Abstract:A novel and efficient approach toward the assembly of the synthetically challenging tetracyclic [6,5,7,5] core structure of calyciphylline A-type alkaloids is developed. The synthetic route features a tandem semipinacol rearrangement/Nicholas reaction that has been devised strategically to construct the spirocyclic A/B ring and the sterically congested vicinal all-carbon quaternary carbon centers in high diastereoselectivity. Late-stage installation of the hydropyrrole ring and the strained 7-membered ring via a double-reductive amination and ring-closing metathesis, respectively, has also been realized.

Abstract:AbstractCyclopianes are novel diterpenes featuring a highly strained 6/5/5/5 tetracyclic core embedded with 6–8 consecutive stereocenters. The concise total syntheses of (−)‐conidiogenone B, (−)‐conidiogenone, and (−)‐conidiogenol have been accomplished in 14–17 steps. The present work features a HAT‐mediated alkene–nitrile cyclization to access the cis‐biquinane, a Nicholas/Pauson–Khand reaction to construct the linear triquinane, and a Danheiser annulation to afford the congested angular triquinane skeleton.

Abstract:The structurally intriguing tetracyclic core of complex harziane diterpenoid was constructed in 14 steps from commercially available 3-ethoxycyclohex-2-en-1-one. The key steps were a Mn/Cu-mediated oxidative 1,3-dicarbonyl radical cascade cyclization reaction, which diastereoselectively formed the core of dimethylbicyclo[3.2.1]octane structure, and a Au-catalyzed diastereoselective formal [2 + 2] cycloaddition for construction of the harziane diterpenoid tetracyclic framework. The developed method paves the way for achieving total synthesis of this type of complex natural product.

Abstract:Enantioselective construction of contiguous quater-nary centers is a daunting challenge in the synthetic community. Substituted 3-(methoxycarbonyl)cyclobutenone, possessing strained C=C double bond, exhibits both good stability and reactivity. Here, we report a chiral oxazaborolidinium ion (COBI)-catalyzed enantioselective Diels-Alder reaction of 2,3-disubsti-tuted cyclobutenones with a variety of dienes to access bicyclo[4.2.0]octane derivatives, bearing two contiguous angular quaternary centers. Further transformations, including alkylation, cross-coupling reaction and oxyallyl cation reaction afforded the molecules with intriguing structural patterns. The formal total synthesis of estrone was also completed in three steps from enantio-enriched cycloadducts.

Abstract:Comprehensive Summary The natural products of dispirocochlearoids are complex Ganoderma meroterpenoids featuring a 6/6/5/6/6/6 ring system and are selective COX-2 inhibitors. Herein, we describe our progress regarding the total synthesis of dispirocochlearoids A-C, in which a hexacyclic skeleton was constructed firstly. The key steps of this work involve an intermolecular [4 + 2] cycloaddition reaction, a ring cleavage approach via TiCl4, and the installation of a profoundly sterically hindering lactone.

Abstract:Tetrahydroisoquinoline alkaloids with a C1 stereogenic center are a common unit in many natural and non-natural compounds of biological importance. Herein we describe a novel Cu(I) -catalyzed highly chemo- and enantioselective synthesis of chiral tetrahydroisoquinoline-alkaloid derivatives from readily available unsubstituted tetrahydroisoquinolines, aldehydes, and terminal alkynes in the presence of the ligand (R,R)-N-pinap. Abstract:A novel method for the construction of a cyclopenta[c]quinoline ring via cyclization of 3-bromoindoles with internal alkynes in the presence of palladium is described. The formation of the cyclopenta[c]quinoline ring is proposed from a double [1,5] carbon sigmatropic rearrangement of the spirocyclic cyclopentadiene intermediate, which is generated in situ from the cyclization of 3-bromoindoles with internal alkynes involving a sequential double alkyne insertion into the carbon-palladium bond and dearomatization of indole. The present studies have developed a novel ring-expansion reaction of the pyrrole ring to pyridine via one carbon insertion into the C2-C3 bond of indoles and provided a simple and distinct route for the construction of tricyclic fused-quinoline derivatives that are not easy to access with conventional methods.

Abstract:Total syntheses of the biologically important and structurally unique tetracyclic diterpenes conidiogenone, conidiogenol, and conidiogenone B of the cyclopiane class are reported. The absolute configuration of naturally occurring conidiogenone B was also corrected. The key step of our strategy involved the highly efficient construction of both ring C and the quaternary carbon center shared by rings A and C through a one‐step regioselective and diastereoselective cycloenlargement in the form of a semipinacol‐type rearrangement. In particular, the desired regioselectivity was made possible by properly adjusting the migratory aptitude of the migrating carbon atom through the introduction of an electron‐donating phenylthio group at this position.

Abstract:A novel one-pot triple-reaction strategy for the asymmetric construction of polycyclic skeletons with multiple consecutive chiral centers through aza-Michael/Michael/Wittig/ketyl radical addition/esterification processes is reported. A wide range of polycyclic tetrahydroquinoline derivatives were smoothly obtained from easily available starting materials with good results (up to 80% yield, >20:1 dr, >99% ee) under mild conditions. Abstract:An efficient protocol for the direct transformation of chrornan-4-ones to tricyclic fused pyridines with the skeleton of cassiarins, a family of alkaloids with antimalarial activity, was developed. Also, a general strategy for modular construction of polyheterocydes with diverse natural product-like skeletons was developed by using ketone-alkyne bifunctional substrates. These reactions involved a one-pot cascade oximation of ketones, rhodium-catalyzed C-H activation, and intermolecular/intramolecular alkyne annulations under mild conditions with high atom, step, and redox economy.

Abstract:A new method for one-step construction of the tetracyclic core structure of the indole alkaloid (+)-minfiensine was developed utilizing a palladium-catalyzed asymmetric indole dearomatization/iminium cyclization cascade. An efficient total synthesis of (+)-minfiensine was realized using this strategy. The present method enables access to the common core structure of a series of monoterpene indole alkaloids, such as vincorine, echitamine, and aspidosphylline A.

Abstract:A novel tandem 1,6-addition/cyclopropanation/rearrangement reaction of vinylogous para -quinone methides with 3-chlorooxindoles has been developed, providing dispirooxindole–cyclopentane–cyclohexadienones with vicinal quaternary carbon centers. A novel tandem 1,6-addition/cyclopropanation/rearrangement reaction of vinylogous para -quinone methides with 3-chlorooxindoles is reported. This method provides an efficient approach to various functionalized dispirooxindole–cyclopentane–cyclohexadienones with vicinal quaternary carbon centers. Computational studies have been conducted to provide insight into the origin of diastereoselectivity and predict a stepwise diradical mechanism. The anti-tumor activities of these structurally unique dispirooxindole derivatives are also reported.
