Abstract:Tetracycline antibiotics (TCs) are extensively used in clinical medicine, animal husbandry, and aquaculture because of their cost-effectiveness and high antibacterial efficacy. However, the presence of TCs residues in the environment poses risks to humans. In this study, an inner filter effect (IFE) fluorescent probe, 2,2'-(ethane-1,2-diylbis((2-((2-methylquinolin-8-yl)amino)-2-oxoethyl)azanediyl))diacetic acid (MQDA), was developed for the rapid detection of Eu3+ within 30 s. And its complex [MQDA-Eu3+] was successfully used for the detection of TCs. Upon coordination of a carboxyl of MQDA with Eu3+ to form a [MQDA-Eu3+] complex, the carboxyl served as an antenna ligand for the effective detection of Eu3+ to intensify the emission intensity of MQDA via "antenna effect", the process was the energy absorbed by TCs via UV excitation was effectively transferred to Eu3+. Fluorescence quenching of the [MQDA-Eu3+] complex was caused by the IFE in multicolor fluorescence systems. The limits of detection of [MQDA-Eu3+] for oxytetracycline, chlorotetracycline hydrochloride, and tetracycline were 0.80, 0.93, and 1.7 μM in DMSO/HEPES (7:3, v/v, pH = 7.0), respectively. [MQDA-Eu3+] demonstrated sensitive detection of TCs in environmental and food samples with satisfactory recoveries and exhibited excellent imaging capabilities for TCs in living cells and zebrafish with low cytotoxicity. The proposed approach demonstrated considerable potential for the quantitative detection of TCs.
Integrated Eu3+ loaded covalent organic framework with smartphone for ratiometric fluorescence detection of tetracycline

Abstract:Developing rapid tetracycline sensing system is of great significance to monitor the illegal addition to drugs and pollution to food and ecosystem. By loading covalent organic frameworks (COFs) with Eu3+, a new hybridized material (COF@Eu3+) was prepared for tetracycline determination. Based on the Schiff base reaction, the COFs were by synthesized through solvent evaporation in 30 min at room temperature. Thereafter, Eu3+ was modified into COFs to develop the COF@Eu3+ sensing platform by adsorption and coordination. In presence of tetracycline, tetracycline can displace water molecules and coordinate with Eu3+ through the antenna effect. As a result, the red fluorescence of Eu3+ was enhanced by tetracycline with green fluorescence of COF as a reference. The developed ratiometric fluorescence sensor exhibits a linear range of 0.1-20 μM for detecting tetracycline with a detection limit of 30 nM. Integrated with a smartphone, the rapid tetracycline detection can be realized in situ, which is potential for high-throughput screening of tetracycline contaminated samples. Furthermore, the COF@Eu3+ fluorescence sensor has been successfully applied to the detection of tetracycline in traditional Chinese medicine compound preparation with satisfied recoveries. Therefore, a smartphone-assisted device was successfully developed based on Eu3+-functionalized COF, which is an attractive candidate for further applications of fluorescence sensing and visual detection.
Ratiometric fluorescence sensing and discrimination of tetracycline analogs by using coumarin-embedded Eu-MOF nanosensor

Abstract:As widely used antibiotics, tetracycline residues exist in food and environmental media, which pose certain hidden dangers and negative effects on public health. Therefore, the sensing and discrimination of tetracycline analogs (TCs) have great significance for improving food safety and preventing environmental pollution. Herein, a 7-hydroxycoumarin-3-carboxylic acid-embedded Eu-MOF (HC@Eu-MOF) material was constructed and then developed for the detection of TCs. Upon addition of TCs, the synthesized sensor displays opposite fluorescence changes at two different wavelengths due to the simultaneous presence of the inner filter effect (IFE) and the antenna effect (AE), and achieves a stable ratio signal response within 90 s. In addition, six important tetracycline analogs, including chlortetracycline (CTC), oxytetracycline (OTC), tetracycline (TC), metacycline (MC), doxycycline (DC) and demeclocycline (DMC) can be discriminated with 100 % accuracy through the principal component analysis even in extremely complicated mixtures. Further, a smartphone-assisted portable device was applied for visual sensing of TCs. The as-developed platform possessed the characteristics of simple synthesis, fast response, high sensitivity, and high stability, which further lays a further foundation for the on-site visual detection and discrimination of TCs in real samples.
Sensitive and selective detection of tetracycline using fluorescence-enhanced Eu(III)-functionalized silver nanoparticles with homocysteine

