Abstract:Cuscuta, commonly known as dodder, is a parasitic plant that thrives by attaching itself to the stems of other plants. It is found across the globe and is notorious for obstructing crop growth as a weed. Over the past decade, Cuscuta has been used to gain insights into plant-plant interactions and molecular trafficking. Here, we report that two plants connected via a Cuscuta bridge can exchange rapid systemic calcium, electric, and reactive oxygen species signals. These findings suggest that plant interactions with Cuscuta may have beneficial effects to plants, enabling them to rapidly communicate with each other.

Orchestrating rapid long-distance signaling in plants with Ca2+ , ROS and electrical signals

Abstract:Plants show a rapid systemic response to a wide range of environmental stresses, where the signals from the site of stimulus perception are transmitted to distal organs to elicit plant-wide responses. A wide range of signaling molecules are trafficked through the plant, but a trio of potentially interacting messengers, reactive oxygen species (ROS), Ca2+ and electrical signaling ('trio signaling') appear to form a network supporting rapid signal transmission. The molecular components underlying this rapid communication are beginning to be identified, such as the ROS producing NAPDH oxidase RBOHD, the ion channel two pore channel 1 (TPC1), and glutamate receptor-like channels GLR3.3 and GLR3.6. The plant cell wall presents a plant-specific route for possible propagation of signals from cell to cell. However, the degree to which the cell wall limits information exchange between cells via transfer of small molecules through an extracellular route, or whether it provides an environment to facilitate transmission of regulators such as ROS or H+ remains to be determined. Similarly, the role of plasmodesmata as both conduits and gatekeepers for the propagation of rapid cell-to-cell signaling remains a key open question. Regardless of how signals move from cell to cell, they help prepare distant parts of the plant for impending challenges from specific biotic or abiotic stresses.
Common mycelial network mediated inter-plant signals modulate plant biotic stress responses and defence against foliar pathogens

Abstract:Arbuscular mycorrhizal fungi (AMF) colonize multiple plant hosts and form common mycelial networks (CMNs) that link multiple plants in nature. CMNs have been hypothesised to function as a highway for interplant information exchange to modulate plant biotic and abiotic stress responses. Here we used AMF Rhizophagus irregularis to inter connect two Medicago truncatula plants and explored the effect of known plant defence elicitor on pathogen tolerance of AMF connected inter-plant signal receivers. We analysed Medicago leaf metabolites (HPLC MS) and emitted volatiles (HS GS MS) together with RNA-seq data to compare responses of the inter-plant signal receivers with intact and cut CMN. The integrity of CMN significantly affected interplant signal receiver responses. Plant defence and signalling pathways were enriched with receiver transcripts that are uniquely changing in the intact vs interrupted CMN along with distinct production of plant isoprenoids: volatile monoterpenes and triterpene saponins. Furthermore, receivers of CMN-mediated signals from stressed senders display increased resistance to Fusarium sporotrichoides and susceptibility to Botrytis cinerea. Our results highlight CMN contribution to both upregulated and downregulated receiver plant gene functions and metabolites which may encode susceptibility and resistance factors that are important in pathogen species specific plant responses. Future dissection of the mechanisms involved in inter-plant signal decoding will yield novel discoveries on genetic regulation of interplant defence priming under pathogen attack.

Abstract:Elevated atmospheric carbon dioxide (eCO(2)) concentrations promote symbiosis between roots and arbuscular mycorrhizal fungi (AMF), modifying plant nutrient acquisition and cycling of carbon, nitrogen and phosphate. However, the biological mechanisms by which plants transmit aerial eCO(2) cues to roots, to alter the symbiotic associations remain unknown. We used a range of interdisciplinary approaches, including gene silencing, grafting, transmission electron microscopy, liquid chromatography tandem mass spectrometry (LC-MS/MS), biochemical methodologies and gene transcript analysis to explore the complexities of environmental signal transmission from the point of perception in the leaves at the apex to the roots. Here we show that eCO(2) triggers apoplastic hydrogen peroxide (H2O2)-dependent auxin production in tomato shoots followed by systemic signaling that results in strigolactone biosynthesis in the roots. This redox-auxin-strigolactone systemic signaling cascade facilitates eCO(2)-induced AMF symbiosis and phosphate utilization. Our results challenge the current paradigm of eCO(2) effects on AMF and provide new insights into potential targets for manipulation of AMF symbiosis for high nutrient utilization under future climate change scenarios.

