Abstract:Nitrate nitrogen, the end product of nitrogen conversion within the biofilter of recirculating aquaculture systems (RAS), has recently been recognized as a potentially toxic pollutant to fish. However, the influence of salinity on nitrate toxicity in juvenile Hexagrammos otakii (H. H. otakii) ) remains an area of uncertainty. In our study, a 5-week chronic expose experiment was conducted on juvenile H. otakii, , with a salinity of 30 PSU and nitrate-N of 12 mg/ Las control treatment (CT). Additionally, three treatment groups were established with salinities of 12, 21, and 30 PSU, all maintained at a nitrate-N concentration of 150 mg/L (T1, T2, and T3 respectively). The findings revealed pronounced decreases in growth and survival across all treatments, with the effects becoming more pronounced at lower salinity levels. Specifically, T1 exhibited elevated plasma nitrite concentrations and significantly reduced white and red blood cell counts, as well as hemoglobin levels. Histopathological evaluations of gill and liver tissues highlighted notable damage, particularly in the T1 group, impacting oxygen uptake and various metabolic functions. Gene ontology (GO) terms showed significant enrichment of categories about ion channel activity, gas transport, wnt signaling pathway and so on in the gill tissue among groups. In liver tissue, significant changes were observed in ion regulation, oxidoreductase activity, and immune response between the T1 and CT groups, though T2 and T3 showed minimal variances from CT. Furthermore, KEGG analysis indicated that PPAR signaling pathway in both gill and liver tissue was significantly inhibited in T1 group, which was a hallmark of tissue damage and disease development. This research demonstrates the protective role of increased salinity against nitrate toxicity in H. otakii culture. These findings have profound implications for understanding the interplay between salinity and nitrate toxicity, with potential applications in improving aquaculture health and management in RAS.
