Abstract:OBJECTIVE:To investigate the pathogenic mechanisms of airway inflammation and recurrent wheezing induced by recurrent respiratory virus infection after respiratory syncytial virus (RSV) infection.METHODS:Sixty-four female BALB/c mice (aged 6-8 weeks) were randomly divided into four groups: control, RSV, Poly(I:C), and RSV+Poly(I:C) (n=16 each). The bronchoalveolar lavage fluid (BALF) was collected on the 3rd day after Poly(I:C) administration, and the total cell number and differential counts in BALF were determined. Hematoxylin-eosin staining was used to observe pulmonary pathological changes. The airway responsiveness was detected. ELISA was used to measure the levels of interferon-γ (IFN-γ), interleukin-4 (IL-4), interleukin-13 (IL-13), matrix metallopeptidase-9 (MMP-9), and tissue inhibitor of metalloproteinase-1 (TIMP-1) in BALF.RESULTS:Compared with the other three groups, the RSV+Poly(I:C) group had significant increases in the total number of inflammatory infiltrating cells in the airway, airway responsiveness, and MMP-9 level in BALF (P<0.05). The RSV+Poly(I:C) group showed more severe pulmonary tissue injuries compared with the control and RSV groups (P<0.01). Compared with the RSV group, the RSV+Poly(I:C) group showed significant reductions in the levels of IL-4 and TIMP-1 in BALF (P<0.01).CONCLUSIONS:Viral re-infection in the late stage of RSV infection may cause an imbalance of MMP-9/TIMP-1 expression and thus contribute to aggravated airway inflammation.

Abstract:Respiratory Syncytial Virus (RSV) infections are the dominant cause of pneumonia in children. In order to determine the epidemiological characteristics and immune status of children with Respiratory Syncytial Virus, a prospective study was performed among patients with RSV infection. Comparisons between RSV pneumonia group and normal control group, RSV pneumonia group had lower IL-2 (median levels, pg/ml: 3.8 vs. 5.1, P < 0.01), and higher IL-4 (median levels, pg/ml: 3.2 vs. 2.4, P < 0.01), IL-10 (median levels, pg/ml: 12.2 vs. 2.3, P < 0.01), and IFN-γ (median levels, pg/ml: 13.4 vs. 4.6, P < 0.01). The level of IgE among pneumonia patients caused by RSV increased sharply (median levels, mg/L: 48.1 vs. 8.8, P < 0.01). Another amazing finding is that after birth, the degree of IgE of the children infected by RSV increases gradually with age. This effect is at its peak in 0.6 years old. The IgE and eosinophil levels were higher when patients suffered from RSV pneumonia with wheeze (IgE median levels, IU/ml: with wheeze: 72.74 vs. without wheeze: 11.5, P < 0.05; eosinophil median levels, ×10(9) /l: with wheeze: 0.21 vs. without wheeze: 0.05, P < 0.05). The main morbidity crowd is the children under the age of 1 year old. The downregulation of IL2 and the upregulation of IL-4, IL-10, IFN-γ, and IgE happen after RSV infection.

Abstract:Background and objectiveInfluenza virus (FLU), rhinovirus (RV) and respiratory syncytial virus (RSV) are the most common acute respiratory infections worldwide. Infection can cause severe health outcomes, while therapeutic options are limited, primarily relieving symptoms without attenuating the development of lesions or impaired lung function. We therefore examined the inflammatory response to these infections with the intent to identify common components that are critical drivers of immunopathogenesis and thus represent potential therapeutic targets. MethodsBALB/c mice were infected with FLU, RV or RSV, and lung function, airway inflammation and immunohistopathology were measured over a 10-day period. Anti-IL-17A mAb was administered to determine the impact of attenuating this cytokine's function on the development and severity of disease. ResultsAll three viruses induced severe airway constriction and inflammation at 2 days post-infection (dpi). However, only FLU induced prolonged inflammation till 10 dpi. Increased IL-17A expression was correlated with the alterations in lung function and its persistence. Neutralization of IL-17A did not affect the viral replication but led to the resolution of airway hyperresponsiveness. Furthermore, anti-IL-17A treatment resulted in reduced infiltration of neutrophils (in RV- and FLU-infected mice at 2 dpi) and lymphocytes (in RSV-infected mice at 2 dpi and FLU-infected mice at 10 dpi), and attenuated the severity of immunopathology. ConclusionIL-17A is a common pathogenic molecule regulating disease induced by three prevalent respiratory viruses. Targeting the IL-17A pathway may provide a unified approach to the treatment of these respiratory infections alleviating both inflammation-induced lesions and difficulties in breathing.

