Abstract:We developed a low-cost multi-core inertial microfluidic centrifuge (IM-centrifuge) to achieve a continuous-flow cell/particle concentration at a throughput of up to 20 mL/min. To lower the cost of our IM-centrifuge, we clamped a disposable multilayer film-based inertial microfluidic (MFIM) chip with two reusable plastic housings. The key MFIM chip was fabricated in low-cost materials by stacking different polymer-film channel layers and double-sided tape. To increase processing throughput, multiplexing spiral inertial microfluidic channels were integrated within an all-in-one MFIM chip, and a novel sample distribution strategy was employed to equally distribute the sample into each channel layer. Then, we characterized the focusing performance in the MFIM chip over a wide flow-rate range. The experimental results showed that our IM-centrifuge was able to focus various-sized particles/cells to achieve volume reduction. The sample distribution strategy also effectively ensured identical focusing and concentration performances in different cores. Finally, our IM-centrifuge was successfully applied to concentrate microalgae cells with irregular shapes and highly polydisperse sizes. Thus, our IM-centrifuge holds the potential to be employed as a low-cost, high-throughput centrifuge for disposable use in low-resource settings.

Abstract:In recent years, point-of-care testing has played an important role in immunoassay, biochemical analysis, and molecular diagnosis, especially in low-resource settings. Among various point-of-care-testing platforms, microfluidic chips have many outstanding advantages. Microfluidic chip applies the technology of miniaturizing conventional laboratory which enables the whole biochemical process including reagent loading, reaction, separation, and detection on the microchip. As a result, microfluidic platform has become a hotspot of research in the fields of food safety, health care, and environmental monitoring in the past few decades. Here, the state-of-the-art application of microfluidics in immunoassay in the past decade will be reviewed. According to different driving forces of fluid, microfluidic platform is divided into two parts: passive manipulation and active manipulation. In passive manipulation, we focus on the capillary-driven microfluidics, while in active manipulation, we introduce pressure microfluidics, centrifugal microfluidics, electric microfluidics, optofluidics, magnetic microfluidics, and digital microfluidics. Additionally, within the introduction of each platform, innovation of the methods used and their corresponding performance improvement will be discussed. Ultimately, the shortcomings of different platforms and approaches for improvement will be proposed.

Abstract:A lab-on-a-CD immunoassay system was designed, fabricated, and tested. The actuator for the centrifugal platform consists of a flyball governor and a group of spring plungers. The microfluidic structures were fabricated using replica processes with poly-dimethylsiloxane (PDMS) as structural material. For better control, the valves in the microfluidic system were designed to be mechanical-pinch ones and actuated. The burst frequencies of the micro-valves can be tuned independently by adjusting the heights and prestresses of corresponding spring plungers. For the research work reported in this paper, four sequential valves and one inward pump were integrated on a centrifugal microfluidic system to develop an automated immunoassay system. Mouse IgG was used as a model analyte to test the feasibility of using the proposed device for immunoassay applications. Sample solution of goat anti-mouse IgG conjugated with magnetic nanoparticles was used for magnetic trapping mechanism to eliminate the blocking process. Rabbit anti-mouse IgG labeled with fluorescent molecules was used as the signal reporter. The entire microfluidic assay process, including beads trapping, immunoassay reaction, washing and pumping was completed in a centrifugal platform. The detection sensitivity may also be improved by using higher quality monoclonal antibodies. This new immunoassay platform offers some significant advantages over those reported in the field: simple fabrication, robust control, easy integration, and lower cost. The system may also be modified for detection of multiple targets. The technology has great potential for applications in complex fluid handling needed in biological, chemical and medical application in Lab-on-a-CD systems. (C) 2017 Elsevier B.V. Abstract:Modular integration of functional components on the chip and increasement in control accuracy through real-time alteration in the force direction of droplets is an effective way to optimize centrifugal microfluidic systems and realize passive components, compact modules, and high-throughput control. Conventional centrifugal microfluidic chips are mainly driven and controlled by centrifugal force and Euler force. The control valves are easily affected by machining precision, making the control unstable. In this study, a novel centrifugal microfluidic system is introduced to improve the freedom and accuracy of chip control while facilitating the design and addition of passive functional components. Furthermore, we modularize the centrifugal microfluidic chip to greatly shorten the period of design and optimization cycle and achieve chip reusability and multi-threaded control. Finally, to verify the feasibility of the modular centrifugal microfluidic chip applied to high-throughput nucleic acid screening, we test the nucleic acid purification and detection colorimetric reactions based on the modular centrifugal microfluidic chip. Among them, Chelex-100 is used to realize the purification of nucleic acid in cell lysate, and the purified solution can realize amplification in the PCR instrument, and the nucleic acid detection results are consistent with the off-chip kit by experimental testing. The system has great flexibility and stability under the acceptable purity of nucleic acid, which indicates that the platform has great potential for large-scale rapid screening applications.

