Abstract:Aptamer with small molecular weight, simple structure, and easy synthesis, which can be used repeatedly and preserved for long time, has important applications in biosensor field. This article describes several commonly used methods for fixing aptamer onto the sensor surfaces, including the gold-sulfur self-assembled monolayer, covalent bond, biotin/avidin affinity, complementary nucleic acid chain connection, as well as current research progress and the characteristics of each method. Development of aptasensors with wide detection range, low detection limit, short detection time, and capability of detection of multitargets will certainly facilitate its application in food quality and safety testing, disease diagnosis, drug analysis, environment monitoring, etc.

Abstract:Graphene and graphene-like two-dimensional nanomaterials have aroused tremendous research interest in recent years due to their unique electronic, optical, and mechanical properties associated with their planar structure. Aptamers have exhibited many advantages as molecular recognition elements for sensing devices compared to traditional antibodies. The marriage of two-dimensional nanomaterials and aptamers has emerged many ingenious aptasensing strategies for applications in the fields of clinical diagnosis and food safety. This review highlights current advances in the development and application of two-dimensional nanomaterials-based aptasensors with the focus on two main signal-transducing mechanisms, i.e. electrochemical and optical. A special attention is paid to graphene, a one-atom thick layer of graphite with exceptional properties, representing a fastgrowing field of research. In view of the unique properties of two-dimensional nanostructures and their inherent advantages of synthetic aptamers, we expect that high-performance two-dimensional nanomaterials-based aptasensing devices will find extensive applications in environmental monitoring, biomedical diagnostics, and food safety.

Abstract:Given the importance of public health, it is crucial to develop quick, targeted, highly sensitive, and accurate technologies to monitor pathogenic microbes in response to the growing concerns of food and environmental safety. Although conventional approaches for microbiological detection are available, they are laborious, and often skill demanding. Therefore, such approaches are incompetent in the on-site or high-throughput assessment of pathogenic microbes. Numerous efforts have been made to develop biosensors that use nucleic acid aptamer as the biorecognition element, which would avoid the abovementioned limitations. Incorporating nanomaterials (NMs) into aptamer-based biosensors (aptasensors) improves their sensitivity and specificity, opening exciting possibilities for various applications, such as bioanalysis of food and environmental samples. Over the last decade, nanomaterial-conjugated aptasensors have seen a steadily rising demand. To this end, the main goal of this study is to demonstrate the novelty in the design of nanomaterial-conjugated aptasensors and how they can be used to detect different pathogenic microbes in water and food. The intent of this paper is to evaluate the cutting-edge techniques that have appeared in nano-aptasensors throughout the past few years, such as manufacturing procedures, analytical credibility, and sensing mechanisms. Additionally, the fundamental performance parameters of aptasensing techniques (such as detection limits, and sensing ranges response) were also used to evaluate their practical applicability. Finally, it is anticipated that this study will inspire innovative ideas and techniques for the construction and use of aptasensors for monitoring pathogenic microorganisms in food, drinks, recreational water, and wastewater.

Abstract:The combination of nanomaterials (NMs) and aptamers into aptasensors enables highly specific and sensitive detection of diverse pollutants. The great potential of aptasensors is recognized for the detection of diverse emerging organic pollutants (EOPs) in different environmental and biological matrices. In addition to high sensitivity and selectivity, NM-based aptasensors have many other advantages such as portability, miniaturization, facile use, and affordability. This work showcases the recent advances achieved in the design and fabrication of NM-based aptasensors for monitoring EOPs (e.g., hormones, phenolic contaminants, pesticides, and pharmaceuticals). On the basis of their sensing mechanisms, the aptasensing systems can be classified as electrochemical, colorimetric, PEC, fluorescence, SERS, and ECL aptasensors. Special attention has been paid to the fabrication processes, analytical reliability, and sensing mechanisms of NM-based aptasensors. Further, the practical utility of aptasensing approaches has also been assessed based on their basic performance metrics (e.g., detection limits, sensing ranges, and response times).

