Abstract:Probiotics, living microorganisms with demonstrated health benefits when administered in sufficient quantities, have a rich history as dietary supplements to benefit human health. Recently, understanding of their mechanisms in the gastrointestinal tract has prompted exploration of probiotics in treating human diseases. However, the effective and precise delivery of probiotics remains a significant challenge in therapeutic applications. Here, we review the mechanisms of action of probiotics in human health and the most advanced strategies for efficient probiotic delivery. We also discuss the potential applications of engineered probiotics in disease treatment. This review contributes insights into the evolving landscape of probiotic research for therapeutic applications.

What problem does this paper attempt to address?
The main problems that this paper attempts to solve are: 1. **Understanding the mechanism of action of probiotics**: Although probiotics have shown great potential in promoting human health, their mechanism of action has not been fully elucidated. The paper aims to deeply explore the mechanism of action of probiotics in human health, especially how they play a role by producing bioactive compounds, regulating the gut microbiota, and modulating the immune and nervous systems. 2. **Improving the effective delivery of probiotics**: One of the main challenges in the application of probiotics in disease treatment is to effectively and precisely deliver them to the target site. The paper reviews the current state - of - the - art probiotic delivery strategies, including chemical engineering and genetic engineering methods, as well as the application of membrane vesicles, in order to improve the delivery efficiency and targeting ability of probiotics. 3. **Exploring the applications of engineered probiotics**: The paper also discusses the potential applications of engineered probiotics in disease treatment, especially the application prospects in fields such as inflammatory bowel disease (IBD), cancer, and neurodegenerative diseases. Through methods such as gene and metabolic engineering, surface modification, and encapsulation technology, engineered probiotics can better meet the complex challenges in disease treatment. 4. **Evaluating the feasibility of clinical applications**: The paper also outlines the applications of functional probiotics in disease treatment, and discusses the results of probiotic interventions in pre - clinical experiments, as well as the prospects for future clinical applications. Through these studies, the paper provides important insights and guidance for the practical application of probiotics in treatment. In summary, this paper aims to comprehensively review the potential of probiotics in disease treatment, from basic mechanisms to delivery strategies, and then to clinical applications, providing theoretical support and technical guidance for future research and treatment.
