Orally Bioavailable STING Antagonist Synthesized via Multi-component Povarov-Doebner Type Reaction

Abstract:The cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) signaling has emerged as the major DNA-sensing pathway and one of the most prominent defensive mechanism in mammals. However, aberrant activation of the cGAS-STING signaling results in innate immune response induction with detrimental effects in host organisms, leading to inflammatory and autoimmune diseases such as STING associated vasculopathy with onset in infancy (SAVI) and Aicardi-Goutier̀es syndrome (AGS). A few STING antagonists have been reported but none has progressed to the clinic. We earlier reported HSD077 as a potent STING antagonist. But HSD1077 suffers from fast clearance in vivo. Herein, we report HSKB142, an orally bioavailable compound containing the 3H-pyrazolo[4,3-f ]quinoline synthesized via a Povarov–Doebner type three-component reaction involving an amine, ketone, and aldehyde. HSKB142 is non cytotoxic towards immune cells and suppresses type-1 interferon expression in human THP-1 and THP-1 dual (KI STING N154S) monocytes upon treatment with 2′-3′ cGAMP, showing the potential as a therapeutic option for treating STING associated inflammatory diseases.
