Abstract:The cultivation and instability of aerobic granular sludge in a sequencing batch reactor under high loading conditions were investigated. Compact bacteria-dominated aerobic granules with a mean diameter of 1 mm were formed at a chemical oxygen demand (COD) loading rate of 6.0 kg m(-3) d(-1) within 30 d. However, the compact bacteria-dominated aerobic granules were not stable and transited to large-sized filamentous ones gradually. With the formation of bacteria-dominated granules, the hydrophobicity and specific gravity of the sludge increased. When the granules were transited to filamentous ones, the hydrophobicity and specific gravity decreased. Both granules had a high COD removal efficiency, excellent settling ability and showed a clear, regular round-shaped outline. After the filamentous granules reached a diameter of 16 mm, due to the mass transfer limitation and the possible presence of anaerobes in the inner part of the granules, they began to disintegrate and be washed out of the reactor, follow by failure of the reactor.

Abstract:One current major hurdle to practical implementation of aerobic granule technology is the frequent occurrence of granule disintegration during long-term operation. However, the mechanism behind this is largely unclear today. Here, 2-decenoic acid, which has been previously demonstrated to be released by Pseudomonas aeruginosa and disperse biofilms, was found to also induce the disintegration of aerobic granules. A comparison of the solution compositions from samples of only trans-2-decenoic acid, only aerobic granules, and granules added with trans-2-decenoic acid shows that bacteria and extracellular polymeric substances (EPS) were stripped from granule surface upon trans-2-decenoic acid dosing. Due to the possible toxicity of trans-2-decenoic acid at a saturation concentration, the disintegrated granules and the milky suspension in the disintegration test showed a significantly lower oxygen uptake rate than the un-integrated granules. This work suggests that trans-2-decenoic acid released by microbes might play a critical role in regulating the disintegration of aerobic granules.
Fluctuation and Re-Establishment of Aerobic Granules Properties during the Long-Term Operation Period with Low-Strength and Low C/N Ratio Wastewater

Abstract:Long-term structure stability of aerobic granules is critical to maintaining stable wastewater treatment performance. In this study, granulation and long-term stability of sludge-treating synthetic wastewater with a low chemical oxygen demand to nitrogen (COD/N) ratio of 4:1 and COD concentration of 400 mg/L in anoxic-oxic conditions were investigated for over 300 days. Inoculated suspended sludge gradually transformed into granules-dominant sludge on day 80. Due to the improved sludge volume index after 30 min settling (SVI30), mixed liquor suspended solids (MLSS) reached 5.2 g/L on day 140. Without any external intervention or disturbance, aerobic granules started to disintegrate from day 140, causing the increase in SVI and the decrease in biomass concentration until day 210, with the average sludge size reduced to 243 μm. From day 210, granular sludge started to be re-established by re-granulation, and the average granule size increased to 500 μm on day 302. During these disintegration and re-granulation periods, there was no obvious difference in terms of COD removal and nitrification, but microbial species were found more diverse after the re-granulation, with Thauera and Sphingomonas dominant. Although there was no external intervention, the food to microorganisms ratio (F/M) varied significantly due to the changes in biomass concentration caused by strong selective pressure and the change of sludge-settling ability in the reactor. F/M ratios should be controlled between 0.3 and 1.0 gCOD/gSS·d to maintain the stable structure of granules to minimize the fluctuation of sludge properties under the conditions used in this study. Although aerobic granular sludge is able to re-establish itself after disintegration, controlling F/M ratios in a certain range would benefit long-term stability. The findings in this study are significant to deepen the understanding of granule stability with low-strength and low COD ratio wastewater and, thus, provide guidance for maintaining the long-term stability of granules.

Long-term storage of aerobic granules in liquid media: viable but non-culturable status

