Abstract:Nitrogen‐containing drug molecules and pharmaceuticals are ubiquitous, rendering the construction of C–N bonds a crucial target in methodology development. The Ritter reaction is a well‐established method for accessing C–N bonds by generating amide functionality through the reaction of carbocations and nitriles. Since its discovery back in 1948, the Ritter reaction has progressively advanced towards milder and more sustainable conditions for carbocation generation. In this regard, notable contributions have been made by means of single electron transfer (SET) chemistry. Nowadays, photo‐ and electrochemistry are established methods of choice for the generation of reactive radical intermediates and their subsequent oxidation via radical‐polar crossover (RPC) mechanism. We review recent examples of tandem RPC and Ritter‐type protocols, demonstrating how photo‐ and electrochemical energies have been effectively harvested to expand the precursor pool for the Ritter reaction and also reimagine the process with novel mechanisms and additives.

Abstract:With the development of organic electrochemical synthesis, a series of notable achievements have been made in electrochemical Ritter amination reactions, which have enriched the methods available for constructing C–N bonds. In this review, electrochemical Ritter amination reactions are introduced based on the classification of reaction substrates, including olefins, aromatics, alkylbenzenes, and the less reported carboxylic acids, ketones, sulfides, and alkanes. The application of electrochemical technology to Ritter reactions has improved the harsh conditions of the traditional reactions, and extended the substrate scope and the structural diversity of the products. The application value of Ritter reactions in organic synthesis has also been further expanded.

Abstract:Ritter reaction is a powerful method for the synthesis of amides, however, photoinduced Ritter reaction of alcohol is rarely reported. We herein report a simple and additive-free photocatalytic dehydration method for efficient direct conversion of benzyl alcohols to corresponding amides using the organic photocatalyst 2,4,6-triphenylpyrylium salt. Experimental and computational mechanistic analyses show that a non-conventional generation process of the carbocation is involved in this photoinduced Ritter reaction.

Abstract:This paper reviewed the recent research progress of organic reactions catalyzed by four typical earth abundant and 3d metal catalysts: Mn, Fe, Co, and Ni complexes and mainly focused on the reactions in which these 3d metal catalysts exhibited obvious advantages over other catalysts. The mechanism that makes these 3d metal catalysts show unusual properties is another focus of this paper. An outlook on 3d metal complexes-catalyzed organic reactions was also considered.

Abstract:In recent years, the development and application of transition metal ruthenium or iridium complexes based photocatalysts have opened up a new research field for organic synthesis, which provides mild and efficient strategies for the construction of chemical bonds and organic transformations. In order to enrich the types of photocatalysts, broaden the application scope of photocatalysis, and develop green and sustainable chemistry, some low-cost and easily available organometallic complexes with copper or iron were applied as photocatalysts recently. Iron-complexes feature non-toxicity, rich variety, and unique property of charge transfer from ligands to metals, which enable them show extraordinary capability in the field of photocatalytic synthesis. According to different reaction types, this review focuses on the recent development of photoinduced iron-catalysis in organic synthesis including C-H bond functionalization, C-C bond functionalization, bifunctionalization of alkenes, cross-coupling reaction, decarboxylative functionalization, selective oxidation and reduction.

Abstract:Minisci-type reactions have been widely known as reactions that involve the addition of carbon-centered radicals to basic heteroarenes followed by formal hydrogen atom loss. While the originally developed protocols for radical generation remain in active use today, in recent years, the new array of radical generation strategies have allowed the use of a wider variety of radical precursors that often operate under milder and more benign conditions. New transformations based on free radical reactivity are now available to a synthetic chemist, to utilize a Minisci-type reaction. Radical-generation methods based on photoredox catalysis and electrochemistry, which utilize thermal cleavage or the in situ generation of reactive radical precursors, have become popular approaches. Our review will cover the remarkable literature that has been reported on this topic in recent 5 years, from 2015-01 to 2020-01, in an attempt to provide guidance to the synthetic chemist on both the challenges that need to be overcome and the applications in organic synthesis.

Abstract:Ritter reaction has been recognized as an elegant strategy to construct the C-N bond. Its key feature is forming the carbocation for nucleophilic attack by nitriles. Herein, we report a complementary visible-light-induced three-component Ritter reaction of alkenes, nitriles, and <i>α</i>-bromo nitriles/esters, thereby providing mild and rapid access to various <i>γ</i>-amino nitriles/acids. Mechanistic studies indicated that traceless fluoride relay, transforming KF into imidoyl fluoride intermediate, is critical for the efficient reaction switch from atom transfer radical addition (ATRA) to the Ritter reaction. This approach to amino-alkylation of alkenes is chemoselective and operationally simple.

Abstract:Carbon–nitrogen cross-coupling reaction is an imperative and vital conversion in organic synthesis since one of its products, arylamines, are key frameworks in several natural products, pharmaceuticals, and other biologically active compounds. For the development of such important C–N coupling reaction, Buchwald and Hartwig made noteworthy contributions by presenting a plethora of ligands in addition to the conventional Pd and Cu catalysts. In this review, we try to update our previous review, covering the recent progresses in applications of Buchwald–Hartwig reaction in organic reactions, in the synthesis of heterocyclic compounds and total synthesis of natural products. This review covers this important advancement from 2018 till date.

