Abstract:A variety of biologically active molecules, pharmaceuticals, and natural products consist of a nitrogen-containing heterocyclic backbone. The majority of them are isoquinolones, indoles, isoquinolines, etc.; thereby the synthesis and derivatization of such heterocycles are synthetically very relevant. Also, certain naphthol derivatives have high synthetic utility as agrochemicals and in dye industries. Previous approaches have utilized ruthenium, rhodium, or iridium which may not be desirable due to the high toxicity, low abundance, and high cost of such 4d and 5d metals. Moreover, the need for an external oxidant during the reaction also adds by-products to the system. A high-valent cobalt-catalyzed redox-neutral C-H functionalization strategy has emerged to be a far better alternative in this regard. The use of the non-noble metal cobalt allows for selectivity and specificity in product formation. Also, the redox-neutral concept avoids the use of an external oxidant either due to the presence of a metal in a non-variable oxidation state throughout the catalytic cycle or due to the presence of an oxidizing directing group or an oxidizing coupling partner. Such an oxidizing directing group not only directs the catalyst to a specific reaction site by chelation but also regenerates the catalyst at the end of the cycle. Certain bonds such as N-O, N-N, N-Cl, N-S, and C-S are the main game-players behind the oxidizing property of such directing groups. In the other case, the directing group only chelates the catalyst to a reaction center, whereas the oxidation is carried out by the upcoming group/coupling partner. Overall, merging the redox-neutral concept with the high-valent cobalt catalysis is paving the way forward toward a sustainable and environmentally friendly approach. This review critically describes the mechanistic understanding, scope, limitations, and synthesis of various biologically relevant heterocycles via the redox-neutral concept in the high-valent Cp*Co(III)-catalyzed C-H functionalization chemistry domain.

Abstract:An N-heterocyclic carbene (NHC) covalently linked to a quinone introduces a novel avenue for internal oxidations within oxidative NHC catalysis. The deployment of this hybrid NHC class promotes intramolecular electronic flow in the oxidation of the Breslow intermediate to acyl azolium. The use of the redox active NHC as a catalyst is facilitated by employing aerobic regeneration, yielding carboxylic esters with efficiencies of ≤99%, while generating water as the sole byproduct.

Abstract:Herein, metal-free oxidative tandem cross-dehydrogenative coupling of 1,3-dicarbonyl compounds with N,N-dimethylanilines is developed. The reaction affords two new C(sp(3))-C(sp(3)) bonds in one pot. A broad range of ss-dicarbonyl compounds and N,N-dimethylanilines were compatible, and the desired products were synthesized in moderate-to-good yields.

Abstract:AbstractA general method for the oxidative substitution of nido‐carborane (7,8‐C2B9H12−) with N‐heterocycles has been developed by using 2,3‐dichloro‐5,6‐dicyanobenzoquinone (DDQ) as an oxidant. This metal‐free B−N coupling strategy, in both inter‐ and intramolecular fashions, gave rise to a wide array of charge‐compensated, boron‐substituted nido‐carboranes in high yields (up to 97 %) with excellent functional‐group tolerance under mild reaction conditions. The reaction mechanism was investigated by density‐functional theory (DFT) calculations. A successive single‐electron transfer (SET), B−H hydrogen‐atom transfer (HAT), and nucleophilic attack pathway is proposed. This method provides a new approach to nitrogen‐containing carboranes with potential applications in medicine and materials.

Abstract:N-Heterocyclic carbene (NHC) catalysis is a by now consolidated organocatalytic platform for a number of synthetic (asymmetric) transformations via diverse reaction modes/intermediates. In addition to the typical umpolung processes involving acyl anion/homoenolate equivalent species, implementation of protocols under oxidative conditions greatly expands the possibilities of this methodology. Oxidative NHC-catalysis allows for oxidative and oxygenative transformations through specific manipulations of Breslow-type species depending upon the oxidant used (external oxidant or O2 /air), the derived NHC-bound intermediates paving the way to non-umpolung processes through activation of carbon atoms and heteroatoms. This review is intended to update the state of the art in oxidative NHC-catalyzed reactions that appeared in the literature from 2014 to present, with a strong focus to crucial intermediates and their mechanistic implications.

Abstract:Herein we report a redox cyclization of amides and sulfonamides with nitrous oxide (N2O) for the direct synthesis of heterocycles. Various amides and sulfonamides could undergo directed ortho metalation (DoM) by treatment with BuLi, and the lithium intermediate could be trapped by N2O gas to achieve redox cyclization. N2O serves as a N-atom donor to mediate the intramolecular coupling of lithium species toward heterocycle formation with free external oxidant. This protocol offers a direct synthesis of heterocycles with features of readily available starting materials, simple operation, and a broad substrate scope.

Abstract:We report herein a direct unsymmetric coupling and controllable aromatization reaction of saturated N-heterocycles enabled by synergistic photoredox and acid catalysis. The reaction furnishes C2-C3 connected biheterocycles in a highly chemo- and regioselective manner under rather mild conditions. Mechanistic studies indicated that the reaction proceeded via enamine-iminium coupling leading to exclusively C2-C3 connection.

