Abstract:Fluorescence resonance energy transfer (FRET) nanoprobes based on aggregated induced emission (AIE) molecules are promising for the monitoring of biomolecules in living cells, but this nanoprobe for reactive oxygen species (ROS) has rarely been reported. Here, a novel AIE-based FRET nanoparticles (NPs) is developed for ratiometric detection and imaging of hypochlorite in live cells. Different to common FRET-based nanosensors, the NPs is prepared via facile co-assembly strategy in water using amphiphilic tetraphenylethene (TPE1, energy donor) and thienyl-diketopyrrolopyrmle (NDPP, energy acceptor). This hybrid NPs are uniform spherical, with size of approximately 120 nm. NDPP maintains bright fluorescence inside the hybrid NPs and acts as the reactive unit for hypochlorite in this FRET nanosystem. As a ratiometric fluorescent nanoprobe, the hybrid NPs shows high selectivity to hypochlorite with a detection limit calculated to be 105 nM, thanks to its stable particle size in hypochlorite sensing. Furthermore, the successful ratiometric fluorescent imaging of ClO- in HeLa cells demonstrates that our NPs can effectively pass through the cell membrane and recognize the ClO- in living cells.

Abstract:A fluorescent hydrazine hydrate probe (DMA) based on 1,4-dihydropyridine derivatives was designed and synthesized. The fluorescence emission peak of this probe is in the near-infrared region (667 nm), which has good selectivity to N2 H4 . H2 O and low detection limit (11 nM). Importantly, the probe exhibits aggregation-induced luminescence (AIE) characteristics. In addition, the probe is prepared with a portable test paper to realize the identification of hydrazine hydrate in the solution and the quantitative detection of hydrazine hydrate gas.

Abstract:A series of AIE molecules (BPO, BPM, BPP) with small conjugation successfully designed and synthesized by combining 6-(dodecylcarbamyl)picolinic acid. AIE gels BPO, BPM, BPP were obtained in different solvents, and the self-assembly process of BPM as a representative sample in 1,4-dioxane was characterized by different technologies. It was interesting that BPM exhibited 'turn-on' fluorescence response towards Cd2+ with excellent selective and the low limit of detection (LOD) of 0.66 nM. The formation of complex BPM-Cd at the stoichiometry of 1:1 was confirmed through Job's plot. By introduction the metal-ligand coordination bonds, complex BPM-Cd shows multiple stimuli to polar solvents including H2O, DMF, DMSO, methanol and ethanol with the LODs of 0.0039%, 0.0042%, 0.002%, 0.088% and 0.12% (v/v), respectively. This work provides a new thought of constructing AIE self-assembly systems and multifunctional organic probe with improved sensitivity and selectivity.

Abstract:A two-photon nanoparticle probe was designed and developed based on the principle of intermolecular interaction of the aggregation-induced locally excited emission luminescence mechanism. The probe has the advantages of simple synthesis, convenient use, strong atomic economy, good biocompatibility, and photobleaching resistance. It can produce a specific and sensitive response to formaldehyde, help detect FA in normal cells and cancer cells, and is expected to become a specific detection probe for FA in vitro and in vivo.

Abstract:Aggregation-induced emission (AIE) refers to a unique photophysical phenomenon observed for some specific fluorophores, which are faintly or non-emissive in solution but become highly emissive in solid and aggregated states. In the cases of fluorescent probes, AIE active molecules possess some unparallel advantages over their classical counterparts. On one hand, it allows more AIE active molecules to bind to the target analyte, without worrying about the aggregation-induced fluorescence quenching; thus favors fluorescence detection. On the other hand, the feature of drastic enhancement in fluorescence intensity associating with an aggregation event can be used as a method of quantitative detection. In this paper, a series of representative AIE molecules are demonstrated; their applications as biological probes in sensing of protein, DNA, G4, and chiral amines are highlighted; their working principles and features of these molecules are described. Finally, a brief outlook on the design of new AIE-active molecules and the application of AIE-molecules in relevant fields is forwarded.

Abstract:Nerve agents are one of the most dangerous and toxic chemical warfare agents (CWAs). The rapid detection for them is very important for specialized persons to take action against these substances. In this work, a salicylaldoximate-based aggregation-induced emission (AIE) probe, 2-hydroxy-5-(1,2,2-triphenylvinyl) benzoxime (TPE-D), was rationally designed for this purpose. The AIE fluorescent sensor has a tetraphenylethylene (TPE) unit combined with a salicylaldoximate moiety as the reaction site for a nerve agent simulant, diethyl chlorophosphite (DCP). TPE-D successively undergoes the formation of nitrile and the phosphorylation of phenolic hydroxyl group after the salicylaldoximate moiety reacting with DCP, which will give a significant fluorescence "light-up" response due to the AIE effect within minutes.

