Abstract:Dr. Guifen Liu, now an Associate Professor at ZhangLab, Tongji University, began her research in epigenetic regulation using zebrafish as a model, first as a postdoctoral researcher and then as a research scientist. She established the zebrafish culturing system at the Department of Informatics, Tongji University, Shanghai, China. Dr. Qi Wang is an Assistant Professor at ZhangLab, Tongji University, and she is in charge of the cell culture part of the lab and has broad expertise in high-throughput experimental research on chromatin structure. They are the only two experimental biologists in a dry lab focusing on different research topics. Enhancing medical students' science communication skills: from the perspective of new media

Abstract:With the development of science over the years, people have increasingly realized the importance of science communication. Unfortunately, very little research has focused on helping medical students develop the capabilities of science communication. To improve medical students' science communication and evaluate the effectiveness of New Media through mobile clients in health science communication, a competition was held among medical undergraduates. Outstanding works were selected for publication on our official health science communication WeChat account. Furthermore, the participants volunteered to complete a questionnaire survey to help us assess students' awareness of science communication. Our analysis revealed that students had a strong willingness to serve society and to participate in science communication work. Students generally agreed that science communication work had excellent effects on professional knowledge and related skills. In addition, the correlation results showed that the greater students' willingness to participate in health science communication was, the greater their sense of gain. New Media effectively expand the influence of students' popular science works. Our findings suggest that competition in science communication has a positive impact on enhancing students' awareness and capabilities in science communication. In addition, New Media are an effective way to improve students' scientific communication efficiency. However, we also noted that students' participation rate and enthusiasm for scientific communication were not high. Further research is needed to determine the reasons for this situation and potential strategies to further improve students' science communication.NEW & NOTEWORTHY The science communication competition had a positive impact on helping medical students develop awareness and capabilities for science communication. In addition, New Media are an effective way to improve students' scientific communication efficiency.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Background</h3><p>Noninvasive monitoring of fetal development and the early detection of pregnancy-associated complications is challenging, largely due to the lack of information about the molecular spectrum during pregnancy. Recently, cell-free DNA in plasma was found to reflect the global nucleosome footprint and status of gene expression, and showed potential for noninvasive health monitoring during pregnancy.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Objective(s)</h3><p>We aimed to test the relationships between plasma cell-free DNA profiles and pregnancy biology, and evaluate the use of cell-free DNA profile as a non-invasive method for physiological and pathological status monitoring during pregnancy.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Study Design</h3><p>We used genome cell-free DNA sequencing data generated from non-invasive prenatal testing in a total of 2937 pregnant women. For each physiological and pathological conditions, features of cell-free DNA profile were identified using the discovery cohort, and support vector machines classifiers were built and evaluated using independent training and validation cohorts.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>We established nucleosome occupancy profiles at transcription start sites in different gestational trimesters, demonstrated the relationships between gene expression and cell-free DNA coverage at transcription start sites, and showed that the cell-free DNA profiles at transcription start sites represented the biological processes of pregnancy. In addition, using cell-free DNA data, nucleosome profiles of transcription factor binding sites were identified to reflect transcription factor footprint, which may help to reveal the molecular mechanisms underlying pregnancy. Finally, by using machine learning models on low coverage non-invasive prenatal testing data, we evaluated the use of cell-free DNA nucleosome profiles for distinguishing gestational trimesters, fetal gender, and fetal trisomy 21, and highlighted its potential utility for predicting physiological and pathological fetal conditions by using low coverage non-invasive prenatal testing data.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusion(s)</h3><p>Our analyses profiled nucleosome footprints and regulatory networks during pregnancy and established a noninvasive, proof-of-principle methodology for health monitoring during pregnancy.</p>

Abstract:Parallel single-cell multimodal sequencing is the most intuitive and precise tool for cellular status research. In this study, we propose AMAR-seq to automate methylation, chromatin accessibility, and RNA expression coanalysis with single-cell precision. We validated the accuracy and robustness of AMAR-seq in comparison with standard single-omics methods. The high gene detection rate and genome coverage of AMAR-seq enabled us to establish a genome-wide gene expression regulatory atlas and triple-omics landscape with single base resolution and implement single-cell copy number variation analysis. Applying AMAR-seq to investigate the process of mouse embryonic stem cell differentiation, we revealed the dynamic coupling of the epigenome and transcriptome, which may contribute to unraveling the molecular mechanisms of early embryonic development. Collectively, we propose AMAR-seq for the in-depth and accurate establishment of single-cell multiomics regulatory patterns in a cost-effective, efficient, and automated manner, paving the way for insightful dissection of complex life processes.
Investigations on career well-being and career development of young scientific researchers in medical college and affiliated hospital

