Abstract:A novel one- and two-photon fluorescent probe CB1 has been developed for discriminating Cys and Hcy in a successive manner with high selectivity. The discrete time-dependent fluorescent responses enable us to sequentially detect Cys and Hcy in different time windows. Two-step reaction and kinetic modes were used to explain the sensing mechanism. As a promising biosensor for cell imaging, CB1 has been confirmed to exhibit membrane permeability to intact cells, low cytotoxicity to viable cells and photostability to ultraviolet light excitation. Furthermore, the results from the control assay have shown that the one- and two-photon fluorescence of CB1 within cells is associated with intracellular mercapto biomolecules but yet there is little interference with physiological pH value, viscosity and common bioanalytes. Finally one- and two-photon fluorescent images of CB1 within living SiHa cells have been presented.

Abstract:In this research, an "off–on" long-wavelength fluorescent probe (DCMN-Cl) based on (E)-2-(3-(2-(6-hydroxynaphthalen-2-yl)vinyl)-5,5-dimethylcyclohex-2-en-1-ylidene) malononitrile (DCMN) is designed and synthesized for cysteine (Cys) detection. DCMN-Cl exhibits a large Stokes shift (211 nm) and shows rapid response and high specificity to Cys. The fluorescence initensity at 635 nm reveals a good linear relationship with Cys concentration in the 0 to 50 μM range, and the detection limit is as low as 159 nM. The probe is also used for fluorescence imaging of Cys in cells and mice. Moreover, the probe provided visual evidence of Cu 2+ and curcumin-induced intracellular Cys fluctuations.

Abstract:The quantification of cysteine (Cys) levels in the organisms holds paramount significance in biological research and disease diagnosis, which can give the correlation between abnormal Cys levels and diseases. In this study, two fluorescent probes, designated as DEA-OH and DEA-AC, featuring a coumarin-aurone backbone specifically engineered for Cys detection, were meticulously designed and synthesized. The diethylamino coumarin-aurone probe DEA-OH and the acrylate-substituted probe DEA-AC demonstrated remarkable sensitivity in detecting cysteine by means of copper displacement (DEA-OH) and acrylate hydrolysis mechanisms (DEA-AC) with fluorescence detection limits of 7.25 μM and 1.65 μM, respectively. Moreover, the fluorescence peak wavelength of the two probes displayed a linear relationship with solvent polarity in the ET (30) range of 30-65 kcal•mol-1, indicating the potential for monitoring changes in environmental polarity within this ET (30) range. The outstanding attributes exhibited by DEA-AC including superior photostability, remarkable selectivity, and swift response (kinetic rate constant: 0.00747 s-1), coupled with the exceptional anti-interference ability, have significantly broadened its scope of applications, for example detecting alterations in Cys within biological systems.
A simple indanone-based red emission fluorescent probe for the rapid detection of cysteine in vitro and in vivo

Abstract:Cysteine is a vital biothiols that plays an important role in numerous physiological and pathological processes. The development of simple molecule tools for detection and analysis Cys in subcellar environment is significant for further exploring their pathophysiological. In this work, a simple but activated fluorescent probe AMIA was constructed with a donor-π-accepter (D- π -A) structure, which using an indanone as the electron-withdrawing unit acting as the fluorophore, dimethylamino group attached to the position 4 of the benzene ring as the electron-donating, two double bonds as the linker group, and the acryloyl ester group as the trigger and response unit. This probe AMIA was exhibited highly selective and sensitive response to Cys over other amino acids and ions under physiological conditions. It was found that AMIA showed a red turn-on fluorescence response at 630 nm towards Cys with a large stroke shift of 170 nm and a very low detection limit of 26.3 nM. HRMS, 1H NMR and TD-DFT calculation further confirmed that the response mechanism is the Cys triggered the addition-cyclization reaction between AMIA' acryloyl group and Cys' sulfhydryl and amino unit, leading to the release of a red fluorescent dye AMIA-OH, which can be identified by naked eyes. Furthermore, AMIA was successfully applied for simultaneous determination of Cys in living cells and zebrafish with lower cytotoxicity and good cell permeability. We hope that this novel indanone-based probe AMIA will provide a new reference for visualized Cys in other complex biological system.