Abstract:Long-term storage and successful reactivation after storage are essential for practical applications of aerobic granules on wastewater treatment. This study cultivated aerobic granules (SI) in sequencing batch reactors and then stored the granules at 4 °C in five liquid media (DI water (SW), acetone (SA), acetone/isoamyl acetate mix (SAA), saline water (SS), and formaldehyde (SF)) for over 1 year. The first four granules were then successfully reactivated in 24h cultivation. The specific oxygen uptake rates (SOUR) of the granules followed SI>SS>SA>SAA>SW>SF; and the corresponding granular strengths (10 min ultrasound) followed SI>SA=SS>SAA>SW>>SF. During storage the granular cells secreted excess quantities of cyclic-diguanylate (c-di-GMP) and pentaphosphate (ppGpp) as responses to the stringent challenges. We proposed that to force cells in granules (Alphaproteobacteria, Flavobacteria, Betaproteobacteria, Gammaproteobacteria, Actinobacteria, Sphingobacteria, and Clostridia) entering viable but non-culturable (VBNC) status is the key of success for extended period storage of granules.
Reproducibility of Aerobic Granules in Treating Low-Strength and Low-C/N-Ratio Wastewater and Associated Microbial Community Structure

Abstract:Long-term stability of the aerobic granular sludge system is essentially based on the microbial community structure of the biomass. In this study, the physicochemical and microbial characteristics of sludge and wastewater treatment performance were investigated regarding formation, maturation, and long-term maintenance of granules in two parallel sequencing batch reactors (SBR), R1 and R2, under identical conditions. The aim was to explore the linkage between microbial community structure of the aerobic granules, their long-term stability, as well as the reproducibility of granulation and long-term stability. The two reactors were operated with a COD concentration of 400 mg/L and a chemical oxygen demand to nitrogen (COD/N) ratio of 4:1 under anoxic–oxic conditions. It was found that although SVI30, sludge size, and distributions in R1 and R2 were different, aerobic granules were formed, and they maintained long-term stability in both reactors for 320 days, implying that a certain level of randomness of granulation does not affect the long-term stability and performance for COD and N removal. In addition, a significant reduction in the richness and diversity of microbial production was observed after the sludge was converted from inoculum or flocs to granules, but this did not negatively affect the performance of wastewater treatment. Among the predominant microbial species in aerobic granules, Zoogloea was identified as the most important bacteria present during the whole operation with the highest abundance, while Thauera was the important genus in the formation and maturation of the aerobic granules, but it cannot be maintained long-term due to the low food-to-microorganisms ratio (F/M) in the system. In addition, some species from Ohtaekwangia, Chryseobacterium, Taibaiella, and Tahibacter were found to proliferate strongly during long-term maintenance of aerobic granules. They may play an important role in the long-term stability of aerobic granules. These results demonstrate the reproducibility of granulation, the small influence of granulation on long-term stability, and the robustness of aerobic granulation for the removal of COD and N. Overall, our study contributes significantly to the understanding of microbial community structure for the long-term stability of aerobic granular sludge in the treatment of low-COD and low-COD/N-ratio wastewater in practice.

Abstract:Aerobic granulation was successfully achieved in a pilot scale sequencing batch reactor within 40 days. Then, stability of different particle size granules was explored according to their activity and resistance to ultrasonic crushing. Different particle size granules (0.3-0.6 mm, 0.6-1 mm, 1-1.43 mm, 1.43-2 mm, 2-3 mm and 3-4 mm) were exposed under different ultrasonic power separately. It was found that the granules with 2-3 mm always had the maximum granulation rates after ultrasonic crushing. Meanwhile, activity data showed that the 2-3 mm granules had the lowest specific oxygen utilization rates, which indicated that they were easier to maintain stability as the increase of their particle sizes was the slowest. So, 500 mL mixed liquid of the reactor were taken out and sieved to obtain the 2-3 mm granules, which were subsequently returned to the reactor to increase their proportion. Through the manual regulation, the proportion of 2-3 mm granules kept increasing which gradually became dominant in the reactor. Under the strategy of 86 days of operation, the aerobic granules were regular and compact, which had good removal effects of the real wastewater. The results indicated that the stability of the system could be greatly enhanced by the method, which provided a new strategy to maintain the granular stability.

