Abstract:The ultimate goal of astrobiology is to determine the distribution and diversity of life in the universe. But as the word "biosignature" suggests, what will be detected is not life itself, but an observation implicating a particular process associated with living systems. Technical constraints and our limited access to other worlds suggest we are more likely to detect an out-of-equilibrium suite of gasses than a writhing octopus. Yet, anything short of a writhing octopus will raise skepticism among astrobiologists about what has been detected. Resolving that skepticism requires a theory to delineate processes due to life and those due solely to abiotic mechanisms. This poses an existential question for the endeavor of life detection: How do astrobiologists plan to detect life via features shared between non-living and living systems? We argue that you cannot without an underlying theory of life. We illustrate this by analyzing the hypothetical detection of an "Earth 2.0" exoplanet. In the absence of a theory of life, we argue the community should focus on identifying unambiguous features of life via four areas of active research: understanding the principles of life on Earth, building life in the lab, detecting life in the solar system and searching for technosignatures. Ultimately, we ask, what exactly do astrobiologists hope to learn by searching for life?

Abstract:For the first time in human history, we will soon be able to apply the scientific method to the question "Are We Alone?" The rapid advance of exoplanet discovery, planetary systems science, and telescope technology will soon allow scientists to search for life beyond our Solar System through direct observation of extrasolar planets. This endeavor will occur alongside searches for habitable environments and signs of life within our Solar System. While the searches are thematically related and will inform each other, they will require separate observational techniques. The search for life on exoplanets holds potential through the great diversity of worlds to be explored beyond our Solar System. However, there are also unique challenges related to the relatively limited data this search will obtain on any individual world. This white paper reviews the scientific community's ability to use data from future telescopes to search for life on exoplanets. This material summarizes products from the Exoplanet Biosignatures Workshop Without Walls (EBWWW). The EBWWW was constituted by a series of online and in person activities, with participation from the international exoplanet and astrobiology communities, to assess state of the science and future research needs for the remote detection of life on planets outside our Solar System.
An Agnostic Biosignature Based on Modeling Panspermia and Terraformation

Abstract:A fundamental goal of astrobiology is to detect life outside of Earth. This proves to be an exceptional challenge outside of our solar system, where strong assumptions must be made about how life would manifest and interact with its planet. Such assumptions are required because of the lack of a consensus theory of living systems, or an understanding of the possible extent of planetary dynamics. Here we explore a model of life spreading between planetary systems via panspermia and terraformation. Our model shows that as life propagates across the galaxy, correlations emerge between planetary characteristics and location, and can function as a population-scale agnostic biosignature. This biosignature is agnostic because it is independent of strong assumptions about any particular instantiation of life or planetary characteristic--by focusing on a specific hypothesis of what life may do, rather than what life may be. By clustering planets based on their observed characteristics, and examining the spatial extent of these clusters, we demonstrate (and evaluate) a way to prioritize specific planets for further observation--based on their potential for containing life. We consider obstacles that must be overcome to practically implement our approach, including identifying specific ways in which better understanding astrophysical and planetary processes would improve our ability to detect life. Finally, we consider how this model leads us to think in novel ways about hierarchies of life and planetary scale replication.

Abstract:Astrobiology, the study of life as a planetary phenomenon, aims to understand the fundamental nature of life on earth and the possibility of life elsewhere. To achieve this goal, astrobiologists have initiated unprecedented communication between the disciplines of astronomy, biology, chemistry, and geology. The Astrobiology Primer has been created as a reference tool for those who are interested in the interdisciplinary field of astrobiology. The field incorporates many diverse research endeavors, but it is our hope that this slim volume will present the reader with all he or she needs to know to become involved and to understand, at least at a fundamental level, the state of the art. Because of the great diversity of material, each section was written by a different author with a different expertise. The Primer was constructed collaboratively. Ninety researchers from around the world contributed information with regard to what they expected from other astrobiologists and what they would like to know themselves but still had difficulty understanding (see Contributors). Those submissions were read and considered by the Editors who produced a list of seven general categories of knowledge, represented by the seven chapters in the Primer: 1) Stellar Formation and Evolution, 2) Planetary Formation and Evolution, 3) Astrobiogeochemistry and the Origin of Life, 4) Evolution of Life through Time, 5) Planet Detection & Characterization, 6) Diversity of Life, and 7) Science in Space. No one volume, of course, can contain the vast amount of information brought to play in astrobiology, but we believe that the Primer will provide a forum and a language around which the community will have the opportunity to develop a consensus about central issues.