Abstract:Centrifugal microfluidic or lab-on-a-disc platforms have many advantages over other microfluidic systems. These advantages include a minimal amount of instrumentation, the efficient removal of any disturbing bubbles or residual volumes, and inherently available density-based sample transportation and separation. Centrifugal microfluidic devices applied to biomedical analysis and point-of-care diagnostics have been extensively promoted recently. This paper presents an up-to-date overview of these devices. The development of biomedical centrifugal microfluidic platforms essentially covers two categories: (i) unit operations that perform specific functionalities, and (ii) systems that aim to address certain biomedical applications. With the aim to provide a comprehensive representation of current development in this field, this review summarizes progress in both categories. The advanced unit operations implemented for biological processing include mixing, valving, switching, metering and sequential loading. Depending on the type of sample to be used in the system, biomedical applications are classified into four groups: nucleic acid analysis, blood analysis, immunoassays, and other biomedical applications. Our overview of advanced unit operations also includes the basic concepts and mechanisms involved in centrifugal microfluidics, while on the other hand an outline on reported applications clarifies how an assembly of unit operations enables efficient implementation of various types of complex assays. Lastly, challenges and potential for future development of biomedical centrifugal microfluidic devices are discussed.

Abstract:Yoon-Kyoung Cho is a full professor in Biomedical Engineering at UNIST. In this contribution she describes a fully integrated and automated lab-on-a-disc using centrifugal microfluidics to provide a "sample-in and answer-out" type of biochemical analysis solution with simple, size-reduced, and cost-efficient instrumentation. She also gives various examples of the fully integrated "lab-on-a-disc" developed for broad applications ranging from medical diagnostics to food, environment, and energy applications.

Abstract:Centrifugal microfluidics has evolved into a mature technology. Several major diagnostic companies either have products on the market or are currently evaluating centrifugal microfluidics for product development. The fields of application are widespread and include clinical chemistry, immunodiagnostics and protein analysis, cell handling, molecular diagnostics, as well as food, water, and soil analysis. Nevertheless, new fluidic functions and applications that expand the possibilities of centrifugal microfluidics are being introduced at a high pace. In this review, we first present an up-to-date comprehensive overview of centrifugal microfluidic unit operations. Then, we introduce the term "process chain" to review how these unit operations can be combined for the automation of laboratory workflows. Such aggregation of basic functionalities enables efficient fluidic design at a higher level of integration. Furthermore, we analyze how novel, ground-breaking unit operations may foster the integration of more complex applications. Among these are the storage of pneumatic energy to realize complex switching sequences or to pump liquids radially inward, as well as the complete pre-storage and release of reagents. In this context, centrifugal microfluidics provides major advantages over other microfluidic actuation principles: the pulse-free inertial liquid propulsion provided by centrifugal microfluidics allows for closed fluidic systems that are free of any interfaces to external pumps. Processed volumes are easily scalable from nanoliters to milliliters. Volume forces can be adjusted by rotation and thus, even for very small volumes, surface forces may easily be overcome in the centrifugal gravity field which enables the efficient separation of nanoliter volumes from channels, chambers or sensor matrixes as well as the removal of any disturbing bubbles. In summary, centrifugal microfluidics takes advantage of a comprehensive set of fluidic unit operations such as liquid transport, metering, mixing and valving. The available unit operations cover the entire range of automated liquid handling requirements and enable efficient miniaturization, parallelization, and integration of assays.

Abstract:Public health events caused by pathogens have imposed significant economic and societal burdens. However, conventional methods still face challenges including complex operations, the need for trained operators, and sophisticated instruments. Here, we proposed a fully integrated and automated centrifugal microfluidic chip, also termed IACMC, for point-of-care multiplexed molecular diagnostics by harnessing the advantages of active and passive valves. The IACMC incorporates multiple essential components including a pneumatic balance module for sequential release of multiple reagents, a pneumatic centrifugation-assisted module for on-demand solution release, an on-chip silicon membrane module for nucleic acid extraction, a Coriolis force-mediated fluid switching module, and an amplification module. Numerical simulation and visual validation were employed to iterate and optimize the chip's structure. Upon sample loading, the chip automatically executes the entire process of bacterial sample lysis, nucleic acid capture, elution quantification, and isothermal LAMP amplification. By optimizing crucial parameters including centrifugation speed, direction of rotation, and silicone membrane thickness, the chip achieves exceptional sensitivity (twenty-five Salmonella or forty Escherichia coli) and specificity in detecting Escherichia coli and Salmonella within 40 min. The development of IACMC will drive advancements in centrifugal microfluidics for point-of-care testing and holds potential for broader applications in precision medicine including high-throughput biochemical analysis immune diagnostics, and drug susceptibility testing.