Abstract:A novel class of curcumin-derived fluorescent probes was designed. This kind of probe introduces easy leaving groups methylsulfonyl and phenylsulfonyl respectively to achieve the detection effect through the nucleophilic attack of amino acids. At the same time, BF2 group is introduced to increase the emission wavelength of the probe. Probes 4 and 5 can respond quickly with amino acids, but can specifically recognize Cys. In UV detection, the maximum absorption wavelength of the probes can be blue-shifted by 81 nm with the addition of Cys and still show a strong fluorescence signal. The detection limits for compounds 4 and 5 were determined to be 0.40 μM and 0.87 μM, respectively, with a goodness-of-fit of 0.99. In addition, a rapid response of the probe to Cys could be observed with the naked eye within 1 min. These results provide a new method for rapid detection of Cys; And this kind of probe has the drug structure of curcumin, which can provide ideas for the design of drug-probe.

Abstract:Cysteine, as a vital endogenous small molecule mercaptan, plays a crucial role in various physiological processes. The high sensitivity and selectivity of fluorescent probes provide a method to monitor cysteine, which is helpful to understand the mechanism of cysteine in physiological processes more comprehensively. However, the detection of cysteine can be interfered by other small molecule biothiols. Therefore, the design of fluorescent probe based on the structural characteristics and reactivity of cysteine has become research focus currently. Given the biological compatibilities, biological targets, the metabolic pathway of 3-hydoxythalidomide, and its unique fluorescent properties, herein, we have designed a chemodosimeter, 2-(2,6-dioxopiperidin-3-yl)-1,3-dioxoisoindolin-4-yl acrylate, for the detection of cysteine based on a tandem reaction of thiol-ene click chemistry and aminolysis involving 3-hydroxythalidomide as a parent compound. Experimental data exhibited that the probe showed unique selectivity and sensitivity for cysteine over other amino acid and biothiols. In addition, the fluorescent intensity at 511 nm increased linearly as a function of cysteine concentration in the range of 0-6 × 10-7 M (regression factor, R2 = 0.999), with a limit of detection of 6.1 nM. The sensing mechanism was confirmed through 1H NMR titration and density functional theory calculations. Additionally, the probe was also successfully utilized for the detection of cysteine in sewage and for bioimaging in HeLa cells.

Abstract:Differentiation of biologically important thiols, such as cysteine (Cys), homocysteine (Hcy), and glutathione (GSH) is still a challenging task. Herein, we present a novel fluorescent chemodosimeter capable of selectively detecting Cys over other biothiols including Hcy and GSH and other amino acids by a facile thiol-Michael addition/transcyclization rearrangement cascade click process. The unique trans-cyclization step is critical for the selectivity as a result of the kinetically favorable formation of a six-membered ring with the Cys Michael adduct. Moreover, the probe adopts a dis-tinctive dual quenching mechanism-photoinduced electron transfer (PET) and photoinduced intramolecular charge transfer (ICT) to deliver a drastic turn-on fluorescence response only at the Cys-selective transcylization step. The judicious selection of strong electron-withdrawing naphthalimide fluorophore with maleimide group enhances the electrophilicity and thus reactivity for the cascade process leading to fast detection and ultrasensitivity with a detection limit of 2.0 nm (S/N=3). The probe has demonstrated its practical utility potential in Cys imaging in live cells.

Abstract:We reported here a turn-on fluorescent probe (1) for the detection of cysteine (Cys) by incorporating the recognition unit of 2,4-dinitrobenzenesulfonyl ester (DNBS) to a coumarin derivative. The structure of the obtained probe was confirmed by NMR and HRMS techniques. The probe shows a remarkable fluorescence off-on response (∼52-fold) by the reaction with Cys in 100% aqueous buffer. The sensing mechanism was verified by the HPLC test. Probe 1 also displays high selectivity towards Cys. The detection limit was calculated to be 23 nM. Moreover, cellular experiments demonstrated that the probe is highly biocompatible and can be used for monitoring intracellular Cys.