Abstract:In 2019, the Atacama Rover Astrobiology Drilling Studies (ARADS) project field-tested an autonomous rover-mounted robotic drill prototype for a 6-Sol life detection mission to Mars (Icebreaker). ARADS drilled Mars-like materials in the Atacama Desert (Chile), one of the most life-diminished regions on Earth, where mitigating contamination transfer into life-detection instruments becomes critical. Our Contamination Control Strategy and Implementation (CCSI) for the Sample Handling and Transfer System (SHTS) hardware (drill, scoop and funnels) included out-of-simulation protocol testing (out-of-sim) for hardware decontamination and verification during the 6-Sol simulation (in-sim). The most effective five-step decontamination combined safer-to-use sterilants (3%_hydrogen-peroxide-activated 5%_sodium-hypochlorite), and in situ real-time verification by adenosine triphosphate (ATP) and Signs of Life Detector (SOLID) Fluorescence Immunoassay for characterization hardware bioburden and airborne contaminants. The 20- to 40-min protocol enabled a 4-log bioburden reduction down to <0.1 fmoles ATP detection limit (funnels and drill) to 0.2-0.7 fmoles (scoop) of total ATP. The (post-cleaning) hardware background was 0.3 to 1-2 attomoles ATP/cm2 (cleanliness benchmark background values) equivalent to ca. 1-10 colony forming unit (CFU)/cm2. Further, 60-100% of the in-sim hardware background was ≤3-4 bacterial cells/cm2, the threshold limit for Class <7 aseptic operations. Across the six Sols, the flux of airborne contaminants to the drill sites was ∼5 and ∼22 amoles ATP/(cm2·day), accounting for an unexpectedly high Fluorescence Intensity (FI) signal (FI: ∼6000) against aquatic cyanobacteria, but negligible anthropogenic contribution. The SOLID immunoassay also detected microorganisms from multiple habitats across the Atacama Desert (anoxic, alkaline/acidic microenvironments in halite fields, playas, and alluvial fans) in both airborne and post-cleaning hardware background. Finally, the hardware ATP background was 40-250 times lower than the ATP in cores. Similarly, the FI peaks (FImax) against the microbial taxa and molecular biomarkers detected in the post-cleaned hardware (FI: ∼1500-1600) were 5-10 times lower than biomarkers in drilled sediments, excluding significant interference with putative biomarker found in cores. Similar protocols enable the acquisition of contamination-free materials for ultra-sensitive instruments analysis and the integrity of scientific results. Their application can augment our scientific knowledge of the distribution of cryptic life on Mars-like grounds and support life-detection robotic and human-operated missions to Mars.
Comparative study of methods for detecting extraterrestrial life in exploration mission of Mars and the solar system

Abstract:The detection and analysis of extraterrestrial life are important issues of space science. Mars is among the most important planets to explore for extraterrestrial life, owing both to its physical properties and to its ancient and present environments as revealed by previous exploration missions. In this paper, we present a comparative study of methods for detecting extraterrestrial life and life-related substances. To this end, we have classified and summarized the characteristics targeted for the detection of extraterrestrial life in solar system exploration mission and the methods used to evaluate them. A summary table is presented. We conclude that at this moment (i) there is no realistic single detection method capable of concluding the discovery of extraterrestrial life, (ii) no single method has an advantage over the others in all respects, and (iii) there is no single method capable of distinguishing extraterrestrial life from terrestrial life. Therefore, a combination of complementary methods is essential. We emphasize the importance of endeavoring to detect extraterrestrial life without overlooking possible alien life forms, even at the cost of tolerating false positives. Summaries of both the targets and the detection methods should be updated continuously, and comparative studies of both should be pursued. Although this study assumes Mars to be a model site for the primary environment for life searches, both the targets and detection methods described herein will also be useful for searching for extraterrestrial life in any celestial environment and for the initial inspection of returned samples.
Technology Needs for Detecting Life Beyond the Solar System: A White Paper in Support of the Astrobiology Science Strategy

