Abstract:A glutathione S-transferase gene (gst) has been introduced into restorers of cytoplasmic male sterility in upland cotton (Gossypium hirsutum L.) using Agrobacterium-mediated transformation. A trans-genie restorer, signed as 'Zheda strong restorer', which has strong restorability to male sterility, was selected from progeny plants of transformants. When compared with an American restorer 'DES-HAF277', the fertility restorability of 'Zheda strong restorer' to male sterility was been enhanced by 25.8% in the percentage of viable pollens of hybrid (sterile line × restorer) F1 The hybrid gave 3.6 more bolls per plant, 10.1% less aborted seeds and 10.6% more lint yield when 'Zheda strong restorer' was used as male parent than when 'DES-HAF277' was. Southern and Northern bloting analysis showed that the foreign gsf gene was detectable and highly expressed in 'Zheda strong restorer'.

Abstract:The present study was conducted during 1999-2000 and 2000-2001 to identify the promising parental lines, more especially the restorer lines of hybrids, for further exploitation in hybrid-breeding programme of upland cotton (Gossypium hirsutum L.). Twentyseven F-1 hybrids were developed using of 'Xiang A', 'Zhong 12A', '1047A', '277A', 'NM1 A', 'NM2 A' and 'NM3 A' cytoplasmic male-sterile (CMS) lines as female parent and '6410 R', '501 R', '613-2 R', '277 R' and 'Z811 R' restorers as male parent. These 27 F-1 hybrids along with a standard check ('Zhang-Mian 12') were grown in randomized block design with 3 replications. The magnitude of useful heterosis was greatest for anther starch content (-73.82 to 168.44%), medium for seed-cotton yield (-11.46 to 15.29%) and relatively small for the remaining traits. Out of 27 combinations, 'Xiang A' x '6410R' and 'Xiang A' x '613-2R' were the most desirable combinations for seed-cotton yield, showing more than 14% heterosis over the standard commercial cultivar. Cytoplasmic male-sterile lines ('Xiang A' and 'Zhang 12A') and restorers ('6410 R', '501 R' and '613-2 R') were identified as the most promising because hybrids developed from the combination of these lines gave higher yield than the others. The correlation studies revealed importance of viable pollen percentage, anther weight, anther starch content, self-pollinated boll percentage and lint percentage among the characters studied for imrovement in yield in CMS-based hybrid cotton.
Genetic basis of fertility restoration to cytoplasmic male sterile lines available in China in upland cotton II. Interactive effects between restorer genes and fertility enhancer gene

Abstract:One male-sterile line DES-HAMS277 and its restorer line DES-HAMF 277 both were introduced from USA and had the cytoplasm of Gossypium harknessii Brandagee, were used as checks to study the genetic machanism of fertility restoration to five China cytoplasmic malesterile lines in upland cotton. The results showed that the fertility restoration of five sterile lines is controlled by two independent dominant restorer genes (Rf1 and Rf2). Rf1 is a completely dominant gene and Rf2 a partial dominant gene, i.e., the genetic effect of Rf1 for fertility restoration is stronger than that of Rf2. The fertility restoration can be enhanced by one dominant gene (E), which may be called fertility enhancer gene, and showed no linkage but interaction with Rf1 and Rf2 in the male-sterile cytoplasm. In order to distinguish the genetic effects between restorer genes and fertility enhancer gene, we suggested two different concepts in the fertility restoration of cytoplasmic male-sterile cotton: restoration of fertility and enhance of fertility restoration. The restoration of fertility is the characteristic that restorer genes (Rf1 and Rf2) suppress cytoplasmic male-sterility and produce pollens in plants of (A x R) F1. The enhance of fertility restoration is the degree of fertility restoration and fertility enhancer gene (E) promotes the expression of Rf1 and Rf2 to increase the quantity and viability of pollens in (A x R) F1 plants.

