Abstract:Compared with the research on DNA damage, there are fewer studies on RNA damage, and the damage mechanism remains mostly unknown. Recent studies have shown that RNA is more vulnerable to damage than DNA when the cells are exposed to endogenous and exogenous insults. RNA injury may participate in a variety of disease occurrence and development. RNA not only has important catalytic functions and other housekeeping functions, it also plays a decisive role in the translation of genetic information and protein biosynthesis. Various kinds of stressors, such as ultraviolet, reactive oxygen species and nitrogen, can cause damage to RNA. It may involve in the development and progression of diseases. In this review, we focused on the relationship between the RNA damage and disease as well as the research progress on the mechanism of RNA damage, which is of great significance for the pathogenesis, diagnosis, and treatment of related diseases.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is RNA oxidative damage and its mechanism of action in various diseases. Specifically, the article focuses on the following aspects: 1. **Research status of RNA oxidative damage**: Compared with DNA damage, the research on RNA damage is relatively scarce, and its damage mechanism has not been fully elucidated. The article points out that RNA is more vulnerable to damage than DNA when cells are exposed to endogenous and exogenous insults. 2. **Relationship between RNA damage and diseases**: RNA not only has important catalytic functions and other housekeeping functions, but also plays a crucial role in the translation of genetic information and protein biosynthesis. Various stressors (such as ultraviolet light, reactive oxygen species (ROS) and reactive nitrogen species (RNS)) can cause damage to RNA, which may be involved in the progression and development of diseases. Therefore, studying the relationship between RNA damage and diseases is of great significance for the pathogenesis, diagnosis and treatment of related diseases. 3. **Specific mechanisms of RNA oxidative damage**: The article elaborately explores the specific mechanisms of RNA oxidative damage, including the formation of 8 - hydroxyguanosine (8 - OHG) and its changes in cells. For example, reactive oxygen species (ROS) are generated through the Fenton reaction (an iron - catalyzed oxidation reaction) and promoted in vivo by mitochondrial dysfunction, which is the main cause of RNA oxidative damage. 4. **RNA damage repair mechanisms**: Although it is still unclear whether cells have specific repair mechanisms for RNA oxidative damage, some studies suggest that stress molecules involved in DNA damage may also be involved in the stress response to RNA oxidative damage. For example, the mRNA monitoring and control mechanism can remove incorrect mRNA transcripts, and some proteins (such as hPNPase) can degrade damaged RNA, reducing the 8 - OHG content, thereby alleviating RNA damage. 5. **Relationship between RNA oxidative damage and neurodegenerative diseases, metabolic diseases, cardiovascular diseases and cancer, etc.**: The article summarizes the specific manifestations and potential mechanisms of RNA oxidative damage in these diseases, emphasizing the importance of RNA oxidative damage as an early marker of diseases. In summary, this paper aims to comprehensively summarize the research progress of RNA oxidative damage, reveal its mechanism of action in various diseases, and provide directions for future research.