Abstract:A new strategy for fast fluorescent detection of cysteine (Cys), based on a response-assisted electrostatic attraction, is demonstrated. By utilizing this strategy, we designed and synthesized three fluorescent probes for the specific detection of Cys under actual physiological conditions. The probe m-CP, a coumarin fluorophore conjugated with a substituted methyl pyridinium group through an unsaturated ketone unit, showed highly selective and sensitive detection for cysteine (Cys) over homocysteine (Hcy) and glutathione (GSH). The kinetic analysis indicated that the sensing process was highly accelerated (a response time less than 1min) by the response-assisted electrostatic attraction. More importantly, control experiments with isomeric probes first demonstrated that the spatial charge configuration of the probe played an important role in Cys-preferred selectivity and kinetic rate acceleration. Furthermore, the practical utility of the probe m-CP in the fluorescent labeling of Cys residues within proteins was demonstrated. Finally, these probes were employed in living cell imaging with HeLa cells, in which it displayed satisfactory cell permeability and enabled us to distinguish active thiols in the cytoplasm, nucleus, and mitochondria.
Ratiometric Fluorescent Nanosensors for Selective Detecting Cysteine with Upconversion Luminescence.

Abstract:Fluorescent sensors based on upconversion (UC) luminescence have been considered as a promising strategy to detect bio-analyte due to their advantages in deep penetration, minimum autofluorescence, and ratiometric fluorescent output. A prototype of nanosensors combined with mesoporous silica coated upconversion nanoparticles (UCNPs) and a fluorescein-based fluorescent probe loaded in pores was therefore designed to detect cysteine (Cys). The silica shell provided loading space for the probe and enabled the nanosensors to disperse in water. In the presence of Cys, the fluorescent probe was transformed into 5(6)-carboxyfluorescein with an emission band centering at 518nm which was secondarily excited by the light at around 475nm from NaYF4:Yb3+, Tm3+ UCNPs driven by 980nm near-infrared (NIR) laser. The intensity ratio between green and blue luminescence (I518/I475) grew exponentially with increasing concentrations of Cys over a range of 20–200μmolL−1. The response of the nanosensors towards Cys was recognizable with naked eyes by luminescence color change. Evidences suggest that these nanosensors are capable of sensing Cys in aqueous solution and distinguishing Cys from homocysteine (Hcy) with kinetically-controlled selectivity. The system was further employed to detect Cys in human serum and the result was in agreement with it tested by high performance liquid chromatography with acceptable recovery.

Abstract:Cysteine (Cys) is a significant biological mercaptan that achieves key roles in several important physiological processes, such as reversible redox homeostasis in living organisms. Abnormal levels of Cys in the human body are directly related to many diseases. In this work, we constructed a sensitive sensor (Cys-NR) by connecting a Cys recognition group to a Nile red derivative. Due to photo-induced electron transfer (PET), the Cys-NR probe showed little fluorescence at 650 nm. With the addition of Cys to the assay solution, the chlorine unit of the probe was substituted by the thiol group of Cys. Further, the amino and sulfhydryl groups in cysteine underwent an intramolecular rearrangement, which led to the Cys-NR probe water solution turning from colorless to pink with an enhancement in fluorescence. The red fluorescence at 650 nm increased about 20 times. Based on the turn-on signal, a selective Cys detection method is developed. The probe signal is not affected by various potential interferences or other competing biothiols and the limit of detection (LOD) is determined to be 0.44 μM. In addition, the probe is further employed for imaging of Cys in live cells, revealing good biological imaging ability that could provide a new way of intracellular Cys detection.

Abstract:We have designed and synthesized a novel ratiometric fluorescent chemodosimeter MHF-based ESIPT process for specific detection of cysteine among the biological thiols. The probe MHF shows very weak blue fluorescence under UV excitation. Upon addition of cysteine (Cys), the reaction of Cys with MHF induces acrylate hydrolysis, thereby enabling the ESIPT process to shift the weak blue emission to a strong green emission with about 20-fold enhancement. We utilized (1)H NMR spectra to elucidate the fluorescence sensing mechanism. Moreover, the cellular imaging experiment indicated the MHF possessed excellent selectivity, low cytotoxicity, and desirable cell permeability for biological applications.

