Abstract:Abstract Mycotoxins are secondary metabolites produced by fungus. Many mycotoxin species are highly toxic and are frequently found in cereals and feedstuffs. So, powerful detection methods are vital and effective ways to prevent feed contamination. Traditional detection methods can no longer meet the needs of massive, real-time, simple, and fast mycotoxin monitoring. Rapid detection methods based on advanced material and sensor technology are the future trend. In this review, we highlight recent progress of mycotoxin rapid detection strategies in feedstuffs and foods, especially for simultaneous multiplex mycotoxin determination. Immunoassays, biosensors, and the prominent roles of nanomaterials are introduced. The principles of different types of recognition and signal transduction are explained, and the merits and pitfalls of these methods are compared. Furthermore, limitations and challenges of existing rapid sensing strategies and perspectives of future research are discussed.

Abstract:<p>Mycotoxin contamination in foods and other commodities has been a global concern due to their serious toxicity and public health threat, as well as fearful international trade loss. Thus, sensitive, rapid and reliable tools for mycotoxin detection are in urgent demand. Recently, immunosensors have become a promising candidate for the highly-efficient determination of diverse mycotoxins thanks to their superior performances. This review initially surveys the current status concerning mycotoxin residues in different plant/animal-derived foods, and other commodities, then, primarily highlights the recent advances of various newly-developed immunosensors for mycotoxin determination over the past five years. Special emphasis is given to the fluorescent, colorimetric, chemiluminescent, electrochemiluminescent, surface plasmon resonance, surface-enhanced Raman spectroscopy, and electrochemical sensors. The real-world application, along with their advantages and potential limits, as well as current challenges and future perspectives related to these developed immunosensors for mycotoxin detection, is comprehensively discussed. It is hoped to provide rewarding insights to the future development and broad application of immunosensors for researchers in different fields.</p>

Abstract:This review focuses on recent developments in immunochemical methods for detection of mycotoxins, with a particular emphasis on simultaneous multiple analyte determination. This includes high-throughput instrumental analysis for the laboratory environment (microtitre plate enzyme-linked immunoabsorbant assay (ELISA), different kinds of immunosensors, fluorescence polarization immunoassay, and capillary electrophoretic immunoassay), as well as rapid visual tests for on-site testing (lateral-flow, dipstick, flow-through and column tests). For each type of immunoassay, perspectives for multiple analyte application are discussed and examples cited.
Rapid Detection of Four Mycotoxins in Corn Using a Microfluidics and Microarray-Based Immunoassay System.

Abstract:Mycotoxins threaten human health seriously because they usually exist in food, fodder and commodities. In this study, a rapid and sensitive immunoassay system for commonly encountered mycotoxins was established based on microfluidics and protein microarrays. Four mycotoxins (T-2 toxin, aflatoxin B1, ochratoxin A, and zearalenone) can be automatically detected in a custom-made microdevice within 30 min under the assistance of a prototype of the instrument with a fluid control system and an imaging system. Once the microdevices are fabricated, they are small-sized and user-friendly. Standard curves for each of the studied mycotoxins were generated with a good logistic correlation (R2 > 0.98). Working ranges from 0.1 to 20 ng/ml were employed in the immunoassay being the limit of detection achieved between 0.03 and 1.24 ng/ml. These values were calculated when the four mycotoxins were present in samples at the same time. Samples of spiked water and field corn were tested to assess the performance of our microfluidic-based detection technique for the mycotoxins. Recovery rates of mycotoxins from spiked water and corn samples were accessed and the results ranged from 80% to 110%, where the intra-assay coefficients of variation were under 15%. In summary, the system can realize rapid and reliable detection of multiple contaminants in actual samples automatically.
Rapid and simultaneous detection of five mycotoxins and their analogs with a gold nanoparticle-based multiplex immuno-strip sensor

