Abstract:Mounting evidence suggests that multiple mechanisms underlie working memory capacity. Using transcranial direct current stimulation (tDCS), the current study aimed to provide causal evidence for the neural dissociation of two mechanisms underlying visual working memory (WM) capacity, namely, the scope and control of attention. A change detection task with distractors was used, where a number of colored bars (i.e., two red bars, four red bars, or two red plus two blue bars) were presented on both sides (Experiment 1) or the center (Experiment 2) of the screen for 100ms, and participants were instructed to remember the red bars and to ignore the blue bars (in both Experiments), as well as to ignore the stimuli on the un-cued side (Experiment 1 only). In both experiments, participants finished three sessions of the task after 15min of 1.5mA anodal tDCS administered on the right prefrontal cortex (PFC), the right posterior parietal cortex (PPC), and the primary visual cortex (VC), respectively. The VC stimulation served as an active control condition. We found that compared to stimulation on the VC, stimulation on the right PPC specifically increased the visual WM capacity under the no-distractor condition (i.e., 4 red bars), whereas stimulation on the right PFC specifically increased the visual WM capacity under the distractor condition (i.e., 2 red bars plus 2 blue bars). These results suggest that the PPC and PFC are involved in the scope and control of attention, respectively. We further showed that compared to central presentation of the stimuli (Experiment 2), bilateral presentation of the stimuli (on both sides of the fixation in Experiment 1) led to an additional demand for attention control. Our results emphasize the dissociated roles of the frontal and parietal lobes in visual WM capacity, and provide a deeper understanding of the neural mechanisms of WM.

Abstract:Working memory is essential for cognition, facilitating the temporary maintenance and manipulation of information to produce goal-directed behavior. While both cortical and subcortical structures are involved, their precise roles and interactions are not fully understood. To investigate this, we simultaneously recorded neural activity from the frontal and parietal cortex, higher-order thalamic nuclei, and core basal ganglia structures during color and spatial working memory tasks in non-human primates. We found widespread yet differential encoding of color and spatial information, marked by area-specific temporal dynamics and modulation according to task demands. Both cortical and subcortical information increased towards working memory-dependent actions, suggesting a task-specific reloading of information. Directed interactions between cortical and subcortical regions were extensive and reciprocal, with dominant directions of information flow, especially from frontoparietal areas. These interactions were dynamically modulated and partially task specific. Our findings provide comprehensive insights into the large-scale circuit dynamics underlying primate working memory and suggest that flexible goal-directed behavior relies on the selective processing of task-relevant memory information within information-specific cortico-subcortical networks.

Mechanisms of distributed working memory in a large-scale network of macaque neocortex

Abstract:Neural activity underlying working memory is not a local phenomenon but distributed across multiple brain regions. To elucidate the circuit mechanism of such distributed activity, we developed an anatomically constrained computational model of large-scale macaque cortex. We found that mnemonic internal states may emerge from inter-areal reverberation, even in a regime where none of the isolated areas is capable of generating self-sustained activity. The mnemonic activity pattern along the cortical hierarchy indicates a transition in space, separating areas engaged in working memory and those which do not. A host of spatially distinct attractor states is found, potentially subserving various internal processes. The model yields testable predictions, including the idea of counterstream inhibitory bias, the role of prefrontal areas in controlling distributed attractors, and the resilience of distributed activity to lesions or inactivation. This work provides a theoretical framework for identifying large-scale brain mechanisms and computational principles of distributed cognitive processes.

Functionally Specific and Sparse Domain-Based Micro-Networks in Monkey V1 and V2

Abstract:The cerebral cortices of human and nonhuman primate brains are characterized by submillimeter functional domains. However, little is known about the connections of single functional domains. Here, in macaque monkey visual cortex, we have developed a targeted focal electrical stimulation method, coupled with functional optical imaging, to map cortical networks with submillimeter precision in vivo. We find that single functional domains are a part of highly specific and sparse intra-areal and inter-areal micro-networks. Across color-related and orientation-related functionalities, these micro-networks exhibit parallel connection patterns, suggesting a common domain-based architecture. Moreover, these micro-networks shift topographically at a submillimeter scale, suggesting that they serve as a fundamental unit for cortical information processing. Our findings establish a domain-based connectional architecture in the primate brain and present new constraints for cortical map representation.

