Abstract:Oncogenic mutations accumulating in many chromatin-associated proteins have been identified in different tumor types. With a mutation rate from 10 to 57%, ARID1A has been widely considered a tumor suppressor gene. However, whether this role is mainly due to its transcriptional-related activities or its ability to preserve genome integrity is still a matter of intense debate. Here, we show that ARID1A is largely dispensable for preserving enhancer-dependent transcriptional regulation, being ARID1B sufficient and required to compensate for ARID1A loss. We provide in vivo evidence that ARID1A is mainly required to preserve genomic integrity in adult tissues. ARID1A loss primarily results in DNA damage accumulation, interferon type I response activation, and chronic inflammation leading to tumor formation. Our data suggest that in healthy tissues, the increased genomic instability that follows ARID1A mutations and the selective pressure imposed by the microenvironment might result in the emergence of aggressive, possibly immune-resistant, tumors.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is about the mechanism of action of ARID1A gene mutations in tumorigenesis. Specifically, the research aims to explore the following points: 1. **Function of the ARID1A gene**: As a subunit of the SWI/SNF chromatin - remodeling complex, ARID1A frequently has inactivating mutations in different types of tumors, especially in gastric cancer, ovarian cancer, uterine cancer, bladder cancer, colon cancer and liver cancer. However, it is still controversial whether ARID1A exerts its tumor - suppressing effect mainly through its transcription - related activities or by maintaining genomic integrity. 2. **Effects of ARID1A deletion**: Researchers generated mouse models with ARID1A knocked out in specific tissues, especially liver - specific ARID1A - knockout mice, to explore the effects of ARID1A deletion on genomic stability and the tissue microenvironment. The study found that ARID1A deletion mainly leads to DNA damage accumulation, activation of type I interferon response and chronic inflammation, and ultimately promotes tumor formation. 3. **Relationship between ARID1A and enhancer activity**: Although a large number of literatures indicate that ARID1A prevents tumors by maintaining enhancer - dependent transcriptional regulation, this study found that ARID1A is not necessary for maintaining tissue - specific enhancer activity in adult tissues. ARID1B can compensate for the loss of ARID1A to a certain extent and maintain cell - specific transcriptional programs. 4. **Early oncogenic events**: The study also revealed early oncogenic events that occur after ARID1A deletion. These events are not related to enhancer - dependent transcriptional regulation, but are related to DNA damage accumulation and increased genomic instability. 5. **Role of the tumor microenvironment**: The study also explored the changes in the tissue microenvironment after ARID1A deletion, such as chronic inflammation and activation of the immune response. These changes may promote the emergence of more aggressive tumors under selective pressure. In summary, the main purpose of this paper is to deeply understand the specific mechanisms of ARID1A gene mutations in tumorigenesis, especially the roles in genomic stability and tissue microenvironment remodeling.
