are developed into various carrier materials for biomedical applications. In particular, advanced polysaccharide-based nano/microcapsules have received extensive attention in biomedical applications due to their good encapsulation ability and tunability. In recent years, polysaccharide-based nano/microcapsules have been widely used in drug carriers, gene carriers, antigen carriers, wound dressings, bioimaging and biosensors. Numerous research results have confirmed the feasibility, safety, and effectiveness of polysaccharide-based nano/microcapsules in the above-mentioned biomedical applications. This review discussed and analyzed the latest research strategies and design considerations for these applications in detail. The preparation methods, application strategies, and design considerations of polysaccharide-based nano/microcapsules are summarized and analyzed, and their challenges and future research prospects in biomedicine are further discussed. It is expected to provide researchers with inspiration and design ideas.

Abstract:With the development of nanotechnology, nanoparticles have played an important role in pharmaceuticals, foods and materials, in particular, protein/polysaccharide based composite nanoparticles have received attention from researchers for safety and green production. This paper summarized in detail the preparation methods, applications of protein/polysaccharide nanoparticles (PPNPs) in recent years, especially the mechanism of stabilizing the oil–water interface. Currently, the polysaccharides applied are more traditional, such as chitosan, pectin and carboxymethyl cellulose, so there is still a lot of room for the development of raw materials that can be used to prepare PPNPs. Based on this, we also proposed three promising polysaccharides: seaweed polysaccharide, lycium barbarum polysaccharide and lactobacillus exopolysaccharides, describing their characteristics as well as their application prospects, this article can serve as a reference for interested researchers.

Abstract:Polysaccharides have drawn a lot of attention due to their potential as carriers for drugs and other bioactive chemicals. In drug delivery systems, natural macromolecules such as polysaccharides are widely utilized as polymers. This utilization extends to various polysaccharides employed in the development of nanoparticles for medicinal administration, with the goal of enhancing therapeutic efficacy while minimizing side effects. This study not only offers an overview of the existing challenges faced by these materials but also provides detailed information on key polysaccharides expertly engineered into nanoparticles. Noteworthy examples include Bael Fruit Gum, Guar Gum , Pectin, Agar, Cellulose, Alginate , Chitin , and Gum Acacia, each selected for their distinctive properties and strategically integrated into nanoparticles. The exploration of these natural macromolecules illuminates their diverse applications and underscores their potential as effective carriers in drug delivery systems. By delving into the unique attributes of each polysaccharide, this review aims to contribute valuable insights to the ongoing advancements in nanomedicine and pharmaceutical technologies. The overarching objective of this review research is to assess the utilization and comprehension of polysaccharides in nanoapplications, further striving to promote their continued integration in contemporary therapeutics and industrial practices.

Abstract:Polysaccharides are natural polymers with hydrophilic, biocompatible and biodegradable characteristics and have many opportunities in the food and pharmaceutical sectors. This review focuses on the field of nano and microstructures whose internal structure is based on networked polysaccharide chains in 3D i.e., polysaccharide nanogels (NGs) and microgels (MGs). As it is observed the number of articles on NGs and MGs in peer reviewed scientific journals has been increasing over the last two decades. At the same time, the relative contribution of polysaccharides in this field is gaining place. This review focuses on the different applied methods for the fabrication of a variety of polysaccharide-based NGs and MGs and aims to highlight the recent advances on the subject and present their potentials and properties with regards to their integration in aspects of medicinal and food sciences. The presentation of the recent advances in the application of polysaccharide NGs and MGs is divided in materials with potential as emulsion stabilizers and materials with potential as carriers of bioactives. For applications in the medical sector the division is based on the fabrication processes and includes self-assembled, electrostatically complexed/ionically crosslinked and chemically crosslinked NGs and MGs. It is concluded that many advances are expected in the application of these polysaccharide-based materials and in particular as nutrient-loaded emulsion stabilizers, viscosity modifiers and co-assembled structures in combination with proteins.

Abstract:Surface engineering of particles based on a polymeric coating is of great interest in materials design and applications. Due to the disadvantages of non-biodegradability and undesirable biocompatibility, the application of petroleum-based synthetic polymers coating in the biomedical field has been greatly limited. In addition, there is lack of a universal surface modification method to functionalize particles of different compositions, sizes, shapes, and structures. Thus, it is imperative to develop a versatile biopolymeric coating with good biocompatibility and tunable biodegradability for the preparation of functional particle materials regardless of their surface chemical and physical structures. Recently, the natural polysaccharide polymers (e.g. chitosan and cellulose), polyphenol-based biopolymers (e.g. polydopamine and tannic acid), and proteins (e.g. amyloid-like aggregates) have been utilized in surface modification of particles, and applications of these modified particles in the field of biomedicine have been also intensively exploited. In this review, the preparation of the above three coatings on particles surface are summarized, and the applications of these materials in drug loading/release, biomineralization, cell immobilization/protection, enzyme immobilization/protection, and antibacterial/antiviral are exemplified. Finally, the challenges and the future research directions on biopolymer coating for particles surface engineering are prospected.