Abstract:Effective enrichment and separation of particles and mixing of multiphase microflows can be technically challenging in microfluidic sample processing and analysis. Acoustofluidic techniques have advantages such as high biocompatibility, versatility, and simple device design to meet these demands. Here, we present a novel tri-symmetrical acoustofluidic centrifuge based on traveling surface acoustic waves (TSAWs), achieving particle enrichment and bidirectional size-based separation, pushing away the larger particles while enriching the smaller ones, in a rapid time (<6 s). A microfluidic chamber was designed for further extraction of treated particles and cells. Experimental results show that our device can achieve enrichment of particles in a range of sizes and high purity separation (e.g. >90 % in 5 µm and 40 µm sample), demonstrating its sample processing capabilities. The acoustic treatment did not affect cells (consistent with the control group above 80 %). TSAW-induced acoustic streaming enabled efficient mixing (mixing efficiency χ >85 %) of fluorescent multi-phase microflows within a short time (<0.5 s). The underlying physical mechanisms have been verified by numerical simulations.
Microfluidic chip electrophoresis for biochemical analysis

Abstract:Microfluidic chip electrophoresis has been widely employed for separation of various biochemical species owing to its advantages of low sample consumption, low cost, fast analysis, high throughput, and integration capability. In this article, we reviewed the development of four different modes of microfluidics-based electrophoresis technologies including capillary electrophoresis, gel electrophoresis, dielectrophoresis, and field (electric) flow fractionation. Coupling detection schemes on microfluidic electrophoresis platform were also reviewed such as optical, electrochemical, and mass spectrometry method. We further discussed the innovative applications of microfluidic electrophoresis for biomacromolecules (nucleic acids and proteins), biochemical small molecules (amino acids, metabolites, ions, etc.), and bioparticles (cells and pathogens) analysis. The future direction of microfluidic chip electrophoresis was predicted.

Abstract:Microfluidic devices especially centrifugal ones have attracted great attention in the nucleic acid testing field, due to their automation, high efficiency, and simple operation. In which, nucleic acid extraction is the basic step, laying a foundation for the downstream amplification and detection procedures. Therefore, the integration of nucleic acid extraction on the chip is expected to achieve cost-efficiency, high-speed automation, diagnostic accuracy, and reaction robustness with the respect to real-time detection. In this work, we employ chitosan-modified magnetic microspheres for pH-induced nucleic acid extraction and integrate this approach into a centrifugal microfluidic chip. The microfluidic system comprises cell lysis, nucleic acids capture and release, isothermal amplification, and real-time fluorescence detection, which is manipulated by centrifugal force and magnetic control. The system exhibits comparable extraction and detection performance in respect of acceptable nucleic acid concentration and purity, high detection specificity and stability, as well as fast detection duration. These efforts to improve the integrated microfluidic detection chip could benefit the portable, efficient and simple nucleic acid diagnosis, especially under the resource-limited circumstance.

Abstract:Microfluidics has been considered as a potential technology to miniaturize the conventional equipments and technologies. It offers advantages in terms of small volume, low cost, short reaction time and highthroughput. The applications in biology and medicine research and related areas are almost the most extensive and profound. With the appropriate scale that matches the scales of cells, microfluidics is well positioned to contribute significantly to cell biology. Cell culture, fusion and apoptosis were successfully performed in microfluidics. Microfluidics provides unique opportunities for rare circulating tumor cells isolation and detection from the blood of patients, which furthers the discovery of cancer stem cell biomarkers and expands the understanding of the biology of metastasis. Nucleic acid amplification in microfluidics has extended to single-molecule, high-throughput and integration treatment in one chip. DNA computer which is based on the computational model of DNA biochemical reaction will come into practice from concept in the future. In addition, microfluidics offers a versatile platform for protein-protein interactions, protein crystallization and high-throughput screening. Although microfluidics is still in its infancy, its great potential has already been demonstrated and will provide novel solutions to the high-throughput applications.

