Design, Synthesis, Biological Evaluation and Molecular Docking Studies of Quinoline-Anthranilic Acid Hybrids as Potent Anti-Inflammatory Drugs

Abstract:In the present study pharmacophore hybridization approach is used for the design and synthesis of three novel anti-inflammatory hybrids (5a-c) by linking medicinally privileged quinoline and anthranilic acid moieties through a methylene. The in silico studies of these hybrids showed that these compounds qualified all the drug-likeness rules with optimum pharmacokinetics and high binding affinity (9.4 to 10.6 kcal/mol) for TNF-α, FLAP, and COX-II in molecular docking. These compounds were synthesized (83-86% yield), and characterized using IR, NMR, and XRD. In vitro investigations of hybrid compounds and the standard at same dose showed that the antiprotease activity of compound 5b was significantly higher than 5a and 5c, and comparable to standard (p < 0.05), while in inhibition of albumin denaturation assay, the activity of 5b was significantly higher than 5a, 5c and standard (p < 0.05). In in vivo model of rat paw edema, results after 6 h indicated that activity of compound 5a-c and standard was good and significantly higher compared to the control (p < 0.05). The activity of 5b was comparable to standard, hence the compound 5b, being more potent may be employed in further studies to deliver a potent drug for effective treatment of inflammatory diseases like Rheumatoid arthritis.
