Abstract:This article provides a detailed review of capillary electrophoresis (CE) technology in 2022, summarizing a total of 881 CE technology-related articles searched from ISI Web of Science using the keywords "capillary electrophoresis mass spectrometry" or "capillary isoelectric focusing" or "micellar electrokinetic chromatography" or "capillary electrophoresis" (excluding "capillary electrochromatography""microchip" "microfluidic" "capillary monolithic column"). The review focuses on 16 articles published in Lancet Global Health, ACS Central Science, Microbiome, Trends in Food Science & Technology, TrAC-Trends in Analytical Chemistry, Journal of Pharmaceutical Analysis, Journal of Cachexia, Sarcopenia and Muscle, Food Hydrocolloids, Science of the Total Environment, and Carbohydrate Polymers with impact factors (IFs) greater than 10.0, and 46 articles published in Analytical Chemistry, Analytica Chimica Acta, Talanta, and Food Chemistry with IFs between 5.0 and 10.0. A comprehensive overview of representative CE works published in Journal of Chromatography A, Electrophoresis, and important Chinese core journals (Peking University) with IFs<5.0 is also provided. Based on IFs, this review introduces representative works on CE to facilitate readers' understanding of important research advances in CE technology over the last year.

Abstract:The process of choosing the most proper technique for studying the molecular interactions is based on critical factors such as instrumentation complexity, automation, experimental procedures, analysis time, consumables, and cost‐value. This review has tracked the use of affinity capillary electrophoresis (ACE) and microscale thermophoresis (MST) techniques in the evaluation of molecular binding between different molecules during the 5 years 2016–2021. ACE has proved to be an attractive technique for biomolecular characterization with high resolution efficiency where small variations in several controlling factors can much improve such efficiency compared to other analytical techniques. Meanwhile, MST has proved its higher sensitivity for smaller amounts of complex non‐purified biosamples without affecting its robustness while providing high through output. However, the main motivation to review both techniques in the proposed review was their capability to carry out all experiments without the need for immobilizing one interacting partner, besides a great flexibility in the use of buffering systems The proposed review demonstrates the importance of both techniques in different areas of life sciences. Abstract:Abstract: The recent development of microfluidics and lab-on-a-chip technology has substantially raised interest in analytical chemistry. Since, they have demonstrated to be extraordinarily adept at precise fluid control, cell manipulation, and signal output, microfluidic chips are a useful tool for quick and in-depth single-cell investigation. This technique is cost-effective, less time-consuming, automatic, high mobility, and fast separation technique. Due to the internal chip sizes, which range from micrometers to millimeters, consumption of the samples and reagents occurs at the nanoliter and picoliter levels. The microfluidic device can fit a variety of functions onto a few centimeter-long chips. In this article, we discussed numerous preparations of microfluidic chip electrophoresis and its recent advancements. This method is useful for the detection of various small amounts of content with less time and greater efficacy. It is also useful in cancer studies, 3D inkjet printing, immunoassay investigation in cell-cell interactions, analysis of nanoparticles, dielectrophoretic particle separation, plant alkaloids, and forensic science applications. This review, therefore, examines the use of various microfluidic chips in electrophoretic separation during 2017–2022. There are various papers found by search, indicating continuous activity in the research area along with studies to explain its material, method, and its efficacy.

Abstract:This paper serves as an annual review of capillary electrophoresis (CE) technology for 2023. The journals were selected based on their impact factor (IF), a universally recognized academic performance metric, combined with experimental work closely related to CE technology, to facilitate the rapid acquisition of significant research and application advancements in CE technology in 2023. A thematic search of the ISI Web of Science database yielded 669 research papers on CE technology published in 2023. This review highlights five experimental papers published in journals with IFs greater than 10.0, including Nature Communications, Nucleic Acids Research, Engineering, Journal of Medical Virology, and Carbohydrate Polymers, and 31 experimental papers from representative journals with IFs between 5.0 and 10.0, such as Analytical Chemistry, Analytica Chimica Acta, Talanta, and Food Chemistry. It also provides an overview of experimental research in journals with focused reporting on CE technology but with IFs less than 5.0, such as Journal of Chromatography A and Electrophoresis, as well as significant experimental research from key domestic Chinese core journals (Peking University). In 2023, all the latest scientific advancements reported in journals with an IF greater than 10.0 utilized previously reported CE methods, offering new breakthroughs for the promotion and application of CE technology. Additionally, new applications of CE in conjunction with mass spectrometry remained a hot topic. An increase in reports on the hardware aspects of CE, such as 3D printing and underwater systems, and significant breakthroughs in the analysis of non-solution samples, such as solid particles, cell vesicles, cells, viruses, and bacteria, was noted. CE is advantageous for the analysis of drugs and their components. In Chinese journals, the number of papers on CE applications exceeded that in previous years, with particular focus on the field of printing for new applications.