Abstract:Mycotoxins, as secondary metabolites produced by fungi, have been the focus of researchers in various countries and are considered to be one of the major risk factors in agricultural products. There is an urgent need for a rapid, simple and high-performance method to detect residues of harmful mycotoxins in agricultural foods. We have developed a gold nanoparticle-based multiplexed immunochromatographic strip biosensor that can simultaneously detect fifteen mycotoxins in cereal samples. With this optimized procedure, five representative mycotoxins, deoxynivalenol (DON), zearalenone (ZEN), T-2 toxin (T-2), tenuazonic acid (TEA) and alternariol (AOH) were detected in the range of 0.91-4.77, 0.04-0.56, 0.11-0.68, 0.12-1.02 and 0.09-0.75 ng/mL, respectively. The accuracy and stability of these measurements were demonstrated by analysis of spiked samples with recoveries of 91.8%-115.3% and coefficients of variation <8.7%. In addition, commercially available samples of real cereals were tested using the strips and showed good agreement with the results verified by LC-MS/MS. Therefore, Our assembled ICA strips can be used for the simultaneous detection of 5 mycotoxins and their analogs (15 mycotoxins in total) in grain samples, and the results were consistent between different types of cereal foods, this multiplexed immunochromatographic strip biosensor can be used as an effective tool for the primary screening of mycotoxin residues in agricultural products.
Antibody Microarray Immunoassay for Simultaneous Quantification of Multiple Mycotoxins in Corn Samples.

Abstract:We developed and tested a prototype of an antibody microarray immunoassay for simultaneous quantitative detection of four typical mycotoxins (aflatoxin B1, ochratoxin A, zearalenone, and fumonisin B1) in corn samples. The test kit consisted of a nitrocellulose membrane layered with immobilized monoclonal antibodies against mycotoxins. During the assay, the mycotoxin-protein conjugates were biotinylated. The signal detection was enhanced by a combination of the biotin-streptavidin system and enhanced chemiluminescence (ECL). This improved the sensitivity of the assay. Under the optimized conditions, four calibration curves with goodness of fit (R2 > 0.98) were plotted. The results showed that the detection limits for aflatoxin B1, ochratoxin A, zearalenone, and fumonisin B1 were 0.21, 0.19, 0.09, and 0.24 ng/mL, with detection ranges of 0.47–55.69, 0.48–127.11, 0.22–31.36, and 0.56–92.57 ng/mL, respectively. The limit of detection (LOD) of this antibody microarray for aflatoxin B1, ochratoxin A, zearalenone, and fumonisin B1 in corn was 5.25, 4.75, 2.25, and 6 μg/kg, respectively. The recovery rates from the spiked samples were between 79.2% and 113.4%, with coefficient of variation <10%. The results of the analysis of commercial samples for mycotoxins using this new assay and the liquid chromatography-tandem mass spectrometry (LC-MS/MS) were comparable and in good agreement. This assay could also be modified for the simultaneous detection of other multiple mycotoxins, as well as low-weight analytes, hazardous to human health.

Abstract:Mycotoxin contamination has become a challenge in the field of food safety testing, given the increasing emphasis on food safety in recent years. Mycotoxins are widely distributed, in heavily polluted areas. Food contamination with these toxins is difficult to prevent and control. Mycotoxins, as are small-molecule toxic metabolites produced by several species belonging to the genera Aspergillus, Fusarium, and Penicillium growing in food. They are considered teratogenic, carcinogenic, and mutagenic to humans and animals. Food systems are often simultaneously contaminated with multiple mycotoxins. Due to the additive or synergistic toxicological effects caused by the co-existence of multiple mycotoxins, their individual detection requires reliable, accurate, and high-throughput techniques. Currently available, methods for the detection of multiple mycotoxins are mainly based on chromatography, spectroscopy (colorimetry, fluorescence, and surface-enhanced Raman scattering), and electrochemistry. This review provides a comprehensive overview of advances in the multiple detection methods of mycotoxins during the recent 5 years. The principles and features of these techniques are described. The practical applications and challenges associated with assays for multiple detection methods of mycotoxins are summarized. The potential for future development and application is discussed in an effort, to provide standards of references for further research.