Abstract:<div><p>Nanobiosensors based on aptamer are extensively being studied as potent analytical tools in clinical analysis. These biosensors provide high sensitivity, fast response, specificity and desired portability in addition to simplicity and decreased cost compared to conventional methods. The purpose of this manuscript is to provide readers with an overview of current advances about electrochemical, electrochemiluminescent and photoelectrochemical aptasensors from the sea of available literature. These are mainly used for determination of protein-based biomarkers, especially for cancer diagnosis. Here in we have given special emphasis on nanosize-based aptasensors which have been reported to show considerable improvement in the analytical performance.</p></div>

Abstract:The past decade has witnessed extensive research in the field of biosensors, where nanoaptasensors achieved widespread interest. Aptamer, the single-stranded DNA or RNA nucleotide, is extensively employed as a bioreceptor due to its stability, ease in a modification that ensures convenient immobilization strategies, reducing the cost of sensor manufacturing. The detection limit and sensitivity were notably improved when modified with nanomaterials such as metal nanoparticles, carbon nanomaterials, graphene, quantum dots, and other nanocomposites. This paper introduces various design strategies for the fabrication of sensors utilizing aptamers and nanomaterials in ¬developing signal-readout mechanism, focusing mainly on the latest research in food, biomedical and environmental applications. Aptasensors that utilize various signal recognition methods such as electrochemistry, colorimetry, luminescence, and fluorescence are highlighted. This review offers a wide range of outlook for future developments of aptamer bioreceptors employing the unique physicochemical nanomaterials as efficient transducers and amplifiers. Furthermore, this review will also provide an insight into aptamer-aided biosensors' technology over the last three to five years of work.

Abstract:Aptamers are single-stranded oligonucleotides (DNA or RNA) capable of folding into well-defined structures and motifs that allow them bind to various target molecules with high specificity and affinity. The crucial ability of aptamers is interaction with a given target from small ions to molecular level that makes them useful in many applications for specific detection of various analytes. Nowadays aptamer is an effective tool not only for in vitro research in diagnostic methods, drug delivery and treatment therapy, but also for the development of nucleic-acid-based sensors in industrial waste management, pollution control and environmental toxicology. This mini review is focused on discussion and updating about the global researches in nucleic-acid-based sensors for environmental issues.

Abstract:The frequent occurrence of food safety incidents has aroused public concern about food safety and key contaminants. Foodborne pathogen contamination, pesticide residues, heavy metal residues, and other food safety problems will significantly impact human health. Therefore, developing efficient and sensitive detection method to ensure food safety early warning is paramount. The aptamer-based sensor (aptasensor) is a novel analytical tool with strong targeting, high sensitivity, low cost, etc. It has been extensively utilized in the pharmaceutical industry, biomedicine, environmental engineering, food safety detection, and in other diverse fields. This work reviewed the latest research progress of aptasensors for food analysis and detection, mainly introducing their application in detecting various key food contaminants. Subsequently, the sensing mechanism and performance of aptasensors are discussed. Finally, the review will examine the challenges and opportunities related to aptasensors for detecting major contaminants in food, and advance implementation of aptasensors in food safety and detection.

Abstract:In view of ever-increasing environmental pollution, there is an immediate requirement to promote cheap, multiplexed, sensitive and fast biosensing systems to monitor these pollutants or contaminants. Aptamers have shown numerous advantages in being used as molecular recognition elements in various biosensing devices. Graphene and graphene-based materials/nanohybrids combined with several detection methods exhibit great potential owing to their exceptional optical, electronic and physicochemical properties which can be employed extensively to monitor environmental contaminants. For environmental monitoring applications, aptamers have been successfully combined with graphene-based nanohybrids to produce a wide range of innovative methodologies. Aptamers are immobilized at the surface of graphene based nanohybrids via covalent and non-covalent strategies. This review highlights the design, working principle, recent developmental advances and applications of graphene based nanohybrid aptasensors (GNH-Apts) (since January 2014 to September 2021) with a special emphasis on two major signal-transduction methods, i.e., optical and electrochemical for the monitoring of pesticides, heavy metals, bacteria, antibiotics, and organic compounds from different environmental samples (e.g., water, soil and related). Lastly, the challenges confronted by scientists and the possible future outlook have also been addressed. It is expected that high-performance graphene-based nanohybrid aptasensors would find broad applications in the field of environmental monitoring.

