Abstract:Metal ions are widely distributed in nature and they have key roles in the environment, bioscience and medical science. Some metal ions,such as Hg2+,Fe3+,Cu2+,Zn2+and Al3+,play important roles in the physiology and pathology of organisms. An excess or deficiency of these metal ions would lead to physiological dysfunction and cause a variety of diseases. Recently,reaction-based fluorescence probes have attracted considerable attention due to their high sensitivity and good selectivity. This review mainly focuses on reaction-based fluorescence probes for the detection of metal ions in the past five years. The design and synthesis of probes,their sensing mechanisms and their applications were summarized. Currently,the reaction-based fluorescence probes are still in their initial stages and hence many problems need to be solved. With the resolution of these problems, reaction-based fluorescence probes would show higher sensitivities with less reaction-time and possess wider applications. In the future,reaction-based fluorescence probes should show efficient development in biological detection and expand practical applications.

Abstract:Metal ions have a significant impact on the environment and the growth and development of organism,therefore,the recognition and detection of metal ions in the environment and organism is gaining widespread attention. Among different analytical methods,fluorescence analysis has the advantages of high sensitivity,good selectivity,real-time and in situ detection,etc.,which is a good tool to achieve the recognition and detection of metal ions in the environment and organism. This paper is aimed to review the coumarin-based fluorescent sensors for the recognition and detection of heavy metal ions (Hg2+、Pb2+、Cd2+、Ni2+、Ag+),biologically important transition metal ions (Cu2+、Zn2+、Fe3+) and their applications in recent five years,focusing on the design and synthesis of sensors, recognition mechanism,sensing characteristics and applications. With the higher requirement for metal ions detection , the design of coumarin-based fluorescent sensors will be developed for better performance with higher sensitivity,selectivity and stronger anti-interference in the future. In addition, applications of coumarin-based fluorescent sensors in biological detection are expected to be further developed.

Abstract:One of the main challenges faced in food safety is the accumulation of toxic heavy metals from environmental sources, which can sequentially endanger human health when they are consumed. It is invaluable to establish a practical assay for the determination of heavy metals for food safety. Among the current detection methods, technology based on fluorescent probes, with the advantages of sensitivity, convenience, accuracy, cost, and reliability, has recently shown pluralistic applications in the food industry, which is significant to ensure food safety. Hence, this review systematically presents the recent progress on novel fluorescent probes in determining heavy metals for food safety over the past five years, according to fluorophores and newly emerging sensing cores, which could contribute to broadening the prospects of fluorescent materials and establishing more practical assays for heavy metal determinations.

Abstract:Mercury (Hg) is a heavy metal with high toxicity and easy migration; it can be enriched through the food chain, and cause serious threats to the natural environment and human health. So, the development of a method that can be used to detect mercury ions (Hg2+) in the environment, in cells, and in organisms is very important. Here, a new 7-hydroxycoumarin-derived carbonothioate-based probe (CC-Hg) was designed and synthesized for detection of Hg2+. After addition of Hg2+, a large fluorescence enhancement was observed due to the formation of 7-hydroxyl, which reinforced the intramolecular charge transfer process. The CC-Hg probe had good water solubility and selectivity. Moreover, the probe was able to detect Hg2+ quantitatively over the concentration range 0-2 mu M and with a detection limit of 7.9 nM. Importantly, we successfully applied the probe to detect Hg2+ in water samples, in living cells, and in zebrafish. The experimental results demonstrated its potential value in practical applications.

Abstract:All forms of life have absolute request for metal elements, because metal elements are instrumental in various fundamental processes. Fluorescent probes have been widely used due to their ease of operation, good selectivity, high spatial and temporal resolution, and high sensitivity. In this paper, the research progress of various metal ion (Fe 3+ ,Fe 2+ ,Cu 2+ ,Zn 2+ ,Hg 2+ ,Pb 2+ ,Cd 2+ ) fluorescent probes in recent years has been reviewed, and the fluorescence probes prepared with different structures and materials in different environments are introduced. It is of great significance to improve the sensing performance on metal ions. This research has a wide prospect in the application fields of fluorescence sensing, quantitative analysis, biomedicine and so on. This paper discusses about the development and applications of metal fluorescent probes in future.

