Abstract:The palladium/isocyanide catalyst is able to facilitate the intermolecular σ-bond exchange between benzocyclobutenones and silacyclobutanes. This reaction cleaves the C–C bond of the benzocyclobutenones and the C–Si bond of the silacyclobutanes, providing a unprecedented access to the eight-membered silacycles with remarkable chemo- and regioselectivities. We studied the mechanism and origins of the chemo- and regioselectivities with density functional theory (DFT) calculations. The reaction proceeds via two sequential oxidative additions (first with benzocyclobutenone and second with silacyclobutane) and two subsequent reductive eliminations (first, C–Si bond formation; second, C–C bond formation). Abstract:Transition-metal-catalyzed C-O bond activation provides a useful strategy for utilizing alcohol- and phenol-derived electrophiles in cross-coupling reactions, which has become a research field of active and growing interest in organic chemistry. The synergy between computation and experiment elucidated the mechanistic model and controlling factors of selectivities in these transformations, leading to advances in innovative C-O bond activation and functionalization methods.Toward the rational design of C-O bond activation, our collaborations with the Jarvo group bridged the mechanistic models of C(sp2)-O and C(sp3)-O bond activations. We found that the nickel catalyst cleaves the benzylic and allylic C(sp3)-O bonds via two general mechanisms: the stereoinvertive SN2 back-side attack model and the stereoretentive chelation-assisted model. These two models control the stereochemistry in a wide array of stereospecific Ni-catalyzed cross-coupling reactions with benzylic or allylic alcohol derivatives. Because of the catalyst distortion, the ligands can differentiate the competing stereospecific C(sp3)-O bond activations. The PCy3 ligand interacts with nickel mainly through σ-donation, and the Ni(PCy3) catalyst can undergo facile bending of the substrate-nickel-ligand angle, which favors the stereoretentive benzylic C-O bond activation. The N-heterocyclic carbene SIMes ligand has additional d(metal)-p(ligand) back-donation with nickel, which leads to an extra energy penalty for the same angle bending. This results in the preference of stereoinvertive benzylic C-O bond activation under Ni/SIMes catalysis. In addition to ligand control, a Lewis acid can increase the selectivity for stereoinvertive C(sp3)-O activation by stabilizing the SN2 back-side attack transition state. The oxygen leaving group complexes with the MgI2 Lewis acid in the stereoinvertive activation, leading to the exclusive stereoinvertive Kumada coupling of benzylic ethers. We also identified that the competing C(sp3)-O bond activation models have noticeable differences in charge separation. This leads to the solvent polarity control of the stereospecificity in C(sp3)-O activations. Low-polarity solvents favor the neutral stereoretentive C-O bond activation, while high-polarity solvents favor the zwitterionic stereoinvertive cleavage.In sharp contrast to the nickel catalysts, the C(sp2)-O bond activation under palladium catalysis mainly proceeds via the classic three-membered ring oxidative addition mechanism instead of the chelation-assisted mechanism. This is due to the lower oxophilicity of palladium, which disfavors the oxygen coordination in the chelation-assisted-type activation. The three-membered ring activation model selectively cleaves the weak C-O bond, resulting in the exclusive chemoselectivity of acyl C-O bond activation in Pd-catalyzed cross-coupling reactions with aryl carboxylic acid derivatives. This explains the overall acylation in the Pd-catalyzed Suzuki-Miyaura coupling with aryl esters. In collaboration with the Szostak group, we revealed that the three-membered ring model applies in the Pd-catalyzed C-O bond activation of carboxylic acid anhydride, which stimulated the development of a series of Pd-catalyzed decarbonylative functionalizations of aryl carboxylic acids.

Abstract:We discovered a cooperative gold/silver catalysis mechanism in the oxidative cross-coupling reaction between 1,2,4,5-tetrafluorobenzene and N-TIPS-indole, using DFT calculations. A silver(I)-catalyzed CMD mechanism is responsible for the C-H activation of 1,2,4,5-tetrafluorobenzene, and C-H acidity determines the chemoselectivity. A gold(III)-catalyzed S(E)2Ar mechanism is responsible for the C3-H activation of N-TIPS-indole, and arene nucleophilicity determines the chemo- and regioselectivity. The orthogonal chemoselectivity control provides a mechanistic guide for dual C-H activation reactions.

