Constructing NO/CO gases-releasing bioactive coating to synergistically enhance corrosion resistance and biocompatibility of magnesium alloy for cardiovascular stents

Abstract:Medical magnesium-based alloys have great potential in the development of new-generation biodegradable intravascular stents, however, the inferior in vivo corrosion resistance and poor biocompatibility usually cause early loss of mechanical properties, as well as complications such as thrombosis and excessive intimal hyperplasia, severely retarding their clinical applications. To address these issues, we constructed a bioactive coating on the magnesium-based alloy surface that can augment anticorrosion properties and improve biocompatibility through the released NO and CO on the surface. This coating could significantly enhance the surface hydrophilicity of magnesium alloys and endow them with the ability to promote albumin adsorption selectively. The results of electrochemical corrosion and degradation behaviors showed that the bioactive coating could significantly improve the thermodynamic stability of the pristine magnesium alloy and reduce the corrosion rate, thereby significantly enhancing the anticorrosion. Due to the excellent corrosion resistance and surface bioactivities, as well as the introduction of bioactive substances (including heparin, chitosan, etc.), this coating could significantly heighten the hemocompatibility and promote endothelial cell (EC) growth on the surface. Meanwhile, our results demonstrated that under the circumstance of the catalytic release of NO and CO, the anticoagulant and EC growth were significantly enhanced compared to the uncatalyzed state. With the increase of CO-releasing molecules in the coating, the biocompatibility was improved significantly, proving that the released CO can synergistically enhance the biocompatibility with NO. Therefore, the strategy of preparing bioactive coatings that can release NO/CO gases in the present study lays the foundation for the development of magnesium alloy stents with synergistic improvement of biocompatibility.