Abstract:Aptamers are synthetic bio-receptors of deoxyribonucleic acid (DNA) or ribonucleic acid (RNA) origin selected by the systematic evolution of ligands (SELEX) process that bind a broad range of target analytes with high affinity and specificity. So far, electrochemical biosensors have come up as a simple and sensitive method to utilize aptamers as a bio-recognition element. Numerous aptamer based sensors have been developed for clinical diagnostics, food, and environmental monitoring and several other applications are under development. Aptasensors are capable of extending the limits of current analytical techniques in clinical diagnostics, food, and environmental sample analysis. However, the potential applications of aptamer based electrochemical biosensors are unlimited; current applications are observed in the areas of food toxins, clinical biomarkers, and pesticide detection. This review attempts to enumerate the most representative examples of research progress in aptamer based electrochemical biosensing principles that have been developed in recent years. Additionally, this account will discuss various current developments on aptamer-based sensors toward heavy metal detection, for various cardiac biomarkers, antibiotics detection, and also on how the aptamers can be deployed to couple with antibody-based assays as a hybrid sensing platform. Aptamers can be used in various applications, however, this account will focus on the recent advancements made toward food, environmental, and clinical diagnostic application. This review paper compares various electrochemical aptamer based sensor detection strategies that have been applied so far and used as a state of the art. As illustrated in the literature, aptamers have been utilized extensively for environmental, cancer biomarker, biomedical application, and antibiotic detection and thus have been extensively discussed in this article.

Abstract:Aptasensors are promising biosensors, which take advantage of using aptamers as a recognition element. The combination of the excellent characteristics of aptamers and the leading detection platform techniques, such as optical, electrochemical with nanomaterial-integrated, or mass-sensitive techniques with high sensitivity and specificity draws a promising view for the application of the aptasensors for the detection of harmful small toxic chemicals and real-time monitoring in the environments. In spite of attraction of aptasensors, application of them is limited to the complex environment due to the facts that how the immobilization of aptamers onto the surface affects the functions of aptamers and their structures for the detection of environmental contaminants are not clearly known. This review examines the most recent update on the selection of aptamers for small molecules, the development and application of aptasensors in the detection of small molecule contaminants in environment. Additionally, their applications to the real samples as environmental monitoring reported in the publications also are reviewed.

Abstract:As a major cause of deaths in developed countries, cardiovascular diseases (CVD) are a steadily growing burden for health systems. In particular, early and rapid detection of CVD is important for on time appropriate therapy so as to reduce the mortality rate. Aptamers, known as single-stranded DNA/RNA or oligonucleotides containing receptors and/or catalytic properties, have been widely employed in biodetection platforms due to their beneficial properties. Like antibodies, aptamers have served as artificial target receptors in affinity biosensors. Currently, advanced biosensors with improved sensitivity and specificity are fabricated by the synergistic combination of aptamers and diverse nanomaterials. Herein, we review the current development and applications of nanomaterial-based aptasensors for the recognition of CVD biomarkers with special emphasis on electrochemical and optical technologies. The performance of aptasensors has been assessed further in terms of key quality assurance metrics along with discussions on recent technologies developed for the amplification of signals with enhanced portability.