Abstract:Utilizing metal luminescence enhancement to design fluorescent probes is a very sensible strategy. Herein, a fluorescent probe based on europium (III)-functionalized silver nanoparticles-conjugated homocysteine (AgNPs-Hcy-Eu3+) was proposed for the selective and sensitive detection of tetracycline (TC). In this probe, Eu(III) was employed as the detection signal unit for TC, while AgNPs-Hcy was used as the ligand of fluorescence enhancement. When TC exists, it can bind to Eu3+ immobilized in AgNPs-Hcy, leading to an enhanced fluorescence signal from Eu3+ through energy transfer. Under optimal conditions, the fluorescence intensity of AgNPs-Hcy-Eu3+ increased linearly with increasing TC concentration in the range of 0.1-30 μM (R2 = 0.9964). The fluorescent probe own fluorescence enhancement, paving the way for sensitive detection with a low detection limit of 0.083 μM. It also has good selectivity for common antibiotics and anions. This work can be applied to the determination of TC in tap water and milk with recoveries of 94-98.5%. We expect AgNPs-Hcy-Eu3+ to have potential applications in environmental testing and food safety.

Abstract:Luminescent lanthanide complexes have attracted growing attention in the visual probe for detection of toxic and dangerous materials. Here, we report an interfacial self-assembly of Eu3+-dipicolinic acid complex functionalized nanoSiO2 composites (nanoSiO2PyTAEu) and their Langmuir-Blodgett (LB) films as well as the luminescent probes to tetracycline (TC) and oxytetracycline (OTC). Organic linkers and Eu3+ ions are anchored on the hy-drophilic nanoSiO2 particles' surface via the surface silanization, substitution and coordination reactions, so they exhibit high stability and well distribution in the solutions and LB films. Both the nanocomposites of nano-SiO2PyTAEu and their LB films show strong and enhanced red fluorescence emissions of Eu3+ ions when they are immersed in the aqueous TC solution, with the detection limit to 3.3 and 0.91 nmol/L, respectively. The present nanocomposites and LB films have the advantages of high sensitivity and stability.

Abstract:In this work, 4-formyl-3-hydroxybenzoic acid (FHBA), which does not contain a large conjugated ring structure, exhibited aggregation-induced emission (AIE) in aqueous solvents. On this basis, FHBA and a metal-organic framework material (i.e., ZIF-8) form into a complex (), which presents the AIE characteristics of FHBA. Then, lanthanide metal (i.e., Eu3+) coordinates with guanosine 5'-monophosphate disodium (GMP) salt to synthesize a coordination polymer (LnCP), which combines with do afford a novel double-emitting fluorescence probe (@Eu-GMP, FZE) for tetracycline antibiotics (TCAs). TCAs coordinate with Eu3+ and act as antenna ligand. Thus, an "antenna effect" comes into being and effectively enhances the emission intensity of Eu3+ at 616 nm. Moreover, the UV-vis spectra of TCAs largely overlap with the excitation spectrum of FHBA, resulting in an inner filter effect (IFE) and quenching of blue-green fluorescence from FHBA molecules at 473 nm. The ratio of fluorescence signal (F616/F473) showed a good linear relationship with the concentration of tetracycline hydrochloride (TC) in the range of 0.1-200.0 μM, and the detection limit was 60 nM (S/N=3). This fluorescence probe can also effectively respond to oxytetracycline hydrochloride (OTC) and chlortetracycline hydrochloride (CTC). The probe was applied to determine TC in milk, and the recoveries ranged from 94.0% to 101.4%, indicating the application potential of this method. This study provides a novel idea for the construction of ratiometric fluorescence sensing platforms and shows the application potential of the AIE effect of FHBA.

