Abstract:Soluble elastin-like peptides (ELPs) can be engineered into a range of physical forms, from hydrogels and scaffolds to fibers and artificial tissues, finding numerous applications in medicine and engineering as “smart polymers”. Elastin-like peptides are attractive candidates as a platform for novel biomaterial design because they exhibit a highly tunable response spectrum, with reversible phase transition capabilities. Here, we report the design of the first virtual library of elastin-like protein models using methods for enhanced sampling to study the effect of peptide chemistry, chain length, and salt concentration on the structural transitions of ELPs, exposing associated molecular mechanisms. We describe the behavior of the local molecular structure under increasing temperatures and the effect of peptide interactions with nearest hydration shell water molecules on peptide mobility and propensity to exhibit structural transitions. Shifts in the magnitude of structural transitions at the single-molecule scale are explained from the perspective of peptide-ion-water interactions in a library of four unique elastin-like peptide systems. Predictions of structural transitions are subsequently validated in experiment. This library is a valuable resource for recombinant protein design and synthesis as it elucidates mechanisms at the single-molecule level, paving a feedback path between simulation and experiment for smart material designs, with applications in biomedicine and diagnostic devices.

Abstract:Elastomeric, robust, and biocompatible hydrogels are rare, while the need for these types of biomaterials in biomedical-related uses remains high. Here, a new family of genetically engineered silk-elastin copolymers (SELPs) with encoded enzymatic crosslinking sites is developed for a new generation of stimuli-responsive yet robust hydrogels. Input into the designs is guided by simulation, and realized via genetic engineering strategies. The avoidance of gamma irradiation or chemical crosslinking during gel fabrication, in lieu of an enzymatic process, expands the versatility of these new gels for the incorporation of labile proteins and cells. In the present study, the new SELP hydrogels offers sequence dependent, reversible stimuli-responsive features. Their stiffness covers almost the full range of the elasticity of soft tissues. Further, physical modification of the silk domains provided a secondary control point to fine-tune mechanical stiffness while preserving stimuli-responsive features, with implications for a variety of biomedical materials and device needs.

Abstract:Supramolecular bonds have been widely used for designing polymers because of their reversible nature. In contrast, utilization of their dynamic equilibrium nature to access materials of unusual mechanical properties has been poorly explored. Taking full advantage of this latter attribute requires the design of polymer networks with high contents of supramolecular bonds. In this work, polymer networks with high contents of self‐complementary hydrogen bonds (ureidopyrimidinone) are synthesized using thiol–acrylate click addition. The excellent tunability of the network allows a range of intriguing mechanical properties to be achieved including the transition from plasticity to elasticity, ultrasoft shape memory polymer, strong strain rate dependence, and high mechanical damping. Materials with such versatile dynamic behaviors may open up a range of new applications.

Abstract:The need for dynamic, elastomeric polymeric biomaterials remains high, with few options with tunable control of mechanical properties, and environmental responses. Yet the diversity of these types of protein polymers pursued for biomaterials-related needs remains limited. Robust high-throughput synthesis and characterization methods will address the need to expand options for protein-polymers for a range of applications. Here, a combinatorial library approach with high throughput screening is used to select specific examples of dynamic protein silk-elastin-like polypeptides (SELPs) with unique stimuli responsive features, including tensile strength and adhesion. Using this approach, 64 different SELPs with different sequences and molecular weights are selected out of over 2000 recombinant E. coli colonies. New understanding of sequence-function relationships with this family of proteins is gained through this combinatorial-screening approach and can provide a guide to future library designs. Further, this approach yields new families of SELPs to match specific material functions.

