Nanoemulsion delivery strategy improves kellerin solubility, stability, and therapeutic efficacy in ischemic stroke

Abstract:Highlights • Preparation of kellerin nanoemulsion via the phase inversion composition. • Characterization of kellerin nanoemulsion physicochemical properties. • Determination of kellerin nanoemulsion release behavior in vitro. • Evaluated the efficacy of kellerin nanoemulsion in the treatment of ischemic stroke. Background Kellerin, an effective component derived from the Traditional Chinese Medicine Awei (resin of Ferula sinkiangensis ), can significantly alleviate cognitive impairment in mice after ischemic stroke. However, the poor water solubility of kellerin severely hinders its further development. Purpose The purpose of this study is to prepare kellerin nanoemulsion to improve its oral bioavailability and enhance therapeutic efficacy. Methods The optimal prescription and process were determined by single-factor analysis and orthogonal experiments. The physicochemical properties, in vitro release, and therapeutic efficacy of kellerin nanoemulsion in middle cerebral artery occlusion model rats were evaluated. Results The optimized kellerin nanoemulsion showed satisfactory particle size, encapsulation rate, consistent, and stable in vitro release behavior. Rats given equal or lower doses of kellerin nanoemulsion showed better results than kellerin suspensions in terms of improved neurobehavior, reduced brain infarct and edema volumes, alleviated neuronal damage, and inhibited microglial activation. Conclusion : These results suggest that kellerin nanoemulsion may be a prospective strategy for improving the efficacy of kellerin in ischemic stroke.