Abstract:The recent progress of Ramberg-Backlund reaction is reviewed both on the side of the reaction itself and its application in synthesis of functional organic molecules and natural products. In case of RB reaction itself, recent developments towards Meyer's modification and Hendrickson's improvement are presented in detail, and then followed by new variants of Ramberg-Backlund rearrangements, including the epoxy-Ramberg-Backlund reaction, tandem Michael-addition-Ramberg-Backlund rearrangement, Diels-Alder-RB reaction. Recent synthetic application of RB reaction to prepare functional organic molecules as medium and large heterocyclic alkenes, optically pure polyoxygenated cycloalkene, chiral allylic and homoallylic compounds, are also discussed. Finally, applications of the Ramberg-Backlund process in synthesis of natural products are reviewed.

Abstract:This review primarily revolves around the photochemical asymmetric three‐component transformations facilitated by transition metal catalysis or organocatalysis. The intricate reaction mechanisms, diverse synthetic applications, and inherent limitations of these methods are thoroughly examined and discussed. Over the past decades, asymmetric photochemical synthesis has garnered significant attention for its sustainability and unique ability to generate enantio‐enriched molecules through distinct reaction pathways. Photochemical asymmetric three‐component reactions have demonstrated significant potential for the rapid construction of chiral compounds with molecular diversity and complexity. However, noteworthy challenges persist, including the participation of high‐energy intermediates such as radical species, difficulties in precise control of stereoselectivity, and the presence of competing background and side reactions. Recent breakthroughs have led to the development of sophisticated strategies in this field. This review explores the intricate mechanisms, synthetic applications, and limitations of these methods. We anticipate that it will contribute towards advancing asymmetric catalysis, photochemical synthesis, and green chemistry.

Abstract:Metal-mediated cyclizations are important transformations in a natural product total synthesis. The Pauson-Khand reaction, particularly powerful for establishing cyclopentenone-containing structures, is distinguished as one of the most attractive annulation processes routinely employed in synthesis campaigns. This review covers Co, Rh, and Pd catalyzed Pauson-Khand reaction and summarizes its strategic applications in total syntheses of structurally complex natural products in the last five years. Additionally, the hetero-Pauson-Khand reaction in the synthesis of heterocycles will also be discussed. Focusing on the panorama of organic synthesis, this review highlights the strategically developed Pauson-Khand reaction in fulfilling total synthetic tasks and its synthetic attractiveness is aimed to be illustrated.

Abstract:Enantioselective transition metal catalysis is an area very much at the forefront of contemporary synthetic research. The development of processes that enable the efficient synthesis of enantiopure compounds is of unquestionable importance to chemists working within the many diverse fields of the central science. Traditional approaches to solving this challenge have typically relied on leveraging repulsive steric interactions between chiral ligands and substrates in order to raise the energy of one of the diastereomeric transition states over the other. By contrast, this Review examines an alternative tactic in which a set of attractive noncovalent interactions operating between transition metal ligands and substrates are used to control enantioselectivity. Examples where this creative approach has been successfully applied to render fundamental synthetic processes enantioselective are presented and discussed. In many of the cases examined, the ligand scaffold has been carefully designed to accommodate these attractive interactions, while in others, the importance of the critical interactions was only elucidated in subsequent computational and mechanistic studies. Through an exploration and discussion of recent reports encompassing a wide range of reaction classes, we hope to inspire synthetic chemists to continue to develop asymmetric transformations based on this powerful concept.This article has not yet been cited by other publications.

Abstract:The use of sulfonium salts in organic synthesis has experienced a dramatic increase during the last years that can arguably be attributed to three main factors; the development of more direct and efficient synthetic methods that make easily available sulfonium reagents of a wide structural variety, their intrinsic thermal stability, which facilitates their structural modification, handling and purification even on large scale, and the recognition that their reactivity resembles that of hypervalent iodine compounds and therefore, they can be used as replacement of such reagents for most of their uses. This renewed interest has led to the improvement of already existing reactions, as well as to the discovery of unprecedented transformations; in particular, by the implementation of photocatalytic protocols. This review aims to summarize the most recent advancements on the area focusing on the work published during and after 2020. The scope of the methods developed will be highlighted and their limitations critically evaluated.

Abstract:Silver, as one of the most ancient coinage metals, has attracted a plethora of attention from chemists in recent years due to the easy availability, high catalytic efficiency and multiple functions displayed in various chemical processes. In particular, the research in this field has gained a great progress since 2007 and several elegant reviews have been documented in the literature. However, there has been no systematic review concentrating on the transformations of electronically unbiased alkenes and alkynes under silver catalysis till now, though a rapid development in this area is witnessed. Therefore, this review mainly focuses on the recent advances in silver‐catalyzed reactions of different electron‐unbiased alkenes and alkynes, covering examples of hydrofunctionalization reactions, difunctionalization reactions, cross‐coupling reactions as well as cyclization reactions since 2015 to February 2020. In addition, some interesting mechanisms were also discussed.

Abstract:Pyridinium salts are valuable building blocks, which have been widely applied in various organic transformations during the past few decades. In particular, N-functionalized pyridinium salts have been explored as convenient radical precursors, which would go through reductive single-electron transfer. As a result, the chemistry of such pyridinium compounds for generating carbon-, nitrogen-, and oxygen-centered radicals has been witnessed, and a remarkable progress has been achieved, making it a hot topic over the last five years. This Review describes recent advances in the area of pyridinium salts as radical precursors, concerning the development of radical reactions involving pyridinium salts in organic synthesis.
