Abstract:The crab Trapezia bidentata is a conspicuous crustacean characterized as an obligate symbiont with branching corals, such as species of Pocillopora. These crustaceans have been identified as strict corallivores with a feeding dependency on the mucus excreted by adult colonies in the genus Pocillopora. However, like other crustaceans, individuals of T. bidentata have a nutritional plasticity that has not been described. We used an integrative approach, including traditional taxonomy and high‐throughput sequencing of hypervariable region V9 of the 18S rRNA gene, to describe the mouthparts and the diet of individuals of Trapezia bidentata associated with adult colonies of Pocillopora verrucosa. The presence of setae on the ambulatory dactylus and maxillipeds, forming food brushes and combs, was evident. Sequencing and analyses of the intestinal content of the crustaceans found evidence of 18 phyla, mostly represented by Brachiopoda (28.1%), Arthropoda (31.7%), and Cnidaria (21.1%), but also small contributions (≤2%) of other animal groups such as Mollusca, Annelida, and Tunicata. In addition, a few traces (≤0.5%) of algae and fungi were identified. The feeding structures and intestinal content showed that individuals of T. bidentata are omnivorous and behave as suspension feeders and scrapers, obtaining nourishment from tissues transported in the water column and in the mucus and tissue of P. verrucosa. The results provide a clearer characterization of the role of T. bidentata, which is a crucial guard crustacean associated with P. verrucosa. We found both corallivorous and suspension‐feeding habits, which demonstrates feeding plasticity, a positive characteristic for the survival of the species.

Morphological convergence from independent crab lineages contributes to the flourishing symbiotic diversity between crabs and branching corals

Abstract:Symbiosis is one of the most important evolutionary processes shaping biodiversity. Phylogenetic and morphological analyses can enhance our understanding of the origin and evolution of symbiosis. The symbiotic relationship between Trapezioidea crabs and branching corals, with high species diversity and flexible symbiosis, is iconic coral/crustacea symbiosis in the marine ecosystem. In this study, the evolutionary history of Trapezioidea crabs was investigated by constructing a phylogenetic relationship based on the whole mitogenome sequences of 66 Brachyura species, including 17 newly sequenced Trapezioidea species. Trapezioidea crabs were split at least into two phylogenetically distant lineages originated at different times. The two lineages, both including facultative and obligate symbionts, convergently evolved smooth and flattened trapezoidal carapaces favorable for specific coral-living. The significantly larger cheliped in obligate species indicated a deeper degree of coral dependence compared with facultative and other associate species.

Abstract:The exact nature of the relationship between symbiont fauna and their hosts is often unclear, but knowing more about these intricate ecological interactions is vital to understand the trophic positions of host-associated fauna, and can aid in accurate constructions of food-webs on coral reefs. Scleractinian corals are hosts to hundreds of symbiont taxa, including fish and many invertebrate species. Some of these associated fauna are beneficial to their coral host(s), whereas other taxa can have detrimental effects, yet their impact is often difficult to determine. Coral-dwelling gall crabs (Cryptochiridae) are obligate, often host-specific, symbionts of scleractinian corals but the nature of this relationship is still under debate. Three Atlantic gall crab species ( Kroppcarcinus siderastreicola , Opecarcinus hypostegus and Troglocarcinus corallicola ) and their coral hosts' tissue/mucus were collected from reefs in Guadeloupe. Stable carbon and nitrogen isotope values were measured for 57 crabs inhabiting host coral colonies belonging to seven different coral species (although only 27 colonies from five coral species were collected), alongside other potential food sources (epilithic algal matrix, plankton and particulate organic matter). The carbon and nitrogen isotope values of gall crabs relative to those of their respective coral host(s) and other possible food sources showed that coral tissue/mucus was the main food source for the crabs. The results of the mixing models further supported this finding, suggesting that corals are responsible for 40–70% of the crabs' diet. In T. corallicola , the isotopic signature differed significantly between sexes, possibly caused by the high sexual dimorphism observed in this species. Here we showed that Atlantic gall crabs mainly dine on coral tissue and/or mucus excreted by their coral hosts, highlighting their nutritional dependence on their host. However, since coral mucus is continuously exuded by scleractinians, hence the energetic or metabolic drain for corals is expected to be minimal. Gall crabs depend on their coral hosts for settlement cues as larvae, for habitat as adults and - highlighted by this study - for food, essential for their subsistence. This obligate dependence on their hosts for all parts of their life makes them extremely vulnerable to reef degradation, and underlines the importance in understanding the exact nature of a relationship between symbiont and coral host.

