Abstract:Phase imaging microscopy, based either on holography or nonholographic methods such as phase retrieval, has seen considerable attention recently. Phase retrieval offers the advantage of being free of a reference arm and enables a more stable and compact setup. We present an optical setup that provides enhanced resolution by implementing synthetic aperture imaging based on phase retrieval using an electrically tunable lens (ETL). The ETL is a more compact and less expensive alternative to computerized translation stages and spatial light modulators. Before applying phase retrieval, we discuss a general calibration algorithm, which performs image registration, corrects for magnifications, and determines the axial locations of image planes. Finally, we obtain resolution-enhanced images of a phase grating and of cells to demonstrate the practical application of our technique. (C) 2015 Optical Society of America

Abstract:The strong need in materials and biological science has prompted the development of high-speed quantitative phase imaging. However, for phase retrieval applying digital micromirror devices (DMDs), the accuracy of the retrieved phase will be disturbed by the DMD-induced aberrations. Here, we propose a phase retrieval method based on measuring and correcting errors caused by phase non-uniformity of the device. Using only four binary amplitude masks and corresponding diffraction intensities, the proposed method achieves rapid convergence and high-quality reconstruction. The experiments prove the practical feasibility for general samples and the effective improvement of the retrieved phase accuracy.
One-Shot In-Line Digital Holography Based Hilbert Phase-Shifting

Abstract:A novel one-shot in-line digital holography based on Hilbert phase-shifting is proposed. By weakening the ratio of object wave to reference wave and applying natural logarithmized operation on the in-line digital hologram, the real part of object wave can be well extracted. Then utilizing Hilbert transform to digitally realize pi/2-phase shift, which would make it possible to reconstruct the object wavefront from a single-exposure in-line digital hologram. Preliminary experimental results are presented to confirm the proposed method. This technique can be used for real time imaging or monitoring moving objects.

Abstract:A new optical microscopy technique, termed high spatial and temporal resolution synthetic aperture phase microscopy (HISTR-SAPM), is proposed to improve the lateral resolution of wide-field coherent imaging. Under plane wave illumination, the resolution is increased by twofold to around 260 nm, while achieving millisecond-level temporal resolution. In HISTR-SAPM, digital micromirror devices are used to actively change the sample illumination beam angle at high speed with high stability. An off-axis interferometer is used to measure the sample scattered complex fields, which are then processed to reconstruct high-resolution phase images. Using HISTR-SAPM, we are able to map the height profiles of subwavelength photonic structures and resolve the period structures that have 198 nm linewidth and 132 nm gap (i.e., a full pitch of 330 nm). As the reconstruction averages out laser speckle noise while maintaining high temporal resolution, HISTR-SAPM further enables imaging and quantification of nanoscale dynamics of live cells, such as red blood cell membrane fluctuations and subcellular structure dynamics within nucleated cells. We envision that HISTR-SAPM will broadly benefit research in material science and biology.

Abstract:A single intensity-only holographic interferogram can records the full amplitude and phase information of optical field. However, current digital holography technologies cannot recover the lossless phase information from a single interferogram. In this paper, we provide an entirely new approach for the full-field quantitative phase imaging technology. We demonstrate that deep learning can be used to replace the entitative phase shifter, and quantitative phase imaging can obtain quantitative phase from a single interferogram in in-line holography. A deep-phase-shift network (DPS-net) is reported, which can be trained with simulation training data. The trained DPS-net can be used to generate multiple interferograms with arbitrary phase shift from a single interferogram as an artificial intelligence phase shifter. The ability and the accuracy of generating arbitrary phase shifts are verified, and the performance of the proposed method is also verified by the experimental interferogram. The results demonstrate that the proposed method can provide a full digital phase shifter with high-accuracy for the technology of dynamic quantitative phase measurement.

Abstract:Holography provides access to the optical phase. The emerging compressive phase retrieval approach can achieve in-line holographic imaging beyond the information-theoretic limit or even from a single shot by exploring the signal priors. However, iterative projection methods based on physical knowledge of the wavefield suffer from poor imaging quality, whereas the regularization techniques sacrifice robustness for fidelity. In this work, we present a unified compressive phase retrieval framework for in-line holography that encapsulates the unique advantages of both physical constraints and sparsity priors. In particular, a constrained complex total variation (CCTV) regularizer is introduced that explores the well-known absorption and support constraints together with sparsity in the gradient domain, enabling practical high-quality in-line holographic imaging from a single intensity image. We developed efficient solvers based on the proximal gradient method for the non-smooth regularized inverse problem and the corresponding denoising subproblem. Theoretical analyses further guarantee the convergence of the algorithms with prespecified parameters, obviating the need for manual parameter tuning. As both simulated and optical experiments demonstrate, the proposed CCTV model can characterize complex natural scenes while utilizing physically tractable constraints for quality enhancement. This new compressive phase retrieval approach can be extended, with minor adjustments, to various imaging configurations, sparsifying operators, and physical knowledge. It may cast new light on both theoretical and empirical studies.