Abstract:In support of the Astrobiology Science Strategy, this whitepaper outlines some key technology challenges pertaining to the remote search for life in exoplanetary systems. Finding evidence for life on rocky planets outside of our solar system requires new technical capabilities for the key measurements of spectral signatures of biosignature gases, and of planetary mass measurement. Spectra of Earth-like planets can be directly measured in reflected stellar light in the visible band or near-infrared using a factor 1e-10 starlight suppression with occulters, either internal (coronagraph) or external (starshade). Absorption and emission (reflected and thermal) spectra can be obtained in the mid-infrared of rocky planets transiting M-dwarfs via spectroscopy of the transit and secondary eclipse, respectively. Mass can be measured from the star's reflex motion, the reflex motion of a star, via either precision radial velocity methods or astrometry. Several technology gaps must be closed to provide astronomers the necessary capabilities to obtain these key measurements for small planets orbiting within the predicted temperate zones around nearby stars. These involved performance improvements, in some cases, 1-2 orders of magnitude from state-of-the-art or involve performances never demonstrated. The technologies advancing to close these gaps have been identified through the NASA Exoplanet Exploration Program's annual Technology Selection and Prioritization Process in collaboration with the larger exoplanet science and technology community

Abstract:The 2-week, virtual Future of the Search for Life science and engineering workshop brought together more than 100 scientists, engineers, and technologists in March and April 2022 to provide their expert opinion on the interconnections between life-detection science and technology. Participants identified the advances in measurement and sampling technologies they believed to be necessary to perform in situ searches for life elsewhere in our Solar System, 20 years or more in the future. Among suggested measurements for these searches, those pertaining to three potential indicators of life termed "dynamic disequilibrium," "catalysis," and "informational polymers" were identified as particularly promising avenues for further exploration. For these three indicators, small breakout groups of participants identified measurement needs and knowledge gaps, along with corresponding constraints on sample handling (acquisition and processing) approaches for a variety of environments on Enceladus, Europa, Mars, and Titan. Despite the diversity of these environments, sample processing approaches all tend to be more complex than those that have been implemented on missions or envisioned for mission concepts to date. The approaches considered by workshop breakout groups progress from nondestructive to destructive measurement techniques, and most involve the need for fluid (especially liquid) sample processing. Sample processing needs were identified as technology gaps. These gaps include technology and associated sampling strategies that allow the preservation of the thermal, mechanical, and chemical integrity of the samples upon acquisition; and to optimize the sample information obtained by operating suites of instruments on common samples. Crucially, the interplay between science-driven life-detection strategies and their technological implementation highlights the need for an unprecedented level of payload integration and extensive collaboration between scientists and engineers, starting from concept formulation through mission deployment of life-detection instruments and sample processing systems.

Abstract:Abstract Astrobiology research seeks to understand how life begins and evolves, and to determine whether life exist elsewhere in the universe. The discovery of diverse ocean worlds has significantly expanded the number of planetary bodies in the Solar System that could potentially contain life. Of the recognized ocean worlds, Saturn's moon Enceladus stands out because it appears to meet all requirements to sustain life. For that reason, robotic mission concepts are being developed to determine whether Enceladus' ocean is inhabited. The theory of organic chemical evolution (OCE) represents an ideal framework to guide this exploration strategy, articulating investigations and associated measurements of organic matter in the subsurface ocean. Within this reference frame, the immediate priority with the lowest science risk would be to understand molecular and structural properties of bulk organic matter in the ocean, and search for metabolic precursors and biochemical building blocks, both free and bound. This could be supplemented with “high‐risk, high‐reward” searches for functional polymers, catalytic activity, and cell‐like objects with traits indicative of evolutionary adaptations. The theory of OCE provides a robust scientific foundation for the astrobiological exploration of ocean worlds, fostering a productive path to discovery with lower mission risk that could be implemented with existing technology. Strong synergies between astrobiology and Earth‐bound research could ensue from this exploration strategy particularly in the context of terrestrial analog studies and laboratory simulations.

