Abstract:Human spaceflight has historically been managed by government agencies, such as the NASA Twins Study 1 , but new commercial spaceflight opportunities have opened spaceflight to a broader population. In 2021, the SpaceX Inspiration4 mission launched the first-ever all civilian crew to low Earth orbit, which included the youngest American astronaut (age 29), novel in-flight experimental technologies (handheld ultrasound imaging, smartwatch wearables, and immune profiling), ocular alignment measurements, and new protocols for in-depth, multi-omic molecular and cellular profiling. Here we report the primary findings from the 3-day spaceflight mission, which induced a broad range of physiological and stress responses, neurovestibular changes indexed by ocular misalignment, and altered neurocognitive functioning, some of which match long-term spaceflight 2 , but almost all of which did not differ from baseline (pre-flight) after return to Earth. Overall, these preliminary civilian spaceflight data suggest that short-duration missions do not pose a significant health risk, and moreover present a rich opportunity to measure the earliest phases of adaptation to spaceflight in the human body at anatomical, cellular, physiologic, and cognitive levels. Finally, these methods and results lay the foundation for an open, rapidly expanding biomedical database for astronauts 3 , which can inform countermeasure development for both private and government-sponsored space missions.

Abstract:As of 2020, fewer than 600 individuals have left Earth to experience work and life in space. That number will grow as government-funded and commercial space programs move forward in countries around the globe. There are however major questions about how humans respond to spaceflight at every level, from the whole body to individual organs to specific cells to molecular pathways. Preparing for a future where longer-duration spaceflights are anticipated and people can begin to contemplate space tourism, researchers are studying astronauts to understand how the human system is affected by and adapts to space. Lara Szewczak got a window on this world, speaking with retired astronaut Scott Kelly about his late-blooming interest in science, what he's learned through the NASA Twins Study, and why space vacations might not be for everyone. They were joined by Chris Mason, a lead investigator looking at the 'omics of spaceflight. Abstract:Our knowledge of the Sun, its atmosphere, long term activity and space weather potential is severely limited by the lack of good observations of the polar and far-side regions. Observations from a polar vantage point would revolutionize our understanding of the mechanism of solar activity cycles, polar magnetic field reversals, the internal structure and dynamics of the Sun and its atmosphere. Only with extended (many day) observations of the polar regions can the polar flows be determined down to the tachocline where the dynamo is thought to originate. Rapid short period polar orbits, using in situ and remote sensing instrumentation, distributed over a small number of spacecraft, will provide continuous 360o coverage of the solar surface and atmosphere in both longitude and latitude for years on end. This unprecedented full coverage will enable breakthrough studies of the physical connection between the solar interior, the solar atmosphere, the solar wind, solar energetic particles and the inner heliosphere at large. A potential implementation, the Solar Polar Diamond Explorer (SPDEx) built upon the Solar Polar Imager mission design, involves up to four small spacecraft in a 0.48-AU orbit with an inclination of 75o. The orbit is achieved using solar sails or ion engines, both technologies already demonstrated in space.

Abstract:Low Earth Orbit (LEO) has emerged as a unique environment for evaluating altered stem cell properties in microgravity. LEO has become increasingly accessible for research and development due to progress in private spaceflight. Axiom Mission 2 (Ax-2) was launched as the second all-private astronaut mission to the International Space Station (ISS). Frozen human induced pluripotent stem cells (hiPSCs) expressing green fluorescent protein (GFP) under the SOX2 promoter, as well as fibroblasts differentiated from SOX2-GFP hiPSCs, were sent to the ISS. Astronauts then thawed and seeded both cell types into commercially available 96-well plates, which provided surface tension that reduced fluid movement out of individual wells and showed that hiPSCs or hiPSC-derived fibroblasts could survive either in suspension or attached to a Matrigel substrate. Furthermore, both cell types could be transfected with red fluorescent protein (RFP)-expressing plasmid. We demonstrate that hiPSCs and hiPSC-fibroblasts can be thawed in microgravity in off-the-shelf, commercially-available cell culture hardware, can associate into 3D spheroids or grow adherently in Matrigel, and can be transfected with DNA. This lays the groundwork for future biomanufacturing experiments in space.