Abstract:Main conclusiondmp1dmp2dmp3 mutants created by CRISPR/Cas9 could trigger maternal haploids in the allotetraploid model plant Nicotiana tabacum L.Double haploid (DH) technology is becoming increasingly important because it can significantly accelerate the breeding process. Haploid induction plays a fundamental role in the production of DH lines. Haploid induction has been realized and applied in diploid plants using DMP genes. However, it has yet to be elucidated whether haploid induction could be established in polyploid plants. In the current study, three homologues of the DMP genes (NtDMP1, 2, and 3) were identified in the allotetraploid plant Nicotiana tabacum, and the encoded proteins localized in the endoplasmic reticulum. Loss-of-function mutations in all three genes triggered maternal haploids with an induction rate of 1.52–1.75%. Compared with wild-type tobacco, the created haploid inducer exhibited differences in pollen vigor and seed germination rate. Furthermore, to rapidly and easily screen haploids, a visible haploid identification system was established based on a powdery mildew resistance phenotype. Findings from this study lay the foundation for the potential application of haploid inducers in allotetraploid plants such as tobacco.

A Cotton Mutant Resist to Glyphosate and Its Utilization in Cotton Heterosis

Abstract:A cotton mutant, R1098 with the trait of resistance to glyphosate, was obtained by the method of somatic cell inducing and continued directional selection. The glyphoste resistance of R1098 was stable, with the lint cotton yield e-quivalent to Sumian 12 and with better fibre characteristies. With the resistance or tolerance to blight and Verticillium wilt, R1098 was an outstanding cotton germplasm. It was shown that, by the genetic studies, the trait of glyphoste resistance of R1098 was controlled by a pair of dominant gene, without the effect of cytoplasm, in that the hybrid FI of the crosses R1098 with the normal upland cotton cultivars was still had the character of the glyophosate, which could be used in cotton heterosis in hybrid seed purity judgement and false hybrid seed e-liminating.

Abstract:The presence of gossypol and its derivatives above the WHO/FAO standards (0.02-0.04%) in cotton seed oil and meal limits its usage as food and feed. To the contrary, the presence of pigment glands filled with gossypol and its derivatives helps to protect cotton plants from phytophageous pests. Thus a desirable cultivar would have glandless seeds on a glanded plant. This paper describes results on the successful introgression of this trait from Gossypium bickii into cultivated upland cotton. Five different tri-specific hybrids (ABH1, ABH2, ABH3, ABH4 and ABH5) were obtained by crossing the amphidiploid F-1 (G. arboreum x G. bickii) with different gland genotypes of G. hirsutum as male parent. The hybrids were highly sterile, and their chromosome configuration at meiosis metaphase 1 (M1) in pollen mother cell (PMC) was 2n = 52 = 41.04 I + 4.54 II + 0.57 III + 0.04 IV. All five hybrids were similar in morphological characters, except for the gland expression and gossypol contents. The hybrid (ABH3) derived from genotype Gl(2)Gl(2)gl(3)gl(3) of upland cotton (a single gene dominant line) had completely introgressed the target trait of G. bickii. While ABH1 and ABH2, which derived from recessive (gl(2)gl(2)gl(3)gl(3)) or dominant (Gl(2)(e)Gl(2)(e) ) glandless upland cotton genotypes, had glandless seeds too, but the density and size of the glands on the plant were reduced significantly.

Abstract:The wild cotton species of G.tomentosum has some outstanding characters such as the higher resistance to pests and tolerance to drought.However,the upland cotton germplasm with the trait has not been obtained through sexual hybridization between G.hirsutum and G.tomentosum due to the distant relationship between the wild species and upland cotton.Protoplast fu-sion technique which depends on the success in the somatic culture of two different species can avoid sexual incompatibility.The callus inducing and somatic embryo germination of G.tomentosum were studied through the regulation of culture conditions such as plant growth regulators(PGRs),PEG,CuSO4and AgNO3.The results indicated that under the same condition the frequency of somatic embryo germination and the plant regeneration ability of G.tomentosum were lower and the plant regeneration was more difficult than Coker 201.But through the regulation of culture conditions,we can get many embryoids and few of them converted to plantlets.The callus of G.tomentosum induced in the medium with the hormone combination of 0.1 mg.L-1 KT + 0.1 mg.L-1 2,4-D held good for cell differentiation,and it was better for proliferation of the embryogenic callus in the medium with 0.5 mg.L-1 IBA + 0.2 mg.L-1 KT.The medium supplied with PEG was not helpful for initiation of somatic embryos.But the medium of MSB5 with 3.5 mg.L-1 CuSO4+0.5 mg.L-1 IBA + 0.2 mg.L-1 KT,or 3 mg.L-1 AgNO3+0.5 mg.L-1 IBA+ 0.2 mg.L-1 KT were good for the proliferation of embryogenic callus and germination of somatic embryo.This report provides an useful technique for somatic culture of the wild cotton species,and also ensures the protoplast fusion feasibly between these wild cotton species and other cotton species to centralize several good traits into a new germplasm.However,it is needed to study further about the various culture factors and conditions for building a quick and high efficient regeneration technique of G.tomentosum.