Abstract:Real-time imaging and monitoring of biothiols in living cells are essential for understanding pathophysiological processes. However, the design of the fluorescent probe that has accurate and repeatable real-time monitoring capabilities for these targets is highly challenging. In this study, we prepared a fluorescent sensor, Lc-NBD - Cu(II) , which contains a N1, N1, N2-tris-(pyridin-2-ylmethyl) ethane-1,2-diamine as a Cu(II) chelating unit and a 7-nitrobenz-2-oxa-1,3-diazole fluorophore to detect Cysteine (Cys). Emission changes promoted by addition of Cys to this probe are distinctive and correspond to a range of processes including Cys induced loss of Cu(II) from Lc-NBD-Cu(II) to form Lc-NBD , Cu(I) oxidation to reform Cu(II), Cys oxidation to form Cys-Cys, Cu(II) binding to Lc-NBD to reform Lc-NBD-Cu(II) , and competitive binding of Cu(II) to Cys-Cys. The study also shows that Lc-NBD-Cu(II) maintains high stability during the sensing process and that it can be utilized over a number of detection cycles. Finally, the findings show that Lc-NBD-Cu(II) can be utilized to repetitively sense Cys in living HeLa cells.

Abstract:Cysteine (Cys) plays a crucial role in numerous physiological and pathological processes. Therefore, it is imperative to design a highly selective and sensitive near-infrared (NIR) fluorescent probe to monitor Cys. In this study, we have developed a novel NIR fluorescent probe XA based on Xanthene hybrid tetrahydro-acridine salt dye for specifically tracking of Cys, where a chlorine-substituted tetrahydro-acridine acts as a high Cys-reactive site and water-soluble group. Probe XA exhibits a remarkable "turn-on" NIR emission (830 nm) with an extra-large Stokes shift (305 nm) for monitoring Cys. It also has a high selectivity, rapid response time (6 min) and high sensitivity (LOD as 0.5 μM). We fully characterized and discussed the sensing mechanism of XA toward Cys using HPLC and MS spectrums, as well as quantum theory calculations. Furthermore, the excellent properties of NIR fluorescent detection allow this novel probe to successfully monitor fluctuations of exogenous and endogenous Cys concentration levels in living cells and in vivo.

Abstract:Cysteine is an essential amino acid for humans and can be generated from the conversion of methionine or the interconversions with cystine. The detection of cysteine in the living system is of great importance for the diagnosis of related diseases. Based on the classical cysteine response mechanism, the sulfhydryl group of cysteine undergoes Michael addition with the acrylate to rapidly respond to form a seven-membered ring that specifically recognizes cysteine. In the present work, we designed a novel ratiometric fluorescent probe N-1, which contained the benzo[cd]indol fluorophore and the response group acrylate. As the concentration of cysteine increased, the fluorescence of I485 was significantly enhanced while the fluorescence intensity at 635 nm was weakened. Moreover, probe N-1 showed excellent sensitivity and selectivity to Cys with the calculated LOD of 4.0 × 10-9 M. Probe N-1 was successfully used for imaging cysteine in the living Hela cells with low cytotoxicity, indicating that this ratiometric fluorescent probe would be a novel and useful tool to monitor cysteine in the physiological or pathological processes.

Abstract:4-Nitro-1,8-naphthalic anhydride (NNA) was used to distinguish cysteine from homocysteine and other potentially interfering thiols through a novel sequential substitution mechanism. The discrimination involves a blue-fluorescent thioether formation via nucleophilic aromatic substitution of the nitro group by thiol, followed by a second intramolecular nucleophilic aromatic substitution of alkylthio with the amino group to give the green-fluorescent 4-amino derivative. NNA is highly selective towards Cys, and the detection limit of Cys by this method is 0.3 μM.

After careful assessment, the probe CL-Cys could detect Cys with high sensitivity and total light photons increased by 28-fold after the probe was treated with Cys, with the detection limit of 7.5 X 10(-8) M. Finally, CL-Cys was further utilized for the chemiluminescent imaging of endogenous Cys in a living mouse. In general, this probe has a remarkable ability to detect Cys, which provides a valuable method for interrogation of the Cys roles in more biological and pathological processes.
