Abstract:Our understanding of the neurobiology of primate behaviour largely derives from artificial tasks in highly controlled laboratory settings, overlooking most natural behaviours that primate brains evolved to produce 1,2,3 . How primates navigate the multidimensional social relationships that structure daily life 4 and shape survival and reproductive success 5 remains largely unclear at the single-neuron level. Here we combine ethological analysis, computer vision and wireless recording technologies to identify neural signatures of natural behaviour in unrestrained, socially interacting pairs of rhesus macaques. Single-neuron and population activity in the prefrontal and temporal cortex robustly encoded 24 species-typical behaviours, as well as social context. Male–female partners demonstrated near-perfect reciprocity in grooming, a key behavioural mechanism supporting friendships and alliances 6 , and neural activity maintained a running account of these social investments. Confronted with an aggressive intruder, behavioural and neural population responses reflected empathy and were buffered by the presence of a partner. Our findings reveal a highly distributed neurophysiological ledger of social dynamics, a potential computational foundation supporting communal life in primate societies, including our own.

Abstract:Primates have rich social lives orchestrated by brain circuits that are still poorly understood, partly because they have not been studied under naturalistic conditions. New wireless technology for recording neuronal activity in freely moving monkeys enabled insights into how neurons track natural social interactions, including reciprocity and social support towards a partner. Recoding the activity of individual neurons in freely moving macaques uncovers a neural population that encodes social behaviours.

Little is known about the neural circuitry that underlies this understanding. Sliwa and Freiwald scanned monkey brains as the monkeys watched static or moving stimuli. A subset of brain areas was exclusively active during monkey-monkey interactions, as opposed to physical interactions between two objects. This network shares some of its components with the monkey mirror neuron system mapped previously by others and with a possible homolog of the human network involved in the theory of mind. Abstract:Event Abstract Back to Event Behavior-dependent responses in primate frontal cortex neurons during natural behavior Primates are typified by their complex social systems and the sophisticated behaviors used to navigate them. Although relatively little research has been directly aimed at examining the neural substrates underlying primate social behaviors, it is clear that producing these behaviors requires an integration of both the sensory and motor information. While data on sensory-motor interactions in the primate cortex are available, relatively little remains known about this process in species - natural behaviors. Given the significance of natural behaviors in the evolutionary history and daily lives of nonhuman primates, empirical study of their underlying neural mechanisms may reveal unique insights into cortical functions. Here we recorded the single-unit activity of frontal cortex neurons in freely-moving common marmoset monkeys (Callithrix jacchus) engaged in a natural, species-typical behavior known as antiphonal calling, a vocal behavior involving the reciprocal exchange of vocalizations between conspecifics. Producing an antiphonal call is dependent upon first hearing a particular vocalization, a phee call, and producing the same call type in response. Importantly, the sequence of sensory and motor events in this behavior occurs over a 10-15s period of time. In our first set of analyses, we compared neural activity during each of the three elements of this behavior: sensory period (phee call stimulus), latency delay, and vocal-motor response (antiphonal call). The aim here was to determine whether neurons across the population showed firing rate changes during any of these three periods individually or in combination. Preliminary analyses revealed neuronal responses during each of these behavioral periods, with a population showing behaviorally modulated activity over the duration of the antiphonal calling behavior. This population of neurons typically showed one of two responses. The first class showed an increased firing rate during the phee stimulus and suppressed activity during the antiphonal call. The second class showed the inverse response, exhibiting suppression during phee stimulus and excitation during the antiphonal response. In our second set of analyses, we examined the effects of behavioral context of the neural response observed during antiphonal calling. Specifically, during test sessions, we presented subjects with phee call stimuli that did not elicit antiphonal callings and subjects produced phee calls spontaneously. As such, we tested whether neurons responded similarly during either the sensory stimulus or motor response alone compared to the same sensory or motor period during antiphonal calls. Our initial analysis suggests that the aforementioned "antiphonal calling" neurons show little response during the sensory stimulus and vocal-motor response independently. Together these data suggest that it is the sensory-motor integration during the antiphonal calling behavior that is driving the neural response. This study builds on our previous work examining the ethology and functional neuroanatomy of antiphonal calling in common marmosets. Network nestedness in primates: a structural constraint or a biological advantage of social complexity?