Abstract:<p>Due to the advantages of <em>in situ,</em> real-time, and non-destructive detection of target molecules, fluorescent probes have become an indispensable tool in the fields of disease diagnosis and environmental and food safety monitoring. However, with the development of modern science and technology, sensing techniques have new challenges and requirements, such as increased sensitivity, better selectivity, and high-throughput visual detection in complicated practical samples. Developing effective fluorescent probes is still ongoing research. Fluorescent carbon dots (CDs) are a novel carbon-based nanomaterial that have attracted tremendous attention because of their excellent biocompatibility, tunable fluorescence properties, good photostability, and easy synthesis and surface modification, and they have shown great potential in fluorescent sensing and imaging applications. In this review, we summarize the recent advances in CD-based fluorescent probes, including the design strategies, sensing mechanisms, and their applications in sensitive and selective detection of public safety substances, organic amines, phenols, bioactive molecules, biological enzymes, and inorganic salts. A future perspective is introduced to highlight the existing challenges and possible strategies to overcome the problems in practical application of currently developed CD-based fluorescent probes, which may be instructive to the design of effective fluorescent probes in the future.</p>

Abstract:Cadmium (Cd) is carcinogenic to humans and can accumulate in the liver, kidneys, and bones. There is widespread presence of cadmium in the environment as a consequence of anthropogenic activities. It is important to detect cadmium in the environment to prevent further exposure to humans. Previous whole-cell biosensor designs were focused on single-sensing constructs but have had difficulty in distinguishing cadmium from other metal ions such as lead (Pb) and mercury (Hg). We developed a dual-sensing bacterial bioreporter system to detect bioavailable cadmium by employing CadC and CadR as separate metal sensory elements and eGFP and mCherry as fluorescent reporters in one genetic construct. The capability of this dual-sensing biosensor was proved to simultaneously detect bioavailable cadmium and its toxic effects using two sets of sensing systems while still maintaining similar specificity and sensitivity of respective signal-sensing biosensors. The productions of double-color fluorescence were directly proportional to the exposure concentration of cadmium, thereby serving as an effective quantitative biosensor to detect bioavailable cadmium. This novel dual-sensing biosensor was then validated to respond to Cd(II) spiked in environmental water samples. This is the first report of the development of a novel dual-sensing, whole-cell biosensor for simultaneous detection of bioavailable cadmium. The application of two biosensing modules provides versatile biosensing signals and improved performance that can make a significant impact on monitoring high concentration of bioavailable Cd(II) in environmental water to reduce human exposure to the harmful effects of cadmium.

Abstract:We report a novel ratiometric and reversible fluorescent probe for Cd(2+) detection utilizing a 6-(dimethylamino)quinaldine derivative as the fluorophore and a 2-hydrazinopyridine derivative as Cd(2+) chelator. This ratiometric fluorescent probe possesses favorable photophysical properties. It shows a large (55 nm) red-shift from 515 nm to 570 nm in the emission spectrum. Moreover, this probe also exhibits an excellent linear relationship of fluorescence intensity ratio (F570/F515) (R(2)=0.989) vs. Cd(2+) concentration in the range of 0-10 µM at physiological pH, which can serve as a "quantitative detecting" probe for Cd(2+). Utilizing this sensitive and selective probe, we have successfully detected Cd(2+) in living cells.

Abstract:Heavy metals, including Hg2+, Cr6+ and Cd2+, have always been a major issue in environmental pollution, leading to abnormal changes in the levels of biologically active molecules including Cys in plants, seriously affecting all aspects of the growth and development of plants. This makes it essential to develop a simple and practical method to study the potential impact of heavy metals on plants. In this paper, our research group has developed near-infrared fluorescent probe WRM-S, which has the advantages of fast response, sensitivity to Cys, and successfully applying it to cells and zebrafish. Moreover, it combined the close relationship between heavy metal stress on plants and Cys, using Cys as the detection target, monitoring the internal environment changes of two plants under Hg2+, Cr6+, and Cd2+ stress in the environment, and then conducting 3D imaging. The results indicated that the probe has strong penetration ability in plant tissues, and revealed abnormal changes in plant Cys levels caused by heavy metal stress-induced cellular oxidative stress or cytotoxicity. Thus, the in-situ imaging detection of this probe provides a direction for the physiological dynamics research of plant environmental stress.

Abstract:A fluorescent sensor for cadmium (CS) based on the BODIPY fluorophore exploiting the PET (Photoinduced Electron Transfer) mechanism was prepared. CS exhibited high selectivity and sensitivity for detecting cadmium in aqueous buffer solution. In addition, the complex of CS with cadmium could detect pyrophosphate (PPI) selectively and sensitively.