Abstract:Rh(III)-catalyzed C-H activation assisted by an oxidizing directing group has evolved to a mild and redox-economic strategy for the construction of heterocycles. Despite the success, these coupling systems are currently limited to cleavage of an oxidizing N-O or N-N bond. Cleavage of an oxidizing C-N bond, which allows for complementary carbocycle synthesis, is unprecedented. In this article, α-ammonium acetophenones with an oxidizing C-N bond have been designed as substrates for Rh(III)-catalyzed C-H activation under redox-neutral conditions. The coupling with α-diazo esters afforded benzocyclopentanones, and the coupling with unactivated alkenes such as styrenes and aliphatic olefins gave ortho-olefinated acetophenoes. In both systems the reactions proceeded with a broad scope, high efficiency, and functional group tolerance. Moreover, efficient one-pot coupling of diazo esters has been realized starting from α-bromoacetophenones and triethylamine. The reaction mechanism for the coupling with diazo esters has been studied by a combination of experimental and theoretical methods. In particular, three distinct mechanistic pathways have been scrutinized by DFT studies, which revealed that the C-H activation occurs via a C-bound enolate-assisted concerted metalation-deprotonation mechanism and is rate-limiting. In subsequent C-C formation steps, the lowest energy pathway involves two rhodium carbene species as key intermediates.

Abstract:Dimethyl sulfoxide (DMSO) promoted catalyst-free oxidative C-N coupling as well as C-O coupling under oxidant-free conditions has been outlined. This protocol is operationally simple and leads to various functionalized substituted imidazoles or oxazoles in good yields. Till date a very limited number of oxidation protocols were established where DMSO solely act as catalyst or oxidant or both. In this report DMSO is not only used as a C-N/C-O coupling agent but also used as the oxidant required for these oxidative transformations. Hence, our demonstrated DMSO promoted catalyst-free coupling transformation has the ability to lead a new dimension in the field of oxidative couplings .

Abstract:The astounding reactivity of tert -butoxides in transition metal-free coupling reactions is driving the scientific community towards a new era of environmental friendly, as well as cost-effective, transformation strategies. Transition metal-catalyzed coupling reactions generate hazardous wastes and require harsh reaction conditions, mostly at elevated temperature, which increases not only costs but also environmental concerns regarding the methodology. Tert -butoxide-catalyzed/mediated coupling reactions have several advantages and potential applications. They can form carbon–carbon, carbon–heteroatom, and heteroatom–heteroatom bonds under mild reaction conditions. Mechanistic insights into these reactions include both ionic and radical pathways, with the fate of the intermediates depending on the reaction conditions and/or additives used in the reactions. Among all of the known tert -butoxides, potassium tert -butoxide has pronounced applications in transition metal-free coupling reactions as compared to other tert -butoxides, such as sodium and lithium tert -butoxides, because of the higher electropositivity of potassium compared to sodium and lithium. Moreover, potassium tert -butoxide can act as a source of base, nucleophile and single electron donors in various important transformations. In this review, we provide an extensive overview and complete compilation of transition metal-free cross-coupling reactions catalyzed/promoted by tert- butoxides during the past 10 years.

Abstract:A rhodium(III)-catalyzed Satoh-Miura type oxidative annulation of N-aryl 2-pyridone derivatives is described using internal alkyne as a coupling partner. A weakly coordinating carbonyl group of the 2-pyridone ring is utilized for this transformation. The reaction proceeds with a broad scope and wide functional group tolerance. The solvent plays an important role in the developed method to furnish a different class of annulated product. A preliminary investigation was carried out to explore the photophysical properties of the obtained polyarylated N-naphthyl 2-pyridones.

Abstract:The cis-MoO2(BHAN)2 (BHAN= β-hydroxy-α-naphthaldehyde) has shown promising potential as a biomimetic catalyst for VHPO (vanadium haloperoxidase) activity. In particular, it has demonstrated effective catalytic activity in the bromination of organic substrates such as phenol red producing the corresponding brominated product, bromo phenol blue. This reaction closely mimics bromoperoxidase activity, highlighting the potential of the synthesized complex as a bio-inspired catalyst. Overall, these findings open exciting avenues for development of novel Mo-based VHPO biomimetic catalysts with potential applications in diverse fields like organic synthesis, catalysis and pharmaceuticals.

Abstract:In this work, we have established a photoredox/Ti dual‐catalyzed dehydroxylation of cyclobutanone oximes. The resulting γ‐cyanoalkyl radicals undergo a cascade radical addition/cyclization with N‐acrylamides to afford cyanoalkyl substituted oxoindolines under mild conditions. This reaction also exhibits broad substrate scope and good functional group tolerance. Furthermore, it is applicable to late‐stage functionalization of complex molecules and gram‐scale synthesis, which illustrates its potential value in organic synthesis.

Abstract:Highly selective divergent oxidative dearomatization coupling reactions, in which the chemoselectivity is controlled by catalysts and bases, are reported herein. Three different kinds of polycyclic cyclohexadienones are produced from the same reactants (41 examples, 85-99% yield). Our method marks a novel copper- and palladium-catalyzed C-H oxidative dearomatization of phenolic derivatives.