Abstract:This study investigates the prevalence and implications of nestedness within primate social networks, examining its relationship with cognitive and structural factors. We analysed data from 51 primate groups across 21 species, employing network analysis to evaluate nestedness and its correlation with modularity, neocortex ratio, and group size. We used Bayesian mixed effects modelling to investigate nestedness in primate social networks, controlling for phylogenetic dependencies and exploring various factors like neocortex ratio and group size. Our findings reveal a significant occurrence of nestedness in 66% of the species studied, exceeding chance expectations. This nestedness was more pronounced in groups with less steep dominance hierarchies, contrary to traditional assumptions linking it to hierarchical social structures. A notable inverse relationship between nestedness and modularity was observed, suggesting a structural trade-off in network formation. This pattern persisted even after controlling for species-specific social behaviours, indicating a general structural feature of primate networks. Surprisingly, our analysis showed no significant correlation between nestedness and neocortex ratio or group size, challenging the social brain hypothesis and suggesting a greater role for ecological factors in cognitive evolution. This study emphasises the importance of weak links in maintaining network resilience. Overall, our research provides new insights into primate social network structures, highlighting complex interplays between network characteristics and challenging existing paradigms in cognitive and evolutionary biology.

Abstract:Primates exhibit complex brain structures that augment cognitive function. The neocortex fulfills high-cognitive functions through billions of connected neurons. These neurons have distinct transcriptomic, morphological, and electrophysiological properties, and their connectivity principles vary. These features endow the primate brain atlas with a multimodal nature. The recent integration of next-generation sequencing with modified patch-clamp techniques is revolutionizing the way to census the primate neocortex, enabling a multimodal neuronal atlas to be established in great detail: (1) single-cell/single-nucleus RNA-seq technology establishes high-throughput transcriptomic references, covering all major transcriptomic cell types; (2) patch-seq links the morphological and electrophysiological features to the transcriptomic reference; (3) multicell patch-clamp delineates the principles of local connectivity. Here, we review the applications of these technologies in the primate neocortex and discuss the current advances and tentative gaps for a comprehensive understanding of the primate neocortex.

The big mixup: Neural representation during natural modes of primate visual behavior

Abstract:The primate brain has evolved specialized visual capacities to navigate complex physical and social environments. Researchers studying cortical circuits underlying these capacities have traditionally favored the use of simplified tasks and brief stimulus presentations in order to isolate cognitive variables with tight experimental control. As a result, operational theories about visual brain function have come to emphasize feature detection, hierarchical stimulus encoding, top-down task modulation, and functional segregation in distinct cortical areas. Recently, however, experimental paradigms combining natural behavior with electrophysiological recordings have begun to offer a distinctly different portrait of how the brain takes in and analyzes its visual surroundings. The present article reviews recent work in this area, highlighting some of the more surprising findings in domains of social vision and spatial navigation along with shifts in thinking that have begun to emanate from this approach.

Abstract:Social behavior is highly complex and adaptable. It can be divided into multiple temporal stages: detection, approach, and consummatory behavior. Each stage can be further divided into several cognitive and behavioral processes, such as perceiving social cues, evaluating the social and non-social contexts, and recognizing the internal/emotional state of others. Recent studies have identified numerous brain-wide circuits implicated in social behavior and suggested the existence of partially overlapping functional brain networks underlying various types of social and non-social behavior. However, understanding the brain-wide dynamics underlying social behavior remains challenging, and several brain-scale dynamics (macro-, meso-, and micro-scale levels) need to be integrated. Here, we suggest leveraging new tools and concepts to explore social brain networks and integrate those different levels. These include studying the expression of immediate-early genes throughout the entire brain to impartially define the structure of the neuronal networks involved in a given social behavior. Then, network dynamics could be investigated using electrode arrays or multi-channel fiber photometry. Finally, tools like high-density silicon probes and miniscopes can probe neural activity in specific areas and across neuronal populations at the single-cell level.
A New Research Trend in Social Neuroscience: Towards an Interactive-Brain Neuroscience

