Abstract:Aza-BODIPY,developed from BODIPYs,have advantages of large molar absorption coefficients,narrow half peak widths,long fluorescence life,good optical stability and tunable fluorescence;hence they have been selected as excellent candidates for near infrared colorimetric and fluorescent probes. However,most of them are poor water solubility and the field of their application is limited. In this paper,the recent researches on Aza-BODIPY-based fluorescent probes were reviewed. Three methods of synthesis of Aza-BODIPYs are summarized briefly including the O'Shea method starting from chalcone,the Carreira method using 2,4-substituted pyrroles as starting materials and the Lukyanets method for fused-ring-Aza-BODIPYs. The sensing performances,mechanisms and practical applications of various probes for pH,H2O2,NH4+,F–,Hg2+,CN–,damtoxin and Cys are discussed. Although there are limited reports about Aza-BODIPY-based probes,they are expected to be developed in the biological and environment areas with the improvement of water solubility,the longer excitation and emission wavelengths in the near-infrared region and the increase of the type and range of identifying species by structural modifications.

Abstract:The development of new techniques to rapidly and accurately detect bacteria has drawn continuous attention due to the potential threats posed by bacteria to human health and safety. Recently, a novel strategy based on fluorescent probes has drawn considerable interest for the detection of bacteria due to its high selectivity, fast response, and simple operation. In this review, we summarize the recent progress on fluorescent probes for the specific recognition and discrimination of Gram-negative and Gram-positive bacteria. In particular, we outline current design strategies, such as targeting of the differences in surface components, cell wall components, endogenous enzymes, surface charge, and hydrophobicity of various kinds of bacteria to develop various fluorescent sensors (organic small-molecule fluorescent probes, nanoprobes, and metal ion probes). We also emphasize the application of organic molecules in probe recognition elements. We hope that this review can stimulate this research area in bacterial detection and imaging in the future.

Abstract:Fluorescent imaging is a multidisciplinary integrated technology for bio-imaging. Application of fluorescent probes allows dynamic visualization of large bio-molecules inside cells and animals, which can benefit the diagnosis and treatment of various diseases.Satisfactory fluorophores and bio-orthogonal reactions play crucial roles in the construction of ideal fluorescent probes for in vivo imaging of cellular biopolymers and biological events. This review summarized the recent advance in these two areas with examples illustrating the applications in fluorescent bio-imaging.

Abstract:Bacterial infection is one of the leading causes of death worldwide and is easy to cause large-scale diseases. It is an urgent need to develop effective methods for the specific detection and treatment of bacterial infections. Recently, small molecular fluorescent probes, bridging the capability of imaging detection and sterilization, have attracted increasing attention. Fluorescence imaging assays have the benefit of being simple, specific, and fast, which is very advantageous in both in vitro and in vivo bacterial detection. Molecularly fluorophores for theranostics provide advantages of non-invasion, high specificity, and fewer side effects. In this review, we summarize the recent advances and design strategies of small molecular fluorescent probes for both targeted detection and treatment of bacteria. We hope that this review will provide guidance for the development of more effective fluorescent dyes in the future as well as encourage preclinical and clinical studies of phototherapy mediated antimicrobial therapy.

Abstract:Bacterial infections remain a global healthcare problem that is particularly attributed to the spread of antibiotic resistance and the evolving pathogenicity. Accurate and swift approaches for infection diagnosis are urgently needed to facilitate antibiotic stewardship and effective medical treatment. Direct optical imaging for specific bacterial labeling and infection detection offers an attractive prospect of precisely monitoring the infectious disease status and therapeutic response in real time. This feature article focuses on the recent advances of small-molecule probes developed for fluorescent imaging of bacteria and infection, which covers the probe design, responsive mechanisms and representative applications. In addition, the perspective and challenges to advance small-molecule fluorescent probes in the field of rapid drug-resistant bacterial detection and clinical diagnosis of bacterial infections are discussed. We envision that the continuous advancement and clinical translations of such a technique will have a strong impact on future anti-infective medicine.

Abstract:In recent years, with the rise of precision therapy and other frontier fields, the demand of the high quality fluorescent probes with extremely low toxicity, good biocompatibility, strong stability and high brightness is more urgent than ever. Proteins that with abundant active groups and special pockets can bind with different probes to alter the fluoresce properties, and some can even greatly enhance the fluorescence signals. Moreover, some proteins can simultaneously bind with two different molecules to fabricate fluorescence resonance energy transfer (FRET) probes. Recent progress witnessed that the protein-mediated strategies have provided a promising way for enhancing the performance of fluorescent probe. This article will review the up-to-date progress of protein-mediated fluorescent probes. We firstly provide a systemically on the principle of protein fluorescent probes fabrication and the interaction mechanisms between proteins and molecular dyes. Then the bioapplications of these fluorescent probes in bioimaging and biosensing were reviewed. And finally the current challenges and perspectives were discussed in-depth. We hope that this review can provide some new ideas and inspiration for relevant researchers to develop a new generation of fluorescent probes.

