Abstract:Bioluminescent dinoflagellates are flow-sensitive marine organisms that produce light emission almost instantaneously upon stimulation by fluid shear in a shear stress dose-dependent manner. In the present study we tested the hypothesis that monitoring bioluminescence by suspended dinoflagellates can be used as a tool to characterize cellular response to hydrodynamic forces in agitated bioreactors. Specific studies were performed to determine: (1) impeller configurations with minimum cell activation, (2) correlations of cellular response and an integrated shear factor, and (3) the effect of rapid acceleration in agitation. Results indicated that (1) at a volumetric mass transfer coefficient of 3 x 10(-4) s(-1), marine impeller configurations were less stimulatory than Rushton configurations, (2) bioluminescence response and a modified volumetric integrated shear factor had an excellent correlation, and (3) rapid acceleration in agitation was highly stimulatory, suggesting a profound effect of temporal gradients in shear in increasing cell stimulation. By using bioluminescence stimulation as an indicator of agitation-induced cell stimulation and/or damage in microcarrier cultures, the present study allows for the verification of hypotheses and development of novel mechanisms of cell damage in bioreactors. Bubble stimulation efficiency of dinoflagellate bioluminescence

Abstract:Dinoflagellate bioluminescence, a common source of bioluminescence in coastal waters, is stimulated by flow agitation. Although bubbles are anecdotally known to be stimulatory, the process has never been experimentally investigated. This study quantified the flash response of the bioluminescent dinoflagellate Lingulodinium polyedrum to stimulation by bubbles rising through still seawater. Cells were stimulated by isolated bubbles of 0.3-3 mm radii rising at their terminal velocity, and also by bubble clouds containing bubbles of 0.06-10 mm radii for different air flow rates. Stimulation efficiency, the proportion of cells producing a flash within the volume of water swept out by a rising bubble, decreased with decreasing bubble radius for radii less than approximately 1 mm. Bubbles smaller than a critical radius in the range 0.275-0.325 mm did not stimulate a flash response. The fraction of cells stimulated by bubble clouds was proportional to the volume of air in the bubble cloud, with lower stimulation levels observed for clouds with smaller bubbles. An empirical model for bubble cloud stimulation based on the isolated bubble observations successfully reproduced the observed stimulation by bubble clouds for low air flow rates. High air flow rates stimulated more light emission than expected, presumably because of additional fluid shear stress associated with collective buoyancy effects generated by the high air fraction bubble cloud. These results are relevant to bioluminescence stimulation by bubbles in two-phase flows, such as in ship wakes, breaking waves, and sparged bioreactors.

Abstract:Bioluminescence is ubiquitous in marine ecosystems and found in uni- and multicellular organisms. Bioluminescent displays can be used to deter predators, attract mates, and lure and hunt prey. Mechanically stimulated flash kinetics of zooplankton and dinoflagellates are life stage-dependent and species-specific, and could prove effective at identification and monitoring biodiversity in bioluminescent species. Here, we provide a comprehensive review of mechanically stimulated bioluminescence for the main dinoflagellate and zooplankton clades in marine environments and assemble known flash kinetics and spectral emission data. Instruments and methods used in measuring bioluminescence are also discussed. Applications, research gaps, perspectives, and biases in approaches to studying bioluminescence are identified. Moreover, emission kinetics of most zooplankton are very poorly known and constitute a critical gap. Lastly, available knowledge is interpreted in terms of potential future changes in global bioluminescence driven by climate change.

Bioluminescence Contributes to the Adaptation of Deep-Sea Bacterium Photobacterium phosphoreum ANT-2200 to High Hydrostatic Pressure

Abstract:Bioluminescence is a common phenomenon in nature, especially in the deep ocean. The physiological role of bacterial bioluminescence involves protection against oxidative and UV stresses. Yet, it remains unclear if bioluminescence contributes to deep-sea bacterial adaptation to high hydrostatic pressure (HHP). In this study, we constructed a non-luminescent mutant of ΔluxA and its complementary strain c-ΔluxA of Photobacterium phosphoreum ANT-2200, a deep-sea piezophilic bioluminescent bacterium. The wild-type strain, mutant and complementary strain were compared from aspects of pressure tolerance, intracellular reactive oxygen species (ROS) level and expression of ROS-scavenging enzymes. The results showed that, despite similar growth profiles, HHP induced the accumulation of intracellular ROS and up-regulated the expression of ROS-scavenging enzymes such as dyp, katE and katG, specifically in the non-luminescent mutant. Collectively, our results suggested that bioluminescence functions as the primary antioxidant system in strain ANT-2200, in addition to the well-known ROS-scavenging enzymes. Bioluminescence contributes to bacterial adaptation to the deep-sea environment by coping with oxidative stress generated from HHP. These results further expanded our understanding of the physiological significance of bioluminescence as well as a novel strategy for microbial adaptation to a deep-sea environment.

