Standardized extract and its compounds from fruits of Piper longum suppress MDA-MB-231 cancer stem cells via down-regulation of intracellular signals

Abstract:We investigated the activities of Piper longum fruit ( PLF ) standardized extract ( PLF-SE ) and its purified components piperine ( 1 ), pipernonaline ( 2 ), and guineesine ( 3 ) against human breast cancer stem cells (CSCs). HPLC-DAD analysis showed that the concentrations of compounds 1–3 in PLF-SE were 25.04 %, 2.91 %, and 0.61 %, respectively. The breast CSC population, identified by flow cytometry using cell-surface markers, was markedly diminished by treatment with PLF-SE (20 μg/mL), piperine (2.5 μM), and guineesine (5 μM) for 48 h. Piperine and guineesine blocked spheroid formation by MDA-MB-231 cells and PLF-SE significantly decreased the number of spheres. Interestingly, the levels of the CSC-related proteins Oct4, Nanog, Sox2, EpCAM, and PARP were significantly reduced by PLF-SE and compounds 1 and 3 . A molecular docking simulation of two active compounds was carried out to support the in vitro results. PL fruits and their components (such as piperine and guineesine) inhibited breast cancer stem cells. Further, in vivo investigations should examine the potential of P. longum fruits as a novel adjunctive treatment for breast cancer.