Abstract:In this study, a novel Eu3+ functionalized zinc metal-organic frameworks (Eu@Zn-MOFs) probe has been prepared by the post-synthetic modification method. Eu@Zn-MOFs exhibit a strong blue fluorescence emission band of Zn-MOFs and a weak characteristic emission of Eu3+ under 390 nm excitation, which were innovatively used to construct a ratiometric fluorescent probe for tetracycline detection. Upon addition of tetracycline, the fluorescence of Zn-MOFs was quenched because of the inner filter effect between tetracycline and Eu@Zn-MOFs, while the fluorescence of Eu3+ was enhanced owing to the "antenna" effect between tetracycline and Eu3+. Based on the ratio of fluorescence intensity, the efficient detection of tetracycline was achieved. The results indicate that the ratiometric fluorescent probe has an excellent selectivity toward tetracycline over various interferents. It was found that a good linear relationship between the fluorescence intensity ratio (F618/F430) and concentrations of tetracycline range from 0.1 to 22.5 μM with a detection limit of 33 nM. Meanwhile, the color change of the solution from blue to light pink under ultraviolet lamp can be observed by naked eyes. The assay is successfully used to detect tetracycline in tap water samples with a recovery of 97.1%-100.6%.

Abstract:Tetracycline hydrochloride (TCH), a prevalent antibiotic in aquaculture for treating bacterial infections, poses challenges for on-site detection. This study employed the reversed-phase microemulsion method to synthesize a uniform nano metal-organic framework (MOF) material, europium-benzene-p-dicarboxylic acid (Eu-BDC), doped with Tb 3+ to form a dual-emission fluorescence probe. By leveraging the combined a-photoinduced electron-transfer (a-PET) and inner filter effect (IFE) mechanisms, high-sensitivity TCH detection in Carassius auratus and Ruditapes philippinarum was achieved. The detection range for TCH is 0.380–75 μM, with a low limit of detection (LOD) at 0.115 μM. Upon TCH binding, Eu-BDC fluorescence rapidly decreased, while Tb 3+ fluorescence remained constant, establishing a ratiometric fluorescence change. Investigation into the TCH quenching mechanism on Eu-BDC was conducted using time-dependent density functional theory (TD-DFT) calculations and fluorescence quenching kinetic equations, suggesting a mixed quenching mechanism. Furthermore, a novel photoelectric conversion fluorescence detection device (FL-2) was developed and evaluated in conjunction with high-performance liquid chromatography-diode-array detection (HPLC-DAD). This is the first dedicated fluorescence device for TCH detection, showcasing superior photoelectric conversion performance and stability that reduces experimental errors associated with smartphone photography methods, presenting a promising avenue for on-site rapid TCH detection.

Abstract:Residues in animal food and drinking water caused by the abuse of antibiotics lead to cell resistance and many chronic diseases in the human body. Therefore, it has become an inevitable trend to develop a fast, easy-to-use, on-site/real-time visualization method for the detection of antibiotics. Herein, we report a dual-response ra-tiometric fluorescence sensor which is fabricated by chelating europium ions (Eu3+) onto cadmium telluride quantum dots (CdTe QDs) for real-time and visible detection of tetracycline (TC). With the TC addition, the fluorescence of probe can be seen by the naked eye, from green to yellow and finally to red, exhibiting a dosage -sensitive and broad-chromatic detection strategy for TC. The fluorescence intensity ratio of I-616/I-512 of Eu/CdTe QDs sensor displays a good linear relation to TC concentrations in the range of 0-80 mu M with a limit of detection (LOD) of 2.2 nM. In addition, the sensor can visually detect 200 nM TC in actual samples, which is lower than the maximum residue limit (MRL) of the safety standard. The methodology reported here opens a window toward the real applications of fluorescent and shows the wide applicability in pursuing the concepts simple, rapid, visual, and real-time for food safety and environmental protection.
Ratiometric fluorescent probes based on carbon dots and europium for rapid detection of tetracycline

