Abstract:Permanent male sterilization has been recognized as useful tools for the development of neuter experimental animals and fattening livestock, as well as efficient control of pet overpopulation. Traditional routes such as surgical ways, chemical injections, and anti-fertility vaccines have addressed these crucial problems with idea outcomes. However, these routes usually bring out serious pain and infection towards animals, as well as induce long-term adverse reaction and immune suppression. Thus, a convenient, but non-surgical strategy for male sterilization under a mild manner is highly desirable. Here, for the first time, we demonstrate a novel platform for male sterilization by using single-layer WO2.72 nanosheets as smart photo-responsive sterilants. Upon a 980 nm irradiation, these nanoagents can possess intrinsic NIR-induced hyperthermia and sensitize the formation of singlet oxygen due to the cooperation of photothermal and photodynamic effects. Mechanism of cellular injury can be attributed to the denaturation of protein and apoptosis-related death. Moreover, long-term toxicity and possible metabolism route after testicular injection are discussed, indicating the neglectable systemic toxicity and high bio-compatibility of our nanoagents. Overall, our strategy can extremely overcome the shortcomings in various routine routes and suggest the new biological application of nanomaterials.
Ferritin-nanocaged aggregation-induced emission nanoaggregates for NIR-II fluorescence-guided noninvasive, controllable male contraception

Abstract:Controllable contraception in male animals was demonstrated through the utilization of gold nanorods' photothermal effect to accomplish mild testicular hyperthermia. However, the challenges arising from testicular administration and the non-biodegradability of nanoparticles hinder further clinical implementation. Therefore, a straightforward, non-invasive, and enhanced contraception approach is required. This study explores the utilization of human heavy chain ferritin (HFn) nanocarriers loaded with aggregation-induced emission luminogens (AIEgens) for noninvasive, controllable male contraception guided by Near-Infrared-II (NIR-II) fluorescence imaging. The HFn-caged AIEgens (HFn@BBT) are delivered via intravenous injection and activated by near-infrared irradiation. Lower hyperthermia treatment induces partial damage to the testes and seminiferous tubules, reducing fertility indices by approximately 100% on the 7th day, which gradually recovers to 80% on the 60th day. Conversely, implementation of elevated hyperthermia therapy causes total destruction of both testes and seminiferous tubules, leading to a complete loss of fertility on the 60th day. Additionally, the use of AIEgens in NIR-II imaging offers improved fluorescence efficiency and penetration depth. The findings of this study hold significant promise for the advancement of safe and effective male contraceptive methods, addressing the need for noninvasive and controllable approaches to reproductive health and population control.

Heterostructured nanorod array with piezophototronic and plasmonic effect for photodynamic bacteria killing and wound healing

Abstract:Bacteria induced infectious diseases have threated the lives and health of millions people each year. Nanosized titanium dioxide (TiO2) have been developed for photodynamic bacterial killing with minimal drug resistance, but the efficiency was restricted by their large band gap, limited light-absorption region, and rapid electron-hole recombination. In this work, we rationally fabricated a TiO2/BTO/Au multilayered coaxial heterostructured nanorod array by inserting a ferroelectric semiconductor barium titanate (BaTiO3) nanolayer between TiO2 nanorod and gold nanoparticles (AuNPs). The TiO2/BTO/Au heterostructure showed greatly enhanced reactive oxygen species (ROS) (superoxide (O2•-) and hydroxyl radicals (•OH)) generation, and incident photo-electron conversion efficiency (IPCE) in UV/visible light region. On the basis of experimental observations, the detailed photodynamic mechanism of the enhanced ROS generation was proposed, mainly ascribed to the piezophototronic effect and plasmonic property of the nanorod array. The nanorod array was used as an antibacterial coating to kill gram-negative bacterium E. coli and gram-positive bacterium S. Aureus with an antibacterial efficiency up to 99.9% under simulated sunlight. It also showed an efficient promotion of infectious wound regeneration in mice with S. aureus infected dermal wounds.
Construction of a 980 nm laser-activated Pt(II) metallacycle nanosystem for efficient and safe photo-induced bacteria sterilization

Abstract:Although metal-based chemical agents have demonstrated promising bacteriostatic effects in phototherapy, their short excitation/emission wavelengths and inadequate phototherapy efficiencies make their application in vivo difficult. We therefore synthesized a novel Pt(II) metallacycle ( Pt1110 ) that can be activated with a 980 nm laser for photodiagnosis/treatment in deep tissue. We found that Pt1110 significantly improved photothermal conversion (95% improvement) and 1 O 2 generation (Φ Δ 75% increase) compared to the ligand itself 1 and was well capable of light-induced sterilization under safe laser irradiation (0.72 W/cm 2 ). In addition, Pt1110 has little to no toxicity to cells. After incorporated into liposome, Pt1110 NPs was effective in wound healing in infection and keratitis models upon laser irradiation, which was accurately observed by NIR-II fluorescence imaging. This novel metal-coordinated supramolecular material has a potential to become a universal platform for phototherapy in deep tissue.