Psychological and Behavioral Changes during Confinement in a 520-Day Simulated Interplanetary Mission to Mars

Abstract:Behavioral health risks are among the most serious and difficult to mitigate risks of confinement in space craft during long-duration space exploration missions. We report on behavioral and psychological reactions of a multinational crew of 6 healthy males confined in a 550 m(3) chamber for 520 days during the first Earth-based, high-fidelity simulated mission to Mars. Rest-activity of crewmembers was objectively measured throughout the mission with wrist-worn actigraphs. Once weekly throughout the mission crewmembers completed the Beck Depression Inventory-II (BDI-II), Profile of Moods State short form (POMS), conflict questionnaire, the Psychomotor Vigilance Test (PVT-B), and series of visual analogue scales on stress and fatigue. We observed substantial inter-individual differences in the behavioral responses of crewmembers to the prolonged mission confinement and isolation. The crewmember with the highest average POMS total mood disturbance score throughout the mission also reported symptoms of depression in 93% of mission weeks, which reached mild-to-moderate levels in >10% of mission weeks. Conflicts with mission control were reported five times more often than conflicts among crewmembers. Two crewmembers who had the highest ratings of stress and physical exhaustion accounted for 85% of the perceived conflicts. One of them developed a persistent sleep onset insomnia with ratings of poor sleep quality, which resulted in chronic partial sleep deprivation, elevated ratings of daytime tiredness, and frequent deficits in behavioral alertness. Sleep-wake timing was altered in two other crewmembers, beginning in the first few months of the mission and persisting throughout. Two crewmembers showed neither behavioral disturbances nor reports of psychological distress during the 17-month period of mission confinement. These results highlight the importance of identifying behavioral, psychological, and biological markers of characteristics that predispose prospective crewmembers to both effective and ineffective behavioral reactions during the confinement of prolonged spaceflight, to inform crew selection, training, and individualized countermeasures.

Abstract:A mission to view the solar poles from high helio-latitudes (above 60$^\circ$) will build on the experience of Solar Orbiter as well as a long heritage of successful solar missions and instrumentation (e.g. SOHO \cite{SOHO}, STEREO \cite{stereo}, Hinode \cite{Hinode}, SDO \cite{SDO}), but will focus for the first time on the solar poles, enabling scientific investigations that cannot be done by any other mission. One of the major mysteries of the Sun is the solar cycle. The activity cycle of the Sun drives the structure and behaviour of the heliosphere and is, of course, the driver of space weather. In addition, solar activity and variability provides fluctuating input into the Earth climate models, and these same physical processes are applicable to stellar systems hosting exoplanets. One of the main obstructions to understanding the solar cycle, and hence all solar activity, is our current lack of understanding of the polar regions. In this White Paper, submitted to the European Space Agency in response to the Voyage 2050 call, we describe a mission concept that aims to address this fundamental issue. In parallel, we recognise that viewing the Sun from above the polar regions enables further scientific advantages, beyond those related to the solar cycle, such as unique and powerful studies of coronal mass ejection processes, from a global perspective, and studies of coronal structure and activity in polar regions. Not only will these provide important scientific advances for fundamental stellar physics research, they will feed into our understanding of impacts on the Earth and other planets' space environment.

Abstract:Seventy-five billion low-mass stars in our galaxy host at least one small planet in their habitable zone (HZ). The stellar ultraviolet (UV) radiation received by the planets is strong and highly variable, and has consequences for atmospheric loss, composition, and habitability. SPARCS is a NASA-funded mission to characterize the quiescent and flare UV emission from low-mass stars, by observing 10 to 20 low-mass stars, over timescales of days, simultaneously in two UV bands: 153-171 nm and 260-300 nm. SPARCS Sun-synchronous terminator orbit allows for long periods of uninterrupted observations, reaching 10s of days for some targets. The payload consists of a 10 cm-class telescope, a dichroic element, UV detectors and associated electronics, a thermal control system, and an on-board processor. The payload is hosted on a Blue Canyon Technologies 6U CubeSat. SPARCS hosts several technology innovations that have broad applicability to other missions. The payload demonstrates the use of "2D-doped" (i.e., delta- and superlattice-doped) detectors and detector-integrated metal dielectric filters in space. This detector technology provides ~5x larger quantum efficiency than NASA's GALEX detectors. In addition, SPARCS' payload processor provides dynamic exposure control, automatically adjusting the exposure time to avoid flare saturation and to time-resolve the strongest stellar flares. A simple passive cooling system maintains the detector temperature under 238K to minimize dark current. The spacecraft bus provides pointing jitter smaller than 6", minimizing the impact of flat-field errors, dark current, and read-noise. All these elements enable competitive astrophysics science within a CubeSat platform. SPARCS is currently in the final design and fabrication phase (Phase C in the NASA context). It will be launched in 2024, for a primary science mission of one year.