Atmospheric Biomarkers and their Evolution over Geological Timescales

Abstract:The search for life on extrasolar planets is based on the assumption that one can screen extrasolar planets for habitability spectroscopically. The first space born instruments able to detect as well as characterize extrasolar planets, Darwin and terrestrial planet finder (TPF-I and TPF-C) are scheduled to launch before the end of the next decade. The composition of the planetary surface, atmosphere, and its temperature-pressure profile influence a detectable spectroscopic signal considerably. For future space-based missions it will be crucial to know this influence to interpret the observed signals and detect signatures of life in remotely observed atmospheres. We give an overview of biomarkers in the visible and IR range, corresponding to the TPF-C and TPF-I/DARWIN concepts, respectively. We also give an overview of the evolution of biomarkers over time and its implication for the search for life on extrasolar Earth-like planets. We show that atmospheric features on Earth can provide clues of biological activities for at least 2 billion years.

Detectability of biosignatures in a low-biomass simulation of martian sediments

Abstract:Discovery of a remnant habitable environment by the Mars Science Laboratory in the sedimentary record of Gale Crater has reinvigorated the search for evidence of martian life. In this study, we used a simulated martian mudstone material, based on data from Gale Crater, that was inoculated and cultured over several months and then dried and pressed. The simulated mudstone was analysed with a range of techniques to investigate the detectability of biosignatures. Cell counting and DNA extraction showed a diverse but low biomass microbial community that was highly dispersed. Pellets were analysed with bulk Elemental Analysis - Isotope Ratio Mass Spectrometry (EA-IRMS), high-resolution Laser-ablation Ionisation Mass Spectrometry (LIMS), Raman spectroscopy and Fourier Transform InfraRed (FTIR) spectroscopy, which are all techniques of relevance to current and future space missions. Bulk analytical techniques were unable to differentiate between inoculated samples and abiotic controls, despite total levels of organic carbon comparable with that of the martian surface. Raman spectroscopy, FTIR spectroscopy and LIMS, which are high sensitivity techniques that provide chemical information at high spatial resolution, retrieved presumptive biosignatures but these remained ambiguous and the sedimentary matrix presented challenges for all techniques. This suggests challenges for detecting definitive evidence for life, both in the simulated lacustrine environment via standard microbiological techniques and in the simulated mudstone via analytical techniques with relevance to robotic missions. Our study suggests that multiple co-incident high-sensitivity techniques that can scan the same micrometre-scale spots are required to unambiguously detect biosignatures, but the spatial coverage of these techniques needs to be high enough not to miss individual cellular-scale structures in the matrix.

Abstract:Although astrobiology is a relatively new field of science, the questions it seeks to answer (e.g., "What is life?" "What does life require?") have been investigated for millennia. In recent decades, formal programs dedicated specifically to the science of astrobiology have been organized at academic, governmental, and institutional scales. Constructing educational programs around this emerging science relies on input from broad expertise and backgrounds. Because of the interdisciplinary nature of this field, career pathways in astrobiology often begin in more specific fields such as astronomy, geology, or biology, and unlike many other sciences, typically involve substantial training outside one's primary discipline. The recent origin of astrobiology as a field of science has led to strong collaborations with education research in the development of astrobiology courses and offers a unique instructional laboratory for further pedagogical studies. This chapter is intended to support students, educators, and early career scientists by connecting them to materials and opportunities that the authors and colleagues have found advantageous. Annotated lists of relevant programs and resources are included as a series of appendices in the supplementary material.
Detecting Earth-like Biosignatures on Rocky Exoplanets around Nearby Stars with Ground-based Extremely Large Telescopes