Abstract:Plants possess systemic signaling networks that allow the perception of local stresses to be translated into plant-wide responses. Although information can be propagated via a variety of molecules such as hormones and RNAs moving within the bulk flow of the phloem or in the transpiration stream, the vasculature also appears to be a major pathway whereby extremely rapid signals move bi-directionally throughout the plant. In these cases, the movement mechanisms are not dependent on redistribution through bulk flow. For example, self-reinforcing systems based around changes in Ca<sup>2+</sup> and reactive oxygen species, coupled to parallel electrical signaling events appear able to generate waves of information that can propagate at hundreds of μm/s. These signals then elicit distant responses that prime the plant for a more effective defense or stress response in unchallenged tissues. Although ion channels, Ca<sup>2+</sup>, reactive oxygen species and associated molecular machineries, such as the NADPH oxidases, have been identified as likely important players in this propagation system, the precise nature of these signaling networks remains to be defined. Critically, whether different stimuli are using the same rapid, systemic signaling network, or whether multiple, parallel pathways for signal propagation are operating to trigger specific systemic outputs remains a key open question.

Abstract:Cuscuta campestris, a stem parasitic plant, has served as a valuable model plant for the exploration of plant-plant interactions and molecular trafficking. However, a major barrier to C. campestris research is that a method to generate stable transgenic plants has not yet been developed. Here, we describe the development of a Cuscuta transformation protocol using various reporter genes (GFP, GUS, or RUBY) and morphogenic genes (CcWUS2 and CcGRF/GIF), ultimately leading to a robust protocol for Agrobacterium-mediated C. campestris transformation. The stably transformed and regenerated RUBY C. campestris plants produced haustoria, the signature organ of parasitic plants, and these were functional in forming host attachments. The locations of T-DNA integration in the parasite genome were confirmed through TAIL-PCR. Transformed C. campestris also produced flowers and transgenic seeds exhibiting betalain pigment, providing proof of germline transmission of the RUBY transgene. Furthermore, the RUBY reporter is not only a useful selectable marker for the Agrobacterium-mediated transformation, but also provides insight into the movement of molecules from C. campestris to the host during parasitism. Thus, the protocol for transformation of C. campestris reported here overcomes a major obstacle to Cuscuta research and opens new possibilities for studying parasitic plants and their interactions with hosts.

Abstract:Parasitic plants use a special organ, the haustorium, to attach to and penetrate host tissues, forming phloem and/or xylem fusion with the host vascular systems. Across this haustorium-host interface, not only water and nutrients are extracted from the host by the parasitic plant, but also secondary metabolites, messenger RNAs, noncoding RNAs, proteins, and systemic signals are transported between the parasite and host and even among different hosts connected by a parasite. Furthermore, mycorrhizal fungi can form common mycelial networks (CMNs) that simultaneously interconnect multiple plants. Increasing lines of evidence suggest that CMNs can function as conduits, transferring stress-related systemic signals between plants. Between-plant signaling mediated by haustoria and CMNs likely has a profound impact on plant interactions with other organisms and adaptation to environmental factors. Here, we summarize the findings regarding between-plant transfer of biomolecules and systemic signals and the current understanding of the physiological and ecological implications of between-plant signaling.

