p53 deficiency induces MTHFD2 transcription to promote cell proliferation and restrain DNA damage

Abstract:Significance Cancer cells acquire distinct metabolic adaptations for survival. However, how metabolic remodeling enables cancer cells to survive genomic instability stress remains poorly understood. This report shows that p53 loss or mutation reprograms folate metabolism through MTHFD2. Moreover, MTHFD2 not only influences reactive oxygen species but also unexpectedly participates in nonhomologous end joining (NHEJ) in response to DNA damage. Down-regulation of MTHFD2 preferably induces apoptosis and cell proliferative arrest in p53-deficient cells but protects p53 wild-type cells by inducing the AICAR-mediated AMPK-p53-p21 activation. Collectively, it reveals both metabolism-dependent and -independent functions of MTHFD2 in controlling p53-dependent cell survival and uncovers a new role for MTHFD2 in NHEJ. MTHFD2 depletion may be a promising therapeutic strategy for the treatment of p53-deficient tumors.