Abstract:The mechanism and origins of selectivities in [Ni(NHC)]-catalyzed intramolecular (5 + 2) cycloadditions and homo-ene reactions of vinylcyclopropanes (VCPs) and alkynes have been studied using density functional theory. The preferred mechanism involves oxidative alkyne-alkene cyclization to form a metallacyclopentene intermediate, in contrast to a cyclopropane cleavage pathway in the reaction with Rh(I) catalysts. The selectivity between the (5 + 2) and homo-ene products is determined in the subsequent competing reductive elimination and β-hydride elimination steps. Two similar-sized N-heterocyclic carbene (NHC) ligands, SIPr and ItBu, yielded reversed product selectivity, favoring the (5 + 2) and homo-ene products respectively. This is attributed to the anisotropic steric environment of these NHC ligands, which positions the bulky substituents on the ligand toward different directions and leads to distinct steric control in the reductive elimination and β-hydride elimination transition states.

Abstract:CONSPECTUS: With new advances in theoretical methods and increased computational power, applications of computational chemistry are becoming practical and routine in many fields of chemistry. In organic chemistry, computational chemistry plays an indispensable role in elucidating reaction mechanisms and the origins of various selectivities, such as chemo-, regio-, and stereoselectivities. Consequently, mechanistic understanding improves synthesis and assists in the rational design of new catalysts. In this Account, we present some of our recent works to illustrate how computational chemistry provides new mechanistic insights for improvement of the selectivities of several organic reactions. These examples include not only explanations for the existing experimental observations, but also predictions which were subsequently verified experimentally. This Account consists of three sections discuss three different kinds of selectivities. The first section discusses the regio- and stereoselectivities of hydrosilylations of alkynes, mainly catalyzed [Cp*Ru(MeCN)(3)](+) or [CpRu(MeCN)(3)](+). Calculations suggest a new mechanism that involves a key ruthenacyclopropene intermediate.This mechanism not only explains the unusual Markovnikov regio-selectivity and anti-addition stereoselectivity observed by Trost and co-workers, but also motivated further experimental investigations. New intriguing experimental observations and further theoretical studies led to an extension of the reaction mechanism. The second section includes three cases of meta-selective C-H activation of aryl compounds. In the case of Cu-catalyzed selective meta-C-H activation of aniline, a new mechanism that involves a Cu(III)-Ar-mediated Heck-like transition state, in which the Ar group acts as an electrophile, was proposed. This mechanism predicted a higher reactivity for more electron-deficient Ar groups, which was supported by experiments. For two template-mediated, meta-selective C-H bond activations catalyzed by Pd(II), different mechanisms were derived for the two templates. One involves a dimeric Pd-Pd or Pd-Ag active catalyst, and the other involves a monomeric Pd catalyst, in which a monoprotected amino acid coordinates in a bidentate fashion and serves as an internal base for C-H activation. The third section discusses a desymmetry strategy in asymmetric synthesis. The construction of rigid skeletons is critical for these catalysts to distinguish two prochiral groups. Overall, fruitful collaborations between computational and experimental chemists have provided new and comprehensive mechanistic understanding and insights into these useful reactions.

Abstract:The three-component coupling of isoprene, an alkenyl triflate, and styrenylboronic acid catalyzed by a palladium pyrox complex affords access to skipped dienes from simple chemical feedstocks. Unfortunately, the transformation proceeds with only moderate selectivity and yields. The reaction mechanism and factors responsible for the resulting regioselectivity were elucidated using M06/SDD/6-311++G(d,p) + SMD calculations. Distortion of the palladium coordination sphere in the transition structure of the migratory insertion step is found to control the 4,1- vs 1,x-selectivity. The calculated ΔΔG⧧ of 1.0 kcal/mol for this step is in excellent agreement with the experimentally observed selectivity of 1:9.9 disfavoring the 4,1-product. The transmetalation was found to be the regioselectivity determining step for the formation of the 1,2- vs 1,4-addition products. Systematic conformational searches for the transmetalation transition structure revealed a series of steric interactions between the t-Bu substituent on the ligand and the substrates in the model system that are balanced by additional repulsive interactions between the substrates and the pyridyl portion of the ligand. The combination of these effects leads to the low to moderate 1,2- vs 1,4-selectivity in the experimentally studied system.