Abstract:Abstract Symbiotic relationships contribute considerably to the high biodiversity found on coral reefs. Coral-dwelling gall crabs (Cryptochiridae) represent a prime example of coral-associated invertebrates that exhibit obligate relationships with their host. The induction of a skeletal modification in the coral, used as a dwelling by the crab, is the most remarkable aspect of this close association. Here we examined Hapalocarcinus marsupialisStimpson, 1859, collected from Pocillopora and Stylophora corals in the Saudi Arabian Red Sea. Specimens were DNA barcoded, and five distinct clades were revealed, providing further evidence for the hypothesis that H. marsupialis is a species complex. Divergence (COI) between the clades ranged from 3.2 to 15.7%. The putative species were tested for differences in morphology and reproduction. Crabs were examined regarding morphometric characters (carapace length and width, pleon (abdomen), chelae, and growth patterns) and reproductive traits. The data were pooled and analysed by host genus and putative species, which revealed significant differences for most of the measured variables in female crabs. Specimens retrieved from Pocillopora were significantly larger (up to 49 %) and had higher fecundity than those inhabiting Stylophora. For reproductive output (RO) no differences at species- or host-genus level were detected. The average RO of ~70% over all specimens is high compared to other brachyurans, supporting the assumption that symbiotic brachyurans invest more energy in reproduction than their free-living counterparts. Discrepancies with published data on growth and reproduction of Hapalocarcinus are discussed. Our results demonstrate the usefulness of morphometric traits and fecundity in separating the clades in the H. marsupialis complex, and prepare the ground for further morphometric studies on the genus and other symbiotic brachyurans. Moreover, it highlights the need to check for the presence of cryptic species when studying aspects of the biology of a species.

Abstract:We report the first observation of coral‐dwelling gall crabs inhabiting corals of the family Poritidae in the central Red Sea. Based on examination of the coral skeletons containing the crab dwellings, we hypothesize that the crabs initially settled on their preferred host (Agariciidae), which was subsequently outcompeted and overgrown by the Porites colonies. These findings give interesting insights into the adaptive potential of gall crabs to changes in their microenvironment and their ability to survive competition for space on coral reefs. Coral reefs accommodate a myriad of species, many of which live in association with a host organism. Decapod crustaceans make up a large part of this associated fauna on coral reefs. Among these, cryptochirid crabs are obligately associated with scleractinian corals, in which they create dwellings where they permanently reside. These gall crabs show various levels of host specificity, with the majority of cryptochirids inhabiting a specific coral genus or species. Here, we report the first records of gall crabs living in association with two different Porites species in the Red Sea. Crescent‐shaped dwellings were observed in Porites rus and a Porites sp. in situ, and colonies with crabs were collected for further study in the laboratory. Using a combination of morphology and DNA barcoding, the crabs were identified as belonging to Opecarcinus, a genus only known to inhabit Agariciidae corals. The coral skeleton was bleached and studied under a stereo microscope, which revealed that the Porites corals overgrew adjoining agariciid Pavona colonies. We hypothesize that the gall crab originally settled on Pavona, its primary host of choice. Due to coral interspecific competition the Porites colony overgrew the adjacent Pavona colonies, resulting in a secondary and never before reported association of Opecarcinus with Porites. These findings suggest that cryptochirid crabs can adapt to the new microenvironment provided by a different coral host and survive competition for space on coral reefs.