Abstract:The co-occurrence and accumulation of mycotoxin in food and feed constitutes a major issue to food safety, food security, and public health. Accurate and sensitive mycotoxins analysis can avoid toxin contamination as well as reduce food wastage caused by false positive results. This mini review focuses on the recent advance in detection methods for multiple mycotoxins, which mainly depends on immunoassay technologies. Advance immunoassay technologies integrated in mycotoxin analysis enable simultaneous detection of multiple mycotoxins and enhance the outcomes' quality. It highlights toxicogenomic as novel approach for hazard assessment by utilizing computational methods to map molecular events and biological processes. Indeed, toxicogenomic is a powerful tool to understand health effects from mycotoxin exposure as it offers insight on the mechanisms by which mycotoxins exposures cause diseases.

Abstract:Mycotoxins are a class of secondary metabolites produced by filamentous fungi that are harmful to humans and animals. Here, a multiplex immunochromatographic assay was reported, which is based on 25 nm colloidal gold for simultaneous qualitative detection of fumonisin B-1 (FB1), deoxynivalenol (DON) and zearalenone (ZEN) in wheat and corn samples. After conditions optimization, the immunochromatographic strip has high sensitivity. The visual detection limits of the immunochromatographic WA strips for FB1, DON, and ZEN reached 60, 12.5 and 6 ng/mL, respectively. In addition, the immunochromatographic strip has high specificity, no reaction to each other, or no cross-reactivity with ochratoxin A (OTA) and aflatoxin B-1 (AEB(1)) Furthermore, eighteen samples (including corn and wheat) were tested using the WA strips and verified by liquid chromatography-mass spectrometry (LC-MS/MS). The results of strips were found to be consistent with those of liquid chromatography-mass spectrometry (LC-MS/MS). In short, this work demonstrates that the spherical colloidal gold-based immunochromatographic test strip we developed can be used to quickly and simultaneously monitor the existence of multiple mycotoxins in agricultural food samples.

Abstract:<p>Mycotoxins can accumulate in different organs or tissues by ingestion or skin contact and cause several health problems to humans. Thus, developing a rapid detection method for mycotoxins is important. Lateral flow immunoassay (LFI), with outstanding advantages of convenience and rapidity, is especially suitable for on-site monitoring of mycotoxin contamination in food. Some existing LFI have been considerably improved by combining different kinds of nanosensors or strategies for increasing sensitivity or efficiency. In this review, we summarized nanoparticles-based LFI (e.g., colored, luminescent, or magnetic nanoparticles) for mycotoxins detection. Enzyme, enhancement, or specific recognition elements-based LFI are also briefly explained. The LFI, which allows multiple detections, has paid more attention to mycotoxins preliminary screening. This review focuses on examples of developed LFI for mycotoxins to illustrate novel concepts as screening tools. We believe this review will attract considerable attention and stimulate more distinctive studies for the rapid detection of mycotoxins.</p>

Abstract:Mycotoxins contaminated in agricultural products are often highly carcinogenic and genotoxic to humans. With the streamlining of the food industry chain and the improvement of food safety requirements, the traditional laboratory testing mode is constantly challenged due to the expensive equipment, complex operation steps, and lag in testing results. Therefore, rapid detection methods are urgently needed in the food safety system. This review focuses on the latest strategies that can achieve rapid and on-site testing, with particular attention to the nanomaterials integrated biosensors. To provide researchers with the latest trends and inspiration in the field of rapid detection, we summarize several strategies suitable for point of care testing (POCT) of mycotoxins, including enzyme-linked immunoassay (ELISA), lateral flow assay (LFA), fluorescence, electrochemistry, and colorimetry assay. POCT-based strategies are all developing towards intelligence and portability, especially when combined with smartphones, making it easier to read signals for intuitive access and analysis of test data. Detection performance of the devices has also improved considerably with the integration of biosensors and nanomaterials.