Abstract:As demonstrated by anatomical and physiological studies, the cerebral cortex consists of groups of cortical modules, each comprising populations of neurons with similar functional properties. This functional modularity exists in both sensory and association neocortices. However, the role of such cortical modules in perceptual and cognitive behavior is unknown. To aid in the examination of this issue we have applied the high spatial resolution optical imaging methodology to the study of awake, behaving animals. In this paper, we report the optical imaging of orientation domains and blob structures, approximately 100-200 micrometer in size, in visual cortex of the awake and behaving monkey. By overcoming the spatial limitations of other existing imaging methods, optical imaging will permit the study of a wide variety of cortical functions at the columnar level, including motor and cognitive functions traditionally studied with positron-emission tomography or functional MRI techniques.

Little is known about the neural circuitry that underlies this understanding. Sliwa and Freiwald scanned monkey brains as the monkeys watched static or moving stimuli. A subset of brain areas was exclusively active during monkey-monkey interactions, as opposed to physical interactions between two objects. This network shares some of its components with the monkey mirror neuron system mapped previously by others and with a possible homolog of the human network involved in the theory of mind. Abstract:The idling brainStudies of brain function tend to measure activity during specific tasks or in response to specific stimuli. Yet most of the brain's time and energy is not devoted to these activities. Functional magnetic resonance imaging now shows that the monkey brain is constantly cycling through elaborate, distributed patterns of activity of a type previously associated with sensory, motor or cognitive phenomena. The fluctuations are present even during anaesthesia-induced unconsciousness, and correspond to underlying patterns of anatomical connection. These neural circuits may represent the underlying structure that makes perception and thought possible. Intriguingly, the templates are similar (but not identical) in monkeys and humans, suggesting that this structure is conserved across primate species.

Optical Imaging Of Short-Term Working Memory In Prefrontal Cortex Of The Macaque Monkey

Abstract:Prefrontal cortex is an area critical for cognitive functions such as planning, decision-making, and reasoning. Working memory is a key aspect to the execution of these functions and has been strongly associated with prefrontal function. This chapter reviews the functional organization of a prefrontal area, area 46, that has been associated with working memory in monkeys. Anatomical and optical imaging studies indicate the presence of a Clustered organization within area 46, similar in nature to clustered organizations found in sensory cortical areas. Although the relationship of these clusters to working memory function is unknown, optical imaging, studies suggest a spatial organization for mnemonic function. This 'spatial memory map' is topographically consistent with electrophysiologically established maps for visual and eye movement response. Interestingly, in trials in which response suppression is required, optical imaging reveals a possible suppressive signal; lack of this signal may underly the perseveration seen in diseases such as schizophrenia. In sum, I suggest that clustered organization in prefrontal cortex provides a scaffold upon which visual, mnemonic, and motor response are organized.

Abstract:Selective attention is central to cognition. Dramatic advances have been made in understanding the neural circuits that mediate selective attention. Forebrain networks, most elaborated in primates, control all forms of attention based on task demands and the physical salience of stimuli. These networks contain circuits that distribute top-down signals to sensory processing areas and enhance information processing in those areas. A midbrain network, most elaborated in birds, controls spatial attention. It contains circuits that continuously compute the highest priority stimulus location and route sensory information from the selected location to forebrain networks that make cognitive decisions. The identification of these circuits, their functions and mechanisms represent a major advance in our understanding of how the vertebrate brain mediates selective attention.

Abstract:The statistical structure of intrinsic parietal and parieto-frontal connectivity in monkeys was studied through hierarchical cluster analysis. Based on their inputs, parietal and frontal areas were grouped into different clusters, including a variable number of areas that in most instances occupied contiguous architectonic fields. Connectivity tended to be stronger locally: that is, within areas of the same cluster. Distant frontal and parietal areas were targeted through connections that in most instances were reciprocal and often of different strength. These connections linked parietal and frontal clusters formed by areas sharing basic functional properties. This led to five different medio-laterally oriented pillar domains spanning the entire extent of the parieto-frontal system, in the posterior parietal, anterior parietal, cingulate, frontal, and prefrontal cortex. Different information processing streams could be identified thanks to inter-domain connectivity. These streams encode fast hand reaching and its control, complex visuomotor action spaces, hand grasping, action/intention recognition, oculomotor intention and visual attention, behavioral goals and strategies, and reward and decision value outcome. Most of these streams converge on the cingulate domain, the main hub of the system. All of them are embedded within a larger eye-hand coordination network, from which they can be selectively set in motion by task demands.

