Abstract:Due to the robust electrophilic properties of the trifluoromethyl group (-CF3), its incorporation into organic compounds can markedly alter their ester affinity, stability, bioavailability, and other properties. The trifluoromethylation reaction is currently experiencing rapid advancement, with an expanding array of substrates and the emergence of novel methodologies. Consequently, compounds containing the -CF3 moiety find extensive utility across diverse fields. This article aims to comprehensively review the latest advancements in trifluoromethylation reaction of olefins, aldehydes, and ketones, encompassing nucleophilic trifluoromethylation, electrophilic trifluoromethylation, and radical trifluoromethylation. The discussion includes an exploration of the types and broadening scope of applicable substrates. Furthermore, this article addresses the associated challenges and delineates prospective directions for future developments in trifluoromethyl reaction.

Abstract:Comprehensive Summary Trifluoromethyl group is one of the most frequently available and promising functional groups in pharmaceutical, agrochemical as well as material molecules. Developing synthetic methods for the synthesis of trifluoromethyl functionalized compounds by trifluoromethylation reactions occupies significant position in modern organic chemistry. On other hand, as a typical tool of sustainable synthesis, transition metal‐free synthesis possesses unique advantages in atom economy, easy product purification as well as excellent eco‐tolerance. Consequently, transition metal‐free trifluoromethylation reactions constitute the most important approaches for accessing trifluoromethylated molecules. Herein, the advances in the transition metal‐free C‐H trifluoromethylation reactions are reviewed for the first time. According to the updated state‐of‐art, both the catalytic reactions by conventional heating chemistry and the more sustainable photocatalysis are included, covering the trifluoromethylation reactions of C(sp3)‐H bond, aromatic C(sp2)‐H bond as well as the non‐aromatic C(sp2)‐H bond in different substrates over the last five years (2017‐2021). Abstract:In the past 5 years, transition-metal-mediated trifluoromethylation for the construction of various CF3-containing building blocks has been the focus of recent research in both industrial and academic communities. Progresses in the construction of C(sp(2))-CF3 bonds and C(sp)-CF3 have been well reviewed. This Letter will focus on the cases of transition-metal-mediated C(sp(3))-CF3 bond formation, which involves the trifluoromethylation of sp(3)-hybridized C-X bonds, alkyl organometallic reagents, sp(3)-hybridized C-H bonds, and alkene derivatives. Abstract:Here we report the synthesis and application of trifluoromethyl thianthrenium triflate (TT-CF3+OTf–) as a novel trifluoromethylating reagent, which is conveniently accessible in a single step from thianthrene and triflic anhydride. We demonstrate the use of TT-CF3+OTf– in electrophilic, radical, and nucleophilic trifluoromethylation reactions.

Abstract:Trifluoromethanesulfonic anhydride has been widely used in synthetic organic chemistry, not only for the conversion of various oxygen-containing compounds to the triflates, but also for the electrophilic activation and further conversion of amides, sulfoxides, and phosphorus oxides. In recent years, the utilization of Tf2O as an activator for nitrogen-containing heterocycles, nitriles and nitro groups has become a promising tool for the development of new valuable methods with considerable success. In addition, Tf2O has been used as an efficient radical trifluoromethylation and trifluoromethylthiolation reagent due to the contained SO2CF3 fragment, and significant progress has been made in this area. This review summarizes the recent progress in the applications of Tf2O in the above two aspects, and aims to illustrate the role and potential application of this reagent in organic synthesis.

Abstract:New trifluoromethylating reagents are designed via computational studies inspired by Togni's experimental research. Trans influence and steric effects are important for this rational design. We have found that the Togni derivative with X = SO2 is the best choice based on its high reactivity and its potential synthesis.

Abstract:The incorporation of fluorine-containing moieties into organic compounds is of great importance in pharmaceutical, agricultural, and materials science. Within these organofluorides, the trifluoromethyl group is one of the most important motifs. In recent years, the trifluoromethyl group has attracted more and more attention, and many trifluoromethylated compounds have been found to possess special activities. However, until now, only a few methods have been developed to achieve this efficiently using Umemoto's reagents. This review highlights recent developments in the direct introduction of a trifluoromethyl group into organic compounds with Umemoto's reagents. Seven approaches to the trifluoromethylation of organic compounds are summarized: (i) trifluoromethylation of arenes, (ii) trifluoromethylation of alkenes, (iii) trifluoromethylation of terminal alkynes, (iv) deoxygenative trifluoromethylation of benzylic xanthates, (v) trifluoromethylation of ketoesters, (vi) trifluoromethylation of aryl boronic acids and aromatic amines (synthesis of ArCF3) and (vii) trifluoromethylation of biphenyl isocyanide derivatives.