Influence of Operational Conditions on the Stability of Aerobic Granules from the Perspective of Quorum Sensing

Abstract:Integrated aerobic granules were first cultivated in two sequencing batch reactors (SBRs) (A1 and A2). Then, A1’s influent organic loading rate (OLR) was changed from alternating to constant (cycling time was still 6 h), while A2’s cycling time varied from 6 to 4 h (influent OLR strategy remained alternating). After 30-day operation since the manipulative alternations, granule breakage happened in two reactors at different operational stages, along with the decrease of granule intensity. Granule diameter in A1 declined from the original 0.84 to 0.32 cm during the whole operation, while granules in A2 dwindled to 0.31 cm on day 22 with similar size to A1. Both the amount of total extracellular polymeric substances (EPSs) and the protein were declining throughout the operation, and the large molecular weight of protein was considered closely related to the stability of aerobic granules. The relative AI-2 level decreased at the same time, and influent OLR strategy might had more evident impact on quorum sensing (QS) ability of sludge compared with starvation period. Combined with microbial results, the decline of total EPS amount in two reactors could be concluded as follows: During the reactor operation, some functional bacteria gradually lost their dominance and were eliminated from the reactors, which finally caused granule disintegration. In summary, the results further confirmed that alternating OLR and proper starvation period were two major factors in effective cultivation and stability of aerobic granules from the perspective of QS.

Abstract:Loss of structural stability of aerobic granular process is the challenge for its field applications to treat wastewaters. The second messenger, cyclic diguanylate (c-di-GMP), is widely used by bacteria to regulate the synthesis of exopolysaccharide. This study for the first time confirmed the correlation between concentration of intracellular c-di-GMP and the granular stability under sequencing batch reactor (MBR) mode. In the presence of manganese ions (Mn(2+)), the concentrations of intracellular c-di-GMP and of extracellular polysaccharides and proteins in granules were declined. Clone library study revealed that the polysaccharide producers. Acinetobacter sp., Thauera sp., Bdellovibrio sp. and Paracoccus sp. were lost after Mn(2+) addition. The findings reported herein confirmed that the c-di-GMP is a key chemical factor epistatic to quorum sensing to determine granular stability. Stimulation of synthesis of intracellular c-di-GMP presents a potential way to enhance long-term stability of aerobic granules.
Disintegration and re-granulation of aged algal-bacterial aerobic granular sludge in wastewater treatment

Abstract:Long-term stability in practical applications is one of the crucial issues challenging the sustainable operation of aerobic granular sludge (AGS)-based wastewater treatment systems. Numerous attempts have been made to improve its long-term stability, among which introducing microalgae to bacterial AGS systems seems promising. This study investigated and recorded the whole process of disintegration and re-granulation of the aged algal-bacterial AGS in a 42-day operation. The disintegration of granules was signaled by broken structure and more effluent biomass discharge, resulting in deterioration of phosphorus removal from 86 % on day 1 to 7 % on day 20 and some slight decrease in chlorophyll a content. The broken granules were found to be filamentous flocs or granule fragments, and the latter with coexisting microalgae possibly accounted for the re-granulation and system recovery. Large proportions of fragments could be developed into new granules on day 30 under no additional strategy adoption. And the effluent biomass, chlorophyll a content and phosphorus removal can recover to their initial levels within 34 days' operation. Interestingly, organics and ammonia removals were not affected, which kept around 99 % during the whole test period. Results from this study provide a more in-depth understanding of the stability of algal-bacterial AGS and its long-term operation management.

Abstract:Abstract:This study elucidates the impact of fluctuating influent conditions and in situ ultrasonic wave exposure on the stability of aerobic granular sludge (AGS) in the treatment of simulated wastewater emanating from rare earth mining operations. During a stable influent period spanning from Day 1 to Day 95, the seed granules underwent an initial disintegration followed by a re-granulation phase. The secondary granulation was achieved on Day 80 and Day 40 for the ultrasonic reactor (R1) and the control reactor (R2), respectively. Notably, granules formed in R1 exhibited a more porous structure compared to those generated in R2. Subsequently, when the ammonia nitrogen in the influent oscillated between 100 and 500 mg/L during Days 96-140, both reactors yielded compact and densely structured granules. Nitrogen removal profiles were comparable between the two reactors: the removal efficiencies for ammonia nitrogen and total inorganic nitrogen escalated from 95% and 80%, respectively, during Days 1-95, to 95% and 90%, respectively, post-Day 140. A suite of performance metrics indicated that steady-state granules from R1 outperformed those from R2 across several parameters. Specifically, the nitrification/denitrification rates, and relative abundance of denitrifying bacteria were all higher in granules from R1. Conversely, the relative abundance of nitrifying bacteria was comparable between granules from both reactors. However, R1 granules demonstrated lower sludge concentration and smaller average particle size than their R2 counterparts. In conclusion, the AGS system demonstrated robust resilience to fluctuating ammonia nitrogen, and the application of ultrasonic waves significantly enhanced granular activity while achieving in situ sludge reduction.