Abstract:In recent decades, heavy metal ions have emerged as a significant global environmental concern, posing threats to the delicate balance of ecosystems worldwide. Their introduction into ecosystems occurs through various activities and poses a serious risk to human health. Among heavy metal ions, Cd 2+ is recognized as a highly toxic pollutant. Its widespread use contributes to its accumulation in the environment. Chronic exposure to Cd 2+ ions present serious risks to both the environment and human health. Therefore, the detection of these metal ions are very important. Organic fluorometric and colorimetric detection have emerged as promising tools for this purpose, offering advantages such as high sensitivity, selectivity, and sometimes reversibility. This review offers a comprehensive overview of the recent advancements in the fluorometric and colorimetric detection of Cd 2+ using organic chemosensors from 2019 to 2024. We delve into key aspects of these studies, including the design strategies employed to design novel chemosensors and the underlying sensing mechanisms. Furthermore, we explore the diverse applications of these organic chemosensors, ranging from environmental monitoring to biomedical diagnostics. By analyzing the latest research findings, this review aims to offer insights into the current state-of-the-art in the field of Cd 2+ detection using organic chemosensors. Abstract:There has been rapid progress in the development of fast, sensitive, cheap and low-cytotoxicity micro-molecule fluorescent probes for application in various fields, including disease diagnosis, food safety and environmental safety. As an analytical tool, dual-function fluorescent probes with dual-emission responses have attracted considerable attention due to their cost-effectiveness and efficiency over single-function sensors. This review primarily describes research progress on multifunctional probes in terms of the reaction type and coordination type, as well as the general design principles of probes. The analytes include reactive oxygen species (ROS), reactive sulfur species (RSS), harmful cations and anions, etc. Multifunctional probes for food, medical and environmental applications are listed for future research. To improve the development of rapid detection methods, trends and strategies in the development of multifunctional fluorescent probes are also discussed.

The Science of The Total Environment

Abstract:Many heavy metals pose significant threats to the environment and human health. Traditional methods for detecting heavy metals are often limited by complex procedures, high costs, and challenges in field monitoring. Carbon quantum dots (CQDs), a novel class of fluorescent carbon nanomaterials, have garnered significant interest due to their excellent biocompatibility, low cost, and minimal toxicity. This paper reviews the primary synthesis methods, luminescence mechanisms, and fluorescence quenching mechanisms of CQDs, as well as their recent applications in detecting heavy metals. In heavy metal sensing applications, the simplest hydrothermal method is commonly employed for the one-step synthesis and surface modification of CQDs. Various green reagents and biomass materials, such as citric acid, glutathione, orange peel, and bagasse, can be used for CQDs' preparation. Quantum confinement effects and surface defects give CQDs their distinctive luminescence properties, enabling the detection of heavy metals through fluorescence quenching or enhancement. Additionally, CQDs can be applied in biological imaging and smart detection, and when combined with adsorption materials, they can offer multifunctional capabilities. This review also discusses the future development prospects of CQDs.

Abstract:Carbon quantum dots (CQDs) are ideal fluorescent probes for rapid detection. This paper reviews the synthesis methods of CQDs, their application in the rapid detection of antibiotics and heavy metals in the environment and food, and the underlying detection mechanisms. The hydrothermal method is the most commonly used for synthesis, and CQDs doped with heteroatoms (such as N, P and S) exhibit superior fluorescence performance. In the presence of antibiotics and heavy metals, the fluorescence of CQDs can be quenched or enhanced. Single-signal and dual-signal probes can be developed using the fluorescence, phosphorescence and absorbance of CQDs, enabling rapid detection of various antibiotics (e.g., tetracycline, quinolone and beta-lactam antibiotics) and heavy metals (e.g., Cd2+, Cr6+, Fe3+, Hg2+, and Pb2+). With the combination of smartphones and fluorescent probe test strips developed based on CQDs, on-the-spot rapid detection can be realized. This review offers new insights into the rapid detection of CQDs.

Abstract:Detecting environmental contaminants is crucial for protecting ecosystems and human health. While traditional carbon dot (CD) fluorescent probes are versatile, they may suffer from limitations like fluctuations in signal intensity, leading to detection inaccuracies. In contrast, ratiometric fluorescent probes, designed with internal self-calibration mechanisms, offer enhanced sensitivity and reliability. This review focuses on the design and applications of ratiometric fluorescent probes based on CDs for environmental monitoring. Our discussion covers construction strategies, ratiometric fluorescence principles, and applications in detecting various environmental contaminants, including organic pollutants, heavy metal ions, and other substances. We also explore associated advantages and challenges and provide insights into potential solutions and future research directions.

Abstract:Arsenic is a toxic non-metallic element that is widely found in nature. In addition, arsenic and arsenic compounds are included in the list of Group I carcinogens and toxic water pollutants. Therefore, rapid and efficient methods for detecting arsenic are necessary. In the past decade, a variety of small molecule fluorescent probes have been developed, which has been widely recognized for their rapidness, efficiency, convenience and sensitivity. With the development of new nanomaterials (AuNPs, CDs and QDs), organic molecules and biomolecules, the conventional detection of arsenic species based on fluorescence spectroscopy is gradually transforming from the laboratory to the portable kit. Therefore, in view of the current research status, this review introduces the research progress of both traditional and newly developed fluorescence spectrometry based on novel materials for arsenic detection, and discusses the potential of this technology in the rapid screening and field testing of water samples contaminated with arsenic. The review also discusses the problems that still exist in this field, as well as the expectations.