Abstract:Protein separation or enrichment is one of the rate-limiting steps in proteomic studies. Specific capture and removal of highly-abundant proteins (HAP) with large sample-handling capacities are in great demand for enabling detection and analysis of low-abundant proteins (LAP). How to grasp and enrich these specific proteins or LAP in complex protein mixtures is also an outstanding challenge for biomarker discovery and validation. In response to these needs, various approaches for removal of HAP or capture of LAP in biological fluids, particularly in plasma or serum, have been developed. Among them, immunoaffinity subtraction methods based upon polyclonal IgY or IgG antibodies have shown to possess unique advantages for proteomic analysis of plasma, serum and other biological samples. In addition, other affinity methods that use recombinant proteins, lectins, peptides, or chemical ligands have also been developed and applied to LAP capture or enrichment. This review discusses in detail the need to put technologies and methods in affinity subtraction or enrichment into a context of proteomic and systems biology as “Separomics” and provides a prospective of affinity-mediated proteomics. Specific products, along with their features, advantages, and disadvantages will also be discussed.

Abstract:Cells are the fundamental unit of life. Owing to cellular heterogeneity, traditional biochemical assays which analyze cells in bulk often overlook the rich information available when single cells are studied. Accurate and reliable determination of the chemical composition in individual cells would greatly improve the probability of discriminating infected cells from healthy ones and provide a solid foundation on study and development in various fields including biochemistry, medicine, and pathology clinic. Microfluidic chip system for single-cell analysis is now attracting broad interests. The ability of integrating the whole process on one microfluidic chip, including single cell injection, lysis, separation and detection of cellular constituents by chip-based CE, offers a new technical platform for single cell analysis. This article provides a review on recent advances in determining intracellular contents in individual cells using microfluidic devices. Focus areas include on-chip single cell manipulation by means of electroosmotic flow (EOF), pressure-driven flow combined with EOF and optical tweezer (OT) to perform sequentially single cell injection, lysis, separation and detection of cellular constituents by chip-based CE with highly sensitive detection. The approaches on on-chip derivatization are also addressed.

Abstract:CE has been a continuously evolving analytical methodology since its first introduction in the 1980s of the last century. The development of new CE separation procedures, the coupling of these systems to more sensitive and versatile detection systems, and the advances in miniaturization technology have allowed the application of CE to the resolution of new and complex analytical problems, overcoming the traditional disadvantages associated with this method. In the present work, different recent trends in CE and their application to the determination of high complexity samples (as biological fluids, individual cells, etc.) will be reviewed: capillary modification by different types of coatings, microfluidic CE, and online microextraction CE. The main advantages and disadvantages of the different proposed approaches will be discussed with examples of most recent applications.

Abstract:The knowledge of elemental speciation and interaction between elemental species and ligands (including biomolecules) can provide an understanding of their transport, transformation, bioavailability, metabolic pathways and toxicity in organisms. By taking advantage of the powerful separation capabilities of capillary electrophoresis (CE) in capillaries/microchannels, and the atomic specificity, multi-elemental characteristics and extremely high sensitivity of inductively coupled plasma spectrometries (ICPs), the CE-ICP hyphenation is one of the most preferred choices in elemental speciation and element-ligand/element-biomolecule interaction as well as the indirect determination of non-metallic compounds. This article provides an overview of electrophoresis in capillaries and microchannels coupled to an ICP with respect to interfaces and applications from elemental speciation, element-ligand interaction to indirect determination.

Abstract:Monoclonal antibodies (mAbs) and their related products as antibody-drug-conjugates (ADCs) or biosimilars represent a constantly growing class of molecules therapeutic proteins used as treatment against numerous diseases. These compounds can undergo several modifications which could alter the efficiency of treatments. In this context, several analytical methods were designed to deliver a comprehensive structural characterization and guarantee the quality of biotherapeutics. Capillary electrophoresis (CE) is considered today as a major technique for the analysis of biotherapeutics due to benefic characteristics as high resolution separation and miniaturized format. Different CE modes have been developed to characterize mAbs at different levels such as capillary gel electrophoresis (CGE), capillary isoelectric focusing (cIEF), and capillary zone electrophoresis (CZE). Recent developments in CE-mass spectrometry (MS) coupling assessed this technology as a promising tool to obtain high level structural characterization of biopharmaceuticals. Moreover, upcoming techniques such as 2D CE-MS and microfluidic systems are now emerging to offer new possibilities beyond actual limits. This review will be dedicated to discuss the state-of-the-art CE-based methods for the characterization of mAbs and ADCs in the period 2016-2018.