Abstract:Mycotoxins, secondary metabolites synthesized by various filamentous fungi genera such as Aspergillus, Penicillium, Fusarium, Claviceps, and Alternaria, are potent toxic compounds. Their production is contingent upon specific environmental conditions during fungal growth. Arising as byproducts of fungal metabolic processes, mycotoxins exhibit significant toxicity, posing risks of acute or chronic health complications. Recognized as highly hazardous food contaminants, mycotoxins present a pervasive threat throughout the agricultural and food processing continuum, from plant cultivation to post-harvest stages. The imperative to adhere to principles of good agricultural and industrial practice is underscored to mitigate the risk of mycotoxin contamination in food production. In the domain of food safety, the rapid and efficient detection of mycotoxins holds paramount significance. This paper delineates conventional and commercial methodologies for mycotoxin detection in ensuring food safety, encompassing techniques like liquid chromatography, immunoassays, and test strips, with a significant emphasis on the role of electrochemiluminescence (ECL) biosensors, which are known for their high sensitivity and specificity. These are categorized into antibody-, and aptamer-based, as well as molecular imprinting methods. This paper examines the latest advancements in biosensors for mycotoxin testing, with a particular focus on their amplification strategies and operating mechanisms.

Abstract:Despite efforts to control fungal contamination, some mycotoxins, including deoxynivalenol, zearalenone, aflatoxin, and ochratoxin, are ubiquitous in the rural China. Microarray Surface Enhanced Raman Scattering Based Immunosensor for Multiplexing Detection of Mycotoxin in Foodstuff

Abstract:A surface enhanced Raman scattering (SERS) based immunosensor was developed for the detection of three mycotoxins (aflatoxin B-1, AFB(1); zearalenone, ZEA; ochratoxin A, OTA) in foodstuff. Gold nanoparticles (GNPs) were labeled with 5,5-dithiobis(succinimidyl-2-nitrobenzoate) (DSNB) as Raman reporter and covalently linked with anti-mycotoxin antibodies as SERS nanoprobes, while AFB(1)-BSA, ZEA-BSA, and OTA-BSA conjugates were covalently linked onto micro-array gold surface as corresponding capture addresses. This design allows three independent immunoreactions multiplexed on a single gold chip. After optimization, the limits of detection of the developed assay are 0.061-0.066 mu g/kg for AFB(1), 0.53-0.57 mu g/kg for ZEA, and 0.26-0.29 mu g/kg for OTA in foodstuff. The spiked experiments presented an acceptable assay accuracy with recovery of 83.8%-108.1% and a reasonable assay precision with variation of coefficient less than 15%. Furthermore, the analysis of actual samples by our method demonstrated consistent results by comparison with conventional instrumental analysis. These results indicate that the developed SERS immunosensor can be a promising tool for simultaneously and rapidly monitoring multiple-mycotoxin levels in foodstuff. (C) 2018 Elsevier B.V. Nanobody Technology for Mycotoxin Detection in the Field of Food Safety: Current Status and Prospects

Abstract:Mycotoxins, which are toxic, carcinogenic, and/or teratogenic, have posed a threat to food safety and public health. Sensitive and effective determination technologies for mycotoxin surveillance are required. Immunoassays have been regarded as useful supplements to chromatographic techniques. However, conventional antibodies involved in immunoassays are difficult to be expressed recombinantly and are susceptible to harsh environments. Nanobodies (or VHH antibodies) are antigen-binding sites of the heavy-chain antibodies produced from Camelidae. They are found to be expressed easily in prokaryotic or eukaryotic expression systems, more robust in extreme conditions, and facile to be used as surrogates for artificial antigens. These properties make them the promising and environmentally friendly immunoreagents in the next generation of immunoassays. This review briefly describes the latest developments in the area of nanobodies used in mycotoxin detection. Moreover, by integrating the introduction of the principle of nanobodies production and the critical assessment of their performance, this paper also proposes the prospect of nanobodies in the field of food safety in the foreseeable future.
Simultaneous Determination of Multi-Component Mycotoxin Contaminants in Foods and Feeds by Ultra-Performance Liquid Chromatography Tandem Mass Spectrometry.