Abstract:Tetracycline (TC) is a typical emerging pharmaceuticals and personal care products (PPCPs) pollutants in the environment. However, TC residues cause severe effects on human health. It is crucial to exploit simple, rapid and sensitive methods to detect TC in water samples. A ratiometric fluorescent probes (CDs-Eu 3+ ) is constructed using carbon dots (CDs) and europium ions (Eu 3+ ) for the visual detection of tetracycline (TC). Under 390 nm excitation, CDs-Eu 3+ shows a strong yellow fluorescence at 545 nm and a weak red fluorescence at 619 nm, respectively. In the presence of TC, the fluorescence intensity at 545 nm decreased while the fluorescence intensity at 619 nm increased, which attributed to the internal filtering effect (IFE) and "antenna effect", respectively. Sensing performance of CDs-Eu 3+ probes is further evaluated with respect to various conditions, including salt concentration, Eu 3+ concentration, pH, and reaction time. Results reveal that the proposed ratiometric fluorescent probes exhibits a specific recognition ability toward TC. An excellent linear relationship is observed between the fluorescence intensity ratio (F 619 /F 545 ) and TC concentrations (1⁓100 μM), with a low detection limit (LOD) of 31 nM. The solution (CDs-Eu 3+ -TC) color gradually changes from yellow to red after adding TC. The probes are successfully applied to determine TC in water with recoveries of 85.43⁓97.53%. Finally, a fluorescent test strip is prepared and used for rapid visual semi-quantitative TC detection.

Abstract:Tetracyclines residues, particularly oxytetracycline (OTC) and tetracycline (TC), have raised extensive concern because of their serious adverse effects on human health. Herein, a dual-response fluorescent probe based on nitrogen-doped carbon dots (N-CDs) and Eu3+ hybrid (N-CDs-Eu3+) was developed to selectively determine OTC and TC. The N-CDs act as ancillary ligands of Eu3+ and recognition units of OTC/TC, while the Eu3+ ions chelated with N-CDs can also specifically recognize OTC/TC. Upon inclusion of OTC/TC, an enhancement in Eu3+ emission occurs due to the energy transfer from OTC/TC to Eu3+ and the efficient elimination of quenching effect caused by H2O molecule, which is attributed to the incorporation of N-CDs; while the blue fluorescence emitted by the N-CDs decreases under the inner filter effect and static quenching effect caused by OTC/TC. Based on the double and reverse response signals, the ratiometric detection of OTC and TC in the range of 0.1-45 μΜ and 0.1-30 μΜ is achieved with a detection limit of 0.017 and 0.041 μM, respectively. In addition, the noticeable variation in fluorescence color of the probe is integrated with a smartphone-assisted analysis device for the rapid on-site quantitative assay of OTC, where the detection limit is 0.15 μΜ. The results show that this probe performs with excellent specificity and anti-interference for both OTC and TC, and satisfactory detection results are obtained in lake water, milk, and honey samples, thereby confirming that the probe exhibits promising application in food safety and environmental monitoring.

Abstract:As a kind of antibiotic, tetracycline (TC) might remain in animal blood and milk products during use, which poses a risk to humans after consumption. Therefore, a ratiometric fluorescence probe was proposed for the detection of TC, which was based on an Eu 3+ functionalized hydrogen-bonded organic framework (HOF). Since there are a large number of N and O atoms in the skeleton of HOF, more Eu 3+ could be loaded onto HOF by forming coordinate bonds, while preserving the fluorescence of luminol monomer in HOF. In the presence of TC, the fluorescence of luminol monomer was attenuated at 425 nm due to inner filter effect (IFE), while TC selectively enhanced the fluorescence peak at 617 nm of Eu 3+ under the influence of antenna effect (AE). This highly sensitive probe could detect TC in the range of 0.1–60 μM and had a low limit of detection of 8.51 nM. Besides, the HOF@Eu probe was able to detect TC in actual samples (milk and tap water) with good recoveries (95.09%-111.51%) and precision (R < 4.78%), indicating this probe has great application potential for the detection of TC in food.

Abstract:Exploring materials with the dual functionality of detecting and removing tetracycline (TC) residues is crucial because of the environmental and health risks posed by antibiotic overuse. This study introduces a dual-emissive luminescent probe, CDs@ZSM-5:Eu3+, created through a solvent-free method combined with subsequent Eu3+ion exchange. The nanocomposite's blue emission, originating from carbon dots (CDs), is quenched by TC via an internal filtering effect, while an antenna effect triggers a strong red fluorescence of a TC-Eu3+chelate. The ratiometric fluorescence changes in CDs@ZSM-5:Eu3+ endow a self-calibrated sensing mechanism for TC, offering a low detection limit of 5.04 nM and a broad detection range of 0.01-50 μM. Demonstrated in real milk samples, the probe exhibits high selectivity and accuracy in detecting TC. The nanocomposite also displayed an impressive TC removal capacity of 238.1 mg g-1 in water, ascribing to the enrichment and electrostatic attraction effects of ZSM-5 toward TC molecules. This research offers a facile strategy for constructing multifunctional zeolite-based hybrids for simultaneous TC detection and removal from aqueous solutions.
Eu3+-functionalized covalent organic framework for ratiometric fluorescence detection and adsorption of tetracycline and information steganography

