Abstract:
Liposomal honokiol isolated from the genus Magnolia has been found to have antiangiogenic, anti-inflammatory and antitumor properties. However, there has no report on its role in hair growth. Hair follicles are life-long cycled organelles that go through from anagen, catagen and telogen stages and are regulated by diverse signaling pathways, including Wnt/β-catenin, Notch, Epidermal growth factor (EGF) and Sonic hegehog (SHH). Wnt signals are essential for the initiation of hair follicle placode development and a new potential target of hair loss treatment. This study was designed to investigate the effect of liposomal honokiol (Lip-honokiol) on inducing hair anagen. We identified the hair grew out in advance in the shaving area of C57BL/6N mice after the treatment of liposomal honokiol (Lip-honokiol) by daily abdominal injection. We first demonstrated that Lip-Honokiol activated the Wnt3a/β-catenin pathway and downregulated the transforming growth factor-β1 (TGF-β1) to promote hair growth in mice via immunohistochemistry and immunofluorescence staining. These findings suggest that Lip-honokiol activated the Wnt/β-catenin pathway and accelerated the transfer from the telogen to anagen stage and finally promoted the hair growth.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to explore the role and mechanism of Liposomal honokiol (Lip - honokiol) in promoting hair growth. Specifically, the researchers want to know whether Lip - honokiol can promote the transition of mouse hair follicles from the telogen phase to the anagen phase by activating the Wnt3a/β - catenin signaling pathway and down - regulating transforming growth factor - β1 (TGF - β1), thereby promoting hair growth. This research not only aims to reveal the specific effects of Lip - honokiol on hair growth, but also attempts to elucidate the underlying molecular mechanisms, providing new potential drug targets for the treatment of alopecia. ### Research Background and Problem The hair follicle is a life - long cycling organ, undergoing three phases: anagen, catagen, and telogen, and is regulated by multiple signaling pathways, including Wnt/β - catenin, Notch, epidermal growth factor (EGF), and Sonic hedgehog (SHH). The Wnt signal is crucial in the initial stage of hair follicle placode development and is a new potential target for the treatment of alopecia. However, there have been no previous reports on the role of Lip - honokiol in hair growth. ### Research Purpose This study aims to investigate the effect of Lip - honokiol on inducing hair follicles to enter the anagen phase. Through experiments, the researchers hope to verify whether Lip - honokiol can promote hair growth in the following ways: - Activating the Wnt3a/β - catenin signaling pathway - Down - regulating the expression of TGF - β1 These findings will help to understand the mechanism of Lip - honokiol in hair growth and provide a scientific basis for the development of new drugs for alopecia.