Abstract:This paper presents an overview of SpaceX which is a private limited United States aerospace maker/producer and space transportation service provider company which was founded by Elon Reeve Musk in 2002. The main goal of SpaceX is to reduce the cost of space transportation and in the end enable the colonization of fourth planet in our solar system that is Mars. Since its inception, SpaceX has achieved many significant milestones in space exploration. One of the most notable achievements of SpaceX was the development of reusable rockets and spacecraft, which has greatly reduced the cost of space transportation. SpaceX has also successfully launched and landed multiple rockets, including the Falcon 9 and Falcon Heavy. These rockets have been used to launch satellites into orbit, resupply the International Space Station, and deliver payloads to deep space. In addition, SpaceX has also developed the Crew Dragon spacecraft, which has successfully transported astronauts to and from the ISS that is International Space Station. Looking towards the future, SpaceX has many ambitious plans for space exploration. The company plans to launch the Starship spacecraft, which is designed for deep space missions and has the potential to transport humans to Mars. Abstract:The China Manned Space Agency (CMSA) has recently unveiled its first lunar spacesuit specifically designed for crewed moon landings, the first of which is planned for 2030. Like all extravehicular activity suits, lunar spacesuits must provide protection from the vacuum of space, extreme temperatures and radiation. However, lunar missions present additional challenges, such as preventing the infiltration of abrasive lunar dust, which can damage both the suit and spacecraft systems over time and pose health risks to astronauts. CMSA's lunar suits are engineered to be lightweight and flexible — enabling astronauts to perform complex movements — and incorporate dust-resistant fabrics that also provide protection against extreme temperatures and radiation. Key innovations include a panoramic, anti-glare visor, an integrated control console and dual-focus cameras embedded in the helmet. In addition to its technical aspects, the design also pays homage to traditional cultural elements; for example, the red stripes on the top part of the suit are said to be inspired by ribbons from the "flying apsaras", a figure in Buddhist mythology. In artistic representations, long ribbons often trail behind apsaras as they fly, symbolizing their fluidity in movement. The CMSA is currently seeking public input to name the new spacesuits. The final name will be unveiled at an unspecified later date.

Abstract:Perovskite solar cells (PSCs), have exhibited potential value for revolutionizing photovoltaic technology for space applications, and they have passed a series of tests in a simulated space environment. Nevertheless, the simulated space environment on land is definitely distinguished from the real‐space conditions, and the results cannot completely represent the real operating state of PSCs in space. Herein, PSCs mounted on a high‐altitude balloon are launched into near‐space and the current–voltage characteristics are measured in situ throughout the 19‐h flight, during which the PSCs are exposed directly to near‐space and experience wide temperature variation, vacuum, and strong irradiation. Thus, the diurnal performance evolution of PSCs is obtained for the first time and a possible variation mechanism is identified. The PSCs present expected stability and a champion power density of ≈12 mW cm−2 at noon, which is demonstrated as the highest result achieved in near‐space for PSCs. Additionally, despite a complex degradation process, the PSCs still deliver a high energy density comparable to that of silicon solar cells. Finally, the challenges facing the space application of PSCs are discussed. This work highlights the feasibility of PSCs in near space and provides direction for further exploration.

Abstract:PolarLight is a compact soft X-ray polarimeter onboard a CubeSat, which was launched into a low-Earth orbit on October 29, 2018. In March 2019, PolarLight started full operation, and since then, regular observations with the Crab nebula, Sco X-1, and background regions have been conducted. Here we report the operation, calibration, and performance of PolarLight in the orbit. Based on these, we discuss how one can run a low-cost, shared CubeSat for space astronomy, and how CubeSats can play a role in modern space astronomy for technical demonstration, science observations, and student training.