Abstract:We present a single-pixel imaging technique that enables phase extraction from objects by complex Fourier spectrum sampling. The technique exploits a digital micromirror device to scan a wavevector-varying plane wave, which interferes with a stationary reference beam to produce time-varying spatial frequencies on the object. Synchronized intensity measurements are made using a single-pixel detector, and four-step phase-shifting is adopted in spectrum acquisition. Applying inverse Fourier transform to the obtained spectrum yields the desired image. The proposed technique is demonstrated by imaging two digital phase objects. Furthermore, we show that the image can be reconstructed from sub-Nyquist measurements via compressive sensing, considerably accelerating the acquisition process. As a particular application, we use the technique to characterize the orbital angular momentum of vortex beams, which could benefit multiplexing techniques in classical and quantum communications. This technique is readily integrated into commercial microscopes for quantitative phase microscopy. Published under license by AIP Publishing.

Abstract:Quantitative phase imaging has been an important labeling-free microscopy modality for many biomedical and material science applications. In which, ultra-fast quantitative phase imaging is indispensable for dynamic or transient characteristics analysis. Conventional wide field optical interferometry is a common scheme for quantitative phase imaging, while its data acquisition rate is usually hindered by the frame rate of arrayed detector. By utilizing novel balanced-photo-detector based digital optics coherent detection techniques, we report on a method of constructing ultra-fast quantitative phase microscopy at the line-scan rate of 100 MHz with ~2 μm spatial resolution.

Abstract:Quantitative phase imaging (QPI) techniques are widely used for the label-free examining of transparent biological samples. QPI techniques can be broadly classified into interference-based and interferenceless methods. The interferometric methods which record the complex amplitude are usually bulky with many optical components and use coherent illumination. The interferenceless approaches which need only the intensity distribution and works using phase retrieval algorithms have gained attention as they require lesser resources, cost, space and can work with incoherent illumination. With rapid developments in computational optical techniques and deep learning, QPI has reached new levels of applications. In this tutorial, we discuss one of the basic optical configurations of a lensless QPI technique based on the phase-retrieval algorithm. Simulative studies on QPI of thin, thick, and greyscale phase objects with assistive pseudo-codes and computational codes in Octave is provided. Binary phase samples with positive and negative resist profiles were fabricated using lithography, and a single plane and two plane phase objects were constructed. Light diffracted from a point object is modulated by phase samples and the corresponding intensity patterns are recorded. The phase retrieval approach is applied for 2D and 3D phase reconstructions. Commented codes in Octave for image acquisition and automation using a web camera in an open source operating system are provided.

Abstract:Digital holography has been widely applied in quantitative phase imaging (QPI) for monolayer objects within a limited depth. For multilayer imaging, compressive sensing is employed to eliminate defocused images but with missing phase information. A phase iteratively enhanced compressive sensing (PIE-CS) algorithm is proposed to achieve phase imaging and eliminate defocused images simultaneously. Linear filtering is first applied to the off-axis hologram in Fourier domain, and an intermediate reconstructed complex image is obtained. A periodic phase mask is then superimposed on the intermediate reconstructed image to iteratively eliminate the defocused images and recover the object with phase information. The experimental recovery of amplitude and phase of a two-layer sample with as little as 7% random measurement is demonstrated. The average phase error of the PIE-CS algorithm is analyzed, and the results show the feasibility for QPI.

Abstract:A different x-ray phase contrast imaging technique based on the combination of structured illumination and an optimized hybrid input-output algorithm for phase and amplitude retrieval is presented and discussed. Based on a modified and flexible experimental setup, compared to standard propagation-based x-ray imaging setups, the method we propose here represents a real advance in the phase-contrast imaging technique relating to the determination of the phase and amplitude distribution. Moreover, in coherent diffractive imaging applications, the proposed technique may yield high spatial resolution with currently available imaging detectors.

Abstract:Quantitative phase imaging and measurement of surface topography and fluid dynamics for objects, especially for moving objects, is critical in various fields. Although effective, existing synchronous phase-shifting methods may introduce additional phase changes in the light field due to differences in optical paths or need specific optics to implement synchronous phase-shifting, such as the beamsplitter with additional anti-reflective coating and a micro-polarizer array. Therefore, we propose a synchronous phase-shifting method based on the Mach–Zehnder interferometer to tackle these issues in existing methods. The proposed method uses common optics to simultaneously acquire four phase-shifted digital holograms with equal optical paths for object and reference waves. Therefore, it can be used to reconstruct the phase distribution of static and dynamic objects with high precision and high resolution. In the experiment, the theoretical resolution of the proposed system was 1.064 μm while the actual resolution could achieve 1.381 μm, which was confirmed by measuring a phase-only resolution chart. Besides, the dynamic phase imaging of a moving standard object was completed to verify the proposed system's effectiveness. The experimental results show that our proposed method is suitable and promising in dynamic phase imaging and measurement of moving objects using phase-shifting digital holography.