Abstract:Wound infections caused by multidrug‐resistant (MDR) bacteria are hard to treat because of tolerance to existing antibiotics, repeated infection, and concomitant inflammation. Herein, zinc atom–doped g‐C3N4 and Bi2S3 nanorod heterojunctions (CN–Zn/BiS) are investigated for disinfection under near‐infrared light (NIR). The photocatalysis of CN–Zn/BiS is enhanced because of efficient charge separation during the interface electron field and increased oxygen adsorption capacity. Then, 99.2% antibacterial efficiency is shown toward methicillin‐resistant Staphylococcus aureus (MRSA) and 99.6% toward Escherichia coli under 10 min NIR irradiation. Meanwhile, a strategy for the combination of lapsed β‐lactam antibiotics with the photosensitizer CN–Zn/BiS is provided to kill MRSA by NIR without observable resistance, suggesting an approach to solve the problem of bacterial infection with NIR light penetrability and for exploiting new anti‐infection methods. The CN–Zn/BiS nanocomposite can also regulate genes and the inflammatory response through inflammatory factors (IL‐1β, IL‐6, TNF‐α, and iNOS) in vivo to accelerate tissue regeneration and thereby promote wound healing.
Achieving a new controllable male contraception by the photothermal effect of gold nanorods.

Abstract:During the process of human civilization, owning household pets has become increasingly popular. However, dogs and cats may be reservoirs or vectors of transmissible diseases to humans. Confronted with the overpopulation of pets, traditional contraception methods, surgical methods of sterilization, for animals are used, namely, ovariohysterectomy and orchidectomy. Therefore, a simple, nonsurgical, controllable, more effective and less expensive contraception method is highly desirable. In this study, we show that in situ testicular injection of methoxy poly(ethylene glycol)-modified gold nanorods with near infrared irradiation in male mice can achieve short-lived or permanent male infertility. In a lower hyperthermia treatment, the morphology of testes and seminiferous tubules is only partly injured, and fertility indices are decreased to similar to 10% at day 7, then recovered to 50% at day 60. In a higher hyperthermia treatment, the morphology of testes and seminiferous tubules are totally destroyed, and fertility indices are decreased to 0 at day 7. Overall, our results indicate a potential application of plasmonic nanomaterials for male contraception.

Abstract:The current trend in tumor research is shifting from monotherapy to multimodal therapy. However, how to achieve on-demand drug delivery and minimize the invasiveness of treatment are still big challenges. Herein, we present a detachable microneedles (MNs) system, which consists of polycaprolactone (PCL) needles and polyvinylpyrrolidone/poly (vinyl alcohol) substrate, to build an implanted drug depot for on-demand photothermo-chemotherapy. Owing to the dissolvability of the substrate, detachable MNs can intradermally implant PCL needles loaded with photothermal conversion agent Prussian blue nanocubes (PB NCs) and chemotherapeutics doxorubicin hydrochloride (Dox center dot HCl). Once near-infrared light irradiates, PB NCs could translate light to local regional hyperthermia, which not only ablates cancer cells but also meltPCL to accelerate the diffusion of Dox center dot HCl. These MNs displayed a stable and repeatable photothermal effect under NIR irradiation. The ex vivo experiments using isolated swine skin demonstrated the as needed Dox center dot HCl delivery triggered by NIR light. Moreover, the robust antitumor efficacy of the MN system was proved in KB tumor-bearing nude mice under three timed NIR irradiation. Therefore, the developed detachable MNs which could build implanted "arsenal" for on-demand photothermo-chemotherapy have a bright future in tumor suppression.