Abstract:Bacterial infections have become a global public health problem. Rapid and sensitive bacterial detection is of great importance for human health. Among various sensor systems, fluorescence sensor is rapid, portable, multiplexed, and cost-efficient. Herein, we reviewed the current trends of fluorescent sensors for bacterial detection from three aspects (response materials, target and recognition way). The fluorescent materials have the advantages of high fluorescent strength, high stability, and good biocompatibility. They provide a new path for bacterial detection. Several recent fluorescent nanomaterials for bacterial detection, including semiconductor quantum dots (QDs), carbon dots (CDs), up-conversion nanoparticles (UCNPs) and metal organic frameworks (MOFs), were introduced. Their optical properties and detection mechanisms were analyzed and compared. For different response targets in the detection process, we studied the fluorescence strategy using DNA, bacteria, and metabolites as the response target. In addition, we classified the recognition way between nanomaterial and target, including specific recognition methods based on aptamers, antibodies, bacteriophages, and non-specific recognition methods based on biological functional materials. The characteristics of different recognition methods were summarized. Finally, the weaknesses and future development of bacterial fluorescence sensor were discussed. This review provides new insights into the application of fluorescent sensing systems as an important tool for bacterial detection.

Abstract:Fluorescent probes have emerged as indispensable chemical tools to the field of chemical biology and medicine. The ability to detect intracellular species and monitor physiological processes has not only advanced our knowledge in biology but has provided new approaches towards disease diagnosis. In this review, we detail the design criteria and strategies for some recently reported fluorescent probes that can detect a wide range of biologically important species in cells and in vivo. In doing so, we highlight the importance of each biological species and their role in biological systems and for disease progression. We then discuss the current problems and challenges of existing technologies and provide our perspective on the future directions of the research area. Overall, we hope this review will provide inspiration for researchers and prove as useful guide for the development of the next generation of fluorescent probes.

Abstract:Bacterial infections remain as a prevailing situation in our today’s society. Owing to the evolving pathogenicity of bacteria, whereby more drug-resistant bacteria has been emerging and bacteria developing their own defense mechanism, traditional antimicrobial approaches like antibiotics treatment have been clinically ineffective. Early and accurate strategies are off dire need to subvert the rapid colonization of bacteria. Hence, the employment of fluorescence imaging for specific bacterial labeling and infection diagnosis could be prospective in providing physiological information in real time for the diagnosis and therapeutic intervention of bacterial infections. Small-molecule fluorescence probes have been an attractive tool in fluorescence imaging due to their inherent properties such as highly tunable emissions, high quantum yield and good biocompatibility. Among the numerous types of small-molecule fluorescence probes, the enzymatic responsive probes have been one that researchers are particularly interested in as such probes were reported to possess great specificity and high signal to noise ratio. Furthermore, bacterial enzymes are found to be closely related to the invasion of host immune system, which makes them a viable target to establish enzyme-activated fluorescent probes for specific bacterial imaging. Abstract:Near-infrared (NIR) fluorescent dyes have emerged as promising modalities for monitoring the levels of various biologically relevant species in cells and organisms. The use of NIR probes enables deep photon penetration in tissue, minimizes photo-damage to biological samples, and produces low background auto-fluorescence from biomolecules present in living systems. The number of new analyte-responsive NIR fluorescent probes has increased substantially in recent years as a consequence of intense research efforts. In this tutorial review, we highlight recent advances (2010-2013) made in the development and applications of NIR fluorescent probes. The review focuses on NIR fluorescent probes that have been devised to sense various biologically important species, including ROS/RNS, metal ions, anions, enzymes and other related species, as well as intracellular pH changes. The basic principles involved in the design of functional NIR fluorescent probes and suggestions about how to expand applications of NIR imaging agents are also described.

Abstract:Fluorescent nanoprobes show similar fluorescence properties to traditional organic dyes, but the addition of nanotechnology accurately controls the size, shape, chemical composition, and surface chemistry of the nanoprobes with unique characteristics and properties, such as bright luminescence, high photostability, and strong biocompatibility. For example, modifying aptamers or antibodies on a fluorescent nanoprobe provides high selectivity and specificity for different objects to be tested. Fluorescence intensity, life, and other parameters of targets can be changed by different sensing mechanisms based on the unique structural and optical characteristics of fluorescent nanoprobes. What's more, the detection of fluorescent nanoprobes is cost-saving, simple, and offers great advantages in rapid food detection. Sensing mechanisms of fluorescent nanoprobes were introduced in this paper, focusing on the application progress in pesticide residues, veterinary drug residues, heavy metals, microbes, mycotoxins, and other substances in food safety detection in recent years. A brief outlook for future development was provided as well.

Antibiotics-based Fluorescent Probes for Selective Labeling of Gram-negative and Gram-positive Bacteria in Living Microbiotas

Abstract:Gram-staining distinguishes bacteria into two major groups,Gram-positive and Gram-negative bacteria,and has become an essential technique in microbiology.However,Gram-staining is not compatible with living cells and thus limits its applications.Here,we report the development of a polymyxin B-based fluorescent probe that enables selective labeling of Gram-negative in the living microbiota samples.We first synthesized the polymyxin B-Cy3 conjugate and confirmed its specificity for labeling Gram-negative bacteria.In combination with a previously developed Gram-positive-specific fluorescent probe,we demonstrate two-color imaging of Gram-positive and Gram-negative bacteria in various kinds living microbiotas,including mouse gut,human oral,soil,and crude oil microbiotas,with high selectivity and coverage.Finally,a pilot use of the probes in staining bacteria on heat-fixed sputum smear was also demonstrated,showing its potentials in clinical microbiology.Our method provides a versatile tool for distinguishing Gram-positive and Gram-negative bacteria in both basic research and clinical settings.

Labels most often have inherent fluorescence regardless of their being flee, nonspecifically bound, or bound to their cellular target.