Abstract:We report a novel turn-on two-photon fluorescent probe (TPNF) for detecting formaldehyde based on aza-Cope rearrangement and intramolecular charge transfer (ICT). The probe, containing a 1,8-naphthalimide dye with a homoallylamino group, has was shown with high selectivity, sensitivity and low cytotoxicity and it was capable of detecting formaldehyde in live cells. Moreover, a time-dependent density functional theory (TD-DFT) calculation was performed to demonstrate the turn on mechanism of the probe via ICT effect. Finally, a zebrafish larvae imaging experiment reveals that TPNF displayed a significant signal change when treated with FA in vivo. Abstract:Two fluorescent probes, Y 1-2 were synthesized from 2-acetonaphthone, 4-acetylbiphenyl, and phenyl hydrazine by Vilsmeier-Haack reaction and Knoevenagel condensation. Their recognition efficacies for N 2 H 4 were tested by UV–visible absorption spectroscopy and fluorescence emission spectroscopy. The recognition mechanism were studies by density-functional theory calculations, and the effect of pH on N 2 H 4 recognition was also studied. The results showed that the probe Y 1-2 has high selectivity and a low detection limit for N 2 H 4 , and the recognition of N 2 H 4 can be accomplished at physiological pH. The probes have had obvious aggregation-induced luminescence effect, large Stokes shift, high sensitivity, and can be successfully applied to live cell imaging.

Abstract:Aggregation-Induced Emission (AIE) active compounds create tremendous interest due to their unique property of restriction of intramolecular rotation which leads to the introduction of luminescence property within itself. Here we have incisively developed a novel AIE active fluorescence probe H2L [(E)-2,4-dibromo-6-(((5-chloropyridin-2-yl)imino)methyl)phenol] with Al(III) and Fe(III) along with aqueous and gas phase picric acid detection ability. UV, fluorescence, and DLS `studies are implemented to give solid proof of the AIE active behavior of the probe. Optical, SEM, and TEM images very nicely prove the morphological adaptability of the probe with structural alteration with the variation of water percentage. It shows AIE active behavior in DMSO – H2O (3:7 v/v) solutions. One remarkable and noteworthy observation is that the ligand's morphology and emission intensity are altered by the presence of water with varying percentages (10-70) when the AIE activity is taken into account. This results in the appearance of two distinct optical color changes and emission intensity changes within the 10-70% water presence range. The probe becomes emissive in the solid state due to the intermolecular H-bonding and keto-enol tautomerism. Interestingly in 9:1 (v/v) DMSO – H2O solvent H2L can selectively detect Al(III) through turn-on fluorescence. In the AIE active medium, it can selectively detect Fe(III) and picric acid by turning off fluorescence. The ESIPT process in the solid state leads to molecular aggregation resulting in the introduction of strong luminescence properties within the probe and impressively the solid state fluorescence is used to detect picric acid in the vapor phase. The LOD is determined as 6.02 × 10-9 (M) for Al(III) 7.5 × 10-9 (M) for Fe(III) and 6 × 10-8 (M) for picric acid. During Al(III) sensing the ESIPT off and "CHEF" -on jointly operate leading to emission enhancement whereas during Fe(III) sensing and picric acid sensing the destruction of the aggregation state is mainly responsible for the reduction of emission intensity of the probe. Impressively the probe can detect picric acid in the solid state also. The job plot analysis from the Uv spectral data and the MASS spectral analysis supports the 1:1 stoichiometry complexation between the probe and the analytes. Moreover, the proposed analytical system with its clear AIEE mechanism demonstrates a promising prognosis for on-site practical applications. Finally, the probe's ability to detect and quantify Al(III) from the market-available drug samples has been checked.

Abstract:Although a set of functional molecules with the D-π-A structure has been explored as optical probes for the detection of target analytes, it remains a great challenge to elaborately design a single probe for distinguishing different analytes by their intrinsic oxidation or reduction capabilities and thus to generate distinct optical responses. Here, a unique TCF-based probe (DMA-CN) containing two unsaturated double bonds in the π-conjugation bridge and TCF with different reaction activities that could be cut off by KMnO4 and NaClO in varying degrees was developed, causing remarkably distinguishable responses for both fluorescence and colorimetric channels to discriminate KMnO4 and NaClO from each other. The fluorescence and colorimetric limits of detection (LODs) of the proposed DMA-CN toward KMnO4 were calculated as 60 and 91 nM, respectively, while those for NaClO were 13.3 and 214 nM, and all the optical signal change can be observed within 1 s with good specificity. Based on the proposed probe design strategy, a well-fabricated test strip was proven to be promising for the rapid, in-field detection and risk management. We expect that the present probe design methodology would provide a powerful strategy for efficient probe exploration, especially for discriminating the substances with similar oxidizing properties.