Abstract:The ability to flexibly modulate our behaviors in social contexts and to successfully interact with other persons is a fundamental, but pivotal, requirement for human survival. Although previous social neuroscience research with single individuals has contributed greatly to our understanding of the basic mechanisms underlying social perception and social emotions, much of the dynamic nature of interactions between persons remains beyond the reach of single-brain studies. This has led to a growing argument for a shift to the simultaneous measurement of the brain activity of two or more individuals in realistic social interactions-an approach termed "hyperscanning." Although this approach offers important promise in unlocking the brain's role in truly social situations, there are multiple procedural and theoretical questions that require review and analysis. In this paper we discuss this research trend from four aspects: hyperscanning apparatus, experimental task, quantification method, and theoretical interpretation. We also give four suggestions for future research: (a) electroen-cephalography and near-infrared spectroscopy are useful tools by which to explore the interactive brain in more ecological settings; (b) games are an appropriate method to simulate daily life interactions; (c) transfer entropy may be an important method by which to quantify directed exchange of information between brains; and (d) more explanation is needed of the results of interbrain synchronization itself.
Representing the dynamics of natural marmoset vocal behaviors in frontal cortex

Abstract:Here we tested the respective contributions of primate premotor and prefrontal cortex to support vocal behavior. We applied a model-based GLM analysis that better accounts for the inherent variance in natural, continuous behaviors to characterize the activity of neurons throughout frontal cortex as freely-moving marmosets engaged in conversational exchanges. While analyses revealed functional clusters of neural activity related to the different processes involved in the vocal behavior, these clusters did not map to subfields of prefrontal or premotor cortex, as has been observed in more conventional task-based paradigms. Our results suggest a distributed functional organization for the myriad neural mechanisms underlying natural social interactions and has implications for our concepts of the role that frontal cortex plays in governing ethological behaviors in primates.

Abstract:To better understand the molecular and cellular differences in brain organization between human and nonhuman primates, we performed transcriptome sequencing of 16 regions of adult human, chimpanzee, and macaque brains. Integration with human single-cell transcriptomic data revealed global, regional, and cell-type-specific species expression differences in genes representing distinct functional categories. We validated and further characterized the human specificity of genes enriched in distinct cell types through histological and functional analyses, including rare subpallial-derived interneurons expressing dopamine biosynthesis genes enriched in the human striatum and absent in the nonhuman African ape neocortex. Our integrated analysis of the generated data revealed diverse molecular and cellular features of the phylogenetic reorganization of the human brain across multiple levels, with relevance for brain function and disease.

Abstract:One of the most exciting new developments in systems neuroscience is the progress being made toward neurophysiological experiments that move beyond simplified laboratory settings and address the richness of natural behavior. This is enabled by technological advances such as wireless recording in freely moving animals, automated quantification of behavior, and new methods for analyzing large data sets. Beyond new empirical methods and data, however, there is also a need for new theories and concepts to interpret that data. Such theories need to address the particular challenges of natural behavior, which often differ significantly from the scenarios studied in traditional laboratory settings. Here, we discuss some strategies for developing such novel theories and concepts and some example hypotheses being proposed.

Abstract:The common marmoset (Callithrix jacchus) has garnered interest recently as a powerful model for the future of neuroscience research. Much of this excitement has centered on the species' reproductive biology and compatibility with gene editing techniques, which together have provided a path for transgenic marmosets to contribute to the study of disease as well as basic brain mechanisms. In step with technical advances is the need to establish experimental paradigms that optimally tap into the marmosets' behavioral and cognitive capacities. While conditioned task performance of a marmoset can compare unfavorably with rhesus monkey performance on conventional testing paradigms, marmosets' social behavior and cognition are more similar to that of humans. For example, marmosets are among only a handful of primates that, like humans, routinely pair bond and care cooperatively for their young. They are also notably pro-social and exhibit social cognitive abilities, such as imitation, that are rare outside of the Apes. In this Primer, we describe key facets of marmoset natural social behavior and demonstrate that emerging behavioral paradigms are well suited to isolate components of marmoset cognition that are highly relevant to humans. These approaches generally embrace natural behavior, which has been rare in conventional primate testing, and thus allow for a new consideration of neural mechanisms underlying primate social cognition and signaling. We anticipate that through parallel technical and paradigmatic advances, marmosets will become an essential model of human social behavior, including its dysfunction in neuropsychiatric disorders.
Multi-dimensional social relationships shape social attention in monkeys