Abstract:Multi-wavelength phase retrieval provides a competitive solution to lensless holographic imaging that features a low-cost, compact design and high data acquisition speed. However, the existence of phase wraps poses a unique challenge for iterative reconstruction, and the resulting algorithms often suffer from limited generalizability and increased computational complexity. Here, we propose a projected refractive index framework for multi-wavelength phase retrieval that directly recovers the amplitude and unwrapped phase of the object. General assumptions are linearized and integrated into the forward model. Based on an inverse problem formulation, physical constraints and sparsity priors are incorporated, which ensures imaging quality under noisy measurements. We experimentally demonstrate high-quality quantitative phase imaging on a lensless on-chip holographic imaging system using three color LEDs.

Abstract:Aberrations in the optical components and misalignments in the optical system cause a background phase in the coherent illumination. To reconstruct the object phase, the background phase illuminating the object must be measured and subtracted. For diffraction imaging and in-line holography, the traditional phase retrieval method reconstructs the phase diffracting from clear edges. However, it falls into stagnation when solving a background phase slowly varying in the spatial domain. In this study, we propose to solve the background phase using a modulation-based phase retrieval method. Alternative structured phase modulation (ASPM) can be the phase constraint to avoid stagnation when solving the background phase without clear edges. With ASPM, the background phase in the experiment can be efficiently retrieved when 16 phase patterns are employed. The ASPM acts as a phase grating to concentrate the intensities and provides robustness to noise. Compared to the conventional random phase modulations, the ASPM method had a smaller error value in the reconstruction iterations, which leads to a better reconstruction quality. After measuring and subtracting the background phase, the object phase was retrieved using a coherent diffraction imaging system. A phase plate can be accurately reconstructed under three different background phases.

Abstract:Incoherent digital holography is a promising solution for acquiring three-dimensional information in the form of interference without a laser. Self-interference is the principal phenomenon of incoherent holography which splits the incident light wave into two waves and interferes with each one. The characteristics of incoherent holograms are defined by the optical configuration of wavefront dividing units. In this paper, we propose the synthetic aperture method to achieve super-resolution of geometric phase lens-based self-interference incoherent digital holography. We analyze the resolution of incoherent holograms acquired from geometric phase lenses and experimentally demonstrate the resolution enhancement via the synthetic-aperture method. Moreover, the sparse synthetic-aperture method is proposed to ensure effective data capturing and the numerical optimization method is also proposed to fill the gap between the empty space of sparsely acquired holograms.

Abstract:Complex field imaging, which captures both the amplitude and phase information of input optical fields or objects, can offer rich structural insights into samples, such as their absorption and refractive index distributions. However, conventional image sensors are intensity-based and inherently lack the capability to directly measure the phase distribution of a field. This limitation can be overcome using interferometric or holographic methods, often supplemented by iterative phase retrieval algorithms, leading to a considerable increase in hardware complexity and computational demand. Here, we present a complex field imager design that enables snapshot imaging of both the amplitude and quantitative phase information of input fields using an intensity-based sensor array without any digital processing. Our design utilizes successive deep learning-optimized diffractive surfaces that are structured to collectively modulate the input complex field, forming two independent imaging channels that perform amplitude-to-amplitude and phase-to-intensity transformations between the input and output planes within a compact optical design, axially spanning ~100 wavelengths. The intensity distributions of the output fields at these two channels on the sensor plane directly correspond to the amplitude and quantitative phase profiles of the input complex field, eliminating the need for any digital image reconstruction algorithms. We experimentally validated the efficacy of our complex field diffractive imager designs through 3D-printed prototypes operating at the terahertz spectrum, with the output amplitude and phase channel images closely aligning with our numerical simulations. We envision that this complex field imager will have various applications in security, biomedical imaging, sensing and material science, among others.

Abstract:Quantitative phase imaging needs of a holographic setup in order to retrieve the sample phase distribution. Often this new phase information is obtained at the cost of reducing the useful range in other parameters of the imaging system such as field of view or resolution. We devised an experimental setup that allows complex amplitude evaluation and phase image quantification of three-dimensional (3-D) samples in wide-field digital holographic microscopy. The basis is a common-path interferometric configuration that can be implemented in two configurations depending on the position and the basic period of the grating: static grating and windowed Fourier filtering method or with phase-shifting moving grating. Both configurations allow complex amplitude recovery and, thus, quantitative phase imaging of the sample under test. Moreover, by combining the basic setup with tilted illumination and time multiplexing, it is possible to generate a synthetic aperture enlargement that improves the final image resolution. Experimental results validated our concepts.