Abstract:Organismal symbioses are fundamental to biodiversity, evolution, and ecosystem functioning. On coral reefs, many decapod species have formed distinct epibiotic symbioses through decoration tendencies that enhance diet, camouflage, and defence. The red decorator crab, Schizophrys aspera (Majidae: Decapoda), has a broad Indo-Pacific distribution and is a successful predator of juvenile crown-of-thorns seastars (CoTS; Acanthaster sp.). However, little is known of the biology and decorating symbioses of S. aspera on the Great Barrier Reef (GBR), where CoTS pose ongoing management challenges. We characterised S. aspera and its epibiont community collected in coral rubble patches on the southern GBR. S. aspera predominantly used sponges (94 ± 1%; mean ± SE) in its decoration, with greater proportions of the carapace covered for juveniles (58 ± 5%) and females (46 ± 4%) compared to males (24 ± 4%). In short-term (8-d) experiments, S. aspera substantially reduced sponge (31%) and algal (47%) cover on rubble pieces, demonstrating its potential to alter sessile communities. The close association of S. aspera with sponges and algae likely reflects its diet and enhances camouflage and chemical defence in its coral rubble niche on the GBR. As sessile taxa are often noxious, we postulate that these symbioses may confer resilience of S. aspera to plancitoxins in its consumption of CoTS. Evaluating how epibiont diversity and biochemistry shape the habitat associations, distribution, and role of S. aspera as predator and prey may be important to understanding its ability to mediate CoTS densities on the GBR and elsewhere.

Dead foundation species create coral rubble habitat that benefits a resilient pest species

Abstract:Critical loss of habitat is the greatest threat to biodiversity, yet some species are inherently plastic to and may even benefit from changes in ecosystem states. The crown-of-thorns sea star (CoTS; Acanthaster spp.) may be one such organism. CoTS are large corallivores native to the tropical Indo-Pacific and in unexplained high densities, can adversely affect entire coral reefs. Proximal causes of CoTS outbreaks remain elusive, so this phenomenon remains a daunting and costly challenge for reef conservation and management. Amplifying anthropogenic impacts and new empirical data point to the degraded reef hypothesis to explain the episodic nature of CoTS population outbreaks. We posit that loss of live coral paradoxically benefits CoTS juveniles, which accumulate in their rubble nursery habitat before conditions trigger their pulsed emergence as coral-eaters. We review trait plasticity across the CoTS life cycle and present the degraded reef hypothesis in an integrative understanding of their propensity to outbreak.

Abstract:Increasingly intense and frequent ocean heatwaves are causing widespread coral mortality. These heatwaves are just one of the many stressors - among for instance ocean acidification, nutrient pollution and destructive fishing practices - that have caused widespread decline of coral reefs over the past century. This destruction of reefs threatens the remarkable biodiversity of organisms that depend upon coral reefs. However, recent research suggests that many of the fishes and invertebrates that inhabit coral reefs may play an underappreciated role in influencing the resistance and recovery of corals to stressors, especially those caused by global climate change such as ocean heatwaves. Unraveling the threads that link these coral inhabitants to the corals' response to stressors has the potential to weave a more comprehensive model of resilience that integrates the plight of coral reefs with the breathtaking diversity of life they host. Here, we aim to elucidate the critical roles that coral-associated fishes and invertebrates play in mediating coral resilience to environmental stressors. By integrating recent research findings, we aim to showcase how these often-overlooked organisms influence coral resilience in the face of climate change.

Abstract:Octocorals are major contributors of terpenoid chemical diversity in the ocean. Natural products from other sessile marine animals are primarily biosynthesized by symbiotic microbes rather than by the host. Here, we challenge this long-standing paradigm by describing a monophyletic lineage of animal-encoded terpene cyclases (TCs) ubiquitous in octocorals. We characterized 15 TC enzymes from nine genera, several of which produce precursors of iconic coral-specific terpenoids, such as pseudopterosin, lophotoxin and eleutherobin. X-ray crystallography revealed that coral TCs share conserved active site residues and structural features with bacterial TCs. The identification of coral TCs enabled the targeted identification of the enzyme that constructs the coral-exclusive capnellane scaffold. Several TC genes are colocalized with genes that encode enzymes known to modify terpenes. This work presents an example of biosynthetic capacity in the kingdom Animalia that rivals the chemical complexity generated by plants, unlocking the biotechnological potential of octocorals for biomedical applications.