Abstract:Explorations of learning and memory, other long-term plastic changes, and additional cognitive functions in the behaving primate brain would greatly benefit from the ability to image the functional architecture within the same patch of cortex, at the columnar level, for a long period of time. We developed methods for long-term optical imaging based on intrinsic signals and repeatedly visualized the same functional domains in behaving macaque cortex for a period extending over 1 year. Using optical imaging and imaging spectroscopy, we first explored the relationship between electrical activity and hemodynamic events in the awake behaving primate and compared it with anesthetized preparations. We found that, whereas the amplitude of the intrinsic signal was much larger in the awake animal, its temporal pattern was similar to that observed in the anesthetized animals. In both groups, deoxyhemoglobin concentration reached a peak 2-3 sec after stimulus onset. Furthermore, the early activity-dependent increase in deoxyhemoglobin concentration (the "initial dip") was far more tightly colocalized with electrical activity than the delayed increase in oxyhemoglobin concentration, known to be associated with an increase in blood flow. The implications of these results for improvement of the spatial resolution of blood oxygenation level-dependent functional magnetic resonance imaging are discussed. After the characterization of the intrinsic signal in the behaving primate, we used this new imaging method to explore the stability of cortical maps in the macaque primary visual cortex. Functional maps of orientation and ocular dominance columns were found to be stable for a period longer than 1 year.

Abstract:Human brain pathways supporting language and declarative memory are thought to have differentiated substantially during evolution. However, cross-species comparisons are missing on site-specific effective connectivity between regions important for cognition. We harnessed functional imaging to visualize the effects of direct electrical brain stimulation in macaque monkeys and human neurosurgery patients. We discovered comparable effective connectivity between caudal auditory cortex and both ventro-lateral prefrontal cortex (VLPFC, including area 44) and parahippocampal cortex in both species. Human-specific differences were clearest in the form of stronger hemispheric lateralization effects. In humans, electrical tractography revealed remarkably rapid evoked potentials in VLPFC following auditory cortex stimulation and speech sounds drove VLPFC, consistent with prior evidence in monkeys of direct auditory cortex projections to homologous vocalization-responsive regions. The results identify a common effective connectivity signature in human and nonhuman primates, which from auditory cortex appears equally direct to VLPFC and indirect to the hippocampus. Probing The Edge Of Synchronization: Cortical Activity Transiently Slips Into Slow Waves In Behaving Monkeys

Abstract:Brain networks oscillate between sleep and wakefulness, following circadian rhythms. Theoretical models suggest distinct phases within this cycle, separated by a critical point where long-range activity patterns emerge, an advantageous condition for information processing in cortical networks. However, the exact nature of this critical dynamics remains elusive. A key question is whether the brain operates at this critical point during cognitive tasks or only during resting wakefulness. Here, we analyzed neural signals from the premotor cortex (PMC) of two macaque monkeys engaged in a delayed-reaching task and under drug-induced unconsciousness. We found evidence of criticality during resting periods at the end of behavioral trials in the awake state. This scale-free activity appeared as coordinated traveling waves, like those observed during anesthesia. As predicted by spiking networks models, activity-dependent adaptation influences wave size, supporting the hypothesis that the PMC operates near a synchronization phase transition while avoiding it during active behaviors.

Abstract:A prominent aspect of primate lateral prefrontal cortex organization is its division into several cytoarchitecturally distinct subregions. Neurophysiological investigations in macaques have provided evidence for the functional specialization of these subregions, but an understanding of the relative representational topography of sensory, social, and cognitive processes within them remains elusive. One explanatory factor is that evidence for functional specialization has been compiled largely from a patchwork of findings across studies, in many animals, and with considerable variation in stimulus sets and tasks. Here, we addressed this by leveraging the common marmoset ( Callithrix jacchus ) to carry out large-scale neurophysiological mapping of the lateral prefrontal cortex using high-density microelectrode arrays, and a diverse suite of test stimuli including faces, marmoset calls, and spatial working memory task. Task-modulated units and units responsive to visual and auditory stimuli were distributed throughout the lateral prefrontal cortex, while those with saccade-related activity or face-selective responses were restricted to 8aV, 8aD, 10, 46 V, and 47. Neurons with contralateral visual receptive fields were limited to areas 8aV and 8aD. These data reveal a mixed pattern of functional specialization in the lateral prefrontal cortex, in which responses to some stimuli and tasks are distributed broadly across lateral prefrontal cortex subregions, while others are more limited in their representation.