Abstract:The widespread use of antibiotics leads to the increasing drug resistance of bacteria and poses a threat to human health. Therefore, there is an urgent need to develop new antibacterial strategies. Herein, based on the good photothermal properties of Copper sulfide (CuS) nanoparticles under near infrared (NIR) laser, we developed a NIR-II window triple-mode synergetic antibacterial cCuS (cationic CuS) @Vancomycin (Van) nanoplatform. In the proposed nanoplatform, the positive charge on the surface makes cCuS@Van nanoplatform show better bacterial uptake and membrane damage; vancomycin induces chemical sterilization and provides a targeting effect to the nanoplatform; combined with the strong photothermal effect and deep tissue penetration at the excitation of 1064 nm laser, cCuS@Van nanoplatform can effectively kill bacterial. The photothermal conversion efficiency of the nanoplatform can reach 49.12 % and in vitro experiments show a sterilizing rate of more than 99.5 % to staphylococcus aureus (S. aureus) at the concentration of 3.0 μM, which also demonstrated the synergistic effect of cCuS@Van nanoplatform. In addition, low cytotoxicity to human cells conforms the good biocompatibility of the as-prepared cCuS@Van nanoplatform, which endows it a good application prospect in the field of antibacterial, such as wound healing and implant sterilization.

NIR-driven Intense UV Upconversion Films for Non-Contact High-Efficiency Prevention of Bacterial Infections

Abstract:Bacterial infections on wounds imposes a significant limitation to the wound healing. Although the conventional drug antibacterial dressing is employed for hindering bacterial infection, the inevitable microbial resistance and secondary damage severely limit the quality and speed of wound healing. Herein, the NIR-driven intense UV upconversion (UC) BaLu2F8:80%Yb3+/0.5%Tm3+ microcrystals are, for the first time, designed and fabricated into solid sterilization film to protect the wounds from infections. Equipped with excessive Yb3+ sensitizers, the emission of Yb3+/Tm3+ co-doped monoclinic BaLu2F8 matrix achieve a 175-fold enhancement at the UV band. The UV UC microcrystals was further fabricated into a NIR-to-UV sterilization film with a diameter of 1.5 cm and a thickness of 150 μm using solution casting method, which can efficiently destroy 97% of E. coli with a NIR irradiation of 5 minutes. Due to the outstanding penetrability of NIR irradiation, the sterilization film under 4 layers gauze dressing can be activated to produce UV photons. The in-vivo mouse tests demonstrate the well-designed NIR-to-UV sterilization film can speed up the wound healing by high-efficient inhibiting E. coli infection under the irradiation of 980 nm NIR laser. The non-contact NIR-driven UV sterilization film require no more drugs and dressing exchange, thus entirely eliminating the effect of microbial resistance and secondary damage from conventional drug dressing. Our work provides a new alternative for constructing highly-efficiency sterilization dressing through the utilization of NIR-to-UV optical conversion materials.
Alternative Copper-Based Single-Atom Nanozyme with Superior Multienzyme Activities and NIR-II Responsiveness to Fight against Deep Tissue Infections

Abstract:Nanozymes are considered to represent a new era of antibacterial agents, while their antibacterial efficiency is limited by the increasing tissue depth of infection. To address this issue, here, we report a copper and silk fibroin (Cu-SF) complex strategy to synthesize alternative copper single-atom nanozymes (SAzymes) with atomically dispersed copper sites anchored on ultrathin 2D porous N-doped carbon nanosheets (CuN x -CNS) and tunable N coordination numbers in the CuN x sites ( x = 2 or 4). The CuN x -CNS SAzymes inherently possess triple peroxidase (POD)-, catalase (CAT)-, and oxidase (OXD)-like activities, facilitating the conversion of H 2 O 2 and O 2 into reactive oxygen species (ROS) through parallel POD- and OXD-like or cascaded CAT- and OXD-like reactions. Compared to CuN 2 -CNS, tailoring the N coordination number from 2 to 4 endows the SAzyme (CuN 4 -CNS) with higher multienzyme activities due to its superior electron structure and lower energy barrier. Meanwhile, CuN x -CNS display strong absorption in the second near-infrared (NIR-II) biowindow with deeper tissue penetration, offering NIR-II-responsive enhanced ROS generation and photothermal treatment in deep tissues. The in vitro and in vivo results demonstrate that the optimal CuN 4 -CNS can effectively inhibit multidrug-resistant bacteria and eliminate stubborn biofilms, thus exhibiting high therapeutic efficacy in both superficial skin wound and deep implant-related biofilm infections.