Three way symbiosis between a goby, a shrimp, and a crab

Abstract:A unique case of triple symbiosis between a goby, a pistol shrimp, and a porcellanid crab sharing the same burrow close to reef patches in Lembeh Strait, Indonesia, is described from direct observations for the first time. The burrow, typically occupied by shrimps and gobies, is shared with the porcellanid Enosteoides lobatus. In the goby–shrimp association, the fish inhabits the burrow, which is constructed and kept clean by the shrimp, and the fish warns the shrimp of potential dangers approaching their refuge. The porcellanid lives in the burrow and also benefits from living in a habitat where it could not survive without the two sentinel species. Because the limiting resource for the suspension-feeding porcellanid is protected space, this liaison is an adaptation by E. lobatus to a symbiotic life in a habitat that protects it from predation, and is optimal for filter feeding.

Abstract:Marine symbioses are integral to the persistence of ecosystem functioning in coral reefs. Solitary corals of the species Heteropsammia cochlea and Heterocyathus aequicostatus have been observed to live in symbiosis with the sipunculan worm Aspidosiphon muelleri muelleri, which inhabits a cavity within the coral, in Zanzibar (Tanzania). The symbiosis of these photosymbiotic corals enables the coral holobiont to move, in fine to coarse unconsolidated substrata, a process termed as "walking." This allows the coral to escape sediment cover in turbid conditions which is crucial for these light-dependent species. An additional commensalistic symbiosis of this coral-worm holobiont is found between the Aspidosiphon worm and the cryptoendolithic bivalve Jousseaumiella sp., which resides within the cavity of the coral skeleton. To understand the morphological alterations caused by these symbioses, interspecific relationships, with respect to the carbonate structures between these three organisms, are documented using high-resolution imaging techniques (scanning electron microscopy and µCT scanning). Documenting multi-layered symbioses can shed light on how morphological plasticity interacts with environmental conditions to contribute to species persistence.
Characteristics of the infestation of Seriatopora corals by the coral gall crab Hapalocarcinus marsupialis Stimpson, 1859 on the great reef of toliara, Madagascar

Abstract:This study describes the association between the obligatory symbiont coral gall crab Hapalocarcinus marsupialis and its stony coral hosts Seriatopora sp. within the Great Reef of Toliara in Madagascar and attempts to discuss their symbiotic status through comparison with previous studies. These corals are inhabited by crabs living in galls that can be categorised in four distinct morphological stages, where the first one corresponds to a small bud and the last one represents a completely closed gall surrounding the crab inside. Within the reef, 563 colonies of Seriatopora species were observed by scuba-diving at ten different stations: 37.8 % of them were infested by H. marsupialis, with a total of 763 galls, and with a majority of stage 4 galls. Galls are monopolised by females that can have different morphologies. Females store the sperm in two spermathecae and are fertilised when their morphology and size are similar to males and the gall is not closed. Histological observations coupled with scanning electronic microscopy analyses show that closed galls are made of an external living tissue, a mid skeletal layer and an internal living tissue. The internal living tissue includes polyps similar to the external tissue, some of them being sexually mature. Nitrogen and carbon isotopic signatures confirmed that these crabs are filter-feeders and do not feed on their host. This association perfectly highlights the difficulties to define the symbiotic status of a symbiont if one considers inflexible the three categories of symbiosis commonly defined.