Abstract:Recent developments in biotechnology offer the new methods for the sensitive detection of heavy metals based on the affinity and specificity of aptamers, as nucleic acid ligands selected from random sequence pools in vitro. Heavy metals have received considerable importance as the most toxic metallic pollutants which may cause serious environmental damages. They are classified as trace elements because of their presence in trace concentrations in various environmental matrices. Thus, the precise and sensitive methods to detect heavy metals are important to ensure human and environment safety. Aptamers as the biological probes, show high binding affinity which can often be directly translated into high detection sensitivity. On the other hand, high selectivity and stability make them possible to detect a wide range of targets, especially metallic ions. This review provides current progress of aptamers for environmental and biological monitoring of heavy metals using the nanomaterials mainly in two groups: (i) aptamer based biosensors (aptasensors) and (ii) aptamer based biosorbents (aptasorbents). The introduction of nanomaterials can efficiently increase the immobilization quantity of aptamers. Furthermore, they play an important role in the orientation and assembly density controlling of aptamers for the optimized recognition ability.

Abstract:Heavy metals ions as metallic pollutants are a growing global issue due to their adverse effects on the aquatic ecosystem, and human health. Unfortunately, conventional detection methods such as atomic absorption spectrometry exhibit a relatively low limit of detection and hold numerous disadvantages, and therefore, the development of an efficient method for in-situ and real-time detection of heavy metal residues is of great importance. The aptamer-based sensors offer distinct advantages over antibodies and emerged as a robust sensing platform against various heavy metals due to their high sensitivity, ease of production, simple operations, excellent specificity, better stability, low immunogenicity, and cost-effectiveness. The nucleic acid aptamers in conjugation with nanomaterials can bind to the metal ions with good specificity/selectivity and can be used for on-site monitoring of metal ion residues. This review aimed to provide background information about nanomaterials-based aptasensor, recent advancements in aptamer conjunction on nanomaterials surface, the role of nanomaterials in improving signal transduction, recent progress of nanomaterials-based aptasening procedures (from 2010 to 2022), and future perspectives toward the practical applications of nanomaterials-based aptasensors against hazardous metal ions for food safety and environmental monitoring.

Abstract:As emerging platforms, nanoporous-based biosensors have potentially useful applications for sensitive and selective detection of various analytes. The integration of nanoporous materials ( e.g. , nanocarbons, mesoporous, metal–organic frameworks (MOFs), and MOF-based nanocomposites, etc. ) with various transduction techniques, such as electrochemical, optical, and mass-sensitized, has led to the development of biosensors for a wide spectrum of applications in healthcare, environmental monitoring, and food safety. These biosensors take advantage of the unique properties of nanoporous materials, such as tunable pore size and selective surface chemistry with high surface area, and enhance the interaction between the analyte and the recognition element. Overall, biosensors based on nanoporous material have very low limits of detection and appropriate linear range. This review focuses on recent developments in the design of biosensors based on nanoporous materials, including sensing mechanisms and applications.

Abstract:Nanomaterials have become one of the most interesting sensing materials because of their unique size- and shape-dependent optical properties, high surface energy and surface-to-volume ratio, and tunable surface properties. Aptamers are oligonucleotides that can bind their target ligands with high affinity. The use of nanomaterials that are bioconjugated with aptamers for selective and sensitive detection of analytes such as small molecules, metal ions, proteins, and cells has been demonstrated. This review focuses on recent progress in the development of biosensors by integrating functional aptamers with different types of nanomaterials, including quantum dots, magnetic nanoparticles (NPs), metallic NPs, and carbon nanotubes. Colorimetry, fluorescence, electrochemistry, surface plasmon resonance, surface-enhanced Raman scattering, and magnetic resonance imaging are common detection modes for a broad range of analytes with high sensitivity and selectivity when using aptamer bioconjugated nanomaterials (Apt-NMs). We highlight the important roles that the size and concentration of nanomaterials, the secondary structure and density of aptamers, and the multivalent interactions play in determining the specificity and sensitivity of the nanosensors towards analytes. Advantages and disadvantages of the Apt-NMs for bioapplications are focused.