Abstract:A piezo‐augmented and photocatalytic nanozyme integrated microneedle is developed for rapid treatment of skin wound infection and inflammation. Apart from the current nanozymes that face uncontrollable catalytic activity, low efficiency, and extended reaction time, this nanozyme can be simply stimulated by ultrasound irradiation to produce ROS for antibacterial purposes, and later can be stimulated by visible light to generate hydrogen gas for anti‐inflammatory treatment. Wound infection is arguably the most common, and potentially the most devastating, complication of the wound healing process. The ideal treatment strategy has to eliminate bacteria, alleviate inflammation, and promote wound healing and skin formation. Herein, a multifunctional heterostructure is designed consisting of ultrasmall platinum–ruthenium nanoalloys and porous graphitic carbon nitride C3N5 nanosheets (denoted as PtRu/C3N5), which concurrently possesses piezoelectric enhanced oxidase ‐mimic nanozyme activity and photocatalytic hydrogen gas production capacity. Moreover, these hybrid nanotherapeutics are integrated in natural hyaluronic acid microneedles, which exhibit almost 100% broad‐spectrum antibacterial efficacy against multiple bacterial strains in vitro and in vivo within 10 min ultrasound treatment, and effectively inhibit inflammation reactions after 1 h visible light irradiation, promising for accelerating the cutaneous wound healing in the bacterial infected mice. This work highlights a competitive strategy for development of all‐in‐one antibacterial and anti‐inflammatory therapies.

Wireless Powered Microwave-Light Conversion Platform with Dual-Stimulus Nanoresponder Coating for Deep-Seated Photodynamic Therapy

Abstract:Traditional external field-assisted therapies, e.g., microwave (MW) therapy and phototherapy, cannot effectively and minimally damage eliminate deep-seated infection, owing to the poor penetrability of light and low reactive oxygen species (ROS) stimulation capability of MW. Herein, an implantable and wireless-powered therapeutic platform (CNT-FeTHQ-TS), in which external MW can be converted into internal light via MW wireless-powered light-emitting chips, is designed to eradicate deep-seated tissue infections by MW-induced deep-seated photodynamic therapy. In application, CNT-FeTHQ-TS is implanted at internal lesions, and the chip emits light under external MW irradiation. Subsequently, CNT-FeTHQ coating in the platform can respond to both MW and light simultaneously to generate ROS and MW-hyperthermia for rapid and precise sterilization at focus. Importantly, MW also improves the photodynamic performance of CNT-FeTHQ by introducing vacancies in FeTHQ to facilitate the photoexcitation process and changing the spin state of electrons to inhibit the complexation of photogenerated electron-hole pairs, which were confirmed by simulation calculations and in situ MW-irradiated photoluminescence experiments. In vivo, CNT-FeTHQ-TS can effectively cure mice with Staphylococcus aureus infection in dorsal subcutaneous tissue. This work overcomes the key clinical limitations of safe energy transmission and conversion for treating deep-seated infections.
Rapid and Highly Effective Noninvasive Disinfection by Hybrid Ag/CS@MnO2 Nanosheets Using Near-Infrared Light.

Abstract:A bacterial infection on the surface of medical apparatus and instruments as well as artificial implants is threatening human health greatly. Antibiotics and traditional bacterial-killing agents, even silver nanoparticles, can induce bacterial resistance during long-term interaction with bacteria. Hence, rapid surface sterilization and prevention of bacterial infection in the long term are urgent for biomedical devices, especially for artificial implant materials. Herein, a hybridized chitosan (CS), silver nanoparticles (AgNPs), and MnO2 nanosheets coating was designed on the surface of titanium plates, which can ensure the implants a rapid and highly effective antibacterial efficacy of 99.00% against Staphylococcus aureus ( S. aureus) and 99.25% against Escherichia coli ( E. coli) within 20 min of 808 nm near-infrared light (NIR) irradiation. The exogenous NIR irradiation can trigger the MnO2 nanosheets to produce enough hyperthermia within 10 min, which can combine with a low concentration of prereleased Ag+ from the coating to achieve superior antimicrobial efficacy through synergistic effects. In contrast, either prereleased Ag ions or a photothermal effect alone can achieve much lower antibacterial efficiency under the same concentration, i.e., 24.00% and 30.01% for the former and 30.00% and 42.54% for the later toward S. aureus and E. coli, respectively. The possible cytotoxicity of coatings could be eliminated owing to the low concentration of AgNPs and chitosan encapsulation. Thus, the novel bifunctional coating Ag/CS@MnO2 can exhibit great potential in deep site disinfection of Ti implants through the synergy of prereleased Ag ions and a photothermal effect within a short time.