Abstract:Abstract Cucurbit crops are suitable models for studying long-distance signaling in horticultural plants. Although thousands of substances are graft transmissible in cucurbits, functional studies have been hampered by the lack of efficient genetic transformation systems. Here, we report a convenient and efficient root transformation method for several cucurbit crops that will facilitate studies of functional genes and shoot–root crosstalk. We obtained healthy plants with completely transformed roots and non-transgenic shoots within 6 weeks. Furthermore, we combined this root transformation method with grafting, which allowed for gene manipulation in the rootstock. We validated our system by exploring salt tolerance mechanisms using a cucumber (Cucumis sativus)/pumpkin (Cucurbita moschata Duch.) (scion/rootstock) graft in which the sodium transporter gene High-affinity K+ transporter1 (CmoHKT1;1) was edited in the pumpkin rootstock, and by overexpressing the pumpkin tonoplast Na+/H+ antiporter gene Sodium hydrogen exchanger4 (CmoNHX4) in cucumber roots.

Abstract:The co-existence of plants and microorganisms throughout their evolution resulted in an intertwined ecological network that serves as the basis for the functioning of today's terrestrial ecosystems. The interactions between plants, bacteria, and fungi are often multipartite and occur both within and among kingdoms. Apart from the textbook examples of tight mutualistic associations such as root nodule symbiosis or mycorrhizas, there are also looser interactions, e.g., those between plants and plant growth-promoting bacteria and fungi. Additionally, plants are also targets of microbial pathogens. Thus, intricate mechanisms of how plants and microorganisms recognize each other have evolved, mediated by chemical signals resulting in a differential gene expression and, eventually, a physiological response. This paper aims to review the molecular basis of such signaling, the apparatus of signal sensing and transduction in bacteria and plants are outlined and exemplified. Moreover, examples of recent discoveries in the field of plant–microbe signaling are presented, i.e., quorum quenching and the involvement of small RNAs. Last but not least, suggestions are provided for the potential exploitation of plant–microbe communication networks for human benefit, e.g., in the field of sustainable agriculture, aiming at crop growth promotion and protection against pathogens.

Integration of electric, calcium, reactive oxygen species and hydraulic signals during rapid systemic signaling in plants

Abstract:The sensing of abiotic stress, mechanical injury or pathogen attack by a single plant tissue results in the activation of systemic signals that travel from the affected tissue to the entire plant. This process is essential for plant survival during stress and is termed systemic signaling. Among the different signals triggered during this process are calcium, electric, reactive oxygen species and hydraulic signals. These are thought to propagate at rapid rates through the plant vascular bundles and to regulate many of the systemic processes essential for plant survival. Although the different signals activated during systemic signaling are thought to be interlinked, their coordination and hierarchy still need to be determined. Here, using a combination of advanced whole-plant imaging and hydraulic pressure measurements, we studied the activation of all four systemic signals in wild-type and different Arabidopsis thaliana mutants subjected to a local treatment of high-light (HL) stress or wounding. Our findings reveal that activation of systemic membrane potential, calcium, reactive oxygen species and hydraulic pressure signals, in response to wounding, is dependent on glutamate receptor-like proteins 3.3 and 3.6. In contrast, in response to HL stress, systemic changes in calcium and membrane potential depended on glutamate receptor-like 3.3 and 3.6, while systemic hydraulic signals did not. We further show that plasmodesmata functions are required for systemic changes in membrane potential and calcium during responses to HL stress or wounding. Our findings shed new light on the different mechanisms that integrate different systemic signals in plants during stress.
A Systemic Small RNA Signaling System in Plants