Abstract:Elastomeric proteins act as the essential functional units in a wide variety of biomechanical machinery and serve as the basic building blocks for biological materials that exhibit superb mechanical properties. These proteins provide the desired elasticity, mechanical strength, resilience, and toughness within these materials. Understanding the mechanical properties of elastomeric protein-based biomaterials is a multiscale problem spanning from the atomistic/molecular level to the macroscopic level. Uncovering the design principles of individual elastomeric building blocks is critical both for the scientific understanding of multiscale mechanics of biomaterials and for the rational engineering of novel biomaterials with desirable mechanical properties. The development of single-molecule force spectroscopy techniques has provided methods for characterizing mechanical properties of elastomeric proteins one molecule at a time. Single-molecule atomic force microscopy (AFM) is uniquely suited to this purpose. Molecular dynamic simulations, protein engineering techniques, and single-molecule AFM study have collectively revealed tremendous insights into the molecular design of single elastomeric proteins, which can guide the design and engineering of elastomeric proteins with tailored mechanical properties. Researchers are focusing experimental efforts toward engineering artificial elastomeric proteins with mechanical properties that mimic or even surpass those of natural elastomeric proteins. In this Account, we summarize our recent experimental efforts to engineer novel artificial elastomeric proteins and develop general and rational methodologies to tune the nanomechanical properties of elastomeric proteins at the single-molecule level. We focus on general design principles used for enhancing the mechanical stability of proteins. These principles include the development of metal-chelation-based general methodology, strategies to control the unfolding hierarchy of multidomain elastomeric proteins, and the design of novel elastomeric proteins that exhibit stimuli-responsive mechanical properties. Moving forward, we are now exploring the use of these artificial elastomeric proteins as building blocks of protein-based biomaterials. Ultimately, we would like to rationally tailor mechanical properties of elastomeric protein-based materials by programming the molecular sequence, and thus nanomechanical properties, of elastomeric proteins at the single-molecule level. This step would help bridge the gap between single protein mechanics and material biomechanics, revealing how the mechanical properties of individual elastomeric proteins are translated into the properties of macroscopic materials.
Engineered Elastomeric Proteins with Dual Elasticity Can Be Controlled by a Molecular Regulator

Abstract:Elastomeric proteins are molecular springs that confer excellent mechanical properties 1 , 2 , 3 , 4 , 5 to many biological tissues and biomaterials. Depending on the role performed by the tissue or biomaterial, elastomeric proteins can behave as molecular springs 1 , 2 , 6 , 7 or shock absorbers 3 , 4 , 5 , 8 , 9 , 10 . Here we combine single-molecule atomic force microscopy and protein engineering techniques to create elastomeric proteins that can switch between two distinct types of mechanical behaviour in response to the binding of a molecular regulator. The proteins are mechanically labile by design and behave as entropic springs with an elasticity that is governed by their configurational entropy. However, when a molecular regulator binds to the protein, it switches into a mechanically stable state and can act as a shock absorber. These engineered proteins effectively mimic and combine the two extreme forms of elastic behaviour found in natural elastomeric proteins, and thus represent a new type of smart nanomaterial that will find potential applications in nanomechanics and material sciences.

Abstract:Elastin is a structural protein with outstanding mechanical properties (e.g., elasticity and resilience) and biologically relevant functions (e.g., triggering responses like cell adhesion or chemotaxis). It is formed from its precursor tropoelastin, a 60-72 kDa water-soluble and temperature-responsive protein that coacervates at physiological temperature, undergoing a phenomenon termed lower critical solution temperature (LCST). Inspired by this behaviour, many scientists and engineers are developing recombinantly produced elastin-inspired biopolymers, usually termed elastin-like polypeptides (ELPs). These ELPs are generally comprised of repetitive motifs with the sequence VPGXG, which corresponds to repeats of a small part of the tropoelastin sequence, X being any amino acid except proline. ELPs display LCST and mechanical properties similar to tropoelastin, which renders them promising candidates for the development of elastic and stimuli-responsive protein-based materials. Unveiling the structure-property relationships of ELPs can aid in the development of these materials by establishing the connections between the ELP amino acid sequence and the macroscopic properties of the materials. Here we present a review of the structure-property relationships of ELPs and ELP-based materials, with a focus on LCST and mechanical properties and how experimental and computational studies have aided in their understanding.

Abstract:The sophisticated, elegant protein-polymers designed by nature can serve as inspiration to redesign and biomanufacture protein-based materials using synthetic biology. Historically, petro-based polymeric materials have dominated industrial activities, consequently transforming our way of living. While this benefits humans, the fabrication and disposal of these materials causes environmental sustainability challenges. Fortunately, protein-based biopolymers can compete with and potentially surpass the performance of petro-based polymers because they can be biologically produced and degraded in an environmentally friendly fashion. This paper reviews four groups of protein-based polymers, including fibrous proteins (collagen, silk fibroin, fibrillin, and keratin), elastomeric proteins (elastin, resilin, and wheat glutenin), adhesive/matrix proteins (spongin and conchiolin), and cyanophycin. We discuss the connection between protein sequence, structure, function, and biomimetic applications. Protein engineering techniques, such as directed evolution and rational design, can be used to improve the functionality of natural protein-based materials. For example, the inclusion of specific protein domains, particularly those observed in structural proteins, such as silk and collagen, enables the creation of novel biomimetic materials with exceptional mechanical properties and adaptability. This review also discusses recent advancements in the production and application of new protein-based materials through the approach of synthetic biology combined biomimetics, providing insight for future research and development of cutting-edge bio-inspired products. Protein-based polymers that utilize nature's designs as a base, then modified by advancements at the intersection of biology and engineering, may provide mankind with more sustainable products.
Solvent-Free Plasticity and Programmable Mechanical Behaviors of Engineered Proteins