Abstract:The primate cerebral cortex is organized into specialized areas representing different modalities and functions along a continuous surface. The functional maps across the cortex, however, are often investigated a single modality at a time (e.g., audition or vision). To advance our understanding of the complex landscape of primate cortical functions, here we develop a polarization-gated wide-field optical imaging method for measuring cortical functions through the un-thinned intact skull in awake marmoset monkeys ( Callithrix jacchus ), a primate species featuring a smooth cortex. Using this method, adjacent auditory, visual, and somatosensory cortices are noninvasively parcellated in individual subjects with detailed tonotopy, retinotopy, and somatotopy. An additional pure-tone-responsive tonotopic gradient is discovered in auditory cortex and a face-patch sensitive to motion in the lower-center visual field is localized near an auditory region representing frequencies of conspecific vocalizations. This through-skull landscape-mapping approach provides new opportunities for understanding how the primate cortex is organized and coordinated to enable real-world behaviors.
The Cognitive Critical Brain: Modulation of Criticality in Perception-Related Cortical Regions

Abstract:The constantly evolving world necessitates a brain that can swiftly adapt and respond to rapid changes. The brain, conceptualized as a system performing cognitive functions through collective neural activity, has been shown to maintain a resting state characterized by near-critical neural dynamics, positioning it to effectively respond to external stimuli. However, how near-criticality is dynamically modulated during task performance remains insufficiently understood. In this study, we utilized the prototypical Ising Hamiltonian model to investigate the modulation of near-criticality in neural activity at the cortical subsystem level during perceptual tasks. Specifically, we simulated 2D-Ising models in silico using structural MRI data and empirically estimated the system's state in vivo using functional MRI data. We first replicated previous findings that the resting state is typically near-critical as captured by the Ising model. Importantly, we observed heterogeneous changes in criticality across cortical subsystems during a naturalistic movie-watching task, with visual and auditory regions fine-tuned closer to criticality. A more fine-grained analysis of the ventral temporal cortex during an object recognition task further revealed that only regions selectively responsive to a specific object category were tuned closer to criticality when processing that object category. In conclusion, our study provides empirical evidence from the domain of perception supporting the cognitive critical brain hypothesis that modulating the criticality of subsystems within the brain's hierarchical and modular organization may be a fundamental mechanism for achieving diverse cognitive functions.
A dopamine gradient controls access to distributed working memory in the large-scale monkey cortex

Abstract:Dopamine is required for working memory, but how it modulates the large-scale cortex is unknown. Here, we report that dopamine receptor density per neuron, measured by autoradiography, displays a macroscopic gradient along the macaque cortical hierarchy. This gradient is incorporated in a connectome-based large-scale cortex model endowed with multiple neuron types. The model captures an inverted U-shaped dependence of working memory on dopamine and spatial patterns of persistent activity observed in over 90 experimental studies. Moreover, we show that dopamine is crucial for filtering out irrelevant stimuli by enhancing inhibition from dendrite-targeting interneurons. Our model revealed that an activity-silent memory trace can be realized by facilitation of inter-areal connections and that adjusting cortical dopamine induces a switch from this internal memory state to distributed persistent activity. Abstract:Abstract Competing theories of consciousness disagree on the role of frontoparietal networks. Here we recorded neural activity from 727 intracerebral contacts in 13 epileptic patients, while they detected near-threshold targets preceded by supra-threshold exogenous cues. Trajectory k -means clustering revealed three patterns: (1) Validly cued seen targets elicit sustained activity in right-hemisphere fronto-temporal regions, connected by superior longitudinal fasciculus (SLF) II/III, and late accumulation activity in bilateral dorsolateral prefrontal cortex and in right-hemisphere orbitofrontal cortex (SLF I/III). (2) Invalidly cued seen targets elicit early, sustained activity in the right-hemisphere reorienting network (SLF III). (3) Seen targets elicit late sustained left dorsolateral prefrontal activity. Task modeling with recurrent neural networks supported the causal contribution of these networks to conscious perception, and elucidated their interplay through excitatory-inhibitory mechanisms. We provide new, compelling evidence on the role of hemisphere-asymmetric frontoparietal networks associated with attentional enhancement and reorienting in conscious perception.