Abstract:Systemic translocation of RNA exerts non-cell-autonomous control over plant development and defense. Long-distance delivery of mRNA has been proven, but transport of small interfering RNA and microRNA remains to be demonstrated. Analyses performed on phloem sap collected from a range of plants identified populations of small RNA species. The dynamic nature of this population was reflected in its response to growth conditions and viral infection. The authenticity of these phloem small RNA molecules was confirmed by bioinformatic analysis; potential targets for a set of phloem small RNA species were identified. Heterografting studies, using spontaneously silencing coat protein (CP) plant lines, also established that transgene-derived siRNA move in the long-distance phloem and initiate CP gene silencing in the scion. Biochemical analysis of pumpkin (Cucurbita maxima) phloem sap led to the characterization of C. maxima Phloem SMALL RNA BINDING PROTEIN1 (CmPSRP1), a unique component of the protein machinery probably involved in small RNA trafficking. Equivalently sized small RNA binding proteins were detected in phloem sap from cucumber (Cucumis sativus) and lupin (Lupinus albus). PSRP1 binds selectively to 25-nucleotide single-stranded RNA species. Microinjection studies provided direct evidence that PSRP1 could mediate the cell-to-cell trafficking of 25-nucleotide single-stranded, but not double-stranded, RNA molecules. The potential role played by PSRP1 in long-distance transmission of silencing signals is discussed with respect to the pathways and mechanisms used by plants to exert systemic control over developmental and physiological processes.

Abstract:Abstract Plants and microbes communicate to collaborate to stop pests, scavenge nutrients, and react to environmental change. Microbiota consisting of thousands of species interact with each other and plants using a large chemical language that is interpreted by complex regulatory networks. In this work, we develop modular interkingdom communication channels, enabling bacteria to convey environmental stimuli to plants. We introduce a “sender device” in Pseudomonas putida and Klebsiella pneumoniae , that produces the small molecule p -coumaroyl-homoserine lactone (pC-HSL) when the output of a sensor or circuit turns on. This molecule triggers a “receiver device” in the plant to activate gene expression. We validate this system in Arabidopsis thaliana and Solanum tuberosum (potato) grown hydroponically and in soil, demonstrating its modularity by swapping bacteria that process different stimuli, including IPTG, aTc and arsenic. Programmable communication channels between bacteria and plants will enable microbial sentinels to transmit information to crops and provide the building blocks for designing artificial consortia.

Abstract:Plant beneficial microorganisms improve the health and growth of the associated plants. Application of beneficial microbes triggers an enhanced resistance state, also termed as induced systemic resistance (ISR), in the host, against a broad range of pathogens. Upon the activation of ISR, plants employ long-distance systemic signaling to provide protection for distal tissue, inducing rapid and strong immune responses against pathogens invasions. The transmission of ISR signaling was commonly regarded to be a jasmonic acid- and ethylene-dependent, but salicylic acid-independent, transmission. However, in the last decade, the involvement of both salicylic acid and jasmonic acid/ethylene signaling pathways and the regulatory roles of small RNA in ISR has been updated. In this review, the plant early recognition, responsive reactions, and the related signaling transduction during the process of the plant–beneficial microbe interaction was discussed, with reflection on the crucial regulatory role of small RNAs in the beneficial microbe-mediated ISR.

Abstract:The genus Cuscuta is stem parasitic angiosperms that parasitize a wide range of vascular plants via de novo formation of a distinctive parasitic organ called a haustorium. In the developing haustorium, meristematic cells, which are initiated from the stem cortical tissue, differentiate into haustorial parenchyma cells, which elongate, penetrate into the host tissues, and finally connect with the host vasculature. This interspecific vasculature connection allows the parasite to uptake water and nutrients from the host plant. Although histological aspects of haustorium development have been studied extensively, the molecular mechanisms underlying vasculature development and the interspecific connection with the host vasculature remain largely unknown. To gain insights into the interspecific cell-to-cell interactions involved in haustorium development, we established an in vitro haustorium induction system for Cuscuta campestris using Arabidopsis thaliana rosette leaves as the host plant tissue. The in vitro induction system was used to show that interaction with host tissue was required for the differentiation of parasite haustorial cells into xylem vessel cells. To further characterize the molecular events occurring during host-dependent xylem vessel cell differentiation in C. campestris, we performed a transcriptome analysis using samples from the in vitro induction system. The results showed that orthologs of genes involved in development and proliferation of vascular stem cells were up-regulated even in the absence of host tissue, whereas orthologs of genes required for xylem vessel cell differentiation were up-regulated only after some haustorial cells had elongated and contacted the host xylem. Consistent results were obtained by another transcriptome analysis of the haustorium development in C. campestris undergoing parasitization of an intact host plant. These findings suggest the involvement of host-derived signals in the regulation of non-autonomous xylem vessel differentiation and suggest that its connection to the host xylem during the haustorium development activates a set of key genes for differentiation into xylem vessel cells.