Abstract:Efficient platforms for cell isolation and analysis play an important role in applied and fundamental biomedical studies. As cells commonly have a size of around 10 microns, conventional handling approaches at a large scale are still challenged in precise control and efficient recognition of cells for further performance of isolation and analysis. Microfluidic technologies have become more prominent in highly efficient cell isolation for circulating tumor cells (CTCs) detection, single-cell analysis and stem cell separation, since microfabricated devices allow for the spatial and temporal control of complex biochemistries and geometries by matching cell morphology and hydrodynamic traps in a fluidic network, as well as enabling specific recognition with functional biomolecules in the microchannels. In addition, the fabrication of nano-interfaces in the microchannels has been increasingly emerging as a very powerful strategy for enhancing the capability of cell capture by improving cell-interface interactions. In this review, we focus on highlighting recent advances in microfluidic technologies for cell isolation and analysis. We also describe the general biomedical applications of microfluidic cell isolation and analysis, and finally make a prospective for future studies.

Detection is an important part in microfluidics-based immunoassays. Recent advances in the studies of micro-systems and detection methods, including electrochemical detection., fluorescence spectrometry, ultraviolet-visible spectrometry, chemiluminescence and bioluminescence, surface-enhanced Raman scattering, fiber-optic sensor, surface plasmon resonance, thermal lens microscopy, colorimetry and other new detection methods applied in microfluidics-based immunoassays, were summarized and reviewed in this paper. The prospects of the detection methods for microfluidics-based immunoassays were presented.

Abstract:Automatic mixing of bio-samples using microchannel and centrifugation is considered. Existing methods for mixing bio-samples for life science applications use micro-well and micro-stirrer which are ineffective when used with highly viscous materials at the microliter or nanoliter level. Our method mixes viscous bio-samples in micro-capsules using micro-channel and centrifugation which minimizes contact with mixing tools. To introduce the method, firstly the design of the micro-capsule along with the micro-channel is presented. Secondly, a hydrodynamic model describing the flow of viscous materials in the micro-channel is presented, and the average sample velocity and the traveling time through the microchannel are analyzed. Thirdly, the relationship between centrifugation speed and time is given to achieve effective and efficient control of the flow. Finally, experimental and theoretical results are compared.

Abstract:Currently, centrifuge apparatus is primarily an end-point sample processing piece of equipment. The lack of real-time active control has imposed an inherent limitation such that many delicate sample processing steps requiring immediate and accurate intervention have never been possible. We report herein a motor-assisted chip-in-a-tube (MACT) platform in which a microfluidic chip placed inside a common centrifuge canister can be rotated through wireless control in order to manipulate the centrifugal force vector in a 3-dimensional (3D) manner. As a demonstration experiment, we have used our MACT prototype to perform the operation for two common biomedical procedures, namely human blood plasma separation and E. coli plasmid DNA extraction. This simple, yet highly effective and versatile approach may serve as a generic one-for-all platform for a wide range of common laboratory experiments and bioassay applications.

Abstract:In this paper, we present a lab-in-a-droplet bioassay strategy for a centrifugal microfluidics or lab-on-a-disc (LOAD) platform with three important advancements including density difference pumping, power to disc and bidirectional flow control. First, with the water based bioassay droplets trapped in a micro-channel filled with mineral oil, centrifugal force due to the density difference between the water and oil phases actuates droplet movement while the oil based medium remains stationary. Second, electricity is coupled to the rotating disc through a split-core transformer, thus enabling on-chip real-time heating in selected areas as desired and wireless programmable functionality. Third, an inertial mechanical structure is proposed to achieve bidirectional flow control within the spinning disc. The droplets can move back and forth between two heaters upon changing the rotational speed. Our platform is an essential and versatile solution for bioassays such as those involving DNA amplification, where localized temperature cycling is required. Finally, without the loss of generality, we demonstrate the functionality of our platform by performing real-time polymerase chain reaction (RT-PCR) in a linear microchannel made with PTFE (Teflon) micro-tubing.

Currently, most traditional analytical devices used for medical are expensive, bulky and require professional operations, which largely limits their use in POCT. However, the microfluidic chip, which integrates the whole biochemical process including reagent loading, reaction, separation and detection, can effectively solve this problem. Therefore, microfluidic chip has been widely studied in the fields of food, environment and medical monitoring. In this paper, the recent advances in immunoassay, biochemical analysis and molecular diagnosis of microfluidic platform are reviewed. In the immunoassay, the latest applications of different types of microfluidic platforms including capillary, gravity, centrifugal, magnetic, digital and optical microfluidic systems were summarized. In biochemical analysis, the research on plasma separation, biological particle extraction and quantitative detection of biochemical substances in the centrifugal microfluidic system was emphasized. In the aspect of molecular diagnosis, the research progress of polymerase chain reaction (PCR), loop mediated isothermal amplification (LAMP) and recombinant polymerase amplification (RPA) were reviewed.
