Abstract:The selective C‐F bonds functionalization in available trifluoromethyl compounds is an economic route for obtaining high‐value organic fluorine compounds. However, due to the high C‐F bonds dissociation energy in trifluoromethyl group, the strength of the bond decreases continuously during defluorination, posing a challenge in modifying such molecules without over‐conversion. Over the past few years, there has been significant progress in developing efficient methods for constructing difluoromethylene unit, using approaches such as radical chemistry, photochemistry, electrochemistry, and organometallic chemistry. These methods involve difluorocarbon radicals, difluorocarbon cations, difluorocarbon anions, and trifluoromethyl carbenes as intermediates. In this review, we summarized the recent five years of research achievements in this field for selective single C‐F bond cleavage in trifluoromethyl aromatic and carbonyl compounds to gain diverse difluoromethylene‐containing organic molecules via different intermediate strategies.

Abstract:Selective functionalization of C–F bonds in trifluoromethyl groups has recently received a growing interest, as it offers atom- and step-economic pathways to access highly valuable mono- and difluoroalkyl-substituted organic molecules using simple and inexpensive trifluoromethyl sources as the starting materials. In this regard, impressive progress has been made on the defluorinative functionalization reactions that proceed via radical intermediates. Nevertheless, it is still a great challenge to precisely control the defluorination process, due to the continuous decrease of the C–F bond strength after the replacement of one or two fluorine atoms with various functionalities. This review article is aimed to provide a brief overview of recently reported methods used to functionalize C–F bonds of CF3 groups via radical intermediates. Abstract:Sequentially snipping off fluorines It is often useful in pharmaceutical or agrochemical research to modify the properties of carbon compounds by appending one or more fluorine atoms. However, the methods used to prepare mono-, di-, or trifluorocarbon centers tend to differ from each other in inconvenient ways. Yu et al. developed a radical reaction that can successively remove one or two fluorine atoms from trifluoromethyl groups adjacent to amides or esters. Abstract:In the past 5 years, transition-metal-mediated trifluoromethylation for the construction of various CF3-containing building blocks has been the focus of recent research in both industrial and academic communities. Progresses in the construction of C(sp(2))-CF3 bonds and C(sp)-CF3 have been well reviewed. Abstract:Trifluoroethyl group served as the bioisosteres of ethyl or ethoxy group widely exists in the fields of pharmaceuticals and agrochemicals. Developing novel strategies of introducing trifluoroethyl group into various skeletons has become a hot topic in organic synthetic chemistry. In this review, the trifluoroethylation reaction of various partners such as alkynes, alkenes, functionalized arenes, amines and so on were summarized. This review was categorized into four parts according to the bond formation: C(sp)-CH2CF3, C(sp2)-CH2CF3, C(sp3)-CH2CF3, and X (X = N, O, S, Se)-CH2CF3. Some representative examples along with reaction mechanisms were also discussed.

Abstract:The incorporation of fluorine-containing moieties into organic compounds is of great importance in pharmaceutical, agricultural, and materials science. Within these organofluorides, the trifluoromethyl group is one of the most important motifs. In recent years, the trifluoromethyl group has attracted more and more attention, and many trifluoromethylated compounds have been found to possess special activities. However, until now, only a few methods have been developed to achieve this efficiently using Umemoto's reagents. This review highlights recent developments in the direct introduction of a trifluoromethyl group into organic compounds with Umemoto's reagents. Seven approaches to the trifluoromethylation of organic compounds are summarized: (i) trifluoromethylation of arenes, (ii) trifluoromethylation of alkenes, (iii) trifluoromethylation of terminal alkynes, (iv) deoxygenative trifluoromethylation of benzylic xanthates, (v) trifluoromethylation of ketoesters, (vi) trifluoromethylation of aryl boronic acids and aromatic amines (synthesis of ArCF3) and (vii) trifluoromethylation of biphenyl isocyanide derivatives.