Abstract:Arsenic (As) is a highly toxic contaminant in the environment and a serious carcinogen for the human being. The toxicity of arsenic significantly threatens environmental and human health. The effective removing technology for arsenic remains challenging, and one of the reasons is due to the lack of powerful detection method in the complex environmental matrix. There is thus an urgent need to develop novel analytical methods for arsenic, preferably with the potential for the field-testing. To combat arsenic pollution and maintain a healthy environment and eco-system, many analytical methods have been developed for arsenic detection in various samples. Among these strategies, biosensors hold great promise for rapid detection of arsenic, in particular, nanomaterials-based aptamer sensors have attracted significant attention due to their simplicity, high sensitivity and rapidness. In this paper, we reviewed the recent development and applications of aptamer sensors (aptasensors) based-on nanomaterial for arsenic detection, in particular with emphasis on the works using optical and electrochemical technologies. We also discussed the recent novel technology in aptasensors development for arsenic detection, including nucleic acid amplification for signal enhancement and device integration for the portability of arsenic sensors. We are hoping this review could inspire further researches in developing novel nanotechnologies based aptasensors for possible on-site detection of arsenic.

Abstract:Due to their relative synthetic and chemical simplicity compared to antibodies, aptamers afford enhanced stability and functionality for the detection of environmental contaminants and for use in environmental monitoring. Furthermore, nucleic acid aptamers can be selected for toxic targets which may prove difficult for antibody development. Of particular relevance, aptamers have been selected and used to develop biosensors for environmental contaminants such as heavy metals, small-molecule agricultural toxins, and water-borne bacterial pathogens. This review will focus on recent aptamer-based developments for the detection of diverse environmental contaminants. Within this domain, aptamers have been combined with other technologies to develop biosensors with various signal outputs. The goal of much of this work is to develop cost-effective, user-friendly detection methods that can complement or replace traditional environmental monitoring strategies. This review will highlight recent examples in this area. Additionally, with innovative developments such as wearable devices, sentinel materials, and lab-on-a-chip designs, there exists significant potential for the development of multifunctional aptamer-based biosensors for environmental monitoring. Examples of these technologies will also be highlighted. Finally, a critical perspective on the field, and thoughts on future research directions will be offered.

Thus, understanding the concept of biosensing has enabled researchers to develop nanobiosensors with different nanomaterials and composites to improve the sensitivity as well as the specificity of pathogen detection. The application of nanomaterials has enabled researchers to use advanced technologies in biosensors for the transfer of signals to enhance their efficiency and sensitivity. Nanomaterials like carbon nanotubes, magnetic and gold, dendrimers, graphene nanomaterials and quantum dots are predominantly used for developing biosensors with improved specificity and sensitivity of detection due to their exclusive chemical, magnetic, mechanical, optical and physical properties. All nanoparticles and new composites used in biosensors need to be classified and categorized for their enhanced performance, quick detection, and unobtrusive and effective use in foodborne analysis. Hence, this review intends to summarize the different sensing methods used in foodborne pathogen detection, their design, working principle and advances in sensing systems.

Abstract:Rapid detection and identification of pathogenic microorganisms is fundamental to minimizing the spread of infectious disease, and informing clinicians on patient treatment strategies. This need has led to the development of enhanced biosensors that utilize state of the art nanomaterials and nanotechnology, and represent the next generation of diagnostics. A primer on nanoscale biorecognition elements such as, nucleic acids, antibodies, and their synthetic analogs (molecular imprinted polymers), will be presented first. Next the application of various nanotechnologies for biosensor transduction will be discussed, along with the inherent nanoscale phenomenon that leads to their improved performance and capabilities in biosensor systems. A future outlook on characterization and quality assurance, nanotoxicity, and nanomaterial integration into lab-on-a-chip systems will provide the closing thoughts.