Abstract:Social relationships guide individual behavior and ultimately shape the fabric of society. Primates exhibit particularly complex, differentiated, and multidimensional social relationships, which form interwoven social networks, reflecting both individual social tendencies and specific dyadic interactions. How the patterns of behavior that underlie these social relationships emerge from moment-to-moment patterns of social information processing remains unclear. Here, we assess social relationships among a group of four monkeys, focusing on aggression, grooming, and proximity. We show that individual differences in social attention vary with individual differences in patterns of general social tendencies and patterns of individual engagement with specific partners. Oxytocin administration altered social attention and its relationship to both social tendencies and dyadic relationships, particularly grooming and aggression. Our findings link the dynamics of visual information sampling to the dynamics of primate social networks.

Abstract:Primate social systems are difficult to characterize, and existing classification schemes have been criticized for being overly simplifying, formulated only on a verbal level or partly inconsistent. Social network analysis comprises a collection of analytical tools rooted in the framework of graph theory that were developed to study human social interaction patterns. More recently these techniques have been successfully applied to examine animal societies. Primate social systems differ from those of humans in both size and density, requiring an approach that puts more emphasis on the quality of relationships. Here, we discuss a set of network measures that are useful to describe primate social organization and we present the results of a network analysis of 70 groups from 30 different species. For this purpose we concentrated on structural measures on the group level, describing the distribution of interaction patterns, centrality, and group structuring. We found considerable variability in those measures, reflecting the high degree of diversity of primate social organizations. By characterizing primate groups in terms of their network metrics we can draw a much finer picture of their internal structure that might be useful for species comparisons as well as the interpretation of social behavior.

Abstract:Social network position in non-human primates has far-reaching fitness consequences. Critically, social networks are both heterogeneous and dynamic, meaning an individual's current network position is likely to change due to both intrinsic and extrinsic factors. However, our understanding of the drivers of changes in social network position is largely confined to opportunistic studies. Experimental research on the consequences of in situ, controlled network perturbations is limited. Here we conducted a food-based experiment in rhesus macaques to assess whether allowing an individual the ability to provide high-quality food to her group changed her social behavioural relationships. We considered both her social network position across five behavioural networks, as well as her dominance and kin interactions. We found that gaining control over a preferential food resource had far-reaching social consequences. There was an increase in both submission and aggression centrality and changes in the socio-demographic characteristics of her agonistic interaction partners. Further, we found that her grooming balance shifted in her favour as she received more grooming than she gave. Together, these results provide a novel, preliminary insight into how in situ , experimental manipulations can modify social network position and point to broader network-level shifts in both social capital and social power.