A New, Nondestructive, Split-Root System for Local and Systemic Plant Responses Studies with Soybean

Abstract:Plants use long-distance signaling mechanisms to coordinate their growth and control their interactions, positive or negative, with microbes. Split-root systems (SRS) have been used to study the relevance of both local and systemic plant mechanisms that participate in the control of rhizobia-legume symbioses. In this work we have developed a modification of the standard split-root system (SRS) used with soybean. This modified method, unlike previous systems, operates in hydroponics conditions and therefore is nondestructive and allows for the continuous monitoring of soybean roots throughout the whole experiment.

Abstract:The process of plant immune response is orchestrated by intracellular signaling molecules. Since plants are devoid of a humoral system, they develop extensive mechanism of pathogen recognition, signal perception, and intricate cell signaling for their protection from biotic and abiotic stresses. The pathogenic attack induces calcium ion accumulation in the plant cells, resulting in calcium signatures that regulate the synthesis of proteins of defense system. These calcium signatures induct different calcium dependent proteins such as calmodulins (CaMs), calcineurin B-like proteins (CBLs), calcium-dependent protein kinases (CDPKs) and other signaling molecules to orchestrate the complex defense signaling. Using advanced biotechnological tools, the role of Ca 2+ signaling during plant-microbe interactions and the role of CaM/CMLs and CDPKs in plant defense mechanism has been revealed to some extent. The Emerging perspectives on calcium signaling in plant-microbe interactions suggest that this complex interplay could be harnessed to improve plant resistance against pathogenic microbes. We present here an overview of current understanding in calcium signatures during plant-microbe interaction so as to imbibe a future direction of research.

Abstract:Every living organism on Earth depends on its interactions with other organisms to live. In the rhizosphere, plants and microorganisms constantly exchange signals and influence each other's behavior. Recent studies have shown that many beneficial rhizosphere microbes can produce specific signaling molecules that affect plant root architecture and may therefore have substantial effects on aboveground growth. This review sorts out these chemical signals and summarizes their mechanisms of action, enhancing our understanding of plant-microbe interactions and providing references for the comprehensive development and utilization of these active components in agricultural production. Finally, we have pointed out future research directions and challenges, such as the searching of microbial signals to induce the primary root development.

Abstract:To elude or minimize the effects of disease and herbivory, plants rely on both constitutive and inducible defenses. In response to attack by pathogens or pests, plants activate signaling cascades leading to the accumulation of endogenous hormones that trigger the induction of defenses. Salicylic acid (SA), jasmonic acid (JA), and ethylene (E) are plant-specific hormones involved in communicating the attack by many pathogens and pests in a broad range of plant species. SA, JA and E signaling cascades do not activate defenses independently, but rather establish complex interactions that determine the response mounted in each condition. Deployment of defenses is energetically costly, so a trade-off between the activation of resistance against a particular pest or pathogen and down regulation of other defenses is common. Conversely, activation of broad range resistance in response to an initial attack may serve to deter opportunistic agents. Thus, the interaction among SA, JA and E defense signaling pathways can be antagonistic, cooperative or synergistic, depending on the plant species, the combination of organisms attacking the plants, and the developmental and physiological state of the plant. A characterization of the interactions among defense signaling pathways and the determination of the molecular components mediating cross-talk between the different pathways will be essential for the rational design of transgenic plants with increased resistance to disease and/or herbivores without critically compromising other agronomic traits.