Association of Cytokine Responses with Disease Severity in Infants with Respiratory Syncytial Virus Infection

Abstract:AIM:To explore the relationship between cytokine responses and severity of respiratory syncytial virus (RSV) infection in infants.METHODS:Intracellular interleukin-4 (IL-4) and interferon-gamma (IFN-gamma) expression in peripheral blood CD3+ and CD8+ lymphocytes was measured by four-colour flow cytometry. Serum IL-12, IL-4 and IFN-gamma levels were also determined by enzyme-linked immunosorbent assay.RESULTS:The frequency of IL-4 and IFN-gamma expression in CD3+CD8- cells was the same in RSV-infected, non-RSV-infected and control infants and in those with RSV bronchiolitis or RSV pneumonia, indicating that no Th2 predominance exists in the acute phase of RSV infection and RSV bronchiolitis. Furthermore, RSV-infected infants had a more frequent IFN-gamma expression in CD3+CD8+ cells than controls, and they also showed a much lower serum IL-4/ IFN-gamma ratio because of decreased IL-4 and elevated IFN-gamma, the latter being most prominent in RSV bronchiolitis. The serum IL-12 level in RSV-infected infants was the same as in control infants, while those with non-RSV infections had a much higher level. Serum IL-12, IFN-gamma and frequency of IFN-gamma expression in CD3+CD8+ cells in mild RSV infection were much higher than in controls, while no difference existed between severe cases and controls.CONCLUSION:Type 2 cytokine predominance was not found in the acute phase of RSV infection and RSV bronchiolitis, but both were accompanied by enhanced production of IFN-gamma and a much higher serum IFN-gamma level than in healthy controls, especially in those with RSV bronchiolitis, suggesting a role in causing airway obstruction. IFN-gamma and IL-12 may also play a protective role in RSV infections by diminishing viral replication, and high levels of IL-12 and IFN-gamma may be associated with lessening of the severity of infection.
The Lung Inflammatory and Immune Responses in IL-12 P40 (-/-) Mice Infected with RSV

Abstract:Objective To explore a possible role for IL-12 during acute RSV infection. Methods IL-12 P40 gene knock-out mice [IL-12P40 (-/-)]and wild-type (WT) mice were intratracheally infected with RSV (2×106 PFUs/mouse). The total inflammatory cells and different types of inflammatory cells in BALF stained with HE, CD3+ cells in lung tissue stained with immunohistochemical method, mucus in the airway stained with AP/PAS method, and Th1 and Th2 cytokines including IL-5, IL-13, IL-18 and IFN-γ in supernatant fluid of lung tissues were detected in both of IL-12 P40(-/-) mice and WT mice on day 3 and day 7 after RSV infection, respectively. Results Total inflammatory cells[(136.0±8.8)×107/L],lymphocytes [(69.98±6.8)×107/L], and neutrophils[(13.1±2.3)×107/L] in BALF were significantly higher in IL-12 P40(-/-) mice than those in WT mice [(88.8±15.2) ×107/L, (32.9±6.1)×107/L and (18.9±3.9) ×107/L] on day 7 after RSV infection (P0.01 for all). CD3+ cells [(113.1±12.3)/×(400 with 10 vision fields] in lung tissue increased obviously in IL-12 P40 (-/-) mice comparing to that in WT mice [(66.8±7.0)/×400 with 10 vision fields] on day 7 after RSV infection (P0.01). Mucus production in the airway was markedly increased in IL-12 P40 (-/-) mice than that in WT mice on day 7 after RSV infection (P0.01). The levels of Th2 cytokines IL-5 [(3.11±1.05) pg/100μg total protein] and IL-13 [(0.32±0.08) pg/100μg total protein] in the lung tissue were higher in IL-12P40 (-/-) mice than those in WT mice on day 7 after RSV infection (P0.05 for all). The level of IFN-γ in lung tissue increased in either IL-12 P40 (-/-) mice or WT mice on day 7 after RSV infection and without significant difference between them (P0.05). The levels of IL-18[(5.51±0.25) pg/100μg total protein] in the lung tissue were significantly higher in IL-12P40(-/-)mice than those in WT mice [(3.19±0.37) pg/100μg total protein] on both of day 3 and day 7 after RSV infection ((P0.01 for all). Furthermore, when IL-18 was blocked with monoclone antibody, the increased level of IL-13 and production of mucus were observed in IL-12P40(-/-) mice on day 7 after RSV infection. However, the level of IFN-γ production was not significantly affected in IL-18 blocked IL-12P40(-/-) mice. ConclusionThese findings indicate that IL-12 and/or IL-18 deficiency can increase lung inflammatory and Th2 immune responses to RSV infection, but does not affect the production of IFN-γ.
The imbalance of pulmonary Th17/Treg cells in BALB/c suckling mice infected with respiratory syncytial virus-mediated intestinal immune damage and gut microbiota changes