Abstract:Biopolymeric networks with plasticity show great competences in diverse fields owing to the combined biocompatible and mechanical characteristics. However, to realize such plasticity external complicated treatments, e.g., UV or organic solvent have to be applied, which in turn impair the biological nature and even mechanical properties of those systems. To address this challenge, one new type of anhydrous protein liquid crystalline (LC) gels, which exhibit flexible morphological plasticity and mechanical programmability is demonstrated. Supramolecular interactions in the smectic biogels play an important role for their high plasticity. Remarkably, the samples exhibit outstanding mechanical behaviors. The tensile strength and Young's modulus at MPa levels are comparable or even higher than chemically cross-linked hydrogels and LC elastomers. More importantly, mechanical programmability of the LC gels is achieved by genetically tuning the charge density of protein backbones. Consequently, the mechanical performance is manipulated in the range of one order of magnitude. Thus, this type of anhydrous protein LC gels offers great opportunities for load-bearing high-tech applications.

Abstract:Protein-based structural biomaterials are of great interest for various applications because the sequence flexibility within the proteins may result in their improved mechanical and structural integrity and tunability. As the two representative examples, protein-based adhesives and fibers have attracted tremendous attention. The typical protein adhesives, which are secreted by mussels, sandcastle worms, barnacles, and caddisfly larvae, exhibit robust underwater adhesion performance. In order to mimic the adhesion performance of these marine organisms, two main biological adhesives are presented, including genetically engineered protein-based adhesives and biomimetic chemically synthetized adhesives. Moreover, various protein-based fibers inspired by spider and silkworm proteins, collagen, elastin, and resilin are studied extensively. The achievements in synthesis and fabrication of structural biomaterials by DNA recombinant technology and chemical regeneration certainly will accelerate the explorations and applications of protein-based adhesives and fibers in wound healing, tissue regeneration, drug delivery, biosensors, and other high-tech applications. However, the mechanical properties of the biological structural materials still do not match those of natural systems. More efforts need to be devoted to the study of the interplay of the protein structure, cohesion and adhesion effects, fiber processing, and mechanical performance.
Rationally Designed Synthetic Protein Hydrogels with Predictable and Controllable Mechanical Properties

Abstract:A key challenge in biomaterials research is to produce synthetic hydrogels that can replicate the diverse mechanical properties of the naturally occurring tissues for various biomedical applications, including tissue engineering, stem cell and cancer research, cell therapy, and immunomodulation. However, currently, the methods that can be used to control the mechanical properties of hydrogels are very limited and are mainly focused only on the elasticity of hydrogels. In this work, combining single molecule force spectroscopy, protein engineering and theoretical modeling, we show that synthetic protein hydrogels with predictable mechanical properties can be rationally designed using protein building blocks with known mechanical properties at the molecular level. We have successfully tuned the elasticity, extensibility, toughness and self-healing of hydrogels using this approach [1]. We have also demonstrated the engineering of physically crosslinked muscle-mimic hydrogels which are strong, tough, elastic and able to function under significant load and spontaneously self-heal when damaged [1]. Next, by using light-responsive proteins as building blocks, we have been able to engineer hydrogels whose mechanical properties are reversibly tunable by light illumination [2]. These hydrogels can capture the dynamic mechanical environments of naturally occurring extracellular matrices and are potentially suitable for cell culture and tissue engineering. Finally, we show that the hydrogel network structure is also important for the mechanical properties of hydrogels. The hydrogels made of the same protein building blocks but with different network structures can have distinct mechanical responses. These studies provide a biomechanical basis for the rational design of synthetic proteins hydrogels with predictable and controllable mechanical properties.
Engineered Protein Fibers with Reinforced Mechanical Properties Via β-Sheet High-Order Assembly