Abstract:As we begin to discover rocky planets in the habitable zone of nearby stars with missions like TESS and CHEOPS, we will need quick advancements on instrumentation and observational techniques that will enable us to answer key science questions, such as What are the atmospheric characteristics of habitable zone rocky planets? How common are Earth-like biosignatures in rocky planets?} How similar or dissimilar are those planets to Earth? Over the next decade we expect to have discovered several Earth-analog candidates, but we will not have the tools to study the atmospheres of all of them in detail. Ground-based ELTs can identify biosignatures in the spectra of these candidate exo-Earths and understand how the planets' atmospheres compare to the Earth at different epochs. Transit spectroscopy, high-resolution spectroscopy, and reflected-light direct imaging on ELTs can identify multiple biosignatures for habitable zone, rocky planets around M stars at optical to near-infrared wavelengths. Thermal infrared direct imaging can detect habitable zone, rocky planets around AFGK stars, identifying ozone and motivating reflected-light follow-up observations with NASA missions like HabEx/LUVOIR. Therefore, we recommend that the Astro2020 Decadal Survey Committee support: (1) the search for Earth-like biosignatures on rocky planets around nearby stars as a key science case; (2) the construction over the next decade of ground-based Extremely Large Telecopes (ELTs), which will provide the large aperture and spatial resolution necessary to start revealing the atmospheres of Earth-analogues around nearby stars; (3) the development of instrumentation that optimizes the detection of biosignatures; and (4) the generation of accurate line lists for potential biosignature gases, which are needed as model templates to detect those molecules.

Abstract:With state-of-the-art imaging and analytical tools onboard the NASA Perseverance Rover Mission, geological information for remote astrobiological analysis is readily available and more widespread than ever. For analysis of such data, there is a need for automated, remote assessment tools, capable of translational and scalable research. To search for evidence of life in the universe, one of the goals of astrobiological research, we must first recognize robust indicators of life on Earth. One method is the study of Earth's early fossil record, dominated by simple organisms similar to that predicted to have existed on Mars - if life ever did develop. We identify a potential morphological biosignature, known as double-rippled bedforms (DRBs), from the Ediacaran Member (Rawnsley Quartzite, 550–555 Ma) of South Australia. We utilize these DRBs to develop a new tool for astrobiological investigation via an improved end-to-end remote, automated biosignature assessment tool, Scene-aware Perception Automation using Composite Embedding for Segmentation (SPACESeg2.0). The proposed tool accurately detects and quantitatively characterizes DRBs in imagery from varied fields-of-view, with practical artifact-ridden data, even when the DRBs constitute less than 1% of the total image. We establish the efficacy of SPACESeg2.0 in detecting desired structures compared with other deep-learning techniques.

Abstract:Life on our planet likely evolved in the ocean, and thus exo-oceans are key habitats to search for extraterrestrial life. We conducted a data-driven bibliographic survey on the astrobiology literature to identify emerging research trends with marine science for future synergies in the exploration for extraterrestrial life in exo-oceans. Based on search queries, we identified 2592 published items since 1963. The current literature falls into three major groups of terms focusing on (1) the search for life on Mars, (2) astrobiology within our Solar System with reference to icy moons and their exo-oceans, and (3) astronomical and biological parameters for planetary habitability. We also identified that the most prominent research keywords form three key-groups focusing on (1) using terrestrial environments as proxies for Martian environments, centred on extremophiles and biosignatures, (2) habitable zones outside of "Goldilocks" orbital ranges, centred on ice planets, and (3) the atmosphere, magnetic field, and geology in relation to planets' habitable conditions, centred on water-based oceans.

Abstract:The search for life on planets outside our solar system has largely been the province of the astrophysics community until recently. A major development since the NASA Astrobiology Strategy 2015 document (AS15) has been the integration of other NASA science disciplines (planetary science, heliophysics, Earth science) with ongoing exoplanet research in astrophysics. The NASA Nexus for Exoplanet System Science (NExSS) provides a forum for scientists to collaborate across disciplines to accelerate progress in the search for life elsewhere. Here we describe recent developments in these other disciplines, with a focus on exoplanet properties and environments, and the prospects for future progress that will be achieved by integrating emerging knowledge from astrophysics with insights from these fields.