Abstract:ABSTRACT The immune response induced by respiratory syncytial virus (RSV) infection is closely related to changes in the composition and function of gastrointestinal microorganisms. However, the specific mechanism remains unknown and the pulmonary-intestinal axis deserves further study. In this study, the mRNA levels of ROR-γt and Foxp3 in the lung and intestine increased first and then decreased. IL-17 and IL-22 reached the maximum on the third day after infection in the lung, and on the second day after infection in the small intestine and colon, respectively. RegIIIγ in intestinal tissue reached the maximum on the third day after RSV infection. Moreover, the genus enriched in the RSV group was Aggregatibacter , and Proteus was reduced. RSV infection not only causes Th17/Treg cell imbalance in the lungs of mice but also leads to the release of excessive IL-22 from the lungs through blood circulation which binds to IL-22 receptors on the intestinal surface, inducing RegIIIγ overexpression, impaired intestinal Th17/Treg development, and altered gut microbiota composition. Our research reveals a significant link between the pulmonary and intestinal axis after RSV infection. IMPORTANCE RSV is the most common pathogen causing acute lower respiratory tract infections in infants and young children, but the complex interactions between the immune system and gut microbiota induced by RSV infection still requires further research. In this study, it was suggested that RSV infection in 7-day-old BALB/c suckling mice caused lung inflammation and disruption of Th17/Treg cells development, and altered the composition of gut microbiota through IL-22 induced overexpression of RegIIIγ, leading to intestinal immune injury and disruption of gut microbiota. This research reveals that IL-22 may be the link between the lung and gut. This study may provide a new insight into the intestinal symptoms caused by RSV and other respiratory viruses and the connection between the lung and gut axis, as well as new therapeutic ideas for the treatment of RSV-infected children.

Abstract:Respiratory syncytial virus (RSV) is the major leading cause of infantile viral bronchiolitis. However, cellular phenotypes contributing to the RSV protection and vaccine-enhanced disease remain largely unknown. Upon RSV challenge, we analyzed phenotypes and cellularity in the lung of mice that were naïve, immunized with formalin inactivated RSV (FI-RSV), or re-infected with RSV. In comparison with naïve and live RSV re-infected mice, the high levels of eosinophils, neutrophils, plasmacytoid and CD11b(+) dendritic cells, and IL-4(+) CD4(+) T cells were found to be contributing to pulmonary inflammation in FI-RSV immune mice despite lung viral clearance. Alveolar macrophages appeared to play differential roles in protection and inflammation upon RSV infection of different RSV immune mice. These results suggest that multiple innate and adaptive immune components differentially contribute to RSV disease and inflammation.
Toll-like receptor 4-mediated respiratory syncytial virus disease and lung transcriptomics in differentially susceptible inbred mouse strains