Abstract:Protein fibers are ideal alternatives to synthetic polymers due to their unique mechanical properties, biocompatibility, and sustainability. However, engineering biomimetic protein fibers with high mechanical properties remains challenging, particularly in mimicking the high molecular weight of natural proteins and regulating their complex hierarchical structures. Here, a modular design and multi-scale assembly strategy is developed to manufacture robust protein fibers using low- or medium-molecular-weight proteins. The distinct functional and structural properties of flexible, rigid, and cross-linked domains in modular proteins are skillfully harnessed. By regulating the ratio of rigid to flexible domains, the formation of high-order β-sheet crystals aligned along the fiber axis is promoted, enhancing both strength and toughness. Furthermore, the dynamic imine cross-linking network, formed by the aldehyde-amine condensation reaction of the cross-linked domains, further reinforces the protein fibers. Remarkably, fibers spun from modular proteins significantly smaller than natural spidroin exhibit outstanding mechanical properties, surpassing those of protein fibers with same or even higher molecular weights. This strategy offers a promising pathway for fabricating protein fibers suitable for diverse applications.
Engineered Protein Hydrogels as Biomimetic Cellular Scaffolds

Abstract:The biochemical and biophysical properties of the extracellular matrix (ECM) play a pivotal role in regulating cellular behaviors such as proliferation, migration, and differentiation. Engineered protein-based hydrogels, with highly tunable multifunctional properties, have the potential to replicate key features of the native ECM. Formed by self-assembly or crosslinking, engineered protein-based hydrogels can induce a range of cell behaviors through bioactive and functional domains incorporated into the polymer backbone. Using recombinant techniques, the amino acid sequence of the protein backbone can be designed with precise control over the chain-length, folded structure, and cell-interaction sites. In this review, the modular design of engineered protein-based hydrogels from both a molecular- and network-level perspective are discussed, and summarize recent progress and case studies to highlight the diverse strategies used to construct biomimetic scaffolds. This review focuses on amino acid sequences that form structural blocks, bioactive blocks, and stimuli-responsive blocks designed into the protein backbone for highly precise and tunable control of scaffold properties. Both physical and chemical methods to stabilize dynamic protein networks with defined structure and bioactivity for cell culture applications are discussed. Finally, a discussion of future directions of engineered protein-based hydrogels as biomimetic cellular scaffolds is concluded.

Abstract:Rubbers are strategically important due to their indispensable applications in the daily life and high-tech fields. For their real-world applications, the covalent cross-linking, reinforcement, and malleability of rubbers are three important issues because they are closely related to the elasticity, mechanical properties, and recycling of the rubber materials. Herein, we demonstrate a simple way to prepare covalently cross-linked yet recyclable and robust elastomeric vitrimer composites by incorporating exchangeable β-hydroxyl ester bonds into the elastomer-nanoparticle interface using epoxy group-functionalized silica (Esilica) as both cross-linker and reinforcement in carboxyl group-grafted styrene-butadiene rubber (CSBR). The Esilica-cross-linked CSBR composites exhibit promising mechanical properties due to the covalent linkages in the interface and fine silica dispersion in the matrix. In addition, the interface can undergo dynamic reshuffling via transesterification reactions to alter network topology at high temperatures, conferring the resulting composites the ability to be reshaped and recycled.

Abstract:The unique biophysical and biochemical properties of intrinsically disordered proteins (IDPs) and their recombinant derivatives, intrinsically disordered protein polymers (IDPPs) offer opportunities for producing multistimuli‐responsive materials; their sequence‐encoded disorder and tendency for phase separation facilitate the development of multifunctional materials. This review highlights the strategies for enhancing the structural diversity of elastin‐like polypeptides (ELPs) and resilin‐like polypeptides (RLPs), and their self‐assembled structures via genetic fusion to ordered motifs such as helical or beta sheet domains. In particular, this review describes approaches that harness the synergistic interplay between order‐promoting and thermoresponsive building blocks to design hybrid biomaterials, resulting in well‐structured, stimuli‐responsive supramolecular materials ordered on the nanoscale.

Abstract:Over the past few decades, polymeric hydrogels have gained significant progress in various fields. As the most biochemically and structurally functional macromolecules in biological organisms, proteins have structural and mechanical properties similar to that of natural extracellular matrices (ECM). By combination of polymers with protein, protein-polymer hydrogel can be constructed as biomaterials with diverse properties and possess a wide range of applications. In this review, we summarized the progress of research on protein-polymer hydrogels and introduced the common fabrication methods. The properties of the protein-polymer hydrogels were briefly described, including biocompatibility, adhesion properties, responsiveness, mechanical properties, swelling properties, and degradability. In addition, we also reviewed the applications of protein-polymer hydrogels in tissue engineering, drug delivery and encapsulation, wearable sensors, adsorption, and other applications. Finally, we summarize the research on protein-polymer hydrogels and highlight the current challenges and future prospects of the field, which may provide a meaningful reference for the field of protein-polymer hydrogels.