Abstract:Stress in sea urchins leads to high mortality and economic losses in both the environment and aquaculture. The green sea urchin Strongylocentrotus droebachiensis has been documented emitting complex biofluorescence, yet how this responds to external stressors is unknown. Adult sea urchins ( n = 210) were divided between control ( n = 30) and experimental groups ( n = 180), using three transport variables: out of water, in water at elevated temperatures, (warm-water) and in water at seawater temperature (cold-water). Hyperspectral imaging of external fluorescence and fluorospectrometric analysis on coelomic fluid was measured at five intervals (hour 0,3,6,9,12). External green emissions ( ∼ 580 nm) responded to all treatments, peaking at h9. External red emissions ( ∼ 680–730 nm) in the cold-water remained low until an h9 peak. The warm water increased emissions at each interval, peaking at h9. The out of water gradually increased, with the highest at h12. The coelomic fluid fluorescence ( ∼ 680 nm) was low to nonexistent except in warm-water, whose elevated levels suggest that fluorescent emissions are a measurable byproduct of internal adaptation(s) to stress. Early detection of fluorescent emissions (broken spines, lesions) may prevent economic losses. The observed link between fluorescence and the applied stressors provides a baseline for developing non-invasive technology for improving echinoderm welfare.

Abstract:Living organisms on the surface biosphere are periodically yet consistently exposed to light. The adaptive or protective evolution caused by this source of energy has led to the biological systems present in a large variety of organisms, including fungi. Among fungi, yeasts have developed essential protective responses against the deleterious effects of light. Stress generated by light exposure is propagated through the synthesis of hydrogen peroxide and mediated by regulatory factors that are also involved in the response to other stressors. These have included Msn2/4, Crz1, Yap1, and Mga2, thus suggesting that light stress is a common factor in the yeast environmental response.
Physiological stress and cell death in marine phytoplankton: Induction of proteases in response to nitrogen or light limitation

Abstract:The physiological processes of natural phytoplankton mortality due to environmental stress (vs. that caused by sedimentation and predation) are poorly understood. Cell survival was examined in batch cultures of the diatom Thalassiosira weissflogii and the chlorophyte Dunaliella tertiolecta during deprivation of fixed nitrogen or light. Despite severe impairment of photosynthetic efficiencies, both species remained viable during 2 weeks of N starvation. Under N stress, a specific protease was induced in the diatom, overall activity of proteases doubled, and there was gradual, selective loss of certain proteins, especially ribulose 1,5‐bisphosphate carboxylase/oxygenase. Light‐deprived diatoms were virtually unaffected, but the chlorophyte underwent catastrophic cell death after about 6 d of darkness. Cell death coincided with a large increase in protease activity and the induction of a specific protease. Although we cannot completely rule out roles for viruses or bacteria in the losses of cells, the consistent timing, the unique response to stress, and the coincident expression of a specific protease strongly suggest that the process is a form of autocatalyzed cell death, such as apoptosis. While of uncertain adaptive significance, phytoplankton cell death may have implications for species succession and cycling of organic matter in aquatic ecosysterns.

Abstract:Photosynthetic algae play a significant role in oceanic carbon capture. Their performance, however, is constantly challenged by fluctuations in environmental light conditions. Here, we show that the non-motile single-celled marine dinoflagellate can internally contract its chloroplast network in response to light. By exposing the cell to various physiological light conditions and applying temporal illumination sequences, we find that network morphodynamics follows simple rules, as established in a mathematical model. Our analysis of the chloroplast structure reveals that its unusual reticulated morphology constitutes properties similar to auxetic metamaterials, facilitating drastic deformations for light-avoidance, while confined by the cell wall. Our study shows how the topologically complex network of chloroplasts is crucial in supporting the dinoflagellate’s adaptation to varying light conditions, thereby facilitating essential life-sustaining processes.