Abstract:Respiratory syncytial virus (RSV) causes severe lower respiratory tract disease in infants, young children, and susceptible adults. The pathogenesis of RSV disease is not fully understood, although toll-like receptor 4 (TLR4)-related innate immune response is known to play a role. The present study was designed to determine TLR4-mediated disease phenotypes and lung transcriptomics and to elucidate transcriptional mechanisms underlying differential RSV susceptibility in inbred strains of mice. Dominant negative Tlr4 mutant (C3H/HeJ, HeJ, Tlr4Lps-d) and its wild-type (C3H/HeOuJ, OuJ, Tlr4Lps-n) mice and five genetically diverse, differentially responsive strains bearing the wild-type Tlr4Lps-n allele were infected with RSV. Bronchoalveolar lavage, histopathology, and genome-wide transcriptomics were used to characterize the pulmonary response to RSV. RSV-induced lung neutrophilia [1 day postinfection (pi)], epithelial proliferation (1 day pi), and lymphocytic infiltration (5 days pi) were significantly lower in HeJ compared with OuJ mice. Pulmonary RSV expression was also significantly suppressed in HeJ than in OuJ. Upregulation of immune/inflammatory (Cxcl3, Saa1) and heat shock protein (Hspa1a, Hsph1) genes was characteristic of OuJ mice, while cell cycle and cell death/survival genes were modulated in HeJ mice following RSV infection. Strain-specific transcriptomics suggested virus-responsive (Oasl1, Irg1, Mx1) and epidermal differentiation complex (Krt4, Lce3a) genes may contribute to TLR4-independent defense against RSV in resistant strains including C57BL/6J. The data indicate that TLR4 contributes to pulmonary RSV pathogenesis and activation of cellular immunity, the inflammasome complex, and vascular damage underlies it. Distinct transcriptomics in differentially responsive Tlr4-wild-type strains provide new insights into the mechanism of RSV disease and potential therapeutic targets.
Respiratory virus-induced TLR7 activation controls IL-17-associated increased mucus via IL-23 regulation

Abstract:The response to respiratory syncytial virus (RSV), negative strand ssRNA virus, depends upon the ability to recognize specific pathogen-associated targets. In the current study, the role of TLR7 that recognizes ssRNA was examined. Using TLR7(-/-) mice, we found that the response to RSV infection in the lung was more pathogenic as assessed by significant increases in inflammation and mucus production. Although there appeared to be no effect of TLR7 deficiency on type I IFN, the pathology was associated with an alteration in T cell responses with increases in mucogenic cytokines IL-4, IL-13, and IL-17. Examination of dendritic cells from TLR7(-/-) animals indicated a preferential activation of IL-23 (a Th17-promoting cytokine) and a decrease in IL-12 production. Neutralization of IL-17 in the TLR7(-/-) mice resulted in a significant decrease in the mucogenic response in the lungs of the RSV-infected mice. Thus, without TLR7-mediated responses, an altered immune environment ensued with a significant effect on airway epithelial cell remodeling and goblet cell hyper/metaplasia, leading to increased mucus production.

Abstract:The specific contribution of interleukin-17/interleukin-17 receptor (IL-17/IL-17R)-mediated responses in regulating host susceptibility against obligatory intracellular Chlamydia infection was investigated in C57BL/6 and C3H/HeN mice during Chlamydia muridarum respiratory infection. We demonstrated that Chlamydia stimulated IL-17/IL-17R-associated responses in both Chlamydia-resistant C57BL/6 and Chlamydia-susceptible C3H/HeN mice. However, C3H/HeN mice developed a significantly greater IL-17/IL-17R-associated response than C57BL/6 mice did. This was reflected by an increase in IL-17 mRNA expression, a higher recall IL-17 production from splenocytes upon antigen restimulation, and higher production of Th17-related cytokines (IL-23 and IL-6) and chemokines (chemokine [C-X-C motif] ligand 2 [CXCL1]/keratinocyte-derived chemokine [KC] and CXCL2/macrophage inflammatory protein 1 [MIP2]) in C3H/HeN mice than in C57BL/6 mice. Furthermore, C3H/HeN mice displayed a massive accumulation of activated and preactivated neutrophils in the airway and lung parenchyma compared to their C57BL/6 counterparts. We further demonstrated that the skewed IL-17/Th17 profile in C3H/HeN mice was predisposed by a higher basal level of IL-17 receptor C (IL-17RC) expression and then further amplified by a higher inducible IL-17RA expression in lungs. Most importantly, in vivo delivery of IL-17RA antagonist that resulted in a 50% reduction in the neutrophilic infiltration in lungs was able to reverse the susceptible phenotype of C3H/HeN mice to respiratory Chlamydia infection. Thus, our data for the first time have demonstrated a critical role for the IL-17/IL-17R axis in regulating host susceptibility to Chlamydia infection in mice.
Respiratory Syncytial Virus Infection Differentiates Airway Dysfunction in the Central and Peripheral Airways in Ova-Sensitized Mice