Abstract:The present study developed an improved analytical method for the simultaneous quantification of 17 kinds of Aspergillus, Fusarium and Penicillium mycotoxin contaminants in foods and feeds by ultra-performance liquid chromatography combined with electrospray ionization triple quadrupole tandem mass spectrometry (UPLC–MS/MS) under the multiple reaction monitoring (MRM) mode, and especially focused on the optimization of extraction, clean-up, UPLC separation and MS/MS parameters of analytes. Homogenized samples were sequentially extracted by 84% (v/v) of acetonitrile aqueous solution with the selected internal standard (zearalanone) spiking, SPE clean-up with Mycosep 226 Aflazon+ Multifunctional cartridges, filtration, concentration and secondary filtration. Using double sample injection method, the analytes were separated by UPLC BEH C18 column (100mm×2.1mm I.D., 1.7μm), and eluted with ammonium acetate/methanol and aqueous ammonia/methanol for the ESI+ and ESI− analysis, respectively. The 10 positive ions and 7 negative ions of mycotoxins were separated by gradient elution with the retention time of 6.5 and 4min, respectively. The LOQ of selected analytes ranged from 0.01 to 0.70μgkg−1, which was lower than the criteria of EU, USA and other countries on the determination of the minimum limiting level of various mycotoxins in foods including baby foods and feed stuffs. Meanwhile, high correlation coefficients (r2>0.99) of 17 mycotoxins were obtained within their respective linear ranges (0.05–20μgkg−1 for 10 positive ions and 0.5–50μgkg−1 for 7 negative ions) and reasonable recoveries (70.6–119.0%) of them were also demonstrated in different spiked levels. This quantitative method has many advantages including simple pretreatment, rapid determination and high sensitivity, which could be applied to the determination and quantification of multi-component mycotoxin contaminants in complex matrixes. Meanwhile, the method successfully fulfilled the minimum limiting level requests from various countries.

Abstract:The coexistence of multiple mycotoxins poses a significant risk to the safety of agricultural products. Hence, a novel and automated magnetic beads-based chemiluminescence immunoassay (MCLIA) system was developed for the simultaneous quantification of typical multi-mycotoxins, including zearalenone (ZEN), Ochratoxin A (OTA), deoxynivalenol (DON), and aflatoxin B 1 (AFB 1 ). It comprises a compact chemiluminescence-based analyzer and a specially designed mycotoxin assay kit. Firstly, antimycotoxin monoclonal antibody (mAb) was coupled with alkaline phosphatase (AP), and the antigen of mycotoxin was then with biotin. After detailed optimization of this indirect competition immunoassay, all necessary reagents were preloaded and sealed into a specialized plastic reagent strip to prepare the assay kit. Secondly, a compact chemiluminescence-based analyzer was developed by programmatically integrating magnetic, thermal, and optical mechanisms. The MCLIA system facilitates the detection of mycotoxins in up to 8 samples simultaneously or various mycotoxins in different samples within 20 min, according to actual needs. The detection limits (LOD) for ZEN, OTA, DON, and AFB 1 were 2.55, 0.64, 15.47, and 0.78 μg/kg, respectively, with the 50 % inhibition concentrations (IC 50 ) in the ranges of 1.84−4.25, 0.95−1.69, 11.36−13.98, and 0.37−0.54 ng/mL. Parallel analysis of commercial agricultural products demonstrated a strong linear correlation (R 2 = 0.9843–0.9962) with UPLC-HRMS, confirming the MCLIA system's reliability for the simultaneous quantitative detection of various mycotoxins. Offering advantages in detection sensitivity, dynamic range, and turnaround time, the compact MCLIA system facilitates automated and parallelized detection in the model of "sample-in to result-out". It presents a versatile platform for the efficient immunodetection of other contaminants.