Abstract:Mechanosensing, the transduction of extracellular mechanical stimuli into intracellular biochemical signals, is a fundamental property of living cells. However, endowing synthetic materials with mechanosensing capabilities comparable to biological levels is challenging. Here, we developed ultrasensitive and robust mechanoluminescent living composites using hydrogels embedded with dinoflagellates, unicellular microalgae with a near-instantaneous and ultrasensitive bioluminescent response to mechanical stress. Not only did embedded dinoflagellates retain their intrinsic mechanoluminescence, but with hydrophobic coatings, living composites had a lifetime of ~5 months under harsh conditions with minimal maintenance. We 3D-printed living composites into large-scale mechanoluminescent structures with high spatial resolution, and we also enhanced their mechanical properties with double-network hydrogels. We propose a counterpart mathematical model that captured experimental mechanoluminescent observations to predict mechanoluminescence based on deformation and applied stress. We also demonstrated the use of the mechanosensing composites for biomimetic soft actuators that emitted colored light upon magnetic actuation. These mechanosensing composites have substantial potential in biohybrid sensors and robotics.

Abstract:Bioluminescence, the ability of organisms to produce visible light, has intrigued scientists for centuries. Studies have examined bioluminescence, using a wide range of approaches and organisms, from its ecological role to its underlying molecular mechanisms, leading to various applications and even a Nobel prize. Over the last ten years, an increasing amount of data has been collected leading to a growing number of recognized marine bioluminescent species. This review provides and describes a referenced listing of the eukaryotic luminous marine species, including information related to: (i) intrinsic versus extrinsic source of the bioluminescence, (ii) the color and maximum wavelength of emission, (iii) the bioluminescent system (substrate and enzyme) and the associated molecules, (iv) the availability of light organ/cell(s) pattern and histological structure, (v) the physiological control of the light production, and (vi) the demonstrated or suggested bioluminescent function(s). This listing provides basic information and references for researchers in or entering in the field of marine bioluminescence. Using a semi-quantitative approach, we then highlight major research gaps and opportunities and reflect on the future of the field.

Abstract:The dynamic movement of cell organelles is an important and poorly understood component of cellular organization and metabolism. In this work we present a non-invasive non-destructive method (Dynamic Cell Imaging, DCI) based on light scattering and interferometry to monitor dynamic events within photosynthetic cells using the diatom Phaeodactylum tricornutum as a model system. For this monitoring we acquire for a few seconds movies of the signals that are related to the motion of dynamic structures within the cell (denoted scatterers), followed by a statistical analysis of each pixel time series. Illuminating P. tricornutum with LEDs of different wavelengths associated with short pulsed or continuous-wave modes of illumination revealed that dynamic movements depend on chloroplast activity, in agreement with the reduction in the number of pixels with dynamic behaviour after addition of photosystem II inhibitors. We studied P. tricornutum under two environmentally relevant stresses, iron and phosphate deficiency. The major dynamic sites were located within lipid droplets and chloroplast envelope membranes. By comparing standard deviation and cumulative sum analyses of the time series, we showed that within the droplets two types of scatterer movement could be observed: random motion (Brownian type) but also anomalous movements corresponding to a drift which may relate to molecular fluxes within a cell. The method appears to be valuable for studying the effects of various environments on microalgae in the laboratory as well as in natural aquatic environments. Light scattering is an alternative to fluorescence to rapidly evidence dynamic processes. Lipid droplets are the major metabolic active sites under stress. A non-destructive visualization method suitable for laboratory microalgae and aquatic samples.