Abstract:Triphenylphosphine-mediated metal-free trifluoromethylthiolation and difluoromethylthiolation of thiols by CF3SO2Cl and CHF2SO2Cl to synthesize trifluoromethyl disulfides and difluoromethyl disulfides, respectively, was achieved at room temperature. Iodine generated in situ from iodide facilitated this reaction via the formation of iodotriphenylphosphonium iodide which could serve as a reducing agent in this transformation. Readily available reagents and mild reaction conditions without transition-metals allow this protocol to be more practical than traditional methods.

Abstract:Transition metal-catalyzed trifluoromethylation of aromatic compounds has been developed rapidly during the last decade. The substrate scope is expanding, new methods and novel trifluoromethyl reagents are continuing to emerge. Recent advances in transition metal-catalyzed trifluoromethylation of aromatic compounds are summarized in this paper. The reactions involve trifluoromethylation of aryl halides, arylboronic acids and C-H trifluoromethylation of arenes. In addition, the mechanisms of transition metal-catalyzed trifluoromethylation are discussed in details.

Abstract:A general and convenient method for the direct transition-metal-free intermolecular chloro- difluoromethylthiolation of a variety of unactivated alkenes and alkynes has been described. This methodology exhibits good functional group tolerance and operationally simple. Mechanistic studies indicated that this is the electrophilic substitution mechanism, and the strong electrophilic difluoromethylthiolating reagent HCF 2 SCl was generated in situ under the standard reaction conditions. The developed protocol fulfills the requirements of sustainable and green chemistry.

Abstract:Recent advances in photoinduced trifluoromethylation and difluoroalkylation under photocatalysis are summarized. Most of the photoredox reactions proceed efficiently under mild conditions with simple operation. Various fluorinated reagents are developed and applied in different transformations. Usually, the photocatalyst in a photocatalytic cycle is initially stimulated to the excited state in the presence of visible light, which then provides an electron to the fluorinated reagent to release a CF3 radical or CF2 radical. The subsequent radical addition to an unsaturated bond with further transformations would produce the corresponding fluorinated products. Alkenes, alkynes, isocyanides, and arenes are frequently employed as substrates for access to fluorochemicals. The advantages of trifluoromethylation and difluoroalkylation under photocatalysis including mild reaction conditions, good functional group tolerance, and experimental ease will make the approaches attractive for further applications.

Abstract:Among the general interest in fluorinated compounds, trifluoromethoxylated molecules play a specific role. However, despite this interest, the development of efficient reagents to perform trifluoromethoxylation reactions remains a challenge. Here, 2,4-dinitro-trifluoromethoxybenzene (DNTFB) is used as a trifluoromethoxylating reagent to perform nucleophilic substitution under mild metal-free conditions with different leaving groups, including direct dehydroxytrifluoromethoxylation. A mechanistic study rationalized the reaction and subsequently proposed only three reaction conditions, depending on the reactivity of the starting substrates.

Abstract:Organic molecules containing trifluoromethyl groups often exhibit unique physical and chemical properties, and have found extensive applications. Trifluoroacetic acid and its derivatives have advantages including low cost, ready availabil-ity and high stability. Furthermore, CO 2 is the byproduct. Therefore, trifluoroacetic acid and its derivatives are ideal trifluoromethylating reagents, and have great application potentials. The progress of trifluoromethylation reactions using trifluoroacetic acid and its derivatives as CF 3 -sources is reviewed, including the reactions of C — X (X ＝ Br and I), C — H, and C ＝ X (X ＝ C, O). The mechanisms are also introduced.

Abstract:A metal-free and cost-effective synthetic protocol for the trifluoromethylation of N,N-disubstituted hydra-zones with Langlois's reagent (CF3SO2Na) to afford the corresponding functionalized trifluoromethyl ketone hydra-zones has been established. It is proposed that a radical/SET mechanism proceeding via a trifluoroalkyl radical may be involved in the reaction. Applications of the methodology in industry will be found and the development of new methods for trifluoromethylation with Langlois's reagent will be continued in our laboratory.

Abstract:Trifluoromethylation using Togni reagents usually releases one equivalent of iodobenzoats as wasteful byproducts. A visible-light-mediated, atom- and step-economical hydrotrifluoromethylation of aromatic alkynes and remote benzoyl-oxvlation of alpha-C(sp(3))-H bond of ether with Togni reagent as a bifunctional reagent by means of hydrogen atom transfer strategy was disclosed. The combination of two organic transformations into one reaction not only brings 100% atom economy but also addresses the challenge of stereoselective hydrotrifluoromethylation of aromatic alkynes. This unprecedented protocol offers an important access to a wide range of highly functionalized CF3-containing alkenes with great potential for post-modification.