Abstract:ABSTRACT Much evidence suggests that respiratory syncytial virus (RSV) infection prolongs airway hyperresponsiveness (AHR) and exacerbates asthma by enhancing airway inflammation. However, the characteristic of airway inflammation and kinetics of airway dysfunction occurred in the central and peripheral airways were not fully delineated. The objective of this study was to investigate the effect of RSV on the allergic airway inflammation in different size airways and to elucidate its possible mechanism. Using a murine model of prior ovalbumin (OVA) sensitization and subsequent RSV challenge, lung resistance (RL), and dynamic compliance (Cdyn) was conducted by barometric whole-body plethysmography. Histological examinations were carried out. Differential cells count in bronchoalveolar lavage (BAL) fluid, serum anti-OVA IgE, and IgG1 were measured. Cytokine mRNA expression in lung tissue were determined. RSV triggered a significant increase in RL and reduction in Cdyn, as well as greatly prolonged the recovery of Cdyn more than that of RL in OVA-sensitized mice. Also, RSV resulted in more severe peripheral airway inflammation which exhibit as globe cell hyperplasia and CD8+ T cell infiltration. Furthermore, the number of lymphocytes, neutrophils and macrophages in BAL fluid, serum anti-OVA IgE and IgG1 were remarkably increased. Additionally, mice increased relative expression of cytokines IL-4, IL-13, and IFN-γ, but not IL-5, IL-17, and IL-17F. These findings demonstrated that RSV could selectively affect pathologic processes that contribute to altered airway function in the central and peripheral airways in OVA-sensitized mice. These processes may be involved in goblet cell hyperplasia and CD8+ T cell infiltration in peripheral airways.

Abstract:MicroRNAs (miRNAs) are small, non-coding RNAs that regulate posttranscription by binding to 3′-untranslated regions of target mRNAs. Recent functional studies have elucidated mechanisms that miRNAs regulate leukotriene synthesis by perturbing arachidonic acid metabolism. Both microarrays and high-throughput sequencing revealed distinct differential expression of miRNAs in children with respiratory syncytial virus (RSV) infection compared with healthy controls. Abnormal miRNA expression may contribute to higher leukotriene levels, which is associated with airway hyperreactivity. Targeting miRNAs may benefit to restore the homeostasis of inflammatory reaction and provide new strategies to alleviate airway hyperreactivity induced by RSV. In this article, we provide an overview of the current knowledge about miRNAs modulating leukotrienes through regulation of arachidonic acid metabolism with a special focus on miRNAs aberrantly expressed in children with RSV infection.

Abstract:Asthma exacerbation could be induced by respiratory syncytial virus (RSV), and the underlying pathogenic mechanism is related to complement activation. Although complement might regulate CD4+T cells immune responses in asthma model, this regulation existed in RSV-induced asthma model remains incompletely characterrized. In this study, we assessed the contribution of C5a-C5aR to CD4+T cell immune responses in RSV-infected asthma mice. Female BALB/C mice were sensitized and challenged with ovalbumin (OVA) while treated with RSV infection and C5a receptor antagonist (C5aRA) during challenge period. RSV enhanced lung damage, airway hyperresponsiveness, and C5aR expressions in asthma mice, while C5aRA alleviated these pathologic changes. The percentages of Th1, Th2 and Th17 cells were increased, while the percentage of Treg cells was decreased in RSV-infected asthma mice compared with asthma mice. IFN-γ, IL-4, IL-10 and IL-17A levels have similar trend with Th1, Th2, Th17 and Treg cells. Notably, above changes of CD4+T cells and their related cytokines were reversed by C5aRA. Together, the data indicates that RSV infection could apparently increase C5a and C5aR expression in the pathogenesis of RSV-infected asthma mice, meanwhile C5aRA prevents some of the CD4+T cells immune changes that are induced by RSV.