Abstract:Coral reef community exhibits high species diversity and a broad range of biological relationships, including widespread symbiosis and complex food utilization patterns. In our study, we investigated the symbiotic relationship between the commonly crinoid host Comaster schlegelii and its ophiuroid obligatory symbiont Gymnolophus obscura. Using a combination of fatty acid biomarkers and stable isotopic compositions, we explored differences in their organic matter utilization strategies and nutritional relationships. The result of stable isotopes revealed that G. obscura had higher δ15N values than its crinoid host. Particulate organic matter and phytoplankton were identified as the primary food sources for both species, however C. schlegelii showed a higher proportional contribution from benthic microalgae. Fatty acid markers showed that C. schlegelii was more dependent on benthic microalgae such as diatoms, and less on debritic organic matter and bacteria than G. obscura. Elevated δ15N values of G. obscura and similar food source contribution rates between the host and symbiont suggest that ophiuroid feeds on materials filtered by crinoids and have similar diet to the host. Our results provide insights into the symbiotic patterns of crinoids and ophiuroids, while also supplying foundational data on how symbiotic reef species select organic matter utilization strategies to adapt to their environment.
Unveiling the molecular choreography: In-depth single-cell transcriptomic exploration of the regenerative dynamics in stony coral

Abstract:The coral reef ecosystem faces increasing threats under global climate challenges. One of the core issues is the inability of fragments to quickly grow into a size that can resist environmental pressures in coral transplantation. The observation of accelerated growth during the early stages of coral regeneration provides new insights for addressing this challenge. To investigate related molecular mechanisms, our study focused on the fast-growing stony coral (with chromosome-scale reference genome). Employing diverse techniques, including single-cell RNA sequencing (scRNA-seq), we unveiled related intricate cellular dynamics. Single-cell analysis revealed notable shifts in calicoblasts and epidermal cells around 2-4 weeks post-injury. Gene expression analysis revealed enrichment in immune response and biomineralization pathways. Pseudotime analysis explained the differentiation of epidermal cells into calicoblasts, while time-course analysis identified key genes associated with dynamic biomineralization changes. This study enhances our understanding of coral regeneration, offering insights for protective strategies to foster coral growth.

Abstract:Coral reefs are one of the most diverse marine ecosystems on Earth and one of the richest in terms of species interactions. Scleractinian corals are usually the most likely to provide numerous different habitats and to support many symbiotic relationships. However, many other invertebrate groups, such as sponges, bryozoans, and other cnidarians, establish strict symbiotic relationships with other marine organisms. Despite the nature of these relationships—as well as the factors that drive their establishment—being unclear in most cases, a few studies have already shown that some associations may increase the resistance of their hosts to external disturbances. Thus, the potential ability of each member of these diverse symbiotic assemblages to influence the fitness and long-term survival of their hosts bring the coral-associated fauna to the top of the list of coral reef studies. Unfortunately, the widespread degradation of coral reef ecosystems may threaten the existence of the intimate relationships that may go unrecognized complicating our understanding of the intricate networks connecting the fates of reef species. Therefore, this unprecedented loss of biodiversity calls for synergic conservation and monitoring actions aimed at significantly increasing our efforts to search for and describe as much of the diversity of coral-associated organisms as possible, shedding new light on the complex, elusive mechanisms controlling coral reef functioning.

Abstract:Corals live in a symbiotic relationship with various bacteria that are fundamental to host fitness, health, and survival. Though the diversity of symbiotic bacteria has been revealed in the early life stages of some corals, the dynamic bacterial community profiles of one coral are still poorly characterized, particularly the stage-specific bacteria. In this study, the bacterial communities in the patent, eggs, and 4-day-old planula larvae of a hermaphrodite coral Dipsastraea favus were investigated by high-throughput sequencing of 16S ribosomal RNA gene. As a result, dynamic profiles of bacterial community in the parent, eggs and larvae of D. favus were suggested. The bacterial diversity in the planula larvae was a bit higher than that in the D. favus parent, and distinct stage-specific symbiotic bacteria were detected, e.g., Oceanospirillaceae, Kordia , and Legionellaceae in D. favus larvae, and Kiloniellales and Prosthecochloris in adult D. favus . The dynamic change of bacterial community in coral adults and larvae may expand our understanding of the complex relationship between coral host and its symbiotic microbiota.