Abstract:Abstract We propose the exploration of polar areas on Mars by a next-generation orbiter mission. In particular, we aim at studying the seasonal and regional variations in snow-deposits, which – in combination with measurements of temporal variations in rotation and gravity field – will improve models of the global planetary CO 2 cycle. A monitoring of polar scarps for rock falls and avalanche events may provide insights into the dynamics of ice sheets. The mapping of the complex layering of polar deposits, believed to contain an important record of climate history, may help us understand the early climate collapse on the planet. Hence, we propose an innovative next-generation exploration mission in polar circular Low Mars Orbit, which will be of interest to scientists and challenging to engineers alike. Schemes will be developed to overcome atmosphere drag forces acting upon the spacecraft by an electric propulsion system. Based on the experience of missions of similar type in Earth orbit we believe that a two-year mission in circular orbit is possible at altitudes as low as 150 km. Such a mission opens new opportunities for novel remote sensing approaches, not requiring excessive telescope equipment or power. We anticipate precision altimetry, powerful radars, high-resolution imaging, and magnetic field mapping.

Abstract:During staffed space flight missions, the astronauts’ circadian timing system and sleep are altered not only due to the microgravity environment but also due to the intense environmental stimulus. Loss of sleep during the preflight, flight, and postflight periods are commonly reported by the space crew. Long periods of enforced wakefulness and poor quality of sleep or insomnia result in fatigue, impaired cognition, and decreased alertness leading to psychological dysfunctions, neurobehavioral performance decrement among astronauts. These may ultimately result in loss of critical thinking and decision-making capabilities, and cause an increased risk of injuries and accidents. Indian Space Research Organisation (ISRO) has established a Human Space Flight Centre (HSFC) in 2019 for undertaking a manned space flight. The success of this interplanetary mission would depend on dedicated and collaboratory efforts of experts from multiple disciplines, including psychology, biology, and medical sciences, apart from experienced engineers and technicians. It is also pertinent to ask whether HSFC of ISRO will have inputs from experts in biological science and whether India will undertake its first manned orbital space flight without conducting a trial run with a trained animal. It is pivotal that ISRO establishes a dedicated Advanced Concepts Team that has expertise in sleep, biological rhythms, and fatigue countermeasures, to support its missions.

Abstract:A main goal of human space exploration is to develop humanity into a multi-planet species where civilization extends beyond planet Earth. Establishing a self-sustaining human presence on Mars is key to achieving this goal. <i>In situ</i> resource utilization (ISRU) on Mars is a critical component to enabling humans on Mars to both establish long-term outposts and become self-reliant. This article focuses on a mission architecture using the SpaceX Starship as cargo and crew vehicles for the journey to Mars. The first Starships flown to Mars will be uncrewed and will provide unprecedented opportunities to deliver ∼100 metric tons of cargo to the martian surface per mission and conduct robotic precursor work to enable a sustained and self-reliant human presence on Mars. We propose that the highest priority activities for early uncrewed Starships include pre-placement of supplies, developing infrastructure, testing of key technologies, and conducting resource prospecting to map and characterize water ice for future ISRU purposes.

Lower Body Negative Pressure Exposure—as Perspective Countermeasure for Moon Missions

Abstract:According to the existing scenarios of interplanetary missions, the Moon is considered as an intermediate base on the way to deep space. In order to prepare for landing and work on the Moon, it is important to study the applicability of countermeasures in such missions. The paper presents the results of a pilot study performed during a short-term spaceflight (12 days). A new experience of using countermeasure impacts of lower body negative pressure (LBNP) at the early stages of adaptation to microgravity conditions has been gained. To assess the effectiveness of LBNP and changes in human physical performance after the spaceflight, we conducted tests on a treadmill, a bicycle ergometer, and testing with model tasks of on-planetary activity "express test". Regulatory mechanisms of the cardiovascular system proved to be quite effective when creating decompression up to -20 mm Hg, which is less than in preparation for returning to Earth. In the treadmill test, a lower speed was achieved after the spaceflight than before (13 km/h and 15 km/h, respectively) and cardiovascular system response to the change in load was slower. At the same time changes in such physiological parameters as oxygen consumption, respiratory rate and pulmonary ventilation were minimal. In the bicycle ergometer test, peak heart rate values were higher after the spaceflight than before, the physiological value of the standard exercise increased. «Express test» showed the positive dynamics from the first day to the third after returning to Earth: the performance of a dual task, the task of controlling the movement of the non-leading hand, and getting up from the supine position improved. Thus, assessment of the state of gravity-dependent physiological systems after short-term flight indicates the decrease of functional reserves of the organism and the necessity to develop appropriate countermeasures. The study was one of the first steps in the development of new medical control operations at the stage of paradigm shift from orbital flights to deep space exploration.