Abstract:It has long been hypothesized that benthic motile pennate diatoms use phototaxis to optimize photosynthesis and minimize photoinhibitory damage by adjusting their position within vertical light gradients in coastal benthic sediments. However, experimental evidence to test this hypothesis remains inconclusive, mainly due to methodological difficulties in studying cell behavior and photosynthesis over realistic spatial microscale gradients of irradiance and cell position. In this study, a novel experimental approach was developed and used to test the hypothesis of photosynthesis optimization through motility, based on the combination of single-cell in vivo chlorophyll fluorometry and microfluidic chips. The approach allows the concurrent study of behavior and photosynthetic activity of individual cells of the epipelic diatom species Craspedostauros britannicus exposed to a light microgradient of realistic dimensions, simulating the irradiance and distance scales of light microgradients in benthic sediments. Following exposure to light, (i) cells explored their light environment before initiating light-directed motility; (ii) cells used motility to lower their light dose, when exposed to the highest light intensities; and (iii) motility was combined with reversible non-photochemical quenching, to allow cells to avoid photoinhibition. The results of this proof-of-concept study not only strongly support the photoprotective nature of photobehavior in the studied species but also revealed considerable variability in how individual cells reacted to a light microgradient. The experimental setup can be readily applied to study motility and photosynthetic light responses of other diatom species or natural assemblages, as well as other photoautotrophic motile microorganisms, broadening the toolset for experimental microbial ecology research.

Abstract:Elastico‐mechanoluminescence technology has shown significant application prospects in stress sensing, artificial skin, remote interaction, and other research areas. Its progress mainly lies in realizing stress visualization and two‐dimensional or even three‐dimensional stress‐sensing effects using a passive sensing mode. However, the widespread promotion of mechanoluminescence (ML) technology has been hindered by issues such as high stress or strain thresholds and a single sensing mode based on luminous intensity. In this study, a highly efficient green‐emitting ML with dual mode stress‐sensing characteristics driven by micro‐scale strain was developed using LiTaO3:Tb3+. In addition to single‐mode sensing based on the luminous intensity, the self‐defined parameter (Q) was also introduced as a dual‐mode factor for sensing the stress velocity. Impressively, the fabricated LiTaO3:Tb3+ film is capable of generating discernible ML signals even when supplied with strains as low as 500 μst. This is the current the minimum strain value that can drive green‐emitting ML. Our study offers an ideal photonic platform for exploring the potential applications of rare‐earth‐doped elastico‐ML materials in remote interaction devices, high‐precision stress sensors, and single‐molecule biological imaging. Abstract:Real-time visualization of stress-sensing can be potentially realized by mechanoluminescence (ML) materials that are able to transform mechanical stimulates into luminescence. The unsatisfactory ML brightness, however, restricts such applications of ML materials mainly to dark-field scenarios. Thanks to the low UV proportion in solar spectrum, a UV ML may afford a suitable signal-to-noise (SNR) to support stress-sensing in bright-field environments. Herein, the ultraviolet-B (UVB) ML material with visible-blind features has been firstly developed to enable a bright-field visualization of stress-sensing. Specifically, an energy-focusing process has been applied via a sensitization strategy to reinforce the ML intensity in UVB region at the cost of that in other regions, which helps provide better SNR of UVB ML. This work provides a new opportunity in terms of the application of ML material with bright fields environment.

Abstract:Directly examining subcellular mechanics whilst avoiding excessive strain of a live cell requires the precise control of light stress on very small areas, which is fundamentally difficult. Here we use a glass nanopipet out of contact with the plasma membrane to both exert the stress on the cell and also accurately monitor cellular compression. This allows the mapping of cell stiffness at a lateral resolution finer than 100 nm. We calculate the stress a nanopipet exerts on a cell as the sum of the intrinsic pressure between the tip face and the plasma membrane plus its direct pressure on any glycocalyx, both evaluated from the gap size in terms of the ion current decrease. A survey of cell types confirms that an intracellular pressure of approximately 120 Pa begins to detach the plasma membrane from the cytoskeleton and reveals that the first 660 +/- 90 nm of compression of a neuron cell body is much softer than previous methods have been able to detect.

Abstract:Bioluminescence is the production of visible light by living organisms thanks to a chemical reaction, implying the oxidation of a substrate called luciferin catalyzed by an enzyme, the luciferase. The luminous brittle star Amphiura filiformis depends on coelenterazine ( i . e ., the most widespread luciferin in marine ecosystems) and a luciferase homologous to the cnidarian Renilla luciferase to produce blue flashes in the arm’s spine. Only a few studies have focused on the ontogenic apparitions of bioluminescence in marine organisms. Like most ophiuroids, A . filiformis displays planktonic ophiopluteus larvae for which the ability to produce light was not investigated. This study aims to document the apparition of the luminous capabilities of this species during its ontogenic development, from the egg to settlement. Through biochemical assays, pharmacological stimulation, and Renilla -like luciferase immunohistological detection across different developing stages, we pointed out the emergence of the luminous capabilities after the ophiopluteus larval metamorphosis into a juvenile. In conclusion, we demonstrated that the larval pelagic stage of A . filiformis is not bioluminescent compared to juveniles and adults.

