Abstract:Abstract Macrotermes barneyi , widely distributed in southern China, is the major fungus-growing termite in the subfamily Macrotermitinae. It has no flagellated protists in the guts. Here, we report occurrence of gregarine, a protozoan parasite in the digestive tract of M. barneyi . The general morphology and ultrastructure of the gregarine gamonts and syzygies by light micrograph and scanning electron micrograph are presented. SSU rDNA sequence analysis showed that the termite gregarine has the highest identity (90.10%) to that of Gregarina blattarum from cockroaches. Phylogenetic analysis based on the SSU rDNA sequences from diverse insect eugregarines indicated that the gregarine from M. barneyi is phylogenetically close to G. blattarus, L. erratica and G. tropica from Gregarinidae and Leidyanidae families, and may represent a novel species. This study expands our knowledge about the diversity of terrestrial eugregarines parasitizing in termites.

What problem does this paper attempt to address?
The main problem this paper attempts to address is the recording and description of the first discovery of gregarine protozoa (belonging to the phylum Apicomplexa) in the fungus-growing termite *Macrotermes barneyi* in China. Specifically, the researchers investigated the presence, morphological characteristics, and taxonomic status of this parasite in the digestive tract of *M. barneyi* through morphological and ultrastructural observations as well as molecular phylogenetic analysis. ### Main Research Content: 1. **Discovery and Distribution**: The presence of gregarine protozoa in the digestive tract of *M. barneyi* worker ants was reported for the first time, but it was not found in soldier ants. 2. **Morphological Characteristics**: The morphological characteristics of the gregarine protozoa, including the structure of gamonts and syzygies, were described in detail through observations using light microscopy and scanning electron microscopy. 3. **Molecular Phylogenetic Analysis**: Through SSU rDNA sequence analysis, the phylogenetic relationship of this gregarine protozoa with known insect gregarine protozoa was determined, revealing the highest homology (90.10%) with *Gregarina blattarum* from cockroaches. 4. **Infection Rate**: The infection rate of *M. barneyi* worker ants was studied, and it was found that approximately 21.45% of worker ants were infected. ### Research Significance: - **Expanding Knowledge**: This study expands the understanding of the diversity of terrestrial gregarine protozoa, particularly their distribution in termites. - **Taxonomic Contribution**: It provides detailed morphological data and molecular evidence that aid in the further identification and classification of this newly discovered gregarine protozoa. - **Ecological Significance**: The ecological role of gregarine protozoa in termite societies was explored, which may be significant for understanding host-parasite coevolution and biological control. In summary, this paper fills a gap in the field through a detailed study of gregarine protozoa in *M. barneyi* and provides important foundational data for future research.