Abstract:Somatic hybrids were produced through protoplast electrofusion between the tetraploid cotton Gossypium hirsutum L. cv. ZDM-3 and the wild diploid cotton G. klotzschianum . Hybrid plants were generated from 3 out of 24 callus lines that were derived from fused protoplasts. Hybrid plants were initially identified as somatic hybrids by ploidy analysis: the plants from the 3 callus lines had chromosome numbers near to sum of the two parents (78 = 52 + 26). The plants from the 3 lines were subsequently confirmed as hybrids by cytological, molecular, histological and morphological analyses. The morphology of hybrids was distinct from that of the parents, with elongated stigmas and malformed anthers lacking microspores and pollen, leading to male sterility. It is expected that the male sterility resulted from the high number of univalent and irregular multivalent chromosome pairings per meiocyte.

Abstract:CRIPSR/Cas9 gene editing system is an effective tool for genome modification in plants. Multiple target sites are usually designed and the effective target sites are selected for editing. Upland cotton ( Gossypium hirsutum L., hereafter cotton) is allotetraploid and is commonly considered as difficult and inefficient to transform, it is important to select the effective target sites that could result in the ideal transgenic plants with the CRISPR-induced mutations. In this study, Agrobacterium rhizogenes -mediated hairy root method was optimized to detect the feasibility of the target sites designed in cotton phytoene desaturase ( GhPDS ) gene. A. rhizogenes showed the highest hairy root induction (30%) when the bacteria were cultured until OD 600 reached to 0.8. This procedure was successfully applied to induce hairy roots in the other three cultivars (TM–1, Lumian–21, Zhongmian–49) and the mutations were detected in GhPDS induced by CRISPR/Cas9 system. Different degrees of base deletions at two sgRNAs (sgRNA5 and sgRNA10) designed in GhPDS were detected in R15 hairy roots. Furthermore, we obtained an albino transgenic cotton seeding containing CRISPR/Cas9-induced gene editing mutations in sgRNA10. The hairy root transformation system established in this study is sufficient for selecting sgRNAs in cotton, providing a technical basis for functional genomics research of cotton.

Abstract:The present work describes the plant regeneration via somatic embryogenesis in two wild cotton species belonging to G genome: Gossypium nelsonii Fryx and Gossypium australe F Muell. The role of plant hormones and carbohydrates was also evaluated for somatic embryogenesis and somatic embryo development. Normal plants were obtained from G. nelsonii Fryx; abnormal plants and somatic embryos were obtained from G. australe F Muell. The best medium for callus induction for these G genome wild cotton species was MSB5 supplemented with 0.1 mg L−1 KT and 0.1 mg L−1 2,4-D. For embryogenic callus proliferation, the best medium used was MSB5 supplemented with 0.2 mg L−1 KT and 0.5 mg L−1 IBA. The medium MSB5 supplemented with 0.15 mg L−1 KT and 0.5 mg L−1 NAA was used successfully for root initiation and plant growth. In addition, adding CuSO4 and AgNO3 in the callus-inducing and proliferation medium resulted in a number of somatic embryos. Glucose and maltose, the carbon sources in somatic culture, were used for callus induction, but maltose worked even better than glucose for proliferation of embryogenic callus and development of somatic embryos.