Abstract:Diatoms are a group of silicified algae that play a major role in marine and freshwater ecosystems. Diatom chloroplasts were acquired by secondary endosymbiosis and exhibit important structural and functional differences from the primary plastids of land plants and green algae. Many functions of primary plastids, including photoacclimation and inorganic carbon acquisition, are regulated by calcium-dependent signalling processes. Calcium signalling has also been implicated in the photoprotective responses of diatoms; however, the nature of calcium elevations in diatom chloroplasts and their wider role in cell signalling remains unknown. Using genetically encoded calcium indicators, we find that the diatom Phaeodactylum tricornutum exhibits dynamic calcium elevations within the chloroplast stroma. Stromal calcium ([Ca2+]str) acts independently from the cytosol and is not elevated by stimuli that induce large cytosolic calcium ([Ca2+]cyt) elevations. In contrast, high light and exogenous hydrogen peroxide (H2O2) induce large, sustained [Ca2+]str elevations that are not replicated in the cytosol. Measurements using the fluorescent H2O2 sensor roGFP2-Oxidant Receptor Peroxidase 1 (Orp1) indicate that [Ca2+]str elevations induced by these stimuli correspond to the accumulation of H2O2 in the chloroplast. [Ca2+]str elevations were also induced by adding methyl viologen, which generates superoxide within the chloroplast, and by treatments that disrupt non-photochemical quenching (NPQ). The findings indicate that diatoms generate specific [Ca2+]str elevations in response to high light and oxidative stress that likely modulate the activity of calcium-sensitive components in photoprotection and other regulatory pathways.

Bioluminescent and Red-Fluorescent Lures in a Deep-Sea Siphonophore

Abstract:Bioluminescence (light production) and fluorescence (re-emission of absorbed radiation as light) are found in an unaccountably diverse array of marine organisms, where their functions are largely unknown. Here we report a deep-sea siphonophore that twitches glowing lures to attract fish. This is rare evidence of bioluminescence used for prey attraction among nonvisual marine organisms. The lures also contain red fluorescent material that shifts the wavelength of emitted light. The existence of a red-luminescent invertebrate suggests that long-wavelength light plays a greater role in marine interactions than previously suspected.

Abstract:Bioluminescence is light produced by organisms through chemical reactions. In most cases, bioluminescent organisms produce light in response to mechanical stimulation, including from shear around objects moving in the water. Many phytoplankton and zooplankton are capable of producing bioluminescence, which is commonly measured as bioluminescence potential, defined as mechanically stimulated light measured inside of a chambered pump-through bathyphotometer. We have developed a numerical model of a pump-through bathyphotometer and simulated flow using Lagrangian particles as an approximation for bioluminescent marine plankton taxa. The results indicate that all particles remain in the detection chamber for a residence time of at least 0.25 s. This suggests that the total first flash of bioluminescent autotrophic and heterotrophic dinoflagellates will be measured based on the existing literature regarding their flash duration. We have found low sensitivity of particle residence time to variations in particle size, density, or measurement depth. In addition, the results show that a high percentage of organisms may experience stimulation well before the detection chamber, or even multiple stimulations within the detection chamber. The results of this work serve to inform the processing of current bioluminescent potential data and assist in the development of future instruments.

Abstract:Significance Phytoplankton form massive blooms in the oceans that are controlled by nutrients, light availability, and biotic interactions with grazers and viruses. Although phytoplankton were traditionally considered passive drifters with currents here we demonstrate how diatom cells sense and respond to oxidative stress through a redox-sensitive protein network. We further demonstrate the redox sensitivity of nitrogen assimilation, which is essential for diatom blooms in the ocean, and provide compelling evidence for organelle-specific oxidation patterns under nitrogen stress conditions using a genetically encoded redox sensor. We propose that redox regulation of metabolic rates in the response to stress provides a mechanism of acclimation to rapid fluctuations in the chemophysical gradients in the marine environment.