Abstract:The development of probes with selectivity and prompt detection of aldehydes molecules is of great importance for protecting human health and public security. Herein, a system based on ethylenediamine (EDA) functionalized and Eu 3+ -doped UiO-66, namely EDA-Eu 3+ @UiO-66, was designed to detect formaldehyde molecules. Based on the "antenna effect" of lanthanide elements, UiO-66 transfers the absorbed energy to Eu 3+ ions and emits characteristic fluorescence belonging to Eu 3+ . By using the fluorescence peaks of UiO-66 and Eu 3+ respectively, a ratiometric fluorescence sensing probe can be constructed. Formaldehyde molecules react with the –NH 2 on the surface of EDA-Eu 3+ @UiO-66 through an aldehyde-amine condensation reaction and connect to the functionalized surface of UiO-66. Due to the absorption of excitation light energy by formaldehyde molecules, the energy transfer efficiency from UiO-66 to Eu 3+ ions is reduced, resulting in the fluorescence quenching of EDA-Eu 3+ @UiO-66, thus achieving selective detection of formaldehyde. The fabricated sensing platform successfully detected residual formaldehyde in frozen shrimp tail samples. The system was also used to respond to formaldehyde vapor, and a significant fluorescence quenching effect was observed. This strategy provides a sensitive, selective, and reliable method for the visual sensing of formaldehyde.

Abstract:Herein, we designed a potent ClO − probe named ETMC , which exhibited aggregation-induced emission (AIE) characteristic in solid state and water/EtOH binary solvent. Probe ETMC exhibited an colormetric from yellow to blue and ratiometric response to ClO − with high selectivity and sensitivity, and a low detection limit of 3.66 × 10 −7 M. Moreover, probe ETMC could be applied to visibly and quantitatively detect the different concentraction of ClO − in test paper. Importantly, probe ETMC could effectively detect endogenous and exogenous ClO − in living A549 cells by an outstanding ratio signal change. Also, the sensing mechanism of probe ETMC for ClO − was confirmed by FT-IR, 1 H NMR, HRMS, DLS, TEM and DFT calculation.

Abstract:Zn2+ and ClO− may be interrelated with a variety of pathologies, their simultaneous measurement is crucial for the disease diagnosis and environmental protection. In this work, we synthesized an independent AIE probe HNTE based on the one step reaction. The probe HNTE displayed a distinctive fluorescence color change from deep yellow to blue for ClO- and green for Zn2+. More importantly, the probe HNTE could simultaneously colormetric detecting endogenous and exogenous ClO- and Zn2+ in living cells via the blue and green channels.

Abstract:Endogenous H2S and HClO play vital roles in regulating the redox status in vivo, and are involved in many pathophysiological events. Accordingly, accurate sensing and imaging of intracellular H2S and HClO have become greatly in demand. However, conventional double-response fluorescent probes for these two substances suffered from the notorious aggregation-caused quenching effect and required multiple excitations to achieve discrimination determination. In this study, we firstly constructed an AIE-active fluorescent probe with three potential reaction sites for individual and continuous detection of H2S and HClO using a single excitation wavelength. The multiple-site probe exhibited distinctive fluorescence behaviors to H2S and HClO with fast response (H2S: 12 min; HClO: 1.8 min), well-separated emission peaks (88 nm), high sensitivity and good selectivity due to H2S-induced thiolysis of 2,4-dinitrophenyl ether assisted by an adjacent aldehyde and HClO-triggered sulfur oxidation. The corresponding recognition mechanisms were fully confirmed by HRMS, 1H NMR and control experiments. Moreover, the probe was successfully utilized for separate and continuous imaging of intracellular H2S and HClO in different fluorescent channels, indicating its potential for further study the biological functions of H2S and HClO in living systems.