Abstract:The ancestor philosophers' dream of thousands of new worlds is finally realised: about 3500 extrasolar planets have been discovered in the neighborhood of our Sun. Most of them are very different from those we used to know in our Solar System. Others orbit their parent star inside the belt known as Habitable Zone where a rocky planet with the appropriate climate could have the availability of liquid water on its surface. Those planets, in HZ or not, will be the object of observation that will be performed by new space-/ground-based instrumentation. Space missions, such as JWST and the very recently proposed ARIEL (ESA M-Class Mission), or ground based instruments (SPHERE@VLT, GPI@GEMINI and EPICS@ELT) have been proposed and built to measure the atmospheric transmission, reflection and emission spectra over a wide wavelength range. Most of exoplanets have local counterparts in the Solar System planets that are available for comparative studies, but there are also interesting outsider cases like super Earths. In our own system, proto-planet evolution was flanked by an active prebiotic chemistry that brought about the emergency of life on the Earth. The search for life signatures requires the knowledge of planet atmospheres, main objective of future exoplanetary space explorations. As, for now, we have only one example of life in the universe, we are bound to study terrestrial organisms to assess possibilities of life on other planets and guide our search for possible extinct or extant life on other planetary bodies. The planet atmosphere characteristics and possible biosignatures will be inferred by studying such composite spectrum in order to identify the emission/absorption lines/bands from atmospheric molecules as water, carbon monoxide, methane, ammonia etc.

Abstract:Despite great advances in our understanding of the formation of the Solar System, the evolution of the Earth, and the chemical basis for life, we are not much closer than the ancient Greeks to an answer of whether life has arisen and persisted on any other planet. The origin of life as a planetary phenomenon will probably resist successful explanation as long as we lack an early record of its evolution and additional examples. It is widely thought that the geologic record shows that life emerged quickly after the end of prolonged bombardment of the Earth. New data and simulations contradict that view and suggest that more than half a billion years of unrecorded Earth history may have elapsed between the origin of life and LUCA. The impact-driven exchange of material between the inner planets may have allowed earliest life to be more cosmopolitan. Indeed, terrestrial life may not have originated on the Earth, or even on any planet. Smaller bodies, e.g. the parent bodies of primitive meteorites, offer alternative environments for the origin of life in our Solar System. The search for past or present life on Mars is an obvious path to greater enlightenment. The subsurface oceans of some icy satellites of the outer planets represent the best locales to search for an independent origin of life in the Solar System because of the high dynamical barriers for transfer, intense radiation at their surfaces, and thick ice crusts. The ``ultimate'' answer to the abundance of life in the Cosmos will remain the domain of speculation until we develop observatories capable of detecting habitable planets - and signs of life - around the nearest million or so stars.
A Mission Simulating the Search for Life on Mars with Automated Drilling, Sample Handling, and Life Detection Instruments Performed in the Hyperarid Core of the Atacama Desert, Chile

Abstract:We report on a field demonstration of a rover-based drilling mission to search for biomolecular evidence of life in the arid core of the Atacama Desert, Chile. The KREX2 rover carried the Honeybee Robotics 1 m depth The Regolith and Ice Drill for Exploration of New Terrains (TRIDENT) drill and a robotic arm with scoop that delivered subsurface fines to three flight prototype instruments: (1) The Signs of Life Detector (SOLID), a protein and biomolecule analyzer based on fluorescence sandwich microarray immunoassay; (2) the Planetary In Situ Capillary Electrophoresis System (PISCES), an amino acid analyzer based on subcritical water extraction coupled to microchip electrophoresis analysis; and (3) a Wet Chemistry Laboratory cell to measure soluble ions using ion selective electrodes and chronopotentiometry. A California-based science team selected and directed drilling and sampling of three sites separated by hundreds of meters that included a light-toned basin area showing evidence of aqueous activity surrounded by a rocky desert pavement. Biosignatures were detected in basin samples collected at depths ranging from 20 to 80 cm but were not detected in the surrounding area. Subsurface stratigraphy of the units drilled was interpreted from drill sensor data as fine-scale layers of sand/clay sediments interspersed with layers of harder material in the basins and a uniform subsurface composed of course-to-fine sand in the surroundings. The mission timeline and number of commands sent to accomplish each activity were tracked. The deepest sample collected (80 cm) required 55 commands, including drilling and delivery to three instruments. Elapsed time required for drilling and sample handling was less than 3 hours to collect sample from 72 cm depth, including time devoted to recovery from a jammed drill. The experiment demonstrated drilling, sample transfer technologies, and instruments that accomplished successful detection of biomolecular evidence of life in one of the most biologically sparse environments on Earth.