Abstract:Aerobic granular sludge technology has been extensively studied over the past 20 years and is regarded as the upcoming new standard for biological treatment of domestic and industrial wastewaters. Aerobic granules (AG) are dense, compact, self-immobilized microbial aggregates that allow better sludge-water separation and thereby higher biomass concentrations in the bioreactor than conventional activated sludge aggregates. This brings potential practical advantages in terms of investment cost, energy consumption and footprint. Yet, despite the relevant advances regarding the process of AG formation, instability of AG during long-term operation is still seen as a major barrier for a broad practical application of this technology. This paper presents an up-to-date review of the literature focusing on AG stability, aiming to contribute to the identification of key factors for promoting long-term stability of AG and to a better understanding of the underlying mechanisms. Operational conditions leading to AG disintegration are described, including high organic loads, particulate substrates in the influent, toxic feed components, aerobic feeding and too short famine periods. These operational and influent wastewater composition conditions were shown to influence the micro-environment of AG, consequently affecting their stability. Granule stability is generally favored by the presence of a dense core, with microbial growth throughout the AG depth being a crucial intrinsic factor determining its structural integrity. Accordingly, possible practical solutions to improve granule long-term stability are described, namely through the promotion of minimal substrate concentration gradients and control of microbial growth rates within AG, including anaerobic, plug-flow feeding and specific sludge removal strategies.

Abstract:The granule floatation is a serious issue of the anammox (anaerobic ammonium oxidation) process when high loading rates are applied that results in instability or even system collapse. The present study reports the granule floatation in an anammox reactor when high loading rates were applied. The comparison of enlarged photos taken for the settling and floating granules showed that the two kinds of granules both contained macroscopic gas pockets accounting for 11 +/- 14% of total volume. The settling granules had gas tunnels that could release the gas bubbles, while the floating granules did not. The presence of gas bubbles enclosed in the gas pockets led to the small density of 979.2 +/- 15.8 mg L(-1) and flotation of anammox granules. Consequently, the flotation caused washout of anammox granules and the deterioration of anammox process (volumetric removal rate decreased from 4.00 to 2.46 kg N m(-3) d(-1)). The collection of floating granules, breaking them into small pieces and then returning to the anammox reactor proved an effective control strategy. The volumetric removal rate was finally up to 16.5 kg N m(-3) d(-1) after the control strategy was put into use.

Abstract:The internal structure of aerobic granules has a significant impact on the hydrodynamic performance and mass transfer process, and severely affects the efficiency and stability of granules-based reactors for wastewater treatment. In this study, for the first time the granule complex structure was correlated with the hydrodynamic performance and substrates reactions process. First, a series of multiple fluorescence stained confocal laser scanning microscopy images of aerobic granules were obtained. Then, the form and structure of the entire granule was reconstructed. A three-dimensional computational fluid dynamics study was carried out for the hydrodynamic analysis. Two different models were developed on the basis of different fluorescence stained confocal laser scanning microscopy images to elucidate the roles of the granule structure in the hydrodynamic and mass transfer processes of aerobic granules. The fluid flow behavior, such as the velocity profiles, the pathlines and hence the hydrodynamic drag force, exerted on the granule in a Newtonian fluid, was studied by varying the Reynolds number. Furthermore, the spatial distribution of dissolved nutrients (e.g., oxygen) was acquired by solving the convection-diffusion equations on the basis of the reconstructed granule structure. This study demonstrates that the reconstructed granule model could offer a better understanding to the mass transfer process of aerobic granules than simply considering the granule structure to be homogeneous.
Drying and Re-Cultivation of Aerobic Granules.