Abstract:Natural occurring polymers, or biopolymers, represent a huge part of our planet biomass. They are formed by long chains of monomers of the same type or a combination of different ones. Polysaccharides are biopolymers characterized by complex secondary structures performing several roles in plants, animals, and microorganisms. Because of their versatility and biodegradability, some of them are extensively used for packaging, food, pharmaceutical, and biomedical industries as sustainable and renewable materials. In the recent years, their manipulation at the nanometric scale enormously increased the range of potential applications, boosting an interdisciplinary research attempt to exploit all the potential advantages of nanostructured polysaccharides. Biomedical investigation mainly focused on nano-objects aimed at drug delivery, tissue repair, and vaccine adjuvants. The achievement of all these applications requires the deep knowledge of polysaccharide nanomaterials’ interactions with the immune system, which orchestrates the biological response to any foreign substance entering the body. In the present manuscript we focused on natural polysaccharides of high commercial importance, namely, starch, cellulose, chitin, and its deacetylated form chitosan, as well as the seaweed-derived carrageenan and alginate. We reviewed the available information on their biocompatibility, highlighting the importance of their physicochemical feature at the nanoscale for the modulation of the immune system.

Abstract:Nanotechnology being undoubtedly an uncut gem over the past few years has been in sighting as a new form of branch with its vigorous discoveries which have led to its divergent evolution giving emergence not only in the pathway of knowledge but also developing technological techniques. The constituting nanoparticles and its versatile properties with dynamic structures have made a major breakthrough in the past few years for its role in biotechnology arising nanobiotechnology, antipollution, renewable polymers, and its biomedical applications. Nanostructure composites forming nanomaterials on the basis of its working are pectin, cellulose, lignin, hyaluronic acid, bacterial cellulose, Arabic gum, and bacterial biosurfactants. In the recent years, it is seen that nanocomposites are giving promising results in medical technology incorporating with useful metal nanoparticles such as silver nanoparticles (AgNPs), gold nanoparticles (AuNPs), diamond nanoparticles, zinc oxide (ZnO), and titanium oxide (TiO). Some useful biomedical applications are in anticancerous, sunscreen, antiageing, and antitumorous. They have shown to be nontoxic at a certain level. Nanoparticle composites have proven with right amount of doping, and experienced techniques have given excellent results. Nanofibers of biodegradable poly(L-lactide) (PLLA)/poly(lactide-co-glycol ide) (PLGA) compounds are used in drug delivery, folate redox-responsive chitosan nanoparticles (FTC-NPs) also as anticancer drug delivery, and mesoporous silica nanoparticles-silver nanoparticles as a tissue growth in vivo processes. The study of a biosynthetic pathway of therapeutic drugs is still much needed. Waste management of renewable nanopolymers are an ultimate goal so that there are less haphazard elements towards the environment.

Abstract:Recently, owing to well-controlled release, enhanced distribution and increased permeability, nanocarriers used for alternative drug and food-delivery strategies have received increasingly attentions. Nanocarriers have attracted a large amount of interest as potential carriers of various bioactive molecules for multiple applications. Drug and food-based delivery via polymeric-based nanocarriers and lipid-based nanocarriers has been widely investigated. Nanocarriers, especially liposomes, are more and more widely used in the area of novel nano-pharmaceutical or food-based design. Herein, we aimed to discuss the recent advancement of different surface-engineered nanocarriers type, along with cutting-edge applications for food and nanomedicine and highlight the alternative of phytochemical as nanocarrier. Additionally, safety concern of nanocarriers was also highlighted.

Abstract:Polysaccharides are biopolymers distinguished by their complex secondary structures executing various roles in microorganisms, plants, and animals. They are made up of long monomers of similar type or as a combination of other monomeric chains. Polysaccharides are considered superior as compared to other polymers due to their diversity in charge and size, biodegradability, abundance, bio-compatibility, and less toxicity. These natural polymers are widely used in designing of nanoparticles (NPs) which possess wide applications in therapeutics, diagnostics, delivery and protection of bioactive compounds or drugs. The side chain reactive groups of polysaccharides are advantageous for functionalization with nanoparticle-based conjugates or therapeutic agents such as small molecules, proteins, peptides and nucleic acids. Polysaccharide NPs show excellent pharmacokinetic and drug delivery properties, facilitate improved oral absorption, control the release of drugs, increases in vivo retention capability, targeted delivery, and exert synergistic effects. This review updates the usage of polysaccharides based NPs particularly cellulose, chitosan, hyaluronic acid, alginate, dextran, starch, cyclodextrins, pullulan, and their combinations with promising applications in diabetes, organ fibrosis and arthritis.

Abstract:Polysaccharides are essential macromolecules which almost exist in all living forms, and have important biological functions, they are getting more attention because they exhibit a wide range of biological and pharmacological activities, such as anti-tumour, immunomodulatory, antimicrobial, antioxidant, anticoagulant, antidiabetic, antiviral, and hypoglycemia activities, making them one of the most promising candidates in biomedical and pharmaceutical fields. Polysaccharides can be obtained from many different sources, such as plants, microorganisms, algae, and animals. Due to their physicochemical properties, they are susceptible to physical and chemical modifications leading to enhanced properties, which is the basic concept for their diverse applications in biomedical and pharmaceutical fields. In this review, we will give insight into the most recent updated applications of polysaccharides and their potentialities as alternatives for traditional and conventional therapies. Challenges and limitations for polysaccharides in pharmaceutical utilities are discussed as well.

