Abstract:Colon adenocarcinoma (COAD) is a prevalent gastrointestinal malignancy and ranks as the third leading cause of cancer-related mortality worldwide. [ 1 ] The disease affects millions of individuals, particularly in developing countries. Current therapeutic approaches for COAD include traditional surgical, chemotherapy, and radiotherapy modalities, along with emerging immunotherapy strategies. [ 2 ] Despite significant advancements in COAD treatment, its incidence is rapidly increasing, and 5-year survival rates remain low, [ 3 , 4 ] particularly among patients with metastatic COAD. Given the high heterogeneity of COAD, [ 5 ] there may be substantial variations in prognosis among patients with similar clinical characteristics. Therefore, it is imperative to establish a multi-gene model that captures patient prognosis and immunotherapy responsiveness to enhance the prognostic outcomes of COAD patients and facilitate personalized treatment.