Abstract:Protein-based polymers are some of the most promising candidates for a new generation of innovative biomaterials as recent advances in genetic-engineering and biotechnological techniques mean that protein-based biomaterials can be designed and constructed with a high degree of complexity and accuracy. Moreover, their sequences, which are derived from structural protein-based modules, can easily be modified to include bioactive motifs that improve their functions and material-host interactions, thereby satisfying fundamental biological requirements. The accuracy with which these advanced polypeptides can be produced, and their versatility, self-assembly behavior, stimuli-responsiveness and biocompatibility, means that they have attracted increasing attention for use in biomedical applications such as cell culture, tissue engineering, protein purification, surface engineering and controlled drug delivery. The biopolymers discussed in this review are elastin-derived protein-based polymers which are biologically inspired and biomimetic materials. This review will also focus on the design, synthesis and characterization of these genetically encoded polymers and their potential utility for controlled drug and gene delivery, as well as in tissue engineering and regenerative medicine.

Abstract:Reinforcement, recycling, and functional applications are three important issues in elastomer science and engineering. It is of great importance, but rarely achievable, to integrate these properties into elastomers. Herein, we report a simple way to prepare covalently cross-linked yet recyclable, robust, and macroscopically responsive elastomer vitrimers by engineering exchangeable bonds into rubber–carbon nanodot (CD) interphase using CD as high-functionality cross-linker. The cross-linked rubbers can rearrange the network topology through transesterification reactions in the interphase, conferring the materials the ability to be recycled, reshaped, and welded. The relatively short chains bridging adjacent CD are highly stretched and preferentially rupture to dissipate energy under external force, resulting in remarkable improvements on the mechanical properties. Moreover, the malleable and welding properties allow the samples to access reconfigurable/multiple shape memory effects.

Abstract:Human beings cannot live without elastomers, which exist almost everywhere in human life nowadays. Although elastomers with a variety of features are developed for specific applications, a timeless topic is how to toughen them, because mechanical properties always determine the reliability and lifespan of the utilized elastomers. Toughening of a soft material has a long history and the main idea is widely accepted, i.e., building energy dissipation into the stretchy networks. Double network (DN) method is the most pioneering and representative practice of materials toughening, which is successfully applied to numerous hydrogel systems. Intensive studies on DN hydrogel systems are implemented and relevant reviews are well documented, whereas important reviews about DN elastomers are absent. In this review, recent progress on toughening elastomers using the DN strategy is reviewed. By reviewing the fabrication methods and relevant extensions, toughening mechanisms at different length scales, functionalities, and applications in sequence, it is shown how one polymer network plus another gives rise to a tough elastomer with superior mechanical properties. It is believed that this review can give some insight into existing DN elastomer systems and inspire the development of next‐generation tough soft materials. Intensive studies on double network (DN) hydrogels are implemented and relevant reviews are well documented, whereas important reviews about DN elastomers are absent. Recent progress in toughening elastomers using the DN strategy reveals how one polymer network plus another gives rise to a tough elastomer with mechanical properties that far exceed that of a simple mixture of two individual networks.

Abstract:<p>Engineered living materials (ELMs) have drawn intense interest from both academia and industry in recent years. The essence of ELMs is the use of living cells to produce molecular building blocks, direct their hierarchical organization, and convert life into functional materials. By doing so, it confers living features on materials, including self-organization, self-maintaining, adaptability and evolvability. As the workhorse of life, proteins play an essential role in current ELM designs. To harness proteins for applications in ELMs, protein engineering is usually required to tailor their assembly, chemistry, functions, and their interactions with abiotic materials. In this review, we discuss the roles of proteins in ELMs and summarize the applications of protein engineering in developing molecular tools toward the creation of advanced ELMs with novel properties. Inspired by systems chemistry, we emphasize that future development of ELMs would benefit from a systems perspective by integrating a rich and versatile interaction network comprising multiple functional components such as genes, RNAs, proteins, and even many other abiotic components.</p>
