Abstract:The improvement of human living conditions has led to an increase in average life expectancy, creating a new social and medical problem—aging, which diminishes the overall quality of human life. The aging process of the body begins with the activation of effector signaling pathways of aging in cells, resulting in the loss of their normal functions and deleterious effects on the microenvironment. This, in turn, leads to chronic inflammation and similar transformations in neighboring cells. The cumulative retention of these senescent cells over a prolonged period results in the deterioration of tissues and organs, ultimately leading to a reduced quality of life and an elevated risk of mortality. Among the most promising methods for addressing aging and age-related illnesses are pharmacological, genetic, and cellular therapies. Elevating the activity of aging-suppressing genes, employing specific groups of native and genetically modified cells, and utilizing senolytic medications may offer the potential to delay aging and age-related ailments over the long term. This review explores strategies and advancements in the field of anti-aging therapies currently under investigation, with a particular emphasis on gene therapy involving adeno-associated vectors and cell-based therapeutic approaches.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is the impact of aging and its related diseases on human health and quality of life. With the improvement of human living conditions, the average lifespan has increased, but the aging problem has ensued, which not only reduces the overall quality of life but also increases the risk of chronic inflammation and other age - related diseases. The paper explores the latest strategies and progress in gene, cell, and drug therapies in the field of anti - aging, especially the use of recombinant adeno - associated virus (AAV) in gene therapy and cell - based interventions. Specifically, the paper focuses on the following aspects: 1. **Molecular and Cellular Mechanisms of Aging**: The paper elaborates on how aging starts at the cellular level, leading to the loss of cell function by activating effector signaling pathways and having an adverse impact on the microenvironment, and ultimately resulting in chronic inflammation and similar changes in other cells. The accumulation of these changes can lead to the degeneration of tissues and organs, thereby reducing the quality of life and increasing the risk of death. 2. **Anti - aging Strategies**: The paper discusses the most promising methods in current research, including: - **Gene Therapy**: Delay the aging process by enhancing the activity of anti - aging genes. - **Cell Therapy**: Use specific native or genetically modified cells to repair or replace damaged cells. - **Drug Therapy**: Use drugs that eliminate senescent cells (such as senolytic drugs) to reduce the negative impact of these cells on surrounding healthy cells. 3. **Specific Mechanisms and Compounds**: The paper lists some specific drugs and compounds, such as dasatinib, quercetin, rapamycin, etc. These drugs induce apoptosis of senescent cells through different molecular targets and signaling pathways, thereby reducing their negative impact on the body. 4. **Future Research Directions**: The paper also mentions the development of second - generation senolytics, including drugs based on lysosome and SA - β - gal activation, nanoparticles, Na+/K+ - ATPase - dependent apoptosis activation, SASP inhibitors, antibody - drug conjugates, vaccination, and CAR T - cell - based cell therapy. In conclusion, this review aims to comprehensively explore and evaluate various strategies and methods in the current anti - aging field, in the hope of finding effective ways to delay aging and prevent age - related diseases.
