What problem does this paper attempt to address?
The problem that this paper attempts to solve is the secondary brain injury caused by oxidative stress and neuronal apoptosis after traumatic brain injury (TBI). Specifically, the study aims to explore the protective effect of astaxanthin (ATX) on TBI and its potential mechanisms, especially whether ATX can reduce oxidative stress and neuronal apoptosis by regulating the SIRT1/Nrf2/Prx2/ASK1/p38 signaling pathway. ### Background and Objectives - **Background**: Traumatic brain injury (TBI) is one of the leading causes of disability and death worldwide. Although the pathophysiology of TBI has not been fully elucidated, increasing evidence suggests that reactive oxygen species (ROS) - related oxidative damage and neuronal apoptosis are key pathological processes in secondary brain injury after TBI. - **Objectives**: This paper studied the protective effect of astaxanthin (ATX) on TBI and attempted to reveal its underlying mechanism of action. ### Research Methods - **Experimental model**: A TBI model was established in mice using the weight - drop method, and a hydrogen peroxide exposure model was used in vitro. - **Experimental design**: The effects of ATX on TBI were evaluated by behavioral tests, Western blotting, biochemical analysis, lesion volume, histopathological studies, and cell viability assays. ### Main Results - **Oxidative stress**: ATX significantly reduced oxidative damage on the 1st, 3rd, and 7th days after TBI. - **Neuronal apoptosis**: ATX significantly improved neuronal apoptosis on the 3rd day after TBI. - **Neurological function**: ATX significantly improved neurological function within 3 weeks after TBI. - **Mechanism**: ATX treatment significantly enhanced the expression of peroxiredoxin 2 (Prx2), nuclear factor - erythroid 2 - related factor 2 (Nrf2), and silent information regulator 1 (SIRT1), while down - regulating the phosphorylation of apoptosis signal - regulating kinase 1 (ASK1) and p38. Injection of small interfering RNA for Prx2 reversed the beneficial effects of ATX on TBI. Nrf2 gene knockout attenuated the neuroprotective effect of ATX. Conversely, overexpression of Prx2 in Nrf2 - knockout mice could reduce secondary brain injury after TBI. In addition, inhibition of SIRT1 using EX527 significantly weakened the neuroprotective effect of ATX and inhibited the SIRT1/Nrf2/Prx2/ASK1/p38 signaling pathway. ### Conclusions and Significance - **Conclusions**: ATX improves neurological function and protects the brain after TBI mainly through the SIRT1/Nrf2/Prx2/ASK1/p38 signaling pathway by reducing oxidative stress and neuronal death. - **Significance**: ATX may be a new candidate drug for the treatment of TBI and other acute brain injuries. ### Keywords - Astaxanthin; TBI; Oxidative damage; Prx2; Nrf2; SIRT1 This paper, through systematic research, reveals the protective effect of ATX in TBI and its underlying molecular mechanisms, providing important scientific basis for the development of new TBI treatment strategies.