Abstract:Capillary electrophoresis (CE) has been abundantly used in the study of molecular interactions owing to such advantages as short analysis time, low sample size requirement, high separation efficiency, and flexible applications. The focus of this paper is to review recent studies and advances (mainly from 1998 to now) in biomolecular interactions using CE. Five CE modes: zone migration CE, affinity CE, frontal analysis (FA), Hummel-Dreyer (HD) and vacancy peak (VP) are cited and compared. Quantitative aspects of the thermodynamics and kinetics of biomolecular interaction are reviewed. Several biomolecular binding systems, including protein-protein (polypeptide), protein-DNA (RNA), protein(polypeptide)-carbohydrate, protein-small molecule, DNA-small molecule, small molecule-small molecule, have been well characterized by CE. CE is shown to be a powerful tool for the determination of the binding parameters of various bioaffinity interactions.

Abstract: Assays employing aptamers in capillary electrophoresis (CE), including competitive and noncompetitive assays, fluorescence polarization (FP) assays, nonequilibrium capillary electrophoresis of equilibrium mixtures, and affinity-polymerase chain reaction-CE assays, are summarized. These assays can be used to estimate dissociation rate and equilibrium binding constants, determine binding stoichiometries, study molecular interactions, and quantitatively determine specific analytes (e.g., proteins) in complex media. They can potentially be completed in under 60 s, detect zeptomol (10−24) amounts of analyte, be utilized in complex media with little or no cross reaction, and target a number of different analytes of biological, environmental, and clinical importance. This chapter briefly overviews the process of aptamer selection using CE and discusses the various CE-based bioanalytical methods that have been used to study biomolecular interactions.

Abstract:Over the past decades, the proteomics field has undergone rapid growth. Progress in mass spectrometry and bioinformatics, together with separation methods, has brought many innovative approaches to the study of the molecular biology of the cell. The potential of affinity chromatography was recognized immediately after its first application in proteomics, and since that time, it has become one of the cornerstones of many proteomic protocols. Indeed, this chromatographic technique exploiting the specific binding between two molecules has been employed for numerous purposes, from selective removal of interfering (over)abundant proteins or enrichment of scarce biomarkers in complex biological samples to mapping the post-translational modifications and protein interactions with other proteins, nucleic acids or biologically active small molecules. This review presents a comprehensive survey of this versatile analytical tool in current proteomics. To navigate the reader, the haphazard space of affinity separations is classified according to the experiment's aims and the separated molecule's nature. Different types of available ligands and experimental strategies are discussed in further detail for each of the mentioned procedures.

[Advances in application of capillary electrophoresis in pharmaceutical analysis]

Abstract:Capillary electrophoresis (CE) is mainly applied in pharmaceutical analysis. All CE separation modes and detection methods show their characteristics and application abilities in the separation and analysis of different drug samples. The present review provides a brief cross section of new advances in CE application in pharmaceutical analysis, including for small molecular drugs and related substances (including chiral drug separation), traditional Chinese medicine and natural products, in vivo pharmaceutical analysis, and biological product analysis. However, the research on physical and chemical constant determination, affinity capillary electrophoresis and binding constant research (drug and receptor interaction, etc.), clinical biomarker analysis, metabolomics, and microchip CE analysis are not included. According to traditional pharmaceutical analysis developments, the recent advances in CE in compliance with pharmaceutical analysis regulatory requirements include CE capacitively coupled contactless conductivity detection (C4D), improved detection sensitivity and precision, CE-sodium dodecyl sulfate (CE-SDS), imaged capillary isoelectric focusing (icIEF), antibody analysis, and so on. Combined with the references, this review also discusses the current requirements in the field of traditional pharmaceutical analysis, as well as the status, challenges and opportunities of CE in it. Some suggestions on CE application as a complementary analysis method for chemical drugs and traditional Chinese medicine analysis are put forward, and the characteristics and ability of CE in biological product analysis is expected to further development. The new development of CE-MS and improvement of CE repeatability may greatly expand the field of application of CE in the future. This review covers the improvements published between January 2017 and February 2020, as well as some important CE papers published in 2016.

Abstract:This review provides an in‐depth exploration of capillary electrophoresis–mass spectrometry (CE–MS) in biomolecular research from 2020 to 2024. CE–MS emerges as a versatile and powerful tool due to its numerous advantages, facilitating the analysis of various biomolecules, including proteins, peptides, oligonucleotides, and other metabolites, such as lipids, carbohydrates, or amines, among others. The review extends to various CE modes and interfaces for the CE–MS coupling, offering comprehensive insights into their applications within biomolecular research. Furthermore, it effectively summarizes the conditions employed in CE–MS while also addressing critical aspects such as sample preparation requirements. Despite its advantages, the review highlights a gap between discovery and practical implementation, underscoring the need for large‐scale validation and method standardization to fully realize the potential of CE–MS in biomolecular research.