Abstract:Objective: To investigate the related factors for career well-being among scientific researchers, and put forward suggestions for the construction of research team. Methods: Young scientific researchers aged between 31 and 45 years were randomly selected for questionnaire survey from Ruijin Hospital affiliated to Shanghai Jiaotong University School of Medicine (n=58), Shanghai Institute of Hematology (n=33) and Basic Medical College of Shanghai Jiaotong University (n=39). The questionnaire was composed of career well-being recognition and mood experience scale. The career well-being recognition dimensions included salary, career development, supervisor, welfare, reward, working procedure, colleague, task and interpersonal coordination, and Positive and Negative Affect Schedule-Revised (PANAS-R) was employed in mood experience test. Besides, in-depth interview was carried out in two core scientific researchers in Shanghai Institute of Hematology. Results: The career well-being index of young scientific researchers ranged between 105 and 120, with the average of 112.48 (124 points in total). The scores of salary, welfare and working procedure were lower. There were significant differences in the degree of satisfaction towards career development, working procedure and supervisor among different age groups (P<0.05). The in-depth interview of researchers in Shanghai Institute of Hematology revealed that both the living pressure and working pressure were high, while there was sense of dignity and achievement in working. Conclusion: More reasonable salary distribution system should be established, and individualized incentive system should be constructed in order to prevent the outflow of talents.

Abstract:Dr. Xiyun Yan obtained her BS degree in 1983, from the Henan Medical College, and then studied cell biology in Professor Shizhang Bei’s lab in the Institute of Biophysics, Chinese Academy of Sciences (CAS) until leaving for Germany in 1989. After receiving her medical doctor's degree from Heidelberg University in Germany, she joined the Memorial Sloan-Kettering Institute in New York as a postdoctoral research fellow. In 1997, Dr. Yan returned to China through her selection in the “Hundred Talents Program” by CAS. She has since studied tumor biology, finding novel targets and developing new methods for tumor diagnosis and therapy. Advances in Transcriptome Analysis of Embryo Development Potential

Abstract:In recent years, with the rapid development of transcriptomics, it has become an important approach to explore the development of germ cells and human embryos at the early stage and the emergence of single-cell sequencing technology has a milestone significance. At the germ cell level, scientists have found that the stabilization of RNA levels in oocytes is important for the development of embryos after fertilization. At the same time, many studies have shown that there are complex RNA molecular regulation systems in sperm, which may be closely related to sperm abnormalities. After sperm-egg binding, the activation and differentiation of fertilized eggs is an extremely precise process involving the timely activation and transcriptional expression of multiple genes, paving the way for late implantation. The discovery of these results is inseparable from the advancement of technology, since the latest technology enables simultaneous multi-layer detection of the same cell, and reveals the relationship among methylation, transcription, and inheritance. With the concept of “non-invasive” deeply rooted in people’s mind, the detection of blastocyst culture fluid has received more and more attention. Among them, several transcription factors have been confirmed to be related to embryonic development, and have the potential to become a new system for evaluating high-quality embryos.

Abstract:Antheraea pernyi is not only an important economic insect, it is increasingly employed as a model organism due to a variety of advantages, including ease of rearing and experimental manipulation compared with other Lepidoptera. Peptidoglycan (PGN) is a major component of the bacterial cell wall, and interactions between PGN and A. pernyi cause a series of physiological changes in the insect. In the present study, we constructed cDNA libraries from a A. pernyi PGN-infected group and a control group stimulated with phosphate-buffered saline (PBS). The transcriptome was de novo assembled using the Trinity platform, and 1698 differentially expressed genes (DEGs) were identified, comprising 894 up-regulated and 804 down-regulated genes. To further investigate immune-related DEGs, gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment were performed. GO analysis identified major immune-related GO terms and KEGG enrichment indicated gene responses to three pathways related to the insect immune system. Several homologous genes related to the immune response of the A. pernyi fat body post-PGN infection were identified and categorised. Taken together, the results provide insight into the complex molecular mechanisms of the responses to bacterial infection at the transcriptional level.