Abstract:Nanoparticles with unique properties have potential applications in food, medicine, pharmacology, and agriculture industries. Accordingly, many significant researches have been conducted to develop novel nanoparticles using chemical and biological techniques. This review focuses on the synthesis of selenium nanoparticles (SeNPs) using polysaccharides as templates. Various instrumental techniques being used to confirm the formation of polysaccharide-SeNPs conjugates and characterize the properties of nanoparticles are also introduced. Finally, the biological activities of the synthesized SeNPs and the influence of structural factors of polysaccharides on the property of synthetic nanocomposites are highlighted. In general, the polysaccharides functionalized SeNPs can be easily obtained using sodium selenite as precursor and ascorbic acid as reductant. The final products having different particle size, morphology, and selenium content exhibit abundant physiological activities. Structural factors of polysacchairdes involving molecular weights, substitution of functional groups, and chain conformation play determinant roles on the properties of nanocomposites, resulting in different biological performances. The review on the achievements and current status of polysaccharides conjugated SeNPs provides insights into this exciting research topic for further studies in the future.

Abstract:Background: Polysaccharides are a type of natural macromolecular polymer that can be found in plants, animals, fungi, algae, and marine organisms. Its activities have piqued the interest of researchers. The internal structure, as well as their chemical and physical properties, dictate how they work. Polysaccharide functionalities are progressively being chemically changed. Using this approach, polysaccharides' structural, physicochemical, and biological properties can all be altered. Aim and Methods: The review sought to provide an overview of polysaccharide modification but also biological use. Recent research has shown that chemically modifying polysaccharides may increase their immunological function as well as their antiviral, antibacterial, antioxidant, as well as other characteristics. There are several chemical modifications, including sulfation, carboxymethylation, acetylation, phosphorylation, and others. Modified polysaccharide recent developments are reviewed. Discussion and Result: Polysaccharide physiochemical properties and biological activity can change as their structural properties change. The structural modifications that occur depend on the source of the polysaccharides. Chemical modification has enormous promise for enhancing biomedical applications. These modified polysaccharides have made significant contributions to tissue engineering and drug delivery applications. Modification of polysaccharides induces therapeutic benefits. The immunomodulation of polysaccharides and their derivatives, as well as their chemical modification, has been studied and discussed. Conclusion: These modified polysaccharides have the potential to be used for wound dressing, gene delivery, drug delivery, etc.

Abstract:Since many nutrients are highly sensitive, they cannot be absorbed and utilized efficiently by the body. Using nano-delivery systems to encapsulate nutrients is an effective method of solving the problems associated with the application of nutrients at this stage. Polysaccharides, as natural biomaterials, have a unique chemical structure, ideal biocompatibility, biodegradability and low immunogenicity. This makes polysaccharides powerful carriers that can enhance the biological activity of nutrients. However, the true role of polysaccharide-based delivery systems requires an in-depth understanding of the structural and physicochemical characteristics of polysaccharide-based nanodelivery systems, as well as effective modulation of the intestinal delivery mechanism and the latest advances in nano-encapsulation. This review provides an overview of polysaccharide-based nano-delivery systems dependent on different carrier types, emphasizing recent advances in the application of polysaccharides, a biocomposite material designed for nutrient delivery systems. Strategies for polysaccharide-based nano-delivery systems to enhance the bioavailability of orally administered nutrients from the perspective of the intestinal absorption barrier are presented. Characterization methods for polysaccharide-based nano-delivery systems are presented as well as an explanation of the formation mechanisms behind nano-delivery systems from the perspective of molecular forces. Finally, we discussed the challenges currently facing polysaccharide-based nano-delivery systems as well as possible future directions for the future.

Abstract:Protein-polysaccharide nanoconjugates are covalently interactive networks that are currently the subject of intense research owing to their emerging applications in the food nanotechnology field. Due to their biocompatibility and biodegradability properties, they have played a significant role as wall materials for the formation of various nanostructures to encapsulate nutraceuticals. The food-grade protein-polysaccharide nanoconjugates would be employed to enhance the delivery and stability of nutraceuticals for their real use in the food industry. The most common edible polysaccharides (cellulose, chitosan, pectin, starch, carrageenan, fucoidan, mannan, glucomannan, and arabic gum) and proteins (silk fibroin, collagen, gelatin, soy protein, corn zein, and wheat gluten) have been used as potential building blocks in nano-encapsulation systems because of their excellent physicochemical properties. This article broadens the discussion of food-grade proteins and polysaccharides as nano-encapsulation biomaterials and their fabrication methods, along with a review of the applications of protein-polysaccharide nanoconjugates in the delivery of plant-derived nutraceuticals.
