Abstract:3,3-Disubstituted oxindoles bearing quaternary and tertiary stereogenic centers are privileged structural motifs, which widely exist in pharmaceutical and natural products. Herein, a highly regio-, enantio-, and diastereoselective allylic alkylation of 3-alkyl oxindoles through synergistic iridium and copper catalysis is described, which provides a series of 3,3-disubstituted oxindole derivatives containing adjacent quaternary and tertiary stereogenic centers in excellent yields, enantiomeric excess, and diastereomeric ratio (for 30 examples, up to 97% yield, > 99% ee, and > 20: 1 dr). This method provides exclusive branched selectivity, excellent enantio- and diastereoselectivities, and good functional compatibility. Control experiments suggested that the chiral copper catalyst is required for achieving high reactivities and diastereoselectivities under mild reaction conditions.

Abstract:A palladium-catalyzed asymmetric chloroenolate allylation with vinyl benzoxazinanones under mild reaction conditions has been developed, affording a series of optically active 3,3-disubstituted oxindoles exhibiting a chloro-group and a linear aryl amino side chain in good yields with up to 96% ee. Versatile functional group tolerance on the benzene ring has been demonstrated, and the utility of this method was probed by a scale-up synthesis and highlighted by product derivatizations.

Abstract:Oxidative rearrangement of 3-substituted indoles is a fundamental organic transformation for the synthesis of 2-oxindole scaffolds. However, this time-consuming and complex task produces a single type of product via two-electron oxidation using stoichiometric chemical oxidants. Herein, we report a unified and efficient copper-catalyzed selective single-electron transfer strategy for three oxidation reactions of 3-substituted indoles using air (O2) as the terminal oxidant, providing a streamlined and practical synthetic approach to access 3-hydroxyl-, 3-alkoxyl-, and 3-hygrogenous-2-oxindoles via a 3-radical-2-hydroxyl indoline intermediate. This copper catalysis protocol demonstrates high chemoselectivity, good functional group tolerance, and broad application potential in the modular synthesis of biologically 3-functionalized oxindole derivatives.

Abstract:A copper-catalyzed enantioselective [3 + 3] cycloaddition of 3-ethynyl-2-oxoindolin-3-yl acetates with 1H-pyrazol-5(4H)-ones for the construction of optically active spirooxindoles bearing a spiro all-carbon quaternary stereocenter has been realized. Abstract:A pentanidium-catalyzed highly enantioselective conjugate addition of 3-alkyloxindoles to phenyl vinyl sulfone has been demonstrated. This approach allows the construction of 3,3-dialkyl-substituted oxindole frameworks with high yield and excellent enantioselectivity (up to 99%) under simple phase-transfer conditions. A variety of oxindoles bearing all-carbon quaternary stereogenic centers were obtained in the presence of 0.25mol% pentanidium. Meanwhile, practicality was illustrated by a gram-scale asymmetric synthesis of two 3,3-dialkyl-substituted oxindoles. The resulting adduct can be smoothly transformed to the natural product analogue in a short synthetic route.

Abstract:The formation of vicinal all-carbon quaternary stereocenters remains a formidable challenge. We report herein the synthesis of such highly congested structural dyads by copper catalyzed decarboxylative propargylation between propargyl carbonates and indanone-based nucleophiles. The implementation of diphenylethylenediamine (DPEN)-based ligands is the key to success. A wide range of functional groups was tolerated, delivering the indanone-based spirolactones in good yields with high diastereo-and enantioselectivity. The mechanistic observations suggest the capability of the new copper complex to enable stereocontrolled addition to copper-allenylidene species.

Abstract:We report a tandem asymmetric Cu-catalyzed propargylic animation (ACPA)/Ag-catalyzed carboxylative cyclization (SCC) with CO 2 to 5-methylidene-2-oxazolidinones, even with tetrasubstituted stereocenters. By varying pyridine bisoxazoline (PYBOX) ligands, a general and highly enantioselective ACPA of unprecedentedly broad scope of secondary propargylic acetates and primary amines is achieved. Both α-aryl and α-aliphatic propargylic acetates could react with either aromatic or aliphatic primary amines to give the corresponding N -aryl or N -aliphatic ethynylamines with α-aliphatic or α-aryl groups in over 90% ee for use in the next step. The key to developing this one-pot sequence is to use a chelator triethylenetetramine (TETA) to mask the copper ion, to avoid its negative effect on Ag-catalyzed cyclization, whilst releasing PYBOX to activate the silver catalyst. With the methylidene moiety, these oxazolidinones can be readily elaborated. The value of the sequence is further shown by the catalytic enantioselective total synthesis of (−)-cytoxazone and the potent ezetimibe analogue.

Abstract:Copper-catalyzed enantioselective propargylic substitution has become an increasingly reliable strategy to construct stereogenic centers over the past two decades. Currently, various catalytic systems and reaction modes have been developed for the synthesis of different chiral propargylic frameworks. Therein, stereochemical control can be achieved either through the nucleophilic addition at the γ-site of the copper-allenylidene intermediate, or the newly proposed nucleophilic addition of copper-vinylvinylidene intermediate comprising two reactive sites (γ, ε). This review briefly summarizes the development of copper-catalyzed asymmetric propargylic substitution on a variety of different substrates, including but not limited to propargylic esters, cyclic carbonates and carbamates, and yne-allylic esters.

