Abstract:LanGui tea, a traditional Chinese medicine formulation comprising of Gynostemma pentaphyllum (Thunb.) Makino, Cinnamomum cassia (L.) J. Presl, and Ampelopsis grossedentata (Hand-Mazz) W.T. Wang, has yet to have its potential contributions to alcoholic liver disease (ALD) fully elucidated. Consequently, the objective of this research is to investigate the protective properties of LanGui tea against binge alcohol-induced ALD and the mechanisms underlying its effects.

What problem does this paper attempt to address?
### Problems the Paper Attempts to Solve This paper aims to explore the protective effects and mechanisms of LanGui tea on acute alcohol-induced liver injury (AALI). Specifically, the researchers hope to experimentally verify whether LanGui tea extract (ELG) can alleviate alcohol-induced hepatic steatosis, inflammatory response, and liver dysfunction by activating the AMPK signaling pathway and inhibiting the NLRP3 signaling pathway. ### Background and Motivation - **Alcoholic Liver Disease (ALD)**: ALD is a common disease caused by excessive alcohol consumption, including conditions such as fatty liver, alcoholic hepatitis, and cirrhosis, which can ultimately lead to liver failure and death. - **AMPK Signaling Pathway**: AMPK plays a key role in energy metabolism. Its activation can promote fatty acid oxidation and inhibit fat synthesis, thereby alleviating fatty liver. Additionally, AMPK activation can reduce liver inflammation, oxidative stress, and liver injury. - **NLRP3 Inflammasome**: The NLRP3 inflammasome is activated upon recognizing pathogen and damaged cell signals, leading to the maturation and release of IL-1β, which triggers an inflammatory response. NLRP3 activation plays a significant role in the progression of ALD. - **LanGui Tea**: LanGui tea is composed of three herbs: Gynostemma pentaphyllum, cinnamon, and Ampelopsis grossedentata. These herbs possess various biological activities, including antioxidant, anti-inflammatory, liver-protective, and metabolic-regulating properties. ### Research Objectives - **Verify the Protective Effects of LanGui Tea**: Evaluate the protective effects of LanGui tea extract (ELG) on acute alcohol-induced liver injury through animal experiments and in vitro cell experiments. - **Explore the Mechanisms**: Investigate whether ELG exerts its protective effects by activating the AMPK signaling pathway and inhibiting the NLRP3 signaling pathway. ### Methods - **Animal Experiments**: Establish an acute alcohol-induced liver injury model using C57BL/6 mice, administer different doses of ELG, and observe its effects on hepatic steatosis, inflammatory response, and liver function. - **In Vitro Experiments**: Use AML-12 hepatocyte cell line to simulate alcohol-induced liver injury, and evaluate the effects of ELG on cell viability, inflammatory factor expression, and liver enzyme levels. - **Molecular Mechanism Studies**: Analyze the effects of ELG on the AMPK and NLRP3 signaling pathways using techniques such as Western blot, qRT-PCR, ELISA, immunohistochemistry, and immunofluorescence. ### Results - **Alleviation of Fatty Liver**: ELG significantly reduced alcohol-induced hepatic fat accumulation and inflammatory cell infiltration. - **Inhibition of Inflammatory Response**: ELG decreased the expression of inflammatory factors such as IL-1β, TNF-α, and MCP-1, alleviating liver inflammation. - **Improvement of Liver Function**: ELG significantly lowered serum ALT and AST levels, indicating its protective effects on liver function. - **Inhibition of Apoptosis and ER Stress**: ELG reduced the expression of CHOP and cleaved caspase-3, suggesting it can alleviate alcohol-induced apoptosis and endoplasmic reticulum stress. - **In Vitro Validation**: ELG also exhibited similar protective effects in vitro, inhibiting alcohol-induced cell damage and the production of inflammatory factors. ### Conclusion LanGui tea effectively alleviates acute alcohol-induced liver injury by activating the AMPK signaling pathway and inhibiting the NLRP3 signaling pathway. These findings provide scientific evidence for the potential use of LanGui tea as a therapeutic agent for ALD.