Abstract:Symbiotic relationships are common in marine ecosystems, especially between cnidarians and crustaceans. This study explored the association of the arrow crab Stenorhynchus lanceolatus with different hosts in the Canary Islands, focusing on the relationship with the sea anemone Telmatactis cricoides . Field surveys and laboratory experiments were conducted to assess crab preference among potential hosts (anemones and sea urchins), association rates and behaviours of crabs interacting with the anemone. Field results revealed similar prevalence on anemones and sea urchins, and that most crabs associated to the anemone were solitary females. However, under laboratory conditions, sea urchins were observed to be preferred as hosts, but association with anemones were more stable in time, although its colonization period was slower, likely due to their toxicity and lack of environmental cues. Additionally, crab intraspecific competition seemed to favour the association with the anemone. This study showed the mutualistic and facultative nature of the relationship between S. lanceolatus and T. cricoides, advancing our understanding of benthic ecosystems dynamics

Abstract:The ecological and evolutionary consequences of partner fidelity and flexibility among coral–dinoflagellate mutualisms are widely debated. Resident symbionts can modulate the resilience of their hosts to environmental stressors, which explains, in part, why host–symbiont combinations differ over broad geographic ranges and across physical–environmental gradients in light and temperature. Therefore, flexibility in these mutualisms may influence the longevity of coral populations and communities subjected to ocean warming. However, despite decades of research, basic knowledge about these mutualisms remains incomplete, hindering the development of predictive ecological theory. In particular, few studies have investigated the long-term composition of symbiont populations within individual colonies. To further examine the extent to which coral colonies have stable relationships with specific symbionts over multiple years, diverse coral taxa (Scleractinia) from a West Indo-Pacific fore reef (Palau) were tagged and sampled at various intervals—ranging from six months to several years—over nine years' time. Symbiont identity was examined using multiple genetic markers that resolved symbiont diversity to species and individual genotypes (i.e., clonal strains). Members of the genus Cladocopium (formerly Symbiodinium Clade C) were prevalent across the host community. Generally, corals with open modes of symbiont acquisition harbored a host–generalist symbiont, while corals with vertical symbiont transmission were associated with co-evolved host-specific symbionts. Consistent with previous colony monitoring studies, symbiont populations in a majority of colonies were dominated by one species and one strain (based on multilocus genotyping) over multiple years. Thus, the distribution of symbiont diversity at the genus, species and clone level, comprising specific and stable partner combinations, scale predictably to reef habitat, host taxon, and individual colony. Recognizing these fundamental ecological patterns establishes a more comprehensive understanding of the population and community structure of these mutualisms.

Abstract:Abstract Background Coral reefs are among the most diverse, complex and densely populated marine ecosystems. To survive, morphologically simple and sessile cnidarians have developed mechanisms to catch prey, deter predators and compete with adjacent corals for space, yet the mechanisms underlying these functions are largely unknown. Here, we characterize the histology, toxic activity and gene expression patterns in two different types of tentacles from the scleractinian coral Galaxea fascilcularis – catch tentacles (CTs), used to catch prey and deter predators, and sweeper tentacles (STs), specialized tentacles used for territorial aggression. Results STs exhibit more mucocytes and higher expression of mucin genes than CTs, and lack the ectodermal cilia used to deliver food to the mouth and remove debris. STs and CTs also express different sensory rhodopsin-like g-protein coupled receptors, suggesting they may employ different sensory pathways. Each tentacle type has a different complement of stinging cells (nematocytes), and the expression in the two tentacles of genes encoding structural nematocyte proteins suggests the stinging cells develop within the tentacles. CTs have higher neurotoxicity to blowfly larvae and hemolytic activity compared to the STs, consistent with a role in prey capture. In contrast, STs have higher phospholipase A2 activity, which we speculate may have a role in inducing tissue damage during territorial aggression. The expression of genes encoding cytolytic toxins (actinoporins) and phospholipases also differs between the tentacle types. Conclusions These results show that the same organism utilizes two distinct tentacle types, each equipped with a different venom apparatus and toxin composition, for prey capture and defense and for territorial aggression.