Abstract:The clustered, regularly interspaced, short palindromic repeats (CRISPR)/CRISPR associated (Cas)9 protein system has emerged as a simple and efficient tool for genome editing in eukaryotic cells. It has been shown to be functional in several crop species, yet there are no reports on the application of this or any other genome editing technologies in the cotton plant. Cotton is an important crop that is grown mainly for its fiber, but its seed also serves as a useful source of edible oil and feed protein. Most of the commercially-grown cotton is tetraploid, thus making it much more difficult to target both sets of homeologous alleles. Therefore, in order to understand the efficacy of the CRISPR/Cas9 system to target a gene within the genome of cotton, we made use of a transgenic cotton line previously generated in our laboratory that had a single copy of the green fluorescent protein (GFP) gene integrated into its genome. We demonstrate, for the first time, the use of this powerful new tool in targeted knockout of a gene residing in the cotton genome. By following the loss of GFP fluorescence, we were able to observe the cells that had undergone targeted mutations as a result of CRISPR/Cas9 activity. In addition, we provide examples of the different types of indels obtained by Cas9-mediated cleavage of the GFP gene, guided by three independent sgRNAs. The results provide useful information that will help us target important native genes in the cotton plant in future.

High Frequency Plant Regeneration from Protoplasts in Cotton Via Somatic Embryogenesis

Abstract:A highly reproducible system for efficient plant regeneration from protoplast via somatic embryogenesis was developed in cotton ( Gossypium hirsutum L.) cultivar ZDM-3. Embryogenic callus, somatic embryos and suspension culture cells were used as explants. Callus-forming frequency (82.86 %) was obtained in protoplast cultures from suspension culture cells in KM 8 P medium with 0.45 µM 2,4-dichlorophenoxyacetic acid (2,4-D), 0.93 µM kinetin (KIN), 1.5 % glucose and 1.5 % maltose. Protocolonies formed in two months with plating efficiency of 14 %. However, the callus-forming efficiencies from other two explants were low. The calli from protoplast culture were transferred to somatic embryo induction medium and 12.7 % of normal plantlets were obtained on medium contained 3 % maltose or 1 % of each sucrose + maltose + glucose, 2.46 µM indole-3-butyric acid (IBA) and 0.93 µM KIN. Over 100 plantlets were obtained from protoplasts derived from three explants. The regenerated plants were transferred to the soil and the highest survival rate (95 %) was observed in transplanting via a new method.
Breeding, introgression and inheritance of delayed gland morphogenesis trait from Gosspium bickii into upland cotton germplasm

Abstract:A tri-specific hybrid with delayed pigment gland morphogenesis was obtained by crossing the amphidiploid of ( G. arboreum X G. bickii) F 1 and an upland cotton germplasm with pigment gland genotype of Gl 2 Gl 2 gl 3 gl 3 . The tri-speciflc hybrid was a typical interspecific hybrid with high sterile, and the chromosome configuration at meiosis MI of PMC was 2n = 52 = 41.04 I + 4.54 II + 0.57III + 0.04. The crossover value of bivalent was 1.19. Two fertile plants with objective character were obtained in BC 8 population by continuously backcrossing with Gl 2 Gl 2 gl 3 gl 3 as recurrent parent to the tri-specific hybrid, and a new upland cotton germplasm, named ABH-0318, with delayed pigment gland morphogenesis trait was developed through selfing and screening. The pigment gland trait of ABH-0318 was stable, and there were almost no pigment glands observed in the dormant seeds, although there were a few pigment glands confined to cotyledon edges, and the gossypol content in the dormant seeds was 0.017% only, being a typical low gossypol cotton type. However, a large quantity of pigment glands emerged in cotyledons and other main organs of plant after seed germination, and the gossypol contents in the upper parts of the plant were similar to that of ordinary glanded cotton types. Genetic analysis demonstrated that the delayed pigment gland morphogenesis trait of this germplasm was controlled by the interaction of the genes located in two pigment gland loci, Gl 2 and Gl 3 . Among them, the gene located in locus of Gl 2 , derived from Gbickii , was dominance to upland cotton pigment gland alleles, Gl 2 and gl 2 , but was recessive epistatic to another glanded gene Gl 3 which was named Gl 2 b temporarily. While the gene located in the locus of Gl 3 was a recessive gene come from upland cotton.