Abstract:The first metal/organo cooperatively catalyzed asymmetric reaction of C-alkynyl N-Boc-protected N,O-acetals with in situ generated oxonium ylides has been developed. This new type of propargylation allows for the efficient synthesis of structurally diverse unreported chiral propargylamines bearing oxa-quaternary stereocenters. The reaction features unprecedented substrate scope and high diastereo- and enantioselectivity. Theoretical studies suggest a novel cooperative catalysis model and the unique transfer of R2OH.

Abstract:Transition-metal-catalyzed asymmetric allylic alkylation is one of the most powerful and well-known strategies for the construction of C–C bonds; nevertheless, propargylation is elusive and remains far less explored. Here, we report the copper-catalyzed asymmetric propargylic substitution of N-acyl phenylglycine N-hydroxyphthalimide (NHP) esters and racemic propargylic carbonates, which afforded multifunctionalized products bearing a terminal alkyne unit in high yields with good stereoselectivities. Moreover, the product can be readily derivatized into other interesting compounds that have great potential for the exploitation of pharmaceutically relevant molecules.

Abstract:Stereoisomers of drugs can have different therapeutic outcomes or side effects, and regulatory agencies require the bioactivity of all stereoisomers of pharmaceutical candidates to be evaluated during the drug discovery and development process. However, relatively few methods can lead to all possible stereoisomers of molecules containing contiguous stereocentres. Here we report a combination of nickel and squaramide catalysis as a versatile strategy for stereodivergent α-propargylation of oxindoles. Effective with both primary and secondary propargylic ammonium salt substrates, the reported process gives a range of α-propargylated oxindole products with excellent regio- and stereocontrol. The stereochemical outcome of the reaction can be programmed through choice of the appropriate enantiomers of the nickel and squaramide catalysts. The synthetic utility of this stereodivergent methodology is demonstrated through its application to the synthesis of a number of natural products, (−)-physovenine, (−)-esermethole and (−)-physostigmine.

Abstract:Catalytic asymmetric sulfenylation of 3-substituted oxindoles has been developed through efficient catalysis by tartaric acid derived chiral iminophosphoranes. With N-(phenylthio)phthalimide as the sulfur source, a wide range of optically active 3-phenylthiooxindoles were obtained in excellent yields (90-99%) and good enantiomeric excess (up to 90% ee). Interestingly, 3-aryl and 3-methyl substituted oxindoles afforded sulfenylated products in S-configuration, whereas substituted oxindoles with 3-arylidene or 3-isobutyl substituents gave the corresponding R-configured sulfenylated products.

Abstract:We report herein a copper-catalyzed straightforward approach for the synthesis of otherwise synthetically challenging enantioenriched dihydroquinoxalinones from propargylic esters and commercially readily available o-phenylenediamines. This catalytic approach could be performed open to air on a gram-scale, with the reaction efficiency being unaffected. The dangling propargylic substituent in the products could be transformed into different functional groups with easy operation, therefore creating much potential for structural diversity.

Abstract:We report a novel strategy for the synthesis of chiral 2-oxazolidinones via a dinuclear copper-catalyzed asymmetric propargylic amination-carboxylative cyclization sequence of propargylic esters with nucleophilic alkyl amines under ambient pressure of carbon dioxide. A variety of chiral 2-oxazolidinones featuring an exocyclic methylene moiety was obtained in good yields with high enantioselectivities via a one-pot operation.

Abstract:A copper-catalyzed intramolecular asymmetric double C-arylation reaction was developed. The method provides a facile approach to chiral spiro bis-oxindoles in high yields and with good to excellent enantioselectivities. It also shows a broad substrate scope and good functional group tolerance. Density functional theory (DFT) calculations were conducted and revealed that the enantioselectivity is determined at the oxidative addition of Cu(I) into the second C-I bond.

Abstract:Chiral trisubstituted vicinal diols are a type of important organic compounds, serving as both common structure units in bioactive natural products and chiral auxiliaries in asymmetric synthesis. Herein, by using siloxypropadienes as the precursors of allyl copper(I) species, a copper(I)-catalyzed diastereoselective and enantioselective reductive allylation of ketones was achieved, providing both syn-diols and anti-diols in good to excellent enantioselectivity. DFT calculations show that cis-γ-siloxy-allyl copper species are generated favorably with either 1-TBSO-propadiene or 1-TIPSO-propadiene. Moreover, the steric difference of TBS group and TIPS group distinguishes the face selectivity of acetophenone, leading to syn-selectivity for 1-TBSO-propadiene and anti-selectivity for 1-TIPSO-propadiene. Easy transformations of the products were performed, demonstrating the synthetic utility of the present method. Moreover, one chiral diol prepared in the above transformations was used as a suitable organocatalyst for the catalytic asymmetric reductive self-coupling of aldimines generated in situ with B2(neo)2.