Physiologically Distinct Neuronal Type in Primate Cortex

Abstract:Unlike other physiological systems, the central nervous system (CNS) of primates evolves dramatically from that of other mammal species. Therefore, neuroscientists are concerned that conclusions derived from studies using common model animals, such as rodents, may not apply to higher primates, especially the humans. Even though studies on primates are strongly restricted by limited subject resource and accessibility, they have been much valued across all the extent of neuroscience. One of the promising means to uncover the detailed functional organization of human neuronal microcircuit is to take advantage of the discarded brain tissue during neurosurgery and directly perform electrophysiological recording from human neurons. Differences in neuronal cell type or synaptic connectivity pattern between CNS circuits of higher and lower mammals, qualitatively and/or quantitatively, would be no doubt one of the most exciting findings in the field of neuroscience. In our recent study, we reported a subgroup of human and monkey cortical inhibitory interneurons possessing distinct intrinsic physiological features, but similar features were not found in rodent neocortical neurons [1]. Those neurons could fire a train of persistent action potentials (APs) after each brief single-AP stimulus, thus we named them persistent-activity neurons (PANs). Stimulation-triggered persistent AP firing is quite widely found in central neurons [2, 3]. However, PANs we found in human and monkey neocortex are distinct in two aspects. First, unlike other forms of persistent activity that require prolonged burst of APs, the persistent firing in PANs is triggered by brief but stopped by strong stimuli. Second, the mechanism for the generation of persistent firing in PANs is attributed to the activation of a persistent Na+ current without the involvement of metabotropic neurotransmitter receptors. We speculate that PANs may contribute to distinct physiological functions in primate brain. Since PANs are inhibitory interneurons, they are most likely responsible for controlling the responsiveness of principal cells by providing inhibitory tone in local circuits. Hypothetically, cortical neurons discriminate between strong and weak stimuli, putatively behavior relevant and irrelevant, by not only the firing frequency during the stimulation but also the following persistent activity. In rodent brain, strong stimuli can trigger long-lasting depolarizing plateau or persistent activity in principal cells, which may elevate their responsiveness to incoming synaptic inputs arrived right after the stimulation. In contrast, with the help of persistent activity in PANs, the responsiveness of principal cells in primate neocortex could be decreased by long-lasting inhibitory tone after a weak stimulus; the cessation of PAN's persistent activity induced by a relatively strong stimulus, however, would increase the responsiveness of principal cells. Thus, PANs may help promote the discriminative ability of primate cortex (Figure (Figure11). Figure 1 Schematic drawing showing the hypothesized behavior relevance of PANs in primates The experimental results provide evidence that neuronal types with distinct physiological properties exist in our human brain, and it is possible that they may contribute to unique human brain functions. However, there remains a huge gap between these unique neuronal properties and the higher-order cognitive functions. For example, it is still unclear whether PANs are providing inhibition or disinhibition to neighboring principal cells. Cortical disinhibitory tone has been demonstrated recently playing important and distinct roles in cortical functions. Future studies may focus on unveiling the local synaptic connectivity of PANs and their contribution to the operation of local circuits. It is also important to investigate the function of PANs in vivo in non-human primates. But first we need to identify some molecular markers that can help in targeting group of PANs in animal models. Single-cell RT-PCR in our experiment could only reveal whether PANs express known markers of cortical neurons, future studies using single-cell transcriptome together with patch-clamp recording [4] may help in identifying new specific markers for PANs. To search for new cell types in the human neocortex, previous studies focusing on distinct morphological features turn out to be unfruitful, though important findings including the observation of spindle neurons (also known as Von Economo neurons) have been reported [5, 6]. Our study, however, opens a new avenue in searching for unique cell types in human brain by paying attention to their distinct physiological properties. The human brain tissues removed during neurosurgery could be very promising resources for studying the substrates of higher-order brain functions. Though unique features of human neurons are the most eye-catching, findings proving consistence between human and lower animals are, to some extent, no less valuable. For example, a recent study from our lab demonstrated that asynchronous GABA release in human neocortex was enhanced in epileptic conditions, and this pathological change was consistent with that occurred in animal disease model [7], greatly promoting the clinical significance of the findings. Together, our studies and those of others using human brain tissues provide direct quantitative morphological and physiological measurements of human neurons and their circuits. We believe the results are very important for comprehensive understanding and reconstruction of human intelligence.
The meso-connectomes of mouse, marmoset, and macaque: network organization and the emergence of higher cognition

Abstract:The recent publications of the inter-areal connectomes for mouse, marmoset, and macaque cortex have allowed deeper comparisons across rodent vs. primate cortical organization. In general, these show that the mouse has very widespread, "all-to-all" inter-areal connectivity (i.e. a "highly dense" connectome in a graph theoretical framework), while primates have a more modular organization. In this review, we highlight the relevance of these differences to function, including the example of primary visual cortex (V1) which, in the mouse, is interconnected with all other areas, therefore including other primary sensory and frontal areas. We argue that this dense inter-areal connectivity benefits multimodal associations, at the cost of reduced functional segregation. Conversely, primates have expanded cortices with a modular connectivity structure, where V1 is almost exclusively interconnected with other visual cortices, themselves organized in relatively segregated streams, and hierarchically higher cortical areas such as prefrontal cortex provide top-down regulation for specifying precise information for working memory storage and manipulation. Increased complexity in cytoarchitecture, connectivity, dendritic spine density, and receptor expression additionally reveal a sharper hierarchical organization in primate cortex. Together, we argue that these primate specializations permit separable deconstruction and selective reconstruction of representations, which is essential to higher cognition.