Abstract:Polyamine plays the important role in human body. Here, the convenient synthesis of AIE material 2-(pyren-1-ylmethylene) malononitrile (L) was capable of fluorescent recognition for 1, 3-diaminopropane in gas-liquid-solid three-phase with good selectivity and high sensitivity. And it's interesting that other amines did not interfere with the recognition between L and 1,3-diaminopropane. The proposed sensing mechanism was supported by Job's plots, 1H NMR titration, FT-IR, electron-spray ionization, and density functional theory calculation analysis. The limits of detection were estimated as 1.34×10−8 M. The self-made test strips and silica gel plates realized the easy detection of 1, 3-diaminopropane. L-1, 3-diaminopropane possessed the potential for fingerprint recognition based on AIEE in the silica gel plates developed within this study. Notably, bioimaging in seeds, plant tissues, Caenorhabditis elegans, and mice revealed that L could be suitable as an effective sensor for detecting 1, 3-diaminopropane in living systems with the strong "turn-on" fluorescence. Importantly, L showed good biocompatibility and could effectively recognize 1, 3-diaminopropane and reveal its location with colored AIEE fluorescence in HeLa cells.

Abstract:By combining leucine (Leu) and tetraphenylethene (TPE), a pH-sensitive aggregation induced emission (AIE) probe TPE-Leu was developed. The aliphatic amine in TPE-Leu was more easily protonated under acidic conditions, which made TPE-Leu more water soluble. Therefore, the protonated AIE probe showed weak fluorescence under acidic conditions. When the pH was changed to basic conditions, it showed strong fluorescence due to the hydrophobic nature of TPE-Leu. We demonstrated that the probe showed high selectivity toward pH changes with the coexistence of other potential species such as metal ions, redox agents, and biomolecules. In contrast, TPE-NH2 did not exhibit obvious pH-sensitive properties. Moreover, TPE-Leu was further utilized to develop a sensitive and selective sensing platform for urease and acetylcholinesterase (AChE) detection. The current study not only provides a new strategy for designing pH-sensitive fluorescent probes for bioassays but also broadens the applications of AIE probes.

Abstract:In the field of optical sensors, small molecules responsive to metal cations are of current interest. Probes displaying aggregation-induced emission (AIE) can solve the problems due to the aggregation-caused quenching (ACQ) molecules, scarcely emissive as aggregates in aqueous media and in tissues. The addition of a metal cation to an AIE ligand dissolved in solution can cause a “turn-on” of the fluorescence emission. Half-cruciform-shaped molecules can be a winning strategy to build specific AIE probes. Herein, we report the synthesis and characterization of a novel L-shaped fluorophore containing a benzofuran core condensed with 3-hydroxy-2-naphthaldehyde crossed with a nitrobenzene moiety. The novel AIE probe produces a fast colorimetric and fluorescence response toward zinc (II) in both in neutral and basic conditions. Acting as a tridentate ligand, it produces a complex with enhanced and red-shifted emission in the DR/NIR spectral range. The AIE nature of both compounds was examined on the basis of X-ray crystallography and DFT analysis.

Abstract:A novel strategy is proposed to boost the sensitivity of aggregation‐induced emission (AIE) probe by regulating van der Waals interactions between the probe and perchlorate through optimizing the alkyl chain length. The present strategy is a promising candidate for detecting trace hazardous substances and AIE‐based functional material conceptualization. The precise regulation of interactions provided by aggregation‐induced emission (AIE) probes is of considerable significance for improving the sensing performance in the field of on‐site detection. Here, a highly sensitive perchlorate detection probe was designed by precisely modulating the van der Waals interactions by adjusting the length of the alkyl chain. The optimized AIE probe demonstrated superior perchlorate detection performance owing to its strong van der Waals interactions with perchlorate, including a low detection limit (53.81 nM), rapid response (<5 s), and excellent specificity even in the presence of 16 interfering anions. In addition, a hydrogel‐based device loaded with the probe was constructed to achieve ultrasensitive recognition of perchlorate particles with a detection limit as low as 15 fg under a fluorescence microscope. Moreover, the practicality of the probe was further verified by employing a sensing chip in a portable detector, and thus the probe has been proven to be highly promising for trace perchlorate monitoring in real scenarios. We expect the present study to be of great value for the efficient design of high‐performance fluorescent probes.

Abstract:Formaldehyde (FA) is a harmful substance commonly found in the environment, so effective testing is essential. Consequently, we designed and synthesized a novel fluorescent probe, which is based on covalent post-synthetic modification (PSM) of UiO-66-NH2 by naphthylamide (NI). The combination of the metal-organic frameworks (MOFs) and the covalent PSM makes the probe with large amount of hydrazine group exposed on the surface, which results excellent FA sensing performance like an ultrafast response time (20 s) and a low detection limit of 0.167 μM. The presence of covalent bonds can lock the fluorophore and prevent strong π-π interactions to exhibit aggregation-induced emission (AIE). Further we confirmed that such probe can be applied to real water samples and portable test strip experiment. In addition, experimental evidence such as fluorescence lifetime decay curves suggest that the main mechanism of fluorescence quenching is Förster resonance energy transfer (FRET).