Abstract:Functional materials with organic/inorganic composites as the main matrix and rare earth ion complexes as the guest have shown a very broad application prospect for antibiotic sensors. However, Eu3+-complex often relies on a single fluorescence response signal, which is susceptible to changes in the detection environment and cannot simultaneously detect and remove tetracycline (TC). Herein, green fluorescent covalent two-dimensional organic framework (COF-TD) is synthesized, followed by modification of Eu3+ to synthesize COF-TD@Eu3+. In the ratiometric sensor, Eu3+ serves as the recognition site and specific response probe for TC, while COF-TD is the fluorescence reference and carrier for Eu3+. Due to the antenna effect, TC enhances the red fluorescence of Eu3+, while the green fluorescence of COF-TD remains almost stable. Based on the change of fluorescence intensity and fluorescence color from green to red, the efficient ratiometric sensing can be finished in 1 min. The developed method shows high sensitivity with a detection limit of 0.3 μM and high selectivity to TC which makes the method applicable to detect TC in traditional Chinese medicine preparations. In addition, due to the high specific surface area of COFs and specific adsorption sites, COF-TD@Eu3+ also shows good performance for TC removal. The findings show that the maximum adsorption capacity is 137.3 mg g-1 and the adsorption equilibrium is reached in 30 min. Smartphone assisted COF-TD@Eu3+ for both ratiometric fluorescence detection and detecting the absorption of TC is proposed for the first time. The molecular cryptosteganography that transforms the selective response of COF-TD@Eu3+ to binary strings is anticipated to advance utilization of nanomaterials in logic sensing and information safety.

Abstract:A multicolor fluorescent nanoprobe has been prepared by loading bovine serum albumin–stabilized copper nanoclusters (BSA-Cu NCs) onto amino clay (AC) and grafting Eu 3+ with auxiliary ligand citric acid (Cit). Tetracycline (TC) can coordinate with Eu 3+ by β-diketone structure and transfer energy to Eu 3+ through antenna effect. When the concentration of TC is in the range 0 to 13 μM, the blue emission intensity of BSA-CuNCs is basically unchanged, and the red emission of Eu 3+ is remarkably enhanced due to the coordination with TC. The emission color gradually changes from blue to red under UV lamp ( λ = 365 nm). However, when the TC concentration is in the range 13 to 350 μΜ, the internal amino acid residues of BSA sensitize TC, and the emission color gradually changed from red to green. The nanoprobe has rich color, is simple to prepare, portable, and provide a wide detection range. The limit of detection (LOD) is 3.04 nM, which could be used for real-time visual analysis of trace TC in actual samples (lake water, milk, honey, and bovine serum albumin). In addition, a visual test paper has been designed and combined with the color scanning APP of a smartphone to complete the qualitative and semi-quantitative test of TC.

Abstract:The widespread presence of tetracyclines in the environment has raised concerns about the potential risks to ecosystems and human health. The ratiometric fluorescence sensor for detecting tetracyclines was developed using europium-doped carbon dots (Eu-CDs) as probes under alkaline conditions. The sensing mechanism of sensor for tetracyclines was considered as inner filter effect (IFE), antenna effect (AE), and self-quenching effect (SQE). The sensor had a wide linear detection range than the reported europium ions-based tetracyclines sensors. The linear detection ranges of oxytetracycline (OTC), tetracycline (TC), doxycycline (DC) and chlorotetracycline (CTC) were respectively 0.00–603.75 μM, 0.00–623.82 μM, 0.00–594.61 μM and 0.00–601.54 μM, and the corresponding detection limits were respectively 9.50 nM, 15.80 nM, 10.4 nM and 90.3 nM. The smartphone with RGB Color Picker was further employed to analyze the concentration of tetracyclines, which provided a new method for visual tetracyclines detection. The application of Eu-CDs test paper was also explored, and the results showed that the Eu-CDs test paper has great potential application in the visual detection of tetracyclines. In addition, the accuracy of the established tetracyclines sensor was compared with that of the China national standard method by high-performance liquid chromatography (HPLC), and the results showed that the established method in this work has similar accuracy to the China national standard method. The sensor has been employed to detect tetracyclines in the actual samples with satisfactory results, which indicated that this method has promising applications in the real-time monitoring tetracyclines of food and environment.