Abstract:Herein, a collaborative precise antibacterial wound healing therapy nanoplatform integrating drug-food homologous bioactive molecule (cinnamaldehyde, CA) with photothermal therapy (PTT) is presented. Copper-gallic acids-cinnamaldehyde-polydopamine nanorods (Cu-GA-CA-PDA NRs) with near-infrared light (NIR)-controlled CA release property are fabricated, which also integrate CA and photothermal synergistic sterilization, as well as antioxidant, anti-inflammatory, and anti-infection capacities. The characteristics of NIR-mediated CA release and photothermal response of Cu-GA-CA-PDA NRs support their excellent sterilization performance in vitro/in vivo. In addition, under the guidance of NIR, Cu-GA-CA-PDA NRs can hinder the formation of inflammatory cells, reduce oxidative stress damage, accelerate the regeneration of skin tissues in S. aureus-infected wound sites, and achieve the goal of promoting wound healing. Therefore, NIR-mediated Cu-GA-CA-PDA NRs with multifunctional biological activities provide a highly competitive strategy for curing bacteria-infected wounds.

Abstract:Spike‐like [Cu(OH)2@Au@Co(OH)2] nanoparticles (CAC NPs) display near‐infrared (NIR) irradiation‐amplified oxidase and peroxidase biocatalytic activities that increase ROS generation and promote bactericidal effects in vitro and in vivo. Antibiotic‐independent artificial nanozymes exhibit excellent biocatalytic activity, and can generate sufficient reactive oxygen species (ROS) for potential applications in the field of anti‐infective therapy. However, single nanozymes often catalyze the production of ROS with low yield and low utilization, resulting in low antibacterial activity. To address the above refractory concern, a spike‐like surface nanoparticles (CAC NPs) platform is built that possesses intrinsic dual enzyme‐like (oxidase and peroxidase‐like) activities and photothermal conversion capacity with near‐infrared irradiation (NIR‐I) to quickly capture and effectively eliminate the bacteria and biofilm. Upon NIR‐I irradiation, CAC NPs have exerted an excellent biocatalytic antibacterial effect through a hyperthermia‐amplified dual enzyme‐like activities strategy. As predicted, the results show CAC NPs can eradicate bacteria in the subcutaneous tissues of mice and promote the healing of inflamed tissues with no significant side effects in vivo. Collectively, this work reveals the potential of NIR‐assisted dual enzyme‐like activities for antimicrobial therapy and provides broad prospects for antimicrobial drugs and strategies.

Construction of Versatile Fibroin/nanozyme Hybrid Microneedles with Controllable Phototherapeutic Sterilization Property Against Periodontitis

Abstract:Periodontitis is a highly prevalent disease that affects up to 90% of the worldwide population. The highly dynamic oral environment, tissue-teeth “pocket” structure, and antimicrobial resistance pose challenges to periodontitis local treatments, which urgently need to be upgraded with advanced biomaterials and novel therapeutics. Herein, a new treatment strategy for periodontitis is proposed by synergistic nanozyme catalyzed phototherapeutic/chemodynamic sterilization and microneedles (MNs) in situ delivery platform. A biodegradable versatile photothermal microneedles (MN-SF/CuS) patch was constructed by the in situ growth of monodispersed ultrasmall CuS nanozyme on silk-fibroin (SF) biomolecule and then hybridization in methacrylated hyaluronic acid (HAMA). The synthetic SF/CuS nanozyme endows microneedles with amplifies phototherapeutic/chemodynamic sterilization, excellent mechanical properties, and promotes periodontal tissue regeneration. The constructed MN-SF/CuS can be conveniently and minimally invasively inserted deeply into periodontal tissue without being affected by saliva, where it delivers thermal action, reactive oxygen species, and copper to the specified depth, achieving rapid sterilization and inhibition of bacterial growth. Meanwhile, MN-SF/CuS achieves on-demand phototherapy effect without damaging surrounding normal tissues. A preclinical large animal model confirmed that the MN-SF/CuS provided excellent irreversible antibacterial activity and promoted tissue regeneration, substantially improving the tolerance and compliance of periodontal treatment.
Nanostructure‐Mediated Photothermal Effect for Reinforcing Physical Killing Activity of Nanorod Arrays