Abstract:Aerobic granules stored in liquid medium can lose structural integrity during storage. This study demonstrated that the aerobic granules cultivated by seeding activated sludge into column-type sequential batch reactors and fed with synthetic wastewater at organic loading rate of 1.5 kg/m3-d can be dried by acetone gradient method to moisture content less than 1%. Then, the dried granules can be reactivated through a re-cultivation process to recover their organic degradation capacity in 12 h, or their appearance in 5 d. During the drying and recovery, the granules experienced volume and weight losses by >80% and >85%, respectively, with minimal loss in structural integrity. The microbial communities of the dried and re-cultivated granules were probed using polymerase chain reaction-denaturing gradient gel electrophoresis technique. The family Xanthomonadaceae and the family Comamonas can survive in dried granules and could contribute to maintain structural integrity in re-cultivation stage.
Microscopic observation of aerobic granulation in sequential aerobic sludge blanket reactor

Abstract:Aims: This paper attempts to provide visual evidence of how aerobic granulation evolves in sequential aerobic sludge blanket reactors. Methods and results: A series of experiments were conducted in two column-type sequential aerobic sludge reactors fed with glucose and acetate as sole carbon source, respectively. The evolution of aerobic granulation was monitored using image analysis and optical and scanning electron microscopy. The results indicated that the formation of aerobic granules was a gradual process from seed sludge to compact aggregates, further to granular sludge and finally to mature granules with the sequential operation proceeding. Glucose- and acetate-fed granules have comparable characteristics in terms of settling velocity, size, shape, biomass density and microbial activity. However, the microbial diversity of the granules was associated with the carbon source supplied. In this work, an important aerobic starvation phase was identified during sequential operation cycles. It was found that periodical aerobic starvation was an effective trigger for microbial aggregation in the reactor and further strengthened cell-cell interaction to form dense aggregates, which was an essential step of granulation. The periodical starvation-induced aggregates would finally be shaped to granules by hydrodynamic shear and flow. Conclusion: Aerobic granules can be formed within 3 weeks in the systems. The periodical starvation and hydrodynamic conditions would play a crucial role in the granulation process. Significance and impact of the study: Aerobic granules have excellent physical characteristics as compared with conventional activated sludge flocs. This research could be helpful for the development of an aerobic granule-based novel type of reactor for handling high strength organic wastewater.

Abstract:The long granulation time and instability of aerobic granular sludge (AGS) have restricted its large-scale application significantly. The objective of this review is to discuss the recent achievements and progress made in the field of rapid granulation and granule stability. To this end, we review more than one hundred studies in this field. In this review, four fast granulation methods, namely the addition of metal ions, the addition of a carrier, seeding with AGS, and inoculation using special strains are considered. Three stability enhancement strategies, namely, the improvement of the influent substrate, the selection of slow-growing microorganisms, and the addition of carrier particles, are also discussed. Finally, the potential of these strategies to accelerate granulation and increase the stability of granules in large-scale applications is discussed. In addition, combining a carrier with other strategies to promote granulation and improve stability is proposed. Furthermore, the prospect of enhancing the transmission of signal molecules in quorum sensing systems to promote sludge granulation and improve granule stability are discussed.

Abstract:The characteristics of aerobic granules for wastewater treatment are greatly related to their complex internal structure. However, due to the limitation of characterizing methods, information about the granule internal morphology and structure is very sparse, and mechanism of mass transfer process is yet unclear. In this work,the internal structure of aerobic granules was explored using nitrogen adsorption method and confocal laser scanning microscopy technique. It was found that aerobic granules had multiporous structure with cross-linked gel matrix structure. With a consideration of the hydrodynamic regime and the porous structure of granules, a two-dimensional percolation model was established to describe the mass transfer in granules. With the approaches, interesting and useful results regarding the pore distribution and mass transfer in aerobic granules have been obtained. The results demonstrate that the intragranule convection could enhance mass transfer, hence ensure an efficient and stable operation of aerobic-granule-based reactors. Such approaches might also be applicable to characterizing the multiporous structure and mass transfer of other microbial aggregates for wastewater treatment
Disruption of granules by hydrodynamic force in internal circulation anaerobic reactor

Abstract:A process of granule disruption by hydrodynamic force is discussed in this paper. Shear force and attrition among granules originated from hydrodynamic force are the main causes of the disruption. Since it is positively correlated to the attrition force, the shear force is utilized to describe the effect of hydrodynamic force on granule disruption. In the experiment, when increase rate of average shear rate (IRgamma) of the 1st stage is about 0.2 s(-1) x d(-1), the granules are disrupted; while re-granulation could develop when IRgamma is about 0.07 s(-1) x d(-1); even when the shear rate is as high as about 30s(-1), the granulation rate keeps stably at a relatively high level, which shows that granules could bear the high hydrodynamic force only if it increases by low increase rate. The experimental results would be valuable for the operation and controlling of the upflow reactors.