Abstract:A ratiometric fluorescence probe is proposed for sensitive and visual detection of tetracyclinee (TC) based on cascade fluorescence signal amplification induced by bovine serum albumin–stabilized copper nanoclusters (BSA-CuNCs) and yttrium ions (Y 3+ ). TC can combine with Y 3+ to form the complex (TC-Y 3+ ) to enhance the fluorescence of TC at 515 nm. Then, positively charged TC-Y 3+ and negatively charged BSA-CuNCs was bonded together by electrostatic interactions to achieve the fluorescence resonance energy transfer (FRET) process. With the increase of TC concentration, the fluorescence intensity of TC-Y 3+ at 515 nm (F 515 ) gradually increased; meanwhile, the fluorescence intensity of BSA-CuNCs at 405 nm (F 405 ) decreased gradually. The ratio of F 515 and F 405 was used for the quantitative determination of TC. The linear range of the constructed fluorescent probe is 1.0 to 60.0 μM, and the limit of detection is 0.22 μM. The method was successfully applied to the determination of TC in spiked milk with recoveries ranging from 94.3 to 112%. Furthermore, the color of this platform can be observed from dark violet to bright green under the UV lamp. Since the response time of the reaction is less than 10 s, an intelligent sensing platform based on the use of the smartphone as image acquisition equipment was also established to realize rapid on-site and portable detection of TC through the colorimetric recognition application.

Abstract:Tetracycline (TC) residues are harmful to the environment and human body, so it is necessary to develop a highly sensitive probe for rapid detection of tetracycline residues. In the present paper, a novel dye-doped porous metal–organic framework (UiO-66)-based multi-color fluorescent nano-probe was designed for sensitive ratiometric detection of tetracycline (TC). In this probe, dye-molecules doped UiO-66 was used as a fluorescent internal standard, and the externally grafted lanthanide Eu3+ complex was used as response signals. The fluorescence of the Eu3+ complex was selectively enhanced with increasing concentrations of TC, which was accompanied by a visual blue-to-red color switch. The nano-probe had a linear response between 0.1 and 6 μM with a lowest detection limit of 17.9 nM, which was much lower than the maximum residue limits set by the United States Food and Drug Administration (676 nM) and the European Union (225 nM). The applicability of this method in the analysis of actual samples was evaluated by the determination of TC in honey and milk samples, indicating satisfactory recovery and good reproducibility. In addition, a cost-effective paper-based probe for rapid and visual detection of TC was developed by fixing the nano-probe on filter papers. With the help of a smartphone camera to capture the fluorescence color, and chromaticity analysis software, the calculation and analysis of red (R) and blue (B) values can be realized, which has the potential for real-time visual detection of TC.

Abstract:The detection of residual tetracycline antibiotics (TCs) in the aquatic environment is important because of the irreversible damage of TCs to the ecosystem and human health. Considering the photostability, sensitivity, and accuracy of the detection systems, mesoporous SBA-15 silica material was an ideal support matrix for improving photostability because it can protect the optical properties of fluorescent signals. To achieve ratiometric detection (with sensitivity and accuracy), pyrene and Eu3+ as two fluorescent signals were implemented into SBA-15, and an organic-inorganic hybrid fluorescent nanoprobe Py/EDTA-SBA-15@Eu3+ was constructed. In the presence of TCs, the red emission of Eu3+ in the nanoprobe was sensitized, and the fluorescence resonance energy transfer occurred from pyrene to Eu3+, resulting in the increased emission of Eu3+ at 617 nm and the decreased emission of pyrene at 384 nm. By monitoring the fluorescent intensity changes at 617 and 384 nm (F617/F384), the nanoprobe achieved the ratiometric detection of oxytetracycline (OTC) and tetracycline (TC) in the range of 0.20-80 and 0.40-80 μM, respectively, with detection limits of 0.18 nM for OTC and 5.83 nM for TC. In addition, the practical application of the nanoprobe was validated in tap water and honey samples.