Aggravated Airway Inflammation and Resistance in a Respiratory Syncytial Virus Infected Murine Asthma Model

Abstract:Objective To explore the role of respiratory syncytial virus(RSV)infection in aggravating airway inflammation and airway resistance in murine asthma model.Methods A total of 30 6-8-week-old male BALB/c mice were equally divided into three groups randomly:the control group,ovalbumin(OVA)group(asthma group),and respiratory syncytial virus(RSV)/OVA group(RSV+asthma group).In OVA group,murine asthma model was established using an OVA sensitization;in OSV/RSV group,mice were firstly sensitized by OVA and subsequently infected with RSV intranasally for three times to make acute viral infection asthma model;and in control group,the mice received equal volume of PBS treatment.Twenty-four hours after the last challenge,the airway resistance was evaluated by mouse ventilator(Buxco RC).Inflammatory cell infiltration was measured in bronchoalveolar lavage fluid(BALF).Pulmonary tissue samples were collected and stained with H-E,PAS or VG to observe inflammation of pulmonary tissues.Results Pulmonary tissue of mice in OVA group had inflammatory cell infiltration,airway mucus secretions were visible,and collagen was seen around the airway.Pulmonary tissue of mice in OVA/RSV group had significant inflammatory cell infiltration,alveolar stenosis,telangiectasia congestion,airway mucus secretions and collagen deposition.Lung function and the proportion of BALF eosinophils(EOS%)in OVA group and OVA/RSV group were significantly different from that of the control group(P<0.05,P<0.01).Airway resistance and the EOS%in OVA/RSV group were significantly different from those of the OVA group(P<0.05).Conclusion RSV infection can aggravate the airway inflammation and result in airway resistance in murine asthma model.
Dual Effects of Respiratory Syncytial Virus Infections on Airway Inflammation by Regulation of Th17/Treg Responses in Ovalbumin-Challenged Mice

Abstract:We investigated the effects of respiratory syncytial virus (RSV) infections on ovalbumin (OVA)-challenged mice via regulation of Th17/Treg cell responses. BALB/c mice were challenged with OVA, followed by RSV infections twice. In OVA-challenged mice, the secretion of Th2/Th17-type cytokines, airway hyperresponsiveness and inflammation were significantly inhibited by initial RSV infection. Moreover, the in vivo findings demonstrated that initial RSV infection reversed the imbalance of Th17/Treg responses. In contrast, RSV re-infection strengthened Th2/Th17-type cytokine secretion, airway hyperresponsiveness, and inflammation, especially for lymphocyte infiltration in OVA-challenged mice. Meanwhile, RSV re-infection enhanced the imbalanced Th17/Treg responses. Upon all results reveal that RSV-induced respiratory infections may lead to dual effects pertaining to allergic airway inflammation by regulation of Th17/Treg responses.

Abstract:Abstract While the anti-viral/protective role of interferon type I (IFN-I) and tumor necrosis factor (TNF)-α in naturally acquired RSV infections remains controversial, human and animal studies suggest the existence of a cross-regulation between these two innate pathways, which may contribute to disease and airway inflammation characteristics of this infection. In this study, we investigated the role of IFN-I:TNF-α axis in a mouse model of RSV infection, which provides robust experimental evidence of clinical disease, airway bronchoconstriction and viral replication, recapitulating aspects of the human infection. BALB/c mice were treated with antibodies targeting the IFN-I receptor (IFNR) and either TNF receptor (TNFR)1 or TNFR2 prior to RSV infection. Significant improvements to body-weight loss, illness score and bronchoconstriction measured by plethysmography were observed in mice treated with either antibody combination vs untreated mice (RSV/IgG control). Additionally, several cytokines were significantly reduced in both RSV-IFNR-TNFR mice including IL-1α (p=0.02), IL-6 (p=0.0002), IL-9 (p=0.003), IL-17 (p=0.01), IFN-γ (p=0.005), Ccl3 (p=0.01), Ccl4 (p=0.0003) and Ccl5 (p=0.02). While lung viral load in RSV-IFNR-TNFR1 mice remained comparable to the RSV control (p=0.30), RSV-IFNR-TNFR2 mice had a significant increase in viral replication (p=0.001). Interestingly, this increase in viral replication was not appreciated in previous works blocking IFNR or TNFR2 alone in a similar experimental RSV model. This data collectively demonstrates a synergism between IFN-I and TNF-α during RSV infection as well as highlights the importance of selective receptor blockade when considering RSV therapeutic development.