Abstract:Comprehensive Summary Trifluoromethyl group is one of the most frequently available and promising functional groups in pharmaceutical, agrochemical as well as material molecules. Developing synthetic methods for the synthesis of trifluoromethyl functionalized compounds by trifluoromethylation reactions occupies significant position in modern organic chemistry. On other hand, as a typical tool of sustainable synthesis, transition metal‐free synthesis possesses unique advantages in atom economy, easy product purification as well as excellent eco‐tolerance. Consequently, transition metal‐free trifluoromethylation reactions constitute the most important approaches for accessing trifluoromethylated molecules. Herein, the advances in the transition metal‐free C‐H trifluoromethylation reactions are reviewed for the first time. According to the updated state‐of‐art, both the catalytic reactions by conventional heating chemistry and the more sustainable photocatalysis are included, covering the trifluoromethylation reactions of C(sp3)‐H bond, aromatic C(sp2)‐H bond as well as the non‐aromatic C(sp2)‐H bond in different substrates over the last five years (2017‐2021). Abstract:This study explores the effects of the −CF 3 group on non-covalent interactions through a comprehensive rotational investigation of the 2-(trifluoromethyl)acrylic acid–water complex. Employing Fourier transform microwave spectroscopy complemented by quantum chemical calculations, two isomers, i.e., s-cis and s-trans structures, have been observed in the pulsed jet. Based on relative intensity measurements, the s-cis to the s-trans population ratio was experimentally estimated to be ∼ 1:1.2. Subsequently, a comparison of the non-covalent interactions was carried out between the three similar complexes, acrylic acid–water, methacrylic acid–water, and 2-(trifluoromethyl)acrylic acid–water, offering quantitative insights into fluorination affecting the strength of the formed hydrogen bonds important, e.g., in molecular recognition.

Abstract:C‐F Insertion reactions, where an organic fragment formally inserts into a carbon‐fluorine bond in a substrate, are highly attractive, yet largely unexplored, methods to prepare valuable fluorinated molecules. The inherent strength of C‐F bonds and the resulting need for a large thermodynamic driving force to initiate C‐F cleavage often leads to sequestering of the released fluoride in an unreactive by‐product. Recently, however, several groups have succeeded in overcoming this challenge, opening up the study of C‐F insertion as an efficient and highly atom‐economical approach to prepare fluorinated compounds. In this article, the recent breakthroughs are discussed focusing on the key conceptual advances that allowed for both C‐F bond cleavage and subsequent incorporation of the released fluoride into the product.

Abstract:Abstract The unique properties of fluorine-containing organic compounds make fluorine substitution attractive for the development of pharmaceuticals and various specialty materials, which have inspired the evolution of diverse C-F bond activation techniques. Although many advances have been made in functionalizations of activated C-F bonds utilizing transition metal complexes, there are fewer approaches available for nonactivated C-F bonds due to the difficulty in oxidative addition of transition metals to the inert C-F bonds. In this regard, using Lewis acid to abstract the fluoride and light/radical initiator to generate the radical intermediate have emerged as powerful tools for activating those inert C-F bonds. Meanwhile, these transition-metal-free processes are greener, economical, and for the pharmaceutical industry, without heavy metal residues. This review provides an overview of recent C-F bond activations and functionalizations under transition-metal-free conditions. The key mechanisms involved are demonstrated and discussed in detail. Finally, a brief discussion on the existing limitations of this field and our perspective are presented.

Abstract:Herein, we describe a novel and efficient photo-redox catalytic difluorinated ester radical addition/defluoroalkylation coupling reaction between trifluoroacetic acid derivatives and α-trifluoromethyl alkenes. A series of polyfluorinated compounds were synthesized by the selective cleavage of the single C–F bond in the trifluoromethyl (CF3) group enabled by spin-center shift (SCS). The reaction is characterized by a wide range of substrates and good functional group tolerance.

Abstract:Organofluorine compounds have greatly benefited the pharmaceutical, agrochemical, and materials sectors. However, they are plagued by concerns associated with PFAS. Additionally, the widespread use of the trifluoromethyl group is facing imminent regulatory scrutiny. Defluorinative functionalization, which converts the trifluoromethyl to the difluoromethyl motifs, represents the most efficient synthetic strategy. However, general methods for robust C(sp3)–F bond transformations remain elusive due to challenges in selectivity and functional group tolerance. Here, we present a method for C(sp3)–F bond defluorinative functionalization of the trifluoromethyl group via difluoromethyl anion in flow. This new approach tames the reactive difluoromethyl anion, enabling diverse functional group transformations. Our methodology offers a versatile platform for drug and agrochemical discovery, overcoming the limitations associated with fluorinated motifs.