Abstract:Doubled haploid technology is widely used to accelerate plant breeding, but its use in the important oilseed crop Brassica napus L. is limited because B. napus haploids could only be obtained through in vitro anther or microspore cultures. Recently, maize (Zea mays) lines containing mutations in Domain of unknown function 679 membrane protein (DMP) were used as haploid inducer lines. This new haploid induction mechanism has been extended to several other plants, including the dicots Arabidopsis thaliana, tomato (Solanum lycopersicum), and tobacco (Nicotiana tabacum). Here, we knocked out four BnaDMP genes in the B. napus cultivar Westar using a clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 vector with an enhanced green fluorescent protein expression cassette. Plants with DMP mutations in B. napus in the T0 , T1 , and T2 generations exhibited a haploid induction rate up to 2.53%. These results suggest that targeting BnaDMP could be useful for haploid induction in B. napus.

Abstract:Cotton (Gossypium spp.) is one of the most important fiber crops worldwide. In the last two decades, transgenesis and genome editing play important roles in cotton improvement. However, genotype dependence is one of the key bottlenecks in generating transgenic and gene‐edited cotton plants through either particle bombardment or Agrobacterium‐mediated transformation. Here, we developed a shoot apical meristem (SAM) cells‐mediated transformation system (SAMT) that allowed transformation of recalcitrant cotton genotypes including widely grown Upland cotton (Gossypium hirsutum), Sea island cotton (G. barbadense) and Asiatic cotton (G. arboreum), respectively. Through SAMT, we successfully introduced two foreign genes, GFP and RUBY, into SAM cells of some recalcitrant cotton genotypes, respectively. Within two to three months, transgenic adventitious shoots generated from the axillary meristem zone could be recovered and grown into whole cotton plants. The GFP fluorescent signal and betalain accumulation could be observed in various tissues in GFP‐ and RUBY‐ positive plants, as well as in their progenies, indicating that the transgenes were stably integrated into the genome and transmitted to the next generation. Furthermore, by using SAMT, we successfully generated edited cotton plants with inheritable targeted mutagenesis in the GhPGF and GhRCD1 genes through CRISPR/Cas9‐mediated genome editing, respectively. Together, the established SAMT transformation system here in this study bypasses the embryogenesis process during tissue culture in a conventional transformation procedure and significantly accelerates generation of transgenic and gene‐edited plants for genetic improvement of recalcitrant cotton varieties. Abstract:Heterosis is a common phenomenon in plants and has been extensively applied in crop breeding. However, the superior traits in the hybrids can only be maintained in the first generation but segregate in the following generations. Maintaining heterosis in generations has been challenging but highly desirable in crop breeding. Recent study showed that maternally produced diploid seeds could be achieved in rice by knocking out three meiosis related genes, namely REC8 , PAIR1 , OSD1 to create MiMe in combination with egg cell specific expression of BBM transcription factor, a technology called clonal seeds. Interestingly, there has been very limited reports indicating the feasibility of this approach in other crops.

Abstract:In plants, in vivo haploid induction has gained increasing attention for its significant potential applications in crop breeding and genetic research. This strategy reduces the chromosome number in progeny after fertilization, enabling the rapid production of homozygous plants through double haploidization, contrasting with traditional inbreeding over successive generations. Haploidy typically initiates at the onset of seed development, with several key genes identified as paternal or maternal factors that play critical roles during meiosis, fertilization, gamete communication, and chromosome integrity maintenance. The insights gained have led to the development of efficient haploid inducer lines. However, the molecular and genetic mechanisms underlying these factors vary considerably, making it challenging to create broadly applicable haploidy induction systems for plants. In this minireview, we summarize recent discoveries and advances in paternal and maternal haploid induction factors, examining their current understanding and functionalities to further develop efficient haploid inducer systems through the application of parental factor manipulation.