Abstract:Experiments indicate that a catalytic amount of CuI plays an important role in the siloxane-mediated Pd-catalyzed cross-coupling reactions with the direct use of organolithium reagents. Addition of organolithium to the siloxane transfer agent generates an organosilicon intermediate. DFT calculations indicate that CuI initially accelerates the Si-Pd(II) transmetalation of the organosilicon intermediate by the formation of CuI2−. Subsequently, CuI2− works as a shuttle between the Si-Cu(I) and Cu(I)-Pd(II) transmetalation processes.

Abstract:The catalytic construction of silicon-carbon bond is one of the most important and key processes in organosilicon chemistry and remains an ongoing challenge in homogeneous catalysis. Herein we report an unprecedent and convenient palladium-catalyzed Si-C(sp3) bond-forming cross-coupling silylation of benzyl halides with structurally diverse hydrosilanes, which provides a direct and chemoselective approach to the facile synthesis of various silanes and their derivatives with moderate to good yields. The phosphine-controlled and B(C6F5)-assisted Pd-catalyzed C(sp3)-Si cross-coupling silylation exhibits good functional group tolerances and demonstrates a powerful potential of silicon-based cross coupling reaction for the preparation of structurally diverse benzyl silanes. This protocol also features simple reaction conditions, wide substrate scope, high chemoselectivity, and environmentally benign, offering a new pathway for the synthesis of tri-or tertiary-substituted functional organosilicon compounds.

Abstract:Direct coupling of Si H bonds has emerged as a promising strategy for designing chemically and biologically useful organosilicon compounds. Heterogeneous catalytic systems sufficiently active, selective, and durable for dehydrosilylation reactions under mild conditions have been lacking to date. Herein, we report that the hydrophobic characteristics of the underlying supports can be advantageously utilized to enhance the efficiency of palladium nanoparticles (Pd NPs) for the dehydrogenative coupling of organosilanes. As a result of this prominent surface wettability control, the modulated catalyst showed a significantly higher level of efficiency and durability characteristics toward the dehydrogenative condensation of organosilanes with water, alcohols, or amines in comparison to existing catalysts. In a broader context, this work illustrates a powerful approach to maximize the performance of supported metals through surface wettability modulation under catalytically relevant conditions.

Abstract:A first principle study of the cross-coupling reaction between 3-chloro-6-iodopyridazine and triphenylbismuth under palladium catalysis has been carried out. The approach selected for this investigation is the density functional theory within the framework of the PBE0 and B3LYP hybrid exchange–correlation functionals, with and without including empirical corrections for the dispersion forces at the GD3 level. For this reaction, a three-step mechanism including an oxidative addition, a transmetalation and a reductive elimination has been proposed. All intermediates and transition states have been fully characterized. The energy profile is deduced in gas phase and in solvent. Calculations suggest that the oxidative addition is the rate-determining step for the entire cross-coupling reaction.

Abstract:Polycyclic conjugated hydrocarbons have acquired increased interests recently because of their potential applications in electronic devices. On metal surfaces, the selective synthesis of four‐ and five‐membered carbon rings remains challenging due to the presence of diverse reaction pathways. Here, utilizing the same precursor molecule, we successfully achieved substrate‐controlled highly selective cycloaddition reactions towards four‐ and five‐membered carbon rings. A 97% yield for four‐membered carbon rings on Au(111), while a 96% yield towards five‐membered carbon rings is achieved on Ag(111). The detailed topological structures of the reaction products are carefully examined by bond‐resolving scanning tunneling microscopy (BR‐STM) imaging with a CO functionalized tip. The underlying mechanism of the novel surface‐directed reaction selectivity is elucidated by extensive density functional theory (DFT) calculations. Our study paves the way for high selective synthesis of polycyclic conjugated hydrocarbons with non‐benzenoid rings.

Abstract:The mechanisms and origins for the Pd- and Ni-catalyzed regioselective hydrosilylation of allene have been investigated by means of density functional theory (DFT) calculations. The free-energy profiles of Pd- and Ni-catalyzed reactions with small and bulky N-heterocyclic carbene (NHC) ligands are calculated to determine the mechanism for regioselectivities. The calculation results show that different metals (Ni vs Pd) lead to regiochemical reversals for the hydrosilylation of allene. The allylsilane is the major product via palladium catalysis with small NHC ligand, while the vinylsilane is the major product via nickel catalysis with bulky NHC ligand. Both electronic and steric factors play a key role in the regioselectivities for the hydrosilylation of allene via Pd and Ni catalysts. The calculation results are in good agreement with observed regioselectivities and could provide insights into the design of new catalysts for the regioselectivity of hydrosilylation reactions.