Abstract:A novel antibacterial strategy utilizes nanostructured "light traps" mediated PTT to enhance the physical puncture of bacteria by nanorod arrays, effectively eliminating biofilm infections without requiring additional photosensitivities. Combined with nitric oxide therapy, this method effectively prevents MRSA infection on the surface of titanium implants, providing a promising approach for sustainable antibacterial implant surfaces. The physical killing of bacteria based on surface topography has attracted much attention due to the sustainable and safe prevention of biofilm formation. However, the antibacterial efficiency of biomedical implants derived solely from nanostructures or microstructures is insufficient to combat bacteria against common infections, such as methicillin‐resistant Staphylococcus aureus with thick cell walls. Herein, photothermal therapy is carried out in the presence of nanorod arrays to mitigate infection of biomedical implants. Different from traditional photothermal therapy relying on a photosensitizer, the photothermal effect is mediated by light traps rendered by the nanorod arrays, and consequently, the photosensitizer is not needed. Finite element simulations and experiments are performed to elucidate the light‐to‐thermal conversion mechanism. This photothermal platform, in conjunction with thermosensitive nitric oxide therapy, is applied to treat titanium implant infection. The nanostructure‐mediated photothermal effect destroys bacterial cell walls by inhibiting peptidoglycan synthesis and increasing the membrane permeability by affecting fatty acid synthesis. Furthermore, the nanorods synergistically puncture the bacterial membrane easily as demonstrated by experiments and transcriptome analysis. The results provide insights into the development of efficient antibacterial treatment of implants by combining nanostructures and photothermal therapy.

Abstract:Recently, bacterial infections have become a global crisis and greatly threatened the health of human beings. Development of non-antibiotic biomaterial is recognized as an alternative way for effective treatment of bacterial infections. In the present work, a multifunctional cupper peroxide (CP) nanodots decorated gold nanostar (GNS)/silica nanorod (SiNR) Janus nanostructure (GNS@CP/SiNR) with excellent antibacterial activity was reported. Due to the formation of Janus nanostructure, GNS@CP/SiNR displayed strong plasmonic resonance absorbance in the near infrared (NIR)-II region that enabled the nanosystem for mild photothermal therapy (MPTT). In acidic conditions, CP decorated on GNS@CP/SiNR dissociated rapidly by releasing Cu2+ and H2O2, which subsequently transformed to •OH via the Fenton-like reaction for chemodynamic therapy (CDT). As a result, GNS@CP/SiNR could effectively inhibit both Gram-negative Escherichia coli (E. coli) and Gram-positive Staphylococcus aureus (S. aureus) and eradicate the associated bacterial biofilms by exerting the synergistic MPTT/CDT antibacterial effect. Moreover, GNS@CP/SiNR was also demonstrated effective in treating wound infection as verified on the S. aureus infected full thickness excision wound rat model. Mechanism study revealed that the synergistic MPTT/CDT effect of GNS@CP/SiNR firstly caused bacterial membrane damage, followed by boosting intracellular ROS via the severe oxidative stress effect, which subsequently caused the depletion of intracellular GSH and DNA damage, finally leading to the death of bacteria.

Abstract:The drug-resistant bacterial infection has seriously threatened human health and thus it is urgent to develop new and effective drugs to overcome the antibiotic resistance crisis. Herein, a near-infrared-II (NIR-II) region light-responsive Pd rough nanoshells on metal-organic framework (MOF) of UiO-66-NH 2 (UiO-66-NH 2 @Pd shell ) has been developed for the first time, which was further loaded with the photosensitizer IR780 to ultimately yield UiO-66-NH 2 @Pd shell -IR780 (UPI) as the core-shell theranostic nanoplatform. The Pd rough nanoshells demonstrate promising photothermal conversion efficiency, superoxide dismutase (SOD)-like activity, and light-enhanced catalase (CAT)-like activity. In the non-healing wound inflammatory microenvironment with overexpression of reactive oxygen species (ROS) caused by subcutaneous bacterial infection, Pd rough nanoshells in UPI transforms ROS, such as O 2 •− , into H 2 O 2 and efficiently catalyzes the decomposition of the formed and endogenous H 2 O 2 to produce O 2 . The O 2 production process could be enhanced by NIR-II light to facilitate photodynamic efficacy of IR780 to prime rapid clearance of methicillin-resistant Staphylococcus aureus (MRSA) through biofilm inhibition, along with down-regulation of hypoxia inducible factor-1α (HIF-1α) and relief of inflammation, thus promote wound healing. Moreover, UPI exhibits negligible biotoxicity in vitro and in vivo . This work provides a method for preparing promising Pd-based nanomaterials toward subcutaneous infection treatment and wound healing.