Abstract:Objectives To discuss the role of IL-13 gene polymorphism Arg110Gln in the pathogenesis of respiratory syncytial virus (RSV)infection. Methods RSV antibody was detected in 210 young children from the pars nasalis pharyngis by means of direct immunofluorescence essay. The patients were divided into two groups based on their clinical presentations: wheezing group(n = 112)and no-wheezing group(n = 98,including those without symptoms). DNA was extracted from buccal mucosa cells,then real-time PCR is employed to analyze IL-13 gene. Results Genotype variants were found. In the wheezing group,Arg110 and Gln110 accounted for 58.92% and 41.07%,while in the no-wheezing group the proportion of Arg110 and Gln110 were 44.39% and 55.61%,respectively. There were significant statistical diffe-rences between the two groups(P < 0.01). Severity of wheezing and duration of hospitalization were positively related to Arg110 of IL-13 gene polymorphism,moreover,level of IgE and eosinophil cationic protein in the children with Arg110 were higher than those with Gln110. Conclusions Arg110 of IL-13 gene polymorphism play a role in pathogenesis of infant wheezing after RSV infection.

Abstract:The type 2 immune response, induced by infection of respiratory syncytial virus (RSV), has been linked to asthma development, but it remains unclear how the response is initiated. Here, we reported that the high-mobility group box-1 (HMGB1) protein promotes the type 2 response in the later stage of RSV infection. In mice, we found that type 2 cytokines were elevated in the later stages, which were strongly diminished after administration of anti-HMGB1 antibodies. Further investigation revealed that HMGB1 expression was localized to CC10+ club cells in the lung. In the clinic, levels of HMGB1 in nasopharyngeal aspirates in hospitalized infants with RSV bronchiolitis [median (interquartile range) 161.20 ng/ml (68.06-221.30)] were significantly higher than those without lower respiratory tract infections [21.94 ng/ml (12.12-59.82); P < 0.001]. Moreover, higher levels of HMGB1 correlated with clinical severity. These results reveal a link between viral infection and the asthma-like type 2 responses that are associated with long-term consequences.
Eosinophils contribute to innate antiviral immunity and promote clearance of respiratory syncytial virus

Abstract:Eosinophils are recruited to the lungs in response to respiratory syncytial virus (RSV) infection; however, their role in promoting antiviral host defense remains unclear. Here, we demonstrate that eosinophils express TLRs that recognize viral nucleic acids, are activated and degranulate after single-stranded RNA (ssRNA) stimulation of the TLR-7-MyD88 pathway, and provide host defense against RSV that is MyD88 dependent. In contrast to wild-type mice, virus clearance from lung tissue was more rapid in hypereosinophilic (interleukin-5 transgenic) mice. Transfer of wild-type but not MyD88-deficient eosinophils to the lungs of RSV-infected wild-type mice accelerated virus clearance and inhibited the development of airways hyperreactivity. Similar responses were observed when infected recipient mice were MyD88 deficient. Eosinophils isolated from infected hypereosinophilic MyD88-sufficient but not MyD88-deficient mice expressed greater amounts of IFN regulatory factor (IRF)-7 and eosinophil-associated ribonucleases EAR-1 and EAR-2. Hypereosinophilia in the airways of infected mice also correlated with increased expression of IRF-7, IFN-beta, and NOS-2, and inhibition of NO production with the NOS-2 inhibitor L-NMA partially reversed the accelerated virus clearance promoted by eosinophils. Collectively, our results demonstrate that eosinophils can protect against RSV in vivo, as they promote virus clearance and may thus limit virus-induced lung dysfunction.
